技术领域
本发明属于生物技术领域,涉及一种神秘果叶复合物及在抗黑色素形成化妆品、保健品中的应用。
背景技术
神秘果(学名:Synsepalum dulcificum Daniell),又名变味果、梦幻果、奇迹果,属于山榄科多年生矮生灌木,神秘果的果肉酸涩,但内含有神秘果蛋白,吃了神秘果半小时内,接着吃其他酸性水果,会觉得这些酸水果不再是酸味,而变为甜味,故名神秘果。另外其籽大而苦,叶可泡茶。神秘果也是有趣的观赏植物,树形美观,也可作盆景,宜在中国高温、高湿的亚热带、热带地区种植,是一种将观赏价值与经济价值集于一身的水果。
目前,神秘果被广泛开采并用于化妆品领域,不过功效主要集中在抗衰老,对于美白方面的作用研究较少。
现有技术中,专利申请号为201510632159.X的发明专利公开一种复方神秘果叶提取物及其在抗衰老化妆品中的应用,其以中草药原料制备而成,主料为神秘果叶,辅料为赤芍、白蒺藜、黄芪、红景天、黄芩中的任意三种的组合。该发明还公开了所述复方神秘果叶提取物的制备方法,包括准备中草药原料、预处理、提取、粗滤、精滤、浓缩、复溶和灭菌。该发明还公开了所述复方神秘果叶提取物在具有抗衰老功效的化妆品中的应用。该发明所述的复方神秘果叶提取物兼具抗氧化活性和酪氨酸酶抑制活性,弥补了神秘果叶仅具有较强的抗氧化活性的不足,可作为抗衰老功效成分应用于化妆品中,酪氨酸抑制活性被认为是体外美白的功效检测方法,并没有进行体内实验的验证;另外,该专利并没有对神秘果美白功效等方面进行详细研究。
发明内容
为了克服现有技术的不足,本发明提供一种神秘果叶复合物,研究了神秘果叶复合物的制备方法以及在抗黑色素(即美白)形成的化妆品、保健品中的应用,填补了研究空白。
本发明提供了如下的技术方案:
一种神秘果叶复合物,用于具有美白功效的化妆品、保健品(具有美白作用的保健药物、保健饮品)中,包括以下重量百分比的组分:神秘果叶40%-50%,樱花8%-15%,茯苓5%-10%,白蒺藜5%-10%,灵芝5%-10%,当归10%-15%,甘草5%-10%。神秘果叶复合物以药食同源的多种原料复配。
优选的是,原料包括以下重量百分比的组分:神秘果叶45%-50%,樱花10%-15%,茯苓8%-10%,白蒺藜8%-10%,灵芝8%-10%,当归12%-15%,甘草8%-10%。
在上述任一方案中优选的是,原料包括以下重量百分比的组分:神秘果叶40%,樱花15%,茯苓10%,白蒺藜10%,灵芝8%,当归10%,甘草7%。
在上述任一方案中优选的是,原料包括以下重量百分比的组分:神秘果叶45%,樱花10%,茯苓5%,白蒺藜10%,灵芝10%,当归15%,甘草5%。

在上述任一方案中优选的是,神秘果叶复合物的制备方法包括以下步骤:
(1)按照重量百分比准备原料,添加酶溶液处理,并进行负压空化提取,酶溶液对神秘果叶重量比为0.2-1:100,得到神秘果叶提取液,神秘果叶提取液浓缩后分离,收集洗脱液;
(2)将洗脱液经过加压浓缩,灭菌后得到神秘果浓缩液;
(3)将其余原料中的樱花、茯苓、白蒺藜、灵芝、当归、甘草洗净后烘干粉碎,混合得到混合粉末;
(4)将混合粉末和60%-96%的乙醇组成的溶剂按照1:20-30的重量比混合,在48-65℃超声提取2小时,得到混合提取液;
