Abstract
Description
本発明は、新規ポリマー、その製造、および表面被覆におけるその使用、被覆表面、ならびにアッセイ装置および方法におけるその使用に関する。本発明は、特に、カルコゲン基を組み込んでいる成分を含有するポリマー、その製造、およびバイオセンサの表面被覆におけるその使用、バイオセンサ自体、およびアッセイ法におけるその使用に関する。
非特異的結合を最小限にしながら、表面に分子を特異的に固定化することは、生体適合性が因子である多くの分野、例えば、バイオセンサの製造、および表面汚損(surface fouling)の防止において、重要であることが示されている。そのような固定化は、特異的結合対の構成員に結合することができる生体適合性被覆剤を使用して、様々な成功度で行われている。適切な条件下に、対のもう一方の構成員は、非特異的結合(例えば、タンパク質、細胞、細菌または他の好ましくない物質の好ましくない結合)を最小限にしながら、アッセイ条件下で被覆剤に結合することができる。これは、バイオセンサ、特に表面プラズモン共鳴および圧電検出器における、被覆物の使用を可能にしている。

本発明は、式−X−Y−Z−Rで示される共有結合側鎖を有するポリマーを提供し、式中、Xは、スペーサ基であり;Yは、硫黄、セレンまたはテルル原子であり;Zは、硫黄、セレンまたはテルル原子であり、それらのいずれかが1個または2個の酸素原子に結合していてよく;Rは、−Z−Rが脱離基を構成するような任意の好適成分である。

本発明は、式−X−Y−Z−Rで示される共有結合側鎖を有するポリマーを提供し、式中、Xは、スペーサ基であり;Yは、硫黄、セレンまたはテルル原子であり;Zは、硫黄、セレンまたはテルル原子であり、それらのいずれかが1個または2個の酸素原子に結合していてよく;Rは、−Z−Rが脱離基を構成するような任意の好適成分である。

使用において、分析すべき溶液は、生物学的源または他の源に由来しうる。例えば、体液、細胞抽出物、食品材料、科学試料等。そのような溶液は、化学物質、薬剤、ステロイド、組織、膜、膜フラグメント、ヌクレオチド、オリゴヌクレオチド、タンパク質、オリゴ糖、細胞、ファージ、細菌、ウイルス、または特異的結合対のもう一方の構成員と特異的相互作用をすることができる基を含有する任意の他の構造物を、分析物として含有しうる。
従って、本発明は、特異的結合対の構成員を分析する方法を提供し、該方法は下記を含んで成る:特異的結合対の構成員を含有すると思われる試料を、本発明のバイオセンサに接触させ(その場合、R1は特異的結合対のもう一方の構成員である)、バイオセンサからの信号の変化を記録する。
ポリマーをp−ニトロフェニルカーボネート誘導体に変換しうる。次に、そのようなp−ニトロフェニルカーボネート誘導体化ポリマーを、アミンNH2−B−Y−Z−R(B、Y、ZおよびRは前記のように定義される)と反応させうる。
実施例
下記の実施例において、シスタミンジスルフィドおよび酸二硫化物に関する実施例は、単に比較を目的とするものである。
中間体1
2−(ピリジニルジチオ)エタンアミン(PDEA)の合成
アルリチオール−2(4.41g、20mmol)を、メタノール20mLおよび酢酸0.8mLに溶解させた。その溶液に、メタノール10mL中の2−アミノエタンチオールヒドロクロリド(1.14g、10mmol)を添加した。反応混合物を2日間撹拌し、次に、真空濃縮して、黄色油状物を得た。次に、ジエチルエーテルを用いた激しい撹拌によって、黄色油状物を充分に洗浄した。透明黄色上澄みをデカンテーションによって除去し、残渣をメタノール(10mL)に溶解させた。得られた溶液に、エーテル50mLを添加し、沈殿物を濾過によって分離した。この手順を3回繰り返し、次に、得られた最終固形物を再結晶(メタノール/ジエチルエーテル)によって精製して、淡白色固形物(1.62g)を得た。
4−ニトロフェニル炭酸化デキストラン(参照番号:AKU34−103)
無水DMSO 18mLおよび無水ピリジン16mL中の、デキストラン T70(1.6g、29.6mmol OH)の溶液に、4−ニトロフェニルクロロホルメート(800mg、4mmol)および触媒量のDMAP(N,N−ジメチルアミノ−4−ピリジン)を、0℃(外部冷却浴)で撹拌しながら添加した。反応混合物を、この温度で1時間、次に、室温でさらに1時間撹拌した。溶液を、メタノールおよびエーテルの混合物(1:1)(150mL)に、激しく撹拌しながらゆっくり添加した。形成された沈殿物を濾過によって収集し、同じ溶媒混合物で3回洗浄した。収集した白色固形物を高真空下で乾燥して、白色粉末1.54gを得た。

