发明背景

不同的靶标和样品对传感器表面具有不同的要求。CMDx对于许多不同类型的样品和测定是有用的,但是当葡聚糖不发挥作用时需要替代基质。因此,需要使漂移和非特异性结合最小化的改进的传感器表面。
众所周知,PEG (聚乙二醇)或OEG (低聚乙二醇)表面具有蛋白排斥特性,并且PEG或OEG已被用于传感器表面上,以减少测定中不必要的结合。在这样的技术中,PEG或OEG部分也被用于连接在相互作用分析中包括的相互作用物的一种(捕获分子)。或者,如WO2006/041392中所述,配体和蛋白抗性化合物被偶联到水凝胶(例如CMDx)的单独部分。然而,该表面可能不适合某些敏感样品,因为它们受到基线波动的影响。

发明概述

使用根据本发明生产的传感器表面运行SPR测定的方法包括在传感器表面上添加可能与捕获分子(配体)相互作用的分析物,并执行动力学测定和/或浓度测定。
在第二个方面,本发明涉及一种传感器表面,其包含常用于SPR的类型的基底或芯片,例如金属镀覆的表面,其具有自组装单层(SAM),在所述SAM上直接提供有官能团,其中蛋白抗性化合物偶联到第一部分的官能团,且其中捕获分子(配体)偶联到表面上的第二部分的官能团,但没有配体偶联到蛋白抗性化合物。
优选地,表面是金属镀覆的表面,优选金镀覆的表面,SAM是硫醇-烷烃C10-C30 SAM,最优选MHA-SAM,和蛋白抗性化合物是聚乙二醇(PEG)或其衍生物。在一个优选的实施方案中,蛋白抗性化合物是浓度为0.05-50 mM,优选0.5-5 mM的PEG mw 5000。

发明详述
在初步研究中,本发明人已经发现,由低聚(乙二醇) (OEG)衍生的烷硫醇化合物或这些化合物与没有OEG的烷硫醇化合物的混合物形成的自组装单层(SAM)显示出高的非特异性结合。得自循环伏安分析的结果不佳,表明不利的填充密度和不佳的表面覆盖率,因此样品的不需要和非特异性结合到金表面/被破坏的SAM的可能性很高。该结果表明,含SAM烷硫醇-OEG的表面不适合于高灵敏度的应用。
本发明提供了一种新的传感器表面,其中捕捉分子将与传感器表面紧密结合,而不与任何基质分子结合,以最大限度地减少基线漂移。捕获分子直接结合到烷硫醇SAM层(或通过SAM上的接头),而不是通过PEG或OEG或任何其它亲水性或蛋白抗性部分。本发明提供了一种方法,其包括用烷烃-SAM建立传感器表面以实现分离性能,其中在第二步中引入亲水性基质以获得蛋白排斥性质,同时提供用于SAM上配体固定的可利用的官能团以实现低的基线漂移。
用硫醇-烷烃(如C10-C30烷烃)试剂,可以在金表面上产生自组装单层(SAM)。SAMC16显示出具有良好分离性能的高密度SAM,其是避免样品组分与金表面以及传感器表面上任何被破坏的SAM组分的非特异性结合所必需的。
蛋白抗性化合物形成基质,如PEG基质,通过SAM层上的官能团共价固定。加入该基质分子仅是因为它的蛋白排斥性质或其它对测定有益的性质。因此,配体和蛋白抗性化合物(基质)二者均直接或通过接头偶联到SAM单层。例如,接头可以是表氯醇。接头是一种短分子,而不是现有技术的亲水化合物。
本发明的优点是没有配体固定到蛋白抗性化合物(基质)上。如果将相同的配体固定在基质和SAM上,那么固定的配体将代表两个不同的配体群体,一个群体固定在SAM层上,而另一个群体固定在基质上。这将对测定结果产生负面影响。使用本发明,所有配体都只被固定在SAM官能团上,因为PEG基质或替代基质或基质混合物在独立的步骤中被固定在SAM官能团上,并且没有官能团被用于配体固定。在失活(任选的)之后,SAM单层上剩余的官能团可用于配体固定。
在本发明中,优选的硫醇-烷烃试剂是MHA 16 (巯基十六烷酸)。如果硫醇-烷烃试剂是MHA,则具有伯胺的PEG分子可以共价地连接至传感器表面上的MHA SAM层上的羧基部分。通过EDC/NHS化学可以将含有伯胺基团的PEG分子共价连接到含有COOH (羧基)的SAM层。含偶联到羧基化烷硫醇SAM的胺官能化PEG-分子(如5 kDa)的传感器表面表现出良好的基质稳定性特性,并被证明对血浆样品和洗涤剂的非特异性结合具有高的抵抗力。