(5)混合提取液经过多级过滤后浓缩、灭菌得到混合浓缩液;
(5)将混合浓缩液和神秘果叶浓缩液按比例混合得到神秘果叶复合物。
在上述任一方案中优选的是,所述步骤(1)中酶溶液为纤维素酶或果胶酶中的至少一种。
在上述任一方案中优选的是,所述步骤(1)中负压空化提取时温度为18-60℃,提取压力为-0.04MPa—-0.07MPa。
本发明还提供上述神秘果叶复合物在抗黑色素形成化妆品、保健品中的应用。化妆品只能外用,保健品可以内服,采用内服加外用,美白效果更好。
在上述任一方案中优选的是,在所述化妆品中,神秘果叶浓缩液占的重量比为0.2%-12%。
在上述任一方案中优选的是,在所述化妆品中,神秘果叶复合物占的重量比为0.1%-10%。
在上述任一方案中优选的是,所述化妆品包括具有美白作用的日、晚霜,精华液、面膜。
在上述任一方案中优选的是,所述保健品包括保健药物、保健饮品。
本发明还提供一种神秘果叶复合物的美白检测方法,进行动物体内模拟实验,包括以下步骤:
步骤(1),将斑马鱼胚胎养在含有不同浓度的神秘果叶复合物的水里,并设置空白对照;
步骤(2),去除神秘果叶复合物的处理,采用解剖立体显微镜对各组斑马鱼进行视觉观察和图像采集,并使用基于图像的形态计量分析进行定量图像分析,计算黑色素抑制率平均值。
优选的是,所述步骤(1)中神秘果叶复合物添加量为10μg/mL,25μg/mL,50μg/mL。
本申请中各组分的功效如下:
神秘果的价值可分为食用价值和药用价值两个方面。从食用价值上来说,神秘果果肉含有丰富奇特的醣蛋白、维生素C、KI、柠檬酸、琥珀酸、草酸等,种子含有天然固醇等,叶子含有钠、钾、钙、镁等微量矿物元素。果肉、种子以及叶子都能够为人体提供足够的营养元素,富含丰富矿物元素也是神秘果的一大特性。从药用价值上来说,神秘果因果实含有特殊的醣蛋白,可将酸度高的水果转化成琼浆玉露般的甘甜。常生吃熟果或浓缩锭剂具有调整高血糖、高血压、高血脂、痛风、尿酸、头痛等,达正常值稳定作用之功效。果汁涂抹于蚊虫叮咬处能消炎消肿。种子可解心绞痛、喉咙痛、痔疮等。叶子泡茶或做菜能调整高血糖、高血压、保护心脏、美颜瘦身、排毒通便、控制尿酸、减轻痛风痛证、且能解酒。
茯苓,除了食用以外,还是一味珍贵的中药材,具有健脾和胃、宁心安神、润肌、美容养颜的等功效。
灵芝含有“锗”,能使人体血液吸收氧的能力增加1.5倍,因此可以促进新陈代谢并有延缓老化的作用,还有增强皮肤本身修护功能的功效,可用于各种慢性病所致的面色黄萎及由于气血不足而致的面部光泽等症。
当归,补血,调经止痛,还可以抗衰老,增强人体的免疫力,防治癌症,消除炎症。
樱花,其主要功效成分以黄酮类化合物为主,含有樱花素、紫云英苷,异槲皮苷、矢车菊素-3-葡萄糖苷,矢车菊素-3-芸香糖甙,樱黄素,染料木黄酮,槲皮素,染料木苷,山柰酚-3-O-葡萄糖苷等黄酮类化合物等,另外还含有天然维生素A、B、E等化学成分,具有良好的抗糖化效果及抑制酪氨酸酶活性,且能够很好的收缩毛孔、平衡油脂和增亮肤色,从而达到美白的功效。。
甘草,美白保湿,用于调和诸药。

一种神秘果叶复合物,所述神秘果叶复合物以药食同源的多种原料制成,原料包括以下重量百分比的组分:神秘果叶40%,樱花15%,茯苓10%,白蒺藜10%,灵芝8%,当归10%,甘草7%。