無水DMSO 12mLおよび無水ピリジン3mL中の、4−ニトロフェニル炭酸化デキストランAKU34−103(750mg)の溶液に、NMM(N−メチルモルホリン)(200L)およびシスタミンジヒドロクロリド(1g、8.8mmol)を添加した。混合物を室温で一晩撹拌し、次に、メタノールおよびエーテルの混合物(1:1)(75mL)に、激しく撹拌しながら添加した。形成された沈殿物を濾過によって収集し、同じ混合物で3回洗浄した。収集した白色固形物を、高真空下でさらに乾燥させて、白色粉末530mgを得た。官能性度:6.6%

デキストラン−酸二硫化物(デキストラン T70)の製造
デキストラン−酸二硫化物(参照番号:AKU34−118)
無水DMSO 12mL中のデキストラン−PDEAジスルフィド AKU34−110(500mg)の溶液に、3−メルカプトプロピオン酸(200μL、2.3mmol)を添加した。混合物を室温で一晩撹拌し、次に、メタノールおよびエーテルの混合物(1:1)(50mL)に、激しく撹拌しながらゆっくり添加した。形成された沈殿物を濾過によって収集し、同じ混合物で3回洗浄した。収集した白色固形物を高真空下に乾燥させて、白色粉末450mgを得た。官能性度10.5%
実施例1
デキストラン−PDEAジスルフィド(デキストラン T70)の製造
4−ニトロフェニル炭酸化デキストラン(参照番号:AKU34−106)
無水DMSO 18mLおよび無水ピリジン16mL中の、デキストラン T70(1.6g、29.6mmol OH)の溶液に、4−ニトロフェニルクロロホルメート(800mg、4mmol)および触媒量のDMAPを、撹拌および外部冷却浴(0℃)下で添加した。反応混合物をこの温度で5時間撹拌した。溶液を、メタノールおよびエーテルの混合物(1:1)(150mL)に、激しく撹拌しながらゆっくり添加した。形成された沈殿物を濾過によって収集し、同じ溶媒混合物で3回洗浄した。収集した白色固形物を高真空下に乾燥させて、白色粉末1.57gを得た。

合成を2回繰り返して、極めて類似した結果を得た。第二合成の生成物はAKU34−175と称し、第三合成の生成物はNo.110105と称した。

この合成を同じ条件下で繰り返し、同様の結果を得た。第二合成の生成物を、AKU34−178とした。
カルボキシメチルデキストラン(428015)
デキストランT70(約10mmolグルコース単位)1.6gを、4M NaOH水溶液(25mL)(水25mL中の4g NaOH)に溶解させた。これに、水20mL中のMCA(モノクロロ酢酸、7.56g)およびNa2CO3(4.24g)の溶液を添加した。得られた溶液を90〜100℃で3時間撹拌した。次に、それを、氷水での冷却浴下に、濃HCl水溶液でpH3.0に酸性化し、pH7.0になるまで蒸留水に対する透析によって精製し、凍結乾燥によって乾燥させて、白色綿毛状固形物を得た。
デキストランT70−COOH(428015、D.S. 0.64)500mgを、無水DMSO 16mLに60℃で溶解させた。次に、得られた透明溶液を、室温に冷却し、その溶液に、HOBt(2.5mmol)383mgおよびジイソプロピルカルボジイミド(DIC)0.78mLを添加した。混合物を室温で10分間撹拌し、次に、ピリジン3mLおよびPDEA 0.55gを添加した。得られた混合物を、室温で一晩撹拌した。次に、それをアセトン50mLに滴下した。得られた混合物に、エーテル20mLおよび石油エーテル200mLを添加した。次に、得られた沈殿物を濾過によって収集し、アセトンで5回洗浄し、高真空下に乾燥させて黄色粉末508.3mgを得た。

メタンチオスルホン酸ナトリウム(参照番号:428028)
メタノール(60mL)中の、ナトリウムメタンスルホネート(1g、9.8mmol)および硫黄(312mg、9.75mmol)の混合物を、全ての硫黄が溶解するまで3時間にわたって還流させながら加熱した。次に、混合物を濾過し、濾液を真空濃縮して乾燥させて、白色固形物を得た。