蛋白抗性化合物(基质)和配体也可以通过SAM层上的其它官能团固定。用于固定的SAM层上的可能官能团的实例是:羟基、氨基、羧基、醛、羰基、环氧、乙烯基和硫醇。
参与SAM层上固定的蛋白抗性化合物(基质)上可能的官能团的实例是:羟基、氨基、羧基、醛、羰基、环氧、乙烯基和硫醇。将蛋白抗性化合物(基质)固定到SAM层后,蛋白抗性化合物(基质)上的任何剩余官能团都必须不能被利用,即不能用于配体/捕获固定,和/或在必要时失活。或者,它们可以不同于SAM上的用于配体固定的官能团。
蛋白抗性化合物(基质)通过自发共价偶联或通过基质上或者SAM层上的官能团(例如SAM上的醛和基质上的肼)的活化,固定到SAM层。对不反应的活化或反应性基团执行失活。可包括基质或接头的稳定化,例如,醛和肼之间的偶联通过NaCNBH4来稳定。
活化和失活程序对本领域的技术人员来说是已知的,可以是例如,通过EDC/NHS化学引入活性酯或通过表氯醇引入环氧化物的SAM或基质的活化。或者,用通过双官能试剂的建立的化学(例如表面上的胺基和双官能试剂上的NHS衍生物)的活化,以及来自双官能试剂的反应性二硫化物用于配体固定。
配体固定
有许多不同的偶联化学能够使配体固定至传感器表面(芯片表面)上的COOH基团。最常见的途径是使用胺偶联,由此芯片表面上的羧基被用于与蛋白(配体)中伯胺基团形成共价酰胺键。然而,这一过程并不是自发发生的,需要活化羧基。用N-乙基-N’-(3-二甲基氨基丙基)碳二亚胺(EDC)和N-羟基-琥珀酰亚胺(NHS)的混合物进行活化。EDC与羧基反应并形成反应中间体,其进而与NHS反应以形成活性NHS酯。当配体在传感器芯片表面上通过时,NHS-部分(其为良好的离去基团)与配体上的伯胺基团自发反应,在配体和芯片表面之间形成共价键。大多数蛋白含有若干伯胺,因此可以在不严重影响配体的生物学活性的情况下实现固定。
为了促进配体固定,在称为预浓缩的过程中使用静电吸引力。将配体溶解在偶联缓冲液中,其pH低于蛋白的等电点(pI),但高于芯片表面的pI。因此,配体和芯片表面获得相反的净电荷。带正电荷的配体分子被静电吸引到负表面,并且表面附近的高配体浓度导致更有效的固定。
配体固定后,用例如乙醇胺或NaOH (其去除残留的NHS酯)使过量的NHS-活化的羧基失活,使得在注射分析物期间蛋白不再可以固定至表面。也可以用带正电荷的表面和带负电荷的配体进行该过程。
配体水平可通过配体浓度、活化或配体接触时间、配体固定/偶联缓冲液的组成和pH或这些参数的组合来控制。本发明人已惊奇地发现,使用根据本发明的具有MHA SAM层的传感器表面,配体水平通常随着活化时间的缩短而增加。这与具有水凝胶(如CMDx)的可利用的传感器表面相比是相反的。对于后者,配体水平随着活化时间(接触时间)的增加而增加。
实验部分
实验1:具有MHA SAM和PEG的传感器表面的合成
对于该实施例,参考图1。
在25℃下,将包含具有金表面的玻璃芯片的基底浸泡在含1 mM MHA的80%/20%乙醇/水中并孵育过夜。金芯片用以下溶液洗涤5分钟,并按所示顺序洗涤:100%乙醇、80%乙醇、50%乙醇、20%乙醇和100%水。
传感器芯片用氮气干燥,并浸入在水中的0.144 M EDC和0.050 M NHS中,在25℃下孵育30分钟。传感器芯片用水洗涤4次,然后用含1.79 mM O-(2-氨基乙基)聚乙二醇mw5000的0.252 M磷酸钠缓冲液(pH8.5)浸泡并在25℃下孵育30分钟。弃去PEG溶液,用1 M乙醇胺(pH 8.5)使残留的活性酯失活。最后,用水洗涤传感器芯片4次。芯片用氮气干燥。
实验2:使用不同活化时间,使配体(抗体)固定至SAM MHA上的羧基
得自实验1的芯片用塑料载体和罩组装。此后,根据制造商在控制软件中的说明,将芯片对接在Biacore 3000中。用200 mM EDC和50 mM NHS在水中的混合物在仪器中活化SAM上的羧基30、60、90和180秒。