神秘果叶复合物的制备方法包括以下步骤:
(1)准备原料,用纤维素酶溶液对神秘果果肉进行负压空化提取,负压空化提取时温度为18℃,提取压力为-0.04MPa,
酶溶液对神秘果叶重量比为0.2:100,得到神秘果叶提取液,神秘果叶提取液浓缩后分离,收集洗脱液,具体的提取方法可以参考本申请人已经公开的专利申请号为201710322530.1神秘果提取物的应用中有详细的记载。
(2)将洗脱液经过加压浓缩,灭菌后得到神秘果叶浓缩液;
(3)将其余原料中的樱花、茯苓、白蒺藜、灵芝、当归、甘草洗净后烘干粉碎,混合得到混合粉末,混合粉末的颗粒度为80目;
(4)将混合粉末和60%的乙醇组成的溶剂按照1:20的重量比混合,在48-℃超声提取2小时,得到混合提取液;
(5)混合提取液经过多级过滤后浓缩、灭菌得到混合浓缩液;过滤和灭菌方法均为本领域常用方法,不再赘述;
(5)将混合浓缩液和神秘果叶浓缩液混合得到到神秘果叶复合物。
实施例2:
一种神秘果叶复合物,所述神秘果叶复合物以药食同源的多种原料制成,原料包括以下重量百分比的组分:神秘果叶45%,樱花10%,茯苓5%,白蒺藜10%,灵芝10%,当归15%,甘草5%。
神秘果叶复合物的制备方法包括以下步骤:
(1)按照重量百分比准备原料,用纤维素酶溶液对神秘果叶进行负压空化提取,负压空化提取时温度为30℃,提取压力为-0.05MPa,
酶溶液对神秘果叶重量比为0.5:100,得到神秘果提取液,神秘果提取液浓缩后分离,收集洗脱液,具体的提取方法可以参考本申请人已经公开的专利申请号为201710322530.1神秘果提取物的应用中有详细的记载。
(2)将洗脱液经过加压浓缩,灭菌后得到神秘果浓缩液;
(3)将其余原料中的樱花、茯苓、白蒺藜、灵芝、当归、甘草洗净后烘干粉碎,混合得到混合粉末,混合粉末的颗粒度为100目;
(4)将混合粉末和92%的乙醇组成的溶剂按照1:25的重量比混合,在55℃超生提取2小时,得到混合提取液;
(5)混合提取液经过多级过滤后浓缩、灭菌得到混合浓缩液;过滤和灭菌方法均为本领域常用方法,不再赘述;
(5)将混合浓缩液和神秘果叶浓缩液混合得到神秘果复合物。
神秘果叶复合物能够用于抗黑色素形成化的产品中,产品包括化妆品、保健品、药物。神秘果叶浓缩液占的重量比为0.2%-12%。
实施例3:
一种神秘果叶复合物,所述神秘果叶复合物与多种药食同源的原料进行制备,原料包括以下重量百分比的组分:神秘果果肉50%,樱花8%,茯苓5%,白蒺藜8%,灵芝9%,当归12%,甘草8%。
神秘果叶复合物的制备方法包括以下步骤:
(1)按照重量百分比准备原料,用纤维素酶溶液对神秘果叶进行负压空化提取,负压空化提取时温度为60℃,提取压力为-0.07MPa,
酶溶液对神秘果叶重量比为1:100,得到神秘果叶提取液,神秘果叶提取液浓缩后分离,收集洗脱液,具体的提取方法可以参考本申请人已经公开的专利申请号为201710322530.1神秘果提取物的应用中有详细的记载。
(2)将洗脱液经过加压浓缩,灭菌后得到神秘果叶浓缩液;
(3)将其余原料中的樱花、茯苓、白蒺藜、灵芝、当归、甘草洗净后烘干粉碎,混合得到混合粉末,混合粉末的颗粒度为120目;