全てのポリマーを、窒素下に−20℃で、凍結乾燥粉末として保存した。超純水中のポリマーの0.1〜2%w/v溶液を、目視的に透明になるまで室温で緩やかに振とうすることによって作製し、次に、Genofuge 16M ベンチトップ遠心器によって14000rpmで遠心分離して、あらゆる不溶質を沈殿させた。次に、上澄みをピペットで注意深く除去し、使用するまで4℃で保存した。
タンパク質および試薬
抗−HSAおよび抗−BSAマウスモノクローナル抗体(AbCam、UK)を、1mg/mL(約、1.5μM)で4℃において保存し、次に、後の結合アッセイ用に、流れ緩衝液(running buffer)で333nMに希釈した。バイオラドタンパク質アッセイ染料試薬を使用するブラッドフォード法によって、タンパク質濃度を測定した。タンパク質純度をSDS−PAGEによって測定した。トリトンX−100、ウシ血清アルブミン、ヒト血清アルブミン、システイン、グリシン、ジチオトレイトール、塩化ナトリウム、水酸化ナトリウム、塩酸、複合緩衝剤(10mM 酢酸ナトリウム緩衝剤 pH4.5、100mM 蟻酸緩衝剤 pH4.3、および100mM ホウ酸緩衝剤 pH8.5)を、Sigma−Aldrich,UKから購入し、その関連溶液は、使用前に0.22μmフィルターで濾過した。
2番コーニングガラススライドを、チタン接着層、次に、47nmの金層で、Showa e−ビーム蒸発器において被覆した。そのガラススライドを、BIACORE(登録商標)2000表面プラズモン共鳴(SPR)バイオセンサ(Biacore, UK)に挿入するのに好適なプラスチックホルダーに取り付けた。e−ビーム蒸発器(Showa)において蒸着によって1.5nmチタン接着層および47nm金層で被覆したAF 45、0.30mm厚ガラススライド(Perfection,Camb. UK)を使用して、SPRガラスチップブランクを作製した。作製したセンサチップは、各フローセルにつき約0.26mm2のSPR信号用プロービングスポットを有する装置中に、寸法2.4 × 0.5 × 0.05mm(l × w × h)の4つのフローセルを形成した。全てのSPR実験を、25℃において0.5秒毎のデータ点で行った。
BIACORE(登録商標)2000バイオセンサシステムに超純水を通し(primed)、次に、100mM NaOH/1% トリトンX−100の溶液40μLを流速10L/分で注入)することによって、金チップの表面を清浄化した。各注入の間において、流れ溶離剤は超純水であった。この注入後直ぐに、活性脱離基ポリマーの溶液50μLを5μL/分で注入した。これによって、装置において得られる最大流速(100μ/
分)までの水の連続流動下に、安定な反応レベルを生じた。典型的な結果を図3に示し、該図は、時間(秒)に対する振動数(dF、Hz)の変化のグラフである。非化学吸着物質は、100mM NaOHのパルスで除去することができ、それによって極めて安定なSPR信号が得られ、該信号は、100mN NaOH、100mM HCl、1M NaCl、1%トリトンX−100、1mM DTTおよび1mM L−システインのいずれの付加的注入によっても影響を受けなかった。PDEA−ポリマーについて、反応は、表面に結合したポリマー約3500RU(約3.5ng/mm2に対応する)のオーダーであり、チオスルホンポリマー(下記の実施例11〜13)については、反応は、表面に結合したポリマー約2200RU(約2.2ng/mm2)のオーダーであった。
PDEA−ポリマーへのタンパク質の結合−SPR
タンパク質スルヒドリル基への直接結合: 100mM ホウ酸緩衝液/1M NaCl、pH8.5中に構成したヒト血清アルブミンの溶液(50μL、1mg/mL)を、ポリマーを付着させた面に注入し、それによってタンパク質1000〜2000RUを固定させた。次に、100mM ホウ酸緩衝液 pH8.5中の50mM システイン(50μL)の注入によって、残留活性ジスルフィド基を不活性化(キャッピング)した。典型的な結果を図4に示し、該図は、時間(秒)に対するdF(Hz)のグラフである。

