Abstract

Description
本发明首先涉及一种确定室内灰尘,引起过敏性反应的潜在可能性的方法。
通常可以确定,在很大范围内越来越多地出现了过敏或变态性反应。特别是在住宅和家庭用品中存在的污染如螨排泄物、霉菌、花粉、草和桦树特定的植物部分以及皮肤鳞屑应对此负责。人们试图用已知的清洁技术来清除这些污染,然后将这些污染积聚作为室内灰尘组份而除掉。因此存在一种需求,即可以对室内灰尘的积聚和去除进行有效地检查。另外还存在一种需求,即确定室内灰尘中上述污染掺入的程度,作为过敏反应潜在可能性的尺度。还可以例如得到一种提示,寻找特殊污染来源。
由此可以看出,本发明要解决的问题是,提出一种检测室内灰尘引起过敏反应的潜在可能性的方法。另一个要解决的技术问题是,提出一种对此适用的手段或设备。
所述的该问题解决方法是为了检测室内灰尘而使用一种蛋白质检测来确定室内灰尘中含有的含蛋白质组份的数量级。具体地说,本发明提供一种确定室内灰尘引起过敏性反应的潜在可能性的方法,其特征是,在室内灰尘中掺入一种用于确定室内灰尘中所含的含蛋白质组份的含量的检测剂。本发明发现,实际上室内灰尘中的上述所有过敏组份都是由蛋白质构成的,或者说具有蛋白质组份。另外还发现,表征特别是这种室内灰尘中的含蛋白质组份的比例表征同时也是所涉及的室内灰尘引起过敏反应的潜在可能性的尺度。另一个要素是,蛋白质检测是一种优选与室内灰尘中的含蛋白质组份发生反应特别是发生变色反应沉淀的染料。该方法的这种方案可以简单的方式借助于颜色的强度确定室内灰尘中可能引起过敏反应的组份的数量级。为此要考虑按照蛋白质检测剂对室内灰尘的作用发生的颜色强度。在该方法的另一个优选的方案中,使用焦倍酚作为染料。具体为焦倍酚-红-Mo-络合物,其中另外在“焦倍酚”上加入。使用染料和特别是使用焦倍酚染料一般来检测室内灰尘也是本发明的另一个独立的主题。在焦倍酚中,蛋白质或含蛋白质的颗粒呈现紫色或蓝色色调的颜色,而染料本身起初是红色的。考虑到检测剂,建议该染料溶解在一种液体中。该液体表明可以作为一种成份特别含有一种缓冲剂和优选稳定剂。缓冲剂的作用是防止液体已经发生的PH值的变化引起染料变色或者颜色强度的变化。在另一个优选的方案中,考虑到应当用于检测最小量的室内灰尘,建议液体含有一种溶剂。优选的溶剂具体的是变性醇。例如50%的变性醇或者0.2mol HCL-甘油缓冲剂与96%的变性醇以1∶1的比例的混合物。该醇同时还具有有利的结合作用,使得有关的组份分解。偶尔存在的脂肪涂层或者脂肪组份可以溶解。
本发明的另一个主题是用于测定室内灰尘中引起过敏反应的可能性的检测剂的应用。该检测剂的特征在于具有一种检测组份,它可以识别室内灰尘中蛋白质成份。该检测剂优选为一种染料,它优选与室内灰尘中的含蛋白质组份发生变色反应或沉淀,其中为了确定该组份的数量级,可以参考所发生的颜色强度。该染料尤其可以是一种焦倍酚-红-Mo-络合物。该检测剂可以是一种染料溶解在其中的液体。该液体可以含有缓冲剂和优选稳定剂作为组份。另外,该液体还可以含有一种溶剂。
作为唯一的附图是一个这里得到的显微镜照片的复印件。用X表示的颗粒彩色复印时显示出明显的蓝-紫色,而用Y表示的颗粒具有其它颜色或者基本上没有颜色。没有颜色的颗粒是例如沙粒或者糖粒。
上述检测液体或者检测剂的一个实施例的组成如下:
1.甘氨酸-HCl,作为缓冲组份,浓度为0.2mol/l,PH值为1.9,以1∶1的比例与96%的甲基乙基酮-变性醇相混合。
2.组份0.2g/l焦倍酚红
-0.3g/l钼酸铵四水合物
-1.0g/l草酸纳
-1.2g/l(L+)-酒石酸
3.以1∶10的比例用0.2mol的与96%的变性醇以1∶1的比例相混合的HCl-甘氨酸稀释。
组份3应当是在液体作为室内灰尘变色试剂在悬浮液时使用。即使不添加组份3,该液体也可以作为浓缩的基本溶液用于例如吸尘器的过滤器的直接变色。
在使用时,例如将2-4滴浓缩的焦倍酚-红-钼-基本溶液滴到约1cm2大的加工成槽形的由聚丙烯组成的过滤件上。该过滤件应当浸透,但不要覆盖有过量的的溶液。根据环境温度和有关的细灰尘的数量和类型,过滤器中的灰尘物料最多5分钟即可根据蛋白质负荷而变色成蓝-紫色。
另一个使用的实例是以1∶10用50%的变性醇稀释的液体,(例如可对比上述实施例的组份3)。将1ml该焦倍酚-红-钼-络合物溶液与一种“刀尖(Messerspitze)”室内灰尘相混合。通过在一个热手中摇动一个含有液体的玻璃试管,加速向蓝-紫色的变色。在通常的室内灰尘中1-2分钟即可检出紫色。
这样表征的室内灰尘颗粒还可以进行显微镜分析。液体或染料对此后进行的分析的影响不大。这既适用于在过滤器中变色的颗粒,也适用于稀释变色的室内灰尘悬浮液。
在上述说明书、附图和权利要求书中公开的特征在既可以单独也可以随意组合来实现本发明时是重要的。所有公开的特征都是本发明的必要特征。在公开本发明时也可以完全包括所属/所附的优先权文件所公开的内容。
Claims (6)
1.一种室内灰尘的检测方法,这些灰尘具有引起过敏性反应的潜在可能性,其特征是,在室内灰尘中掺入一种用于确定室内灰尘中所含的含蛋白质组份的数量级的检测剂。
2.按照权利要求1所述的方法,其特征是,蛋白质检测剂是一种染料,它与含蛋白质组份沉淀发生变色反应,借助于所发生的颜色强度来确定数量级。
3.按照权利要求2所述的方法,其特征是,该染料是焦倍酚。
4.按照权利要求2或3所述的方法,其特征是,染料溶解在一种液体中,它含有缓冲剂和稳定剂作为组份。
5.按照权利要求4所述的方法,其特征是,液体含有一种溶剂。
6.按照权利要求5所述的方法,其特征是,溶剂是约50%的变性醇。
