Description
Technical Field
The invention relates to the technical field of skin care products, in particular to a composition with skin repairing and anti-aging effects and a skin care product thereof.
Background
Mitochondria are organelles that generate energy (ATP) by consuming oxygen, and although they have antioxidant defense systems, ROS produced continuously still damage mitochondrial DNA
(mtDNA). mtDNA mutates more frequently because it lacks histone protection and it is generally less efficient than nuclear DNA repair. Increased variation impairs mitochondrial function, thereby reducing energy production and inducing a vicious cycle, and defective respiratory chains produce more ROS, which in turn impairs the whole cell and accelerates skin aging. Therefore, mitochondria are not only the main site where cells perform oxidative phosphorylation and generate energy, but also are closely related to ROS generation, apoptosis, telomere, signal transduction and the like, are considered to play a key role in relieving oxidative stress and aging, and are important breakthrough for delaying aging of organisms.
Experiments show that the radix scutellariae flavone can reduce the level of ROS and Lipid Peroxide (LPO) by enhancing a mitochondrial enzyme system and an antioxidant system, protect nuclear DNA and mtDNA from free radical damage, enhance voltage-dependent anion channels and adenine nucleotide translocase protein expression, improve mitochondrial membrane permeability transition pore dysfunction and play a stronger role in mitochondrial protection.
Many researches show that the small molecular polypeptide has the effects of repairing and resisting aging from a plurality of targets due to small molecular weight, high activity and easy absorption. Wherein, palmitoyl tripeptide-5, also called collagen peptide, is the cosmetic short peptide with the most extensive application. The palmitoyl tripeptide-5 serving as a signal peptide can promote the synthesis of collagen and the generation of extracellular matrix elastin by increasing the activity of stromal cells, improve the facial elasticity, remarkably reduce and lighten wrinkles, enhance the cell activity, improve the moisture content of skin, enhance the skin luster and generate the effects of repairing and resisting aging.
At present, no report on the combined use of the flavonoids of scutellaria and the active polypeptide palmitoyl tripeptide-5 for repairing and resisting aging exists.

the massive production of ROS is the early reaction of apoptosis, mitochondria are the main production site of ROS, more than 90% of oxygen molecules in organisms are consumed in mitochondria, and the probe H is utilized in the test example2DCFDA detects ROS scavenging ability of Scutellariae radix flavone. H2DCFDA excites green light, the brighter the green light, the higher the active oxygen content. Cells were cultured in 96-well plates, pretreated for 16h by adding different concentrations of composition solutions (1, 2, 4, 6, 8, 10. mu.g/ml) followed by 400. mu. M H2O2And (5) incubating for 24 h. Addition of H2DCFDA (final concentration 50. mu.M) was cultured for another 30min and excited by 488nM in a multifunctional microplate reader, and fluorescence intensity of each well of the plate was measured at 525nM emission wavelength. Each set is provided with 6 multiple holes, refer to fig. 1.
As can be seen from FIG. 1, the relative amount of ROS treated with the composition is significantly reduced compared to the model group, and the relative amount of ROS treated with the sample at a concentration of 10 μ g/ml is about 50% of that of the model group, which is statistically significant (p <0.01), indicating that the composition of example 3 has a significant ROS scavenging effect. In addition, the relative content of ROS is reduced along with the increase of the concentration of the sample, and a remarkable dose-effect relationship is shown. The result shows that the composition consisting of the flavonoids of the scutellaria and the active polypeptide palmitoyl tripeptide-5 has antioxidant activity and can eliminate redundant free radicals in cells.

2. The test method comprises the following steps: cells were cultured in 24-well plates and after 24h, different concentrations of composition solutions (1, 5, 10. mu.g/ml) were added and incubation continued for 48 h. Adding mitochondria extraction medium, extracting cell mitochondria by adopting a repeated freeze-thaw method, measuring protein content by using a BCA method, and adjusting each group of mitochondria extracting solution to ensure that the protein content is basically consistent. Mitochondria are the main site for aerobic respiration and provide energy for cell life activities. The respiratory chain complex is the site for the transfer of electrons and the execution of mitochondrial functions, and its activity has an important influence on mitochondrial function and cell status. As shown in figure 2, the 10 mu g/ml composition solution can obviously improve the activity of MRCC I, III and IV (p is less than 0.01), and has no obvious influence on MRCC II, and the result shows that the composition consisting of the radix scutellariae flavone and the active polypeptide palmitoyl tripeptide-5 has a certain promotion effect on the function of the mitochondrial respiratory chain.
Test example four measurement of mitochondrial Membrane potential by the composition having skin repairing and anti-aging effects of the present invention
1. Test materials:
example 3 composition with skin repair anti-aging effect.
2. The test method comprises the following steps: the transmembrane potential of mitochondria is an important indicator of the state of mitochondrial function, and its decline is considered to be the earliest event in the course of the apoptosis cascade. Rhodamine 123(Rh123) is a dye capable of staining mitochondria of living cells, has fluorescent cations, can penetrate cell membranes, enters a mitochondrial matrix by depending on the transmembrane potential of the mitochondria, and is neutralized with H + to quench fluorescence. When the mitochondrial transmembrane potential is reduced or lost, Rh123 can be released from the mitochondria again to emit yellow-green fluorescence. Cells were cultured in 24-well plates, and after 16h, the cells were pretreated by adding composition solutions (1, 2, 4, 6, 8, 10. mu.g/ml) at different concentrations for 1h, and then incubated for 24h by adding 400. mu. M H2O2, and the change of mitochondrial membrane potential was detected by using the fluorescent dye Rh123, as shown in FIG. 3.
The larger the fluorescence value, the lower the mitochondrial membrane potential, i.e., the lower the mitochondrial activity. The upper graph shows that the fluorescence of the cells is obviously enhanced and the mitochondrial membrane potential is reduced after the H2O2 treatment compared with the normal cells, while the fluorescence value of the solution of the composition of example 3 of 6, 8 and 10 mug/ml is lower than that of the model group after the pretreatment, and the difference has statistical significance (p is less than 0.01), which indicates that the mitochondrial membrane potential is increased, and the composition consisting of the baical skullcap root flavone and the active polypeptide palmitoyl tripeptide-5 can obviously maintain the mitochondrial function activity.

1. Test materials:
example 3 composition with skin repair anti-aging effect.
2. The test method comprises the following steps: intracellular ATP levels are an important indicator of cellular energy metabolism, and ATP levels are markedly reduced in senescent cells. The determination of cellular ATP content by luciferase (luciferase) -luciferin (luciferin) system is one of the most sensitive and specific methods in the current bioenergy research, and the principles are luciferase (E), reduced luciferin (LH2) and in Mg2+And reacting under the participation of oxygen, wherein the generated light quantum h is in positive correlation with a certain amount of ATP. Cells were cultured in 24-well plates, pretreated for 16h by adding different concentrations of composition solutions (1, 2, 4, 6, 8, 10. mu.g/ml) followed by 400. mu. M H2O2And (5) incubating for 24 h. The intracellular ATP content was measured by the ATP detection kit, and FIG. 4 shows the results.
The experimental results show that H is compared with normal cells2O2After treatment, the ATP in the cells is obviously reduced, and after the cells are pretreated by 6, 8 and 10 mu g/ml composition solution, the ATP concentration in the cells is increased in a gradient manner. The experimental results show that the composition of flavonoids of scutellaria and active polypeptide palmitoyl tripeptide-5 can maintain normal ATP synthesis under the condition of cell oxidative damage.