将在10 mM醋酸钠(pH 5)中的50 µg/ml小鼠IgG1抗体在活化的芯片表面上注射7分钟。用1M乙醇胺(pH 8.5)使芯片表面(未反应的琥珀酰亚胺酯)失活5分钟。在不同的流动池中用不同活化时间进行固定。运行缓冲液HBS-EP (BR100188) (GE Healthcare)。
对于这个实施例,参考图3中的图,它表明固定水平与30秒后的活化时间成反比。
实验3:使用本发明的传感器表面的测定
得自实验1的芯片用塑料载体和罩组装。根据控制软件中的说明,将芯片对接在Biacore® 8K中。SAM上的羧基在仪器中用200 mM EDC和50 mM NHS在水中的混合物活化30秒。
将在10 mM醋酸钠(pH 5)中的30 µg/ml小鼠抗-β2µ在活化的芯片表面上注射7分钟。用1M乙醇胺(pH 8.5)使芯片表面(未反应的琥珀酰亚胺酯)失活5分钟。将在运行缓冲液中的不同浓度的β2µ (1、2、4、8、16 nM)在固定的表面和无配体的参考点/表面上注射。运行缓冲液为HBS-P+。确定β2µ对小鼠抗-β2µ的结合动力学是可能的。应用至一对一结合模型的拟合。
对于这个实施例,参考图4,其显示了减去参考的传感图。通过Biacore® 8K评价软件,计算了缔合常数(Ka)、解离常数(Kd)和亲和常数(KD)。结果与先前确定的可获得的结果一致。
Claims (14)
1.一种制备用于SPR仪器的传感器表面的方法,其包括通过使所述表面与一种或多种具有至少一种类型的官能团的硫醇C10-C30烷烃试剂反应,在表面上形成自组装单层(SAM),其中一种或多种蛋白抗性化合物与第一部分的所述官能团偶联;和其中一种或多种捕获分子直接或经由接头偶联到所述表面上的第二部分的所述官能团,所述接头不是所述蛋白抗性化合物,其中没有捕获分子偶联到蛋白抗性化合物。
2.依据权利要求1的方法,其中第二部分的官能团被包括在通过第一部分的官能团偶联蛋白抗性化合物之后在表面上剩余的那些中,并且其中第一和第二部分的官能团具有相同或不同的类型。
3.依据权利要求1或2的方法,其中所述表面是金属镀覆的表面,优选金或银镀覆的表面。
4.依据权利要求1、2或3的方法,其中C10-C30硫醇烷烃试剂的官能团是羧基、羟基、氨基、醛基、环氧基、乙烯基、羰基或硫醇基的一种或多种。
5. 依据上述权利要求的一项或多项的方法,其中C10-C30硫醇烷烃试剂是MHA (巯基十六烷酸)或其衍生物。
6.依据上述权利要求的一项或多项的方法,其中蛋白抗性化合物是选自聚乙二醇或其衍生物的亲水性聚合物,例如具有<20000的mw的甲氧基聚乙二醇胺。
7.依据上述权利要求的一项或多项的方法,其中蛋白抗性化合物是具有100-20000的mw,优选2000-10000的mw,最优选4000-6000的mw的PEG。
8.依据上述权利要求的一项或多项的方法,其中第二部分的官能团在捕获分子(配体)的偶联之前被活化,并且偶联的捕获分子(配体)的量与活化时间长度成反比;并且其中活化试剂选自EDC/NHS、表氯醇或双官能试剂。
9.使用依据权利要求1-8的一项或多项生产的传感器表面运行SPR测定的方法,其包括在传感器表面上加入可能与捕获分子(配体)相互作用的分析物,并执行动力学测定和/或浓度测定。
10.依据权利要求1生产的传感器表面,其包括具有自组装单层(SAM)的基底,其提供有在所述SAM上可利用的官能团,其中蛋白抗性化合物偶联到第一部分的官能团和其中捕获分子(配体)偶联到表面上的第二部分的官能团,并且其中没有捕获分子(配体)偶联到蛋白抗性化合物。
11.依据权利要求10的传感器表面,其中蛋白抗性化合物和捕获分子偶联到相同类型的官能团。
12.依据权利要求10的传感器表面,其中蛋白抗性化合物和捕获分子偶联到不同类型的官能团。
13.依据权利要求10、11或12的传感器表面,其中所述表面是金属镀覆的表面,优选金镀覆的表面,和SAM是硫醇-烷烃C10-C30,优选MHA-SAM,和蛋白抗性化合物是聚乙二醇(PEG)或其衍生物。
14. 依据权利要求10、11、12或13的传感器表面,其中蛋白抗性化合物是浓度为0.5-5mM的PEG mw 5000。