(4)将混合粉末96%的乙醇组成的溶剂按照1:30的重量比混合,在65℃超生提取2小时,得到混合提取液;
(5)混合提取液经过多级过滤后浓缩、灭菌得到混合浓缩液;过滤和灭菌方法均为本领域常用方法,不再赘述;
(5)将混合浓缩液和神秘果叶浓缩液混合得到神秘果叶复合物。
神秘果叶复合物能够用于抗黑色素形成化的产品中,产品包括化妆品、保健品、药物。神秘果叶浓缩液占的重量比为0.2%-12%。
实施例4:
一种神秘果叶复合物,制备方法和实施例1相同,不同的是,由神秘果叶与多种药食同源的原料进行制备,原料包括以下重量百分比的组分:神秘果叶35%,樱花15%,茯苓10%,白蒺藜10%,灵芝10%,当归13%,甘草7%。
实施例5:
一种神秘果叶复合物,制备方法和实施例1相同,不同的是,由神秘果叶与多种药食同源的原料进行制备,原料包括以下重量百分比的组分:神秘果叶55%,樱花8%,茯苓5%,白蒺藜8%,灵芝7%,当归10%,甘草7%。
采用上述实施例1-5制备的神秘果叶复合物对斑马鱼进行试验。
以斑马鱼进行色素抑制试验和色素恢复试验。
色素抑制试验:斑马鱼正常发育会形成黑色斑点,在斑马鱼胚胎发育初期加入神秘果叶复合物(9hpf,hpf为受精后时间,以小时计算),将斑马鱼胚胎养在含有复合物的水里(即通过体内模型实验验证),神秘果叶复合物添加量为10μg/mL,25μg/mL,50μg/mL,空白对照。48hpf时去除神秘果复合物的处理,采用解剖立体显微镜对各组斑马鱼进行视觉观察和图像采集,并使用基于图像的形态计量分析进行定量图像分析,结果表明斑马鱼胚胎发育过程中的黑色素合成会受到抑制,黑色减弱。单个试验共分为四组,每组30个斑马鱼胚胎为一组,黑色素抑制率平均值,如表1所示:

上述实验结果表明,本发明提供一种神秘果叶复合物,所述神秘果叶复合物以药食同源的多种原料制成,原料包括以下重量百分比的组分:神秘果叶40%-50%,樱花8%-15%,茯苓5%-10%,白蒺藜5%-10%,灵芝5%-10%,当归10%-15%,甘草5%-10%。本申请各组分按照君臣佐使的原则,神秘果叶起到主导作用,选择药食同源的樱花、茯苓、白蒺藜、灵芝、当归做种组分相互辅佐,增强药效。本申请中的神秘果叶也可以单处作为美白成分加入化妆品或保健品(保健饮品等)中,当然结合本申请的其他多种组分后,比单独使用神秘果叶的黑色素抑制率更高,将神秘果叶及樱花,茯苓,白蒺藜,灵芝,当归进行复配,可作为美白成分应用于化妆品或保健品(保健饮品等)中,各药用成分的相互辅助,美白效果更加显著。
由表1可知,实施例1-3的黑色素抑制率相对较高,实施例2在不同浓度下,黑色素抑制率效果明显优于其他实施例,实施例4和5由于神秘果叶含量不在本申请的40%-50%范围内,因此势必影响各组分的协同作用,黑色素抑制率明显降低。对实施例2进行试验的一组斑马鱼进行拍照(52hpf),侧视图和俯视图如图1所示。
采用上述实施例1-5制备的神秘果叶复合物对斑马鱼进行色素恢复试验。
具体的,色素恢复试验:同样,在斑马鱼胚胎发育初期(9hpf)就加入复合物,将斑马鱼胚胎养在含有复合物的水里,神秘果叶复合物添加量为10μg/mL,25μg/mL,50μg/mL,抑制黑色素合成,处理值48hpf时,将神秘果叶复合物去除,在不含复合物的培养水中恢复24h,其他方法同色素抑制试验。黑色素相对含量结果平均值如表2所示:

由表2可知,各实施例的黑色素均快速恢复甚至基本达到对照组水平,。上述实验也进一步说明本申请中的斑马鱼色素合成之所以受到抑制是由于神秘果复合物所产生的效果,同时斑马鱼发育情况良好,这也说明本申请是无副作用的。对实施例2进行色素恢复试验的一组斑马鱼进行拍照(72hpf),侧视图和俯视图如图2所示。根据上述实验结果,可以将本发明的神秘果叶复合物应用在具有抗黑色素形成即具有美白作用的化妆品、或者内服保健品。内服保健品可以为保健药物、保健饮品、保健饮料等。
Claims (10)
1.一种神秘果叶复合物,其特征在于:由神秘果叶与多种药食同源的原料进行制备,原料包括以下重量百分比的组分:神秘果叶40%-50%,樱花8%-15%,茯苓5%-10%,白蒺藜5%-10%,灵芝5%-10%,当归10%-15%,甘草5%-10%。
2.根据权利要求1所述的神秘果叶复合物,其特征在于:原料包括以下重量百分比的组分:神秘果叶45%-50%,樱花10%-15%,茯苓8%-10%,白蒺藜8%-10%,灵芝8%-10%,当归12%-15%,甘草8%-10%。
3.根据权利要求1所述的神秘果叶复合物,其特征在于:神秘果叶复合物的制备方法包括以下步骤:
(1)按照重量百分比准备原料,添加酶溶液处理,并进行负压空化提取,酶溶液与神秘果叶重量比为0.2-1:100,得到神秘果叶提取液,神秘果叶提取液浓缩后分离,收集洗脱液;
(2)将洗脱液经过加压浓缩,灭菌后得到神秘果叶浓缩液;
(3)将其余原料中的樱花、茯苓、白蒺藜、灵芝、当归、甘草洗净后烘干粉碎,混合得到混合粉末;
(4)将混合粉末和60%-96%的乙醇组成的溶剂按照1:20-30的重量比混合,在48-65℃超声提取2小时,得到混合提取液;
(5)混合提取液经过多级过滤后浓缩、灭菌得到混合浓缩液;
(5)将混合浓缩液和神秘果叶浓缩液按比例混合得到神秘果叶复合物。
4.根据权利要求3所述的神秘果叶复合物,其特征在于:所述步骤(1)中酶溶液为纤维素酶或果胶酶中的至少一种。
5.根据权利要求3所述的神秘果叶复合物,其特征在于:所述步骤(1)中负压空化提取时温度为18-60℃,提取压力为-0.04MPa—-0.07MPa。
6.根据权利要求1-7中任一项所述的神秘果叶复合物在抗黑色素形成的化妆品、保健品中的应用。
7.根据权利要求6所述的应用,其特征在于:在所述化妆品中,神秘果叶浓缩液占的重量比为0.2%-12%。
8.根据权利要求6所述的应用,其特征在于:在所述化妆品中,神秘果叶复合物占的重量比为0.1%-10%。
9.一种如权利要求1-5中任一项所述的神秘果叶复合物的美白检测方法,其特征在于:采用动物体内模拟实验进行,包括以下步骤:
步骤(1),将斑马鱼胚胎养在含有不同浓度的神秘果复合物的水里,并设置空白对照;
步骤(2),去除神秘果叶复合物的处理,采用解剖立体显微镜对各组斑马鱼进行视觉观察和图像采集,并使用基于图像的形态计量分析进行定量图像分析,计算黑色素抑制率平均值。
10.根据权利要求9所述的神秘果叶复合物的美白检测方法,其特征在于:所述步骤(1)中神秘果叶复合物添加量为10μg/mL,25μg/mL,50μg/mL。