市販BIACORE(登録商標)CM5表面へのタンパク質の結合−SPR
同量のNHS(50μL、水中50mM)およびEDC(50μL、水中200mM)を混合し、次に、この溶液50μLを10μ/分でBIACORE(登録商標)CM5 カルボキシメチルデキストランセンサーチップに注入した。このあと直ぐに、10mM NaOAc緩衝液、pH4.5中のHSA(50μL、50μg/mL)を注入し、それによって、タンパク質約8000RUを固定させた。次に、残留NHSエステルを、エタノールアミン(50μL、1M、pH8.0)の注入によって不活性化した。
ポリマー捕捉タンパク質受容体への抗体の結合−SPR
マウス抗−HSA IgGを、流れ緩衝液(PBS: 10mM Na2HPO4/NaH2PO4、137mM NaCl、2.7mM KCl、pH7.4)において、333〜4.1nMに3倍希釈し、次に、流速20μ/分で、固定BSA含有フローセル、次に、固定HSA含有フローセルに連続的に通した。IgGの5分間注入および固定アルブミン約1500RUを使用して、カイネテッィクアッセイを行った。次に、試料溶液を流れ緩衝液と置き換え、抗体−リガンド複合体を5分間解離させた。遊離タンパク質受容体の再生は、塩溶液(5μL、10mM NaCl、pH2.0)の注入によって行った。
水晶共鳴検出器
水晶実験を、下記のような機器で行った。基材にエッチングされた2つの共鳴器を有するメサ構造を有するATカット水晶を、従来の蒸発法によってチタン付着層(5nm)および金(200nm)で被覆した。該検出を、図6に模式的に示す。
PDMS)インサートを有する。インサートは、取替え可能であり、2つのバージョンを有し、該バージョンを使用して、2つの検出器に、別々に(6)、または共通に(8)、溶液中の試薬を送達しうる。多方向Rheodyneバルブ(10)を使用して、緩衝液または試薬をセンサに供給する。緩衝液および試料マクロ流体工学(macrofluidic)システムは、2つのハミルトンシリンジポンプユニット、1対のYコネクタ(12、14)、分離空気圧縮(または「サンパー」(‘thumper’))バルブ(16)、試料装填/注入多方向バルブ(10)およびフローセルセンサセレクタバルブ(18)を有する。緩衝液ポンプユニット上の2つのシリンジ(一般に、500μLガラスシリンジ)は、交互に吐出して、一定の緩衝液流れを維持する。試料ポンプ(4)は、注入すべき試料を試料ループに引き込み、次に、流れ緩衝液の分岐によって試料ループを通って注入するための、1つの5mLシリンジを有する。
下記の実施例において、先ず、シングルセルアドレッシングによって2つのセンサを共にポリマーで被覆した。被覆の終了後、センサを緩衝液で充分に洗浄して、遊離PDMAの全痕跡を除去し、次に、セルに再配置した。
次に、セルを個々に、HSAで処理して特異的結合対の第一構成員(第二構成員はHSAに対する抗体である)を担持する表面を形成し、BSAで処理して対照表面を形成した。HSAへの抗HSAの固定化を測定するために、次に、2つのセンサを再びシングルアドレッシングモードで使用し、抗HSA溶液を両センサ上に流した。試料および対照チャンネルについて、リアルタイムの振動数の変化を記録した。種々の濃度の抗HSA分析物を使用して、試験を繰り返した。試料センサの振動数シフトを、対照センサのバックグラウンドシフトに関して補正し、この補正値を、攻撃された抗HSAの量と見なした。分析を下記のように行った。
15%PDEA置換デキストラン表面(AKU34−178)を、下記のように、ビアコアチップ上に作製した。
15%PDEA置換デキストラン表面(AKU 34−178)を、前記のように作製し、残留PDEA基をキャッピングし、実施例7のようにデキストラン上のOH基をCOOH反応性基に変換した。表面をEDC/NHSで活性化し、カルボニックアンヒドラーゼII(CAII)に曝露した。COOHとCAIIのアミン基との間に反応が起き、攻撃タンパク質による1800Hzの振動数シフトが生じた。対照表面を活性化し、エタノールアミンでキャッピングした。表面作製の典型的結果を、図9に示し、該図は、時間(秒)に対する振動数変化(Hz)のグラフである。
実施例7に記載した表面を、10μg/mL ビオチン化BSAに曝露し、結合を図11に示し、該図は、時間(秒)に対する応答(任意の応答単位)のグラフである。この場合、フローセル(FC)3は、マウス抗ビオチンIgGへのビオチン化BSAの結合を示し、FC2は、マウスIgG対照表面への非特異的結合を示し、FC1は、キャッピングしたポリマーへの結合を示し、FC4は、遊離ポリマーへの結合を示す。
実施例7と同じ手順を使用して、デキストランT70−COOH−PDEA(428018)を付着させ、マウス抗ビオチンIgGを固定化した。固定化の測定値を図12に示
し、該図も時間(秒)に対する応答(RU)のグラフである。
タンパク質、試薬、SPR分析に好適な金表面、および抗体を、実施例2と同様に準備した。代替脱離基ポリマー、デキストラン−T70チオスルホン(実施例1からの428034)を、1mg/mL溶液として調製し、実施例2に記載のようにSPRセンサチップの金表面に付着させた。この結果、2200RU(表面に結合した約2.2ng/mm2ポリマーに対応する)のオーダーで付着を生じた。このポリマーの二重注入によって、図14(時間(秒)に対するRU)に示す結果を得た。
実施例11のチオスルホンポリマーで被覆した(428034)金表面に、ヒト血清アルブミン(HSA)またはウシ血清アルブミン(BSA)を、実施例3に記載のように結合させた。該タンパク質は、それらのスルフヒドリル基を介してチオスルホンに直接的に結合し、その結果を図15(時間に対するRUのグラフ)に示す。
抗HSA IgGを準備し、希釈し、次に、実施例5に記載の方法を使用して、実施例12の両タンパク質結合ポリマー表面に通し、但し、遊離タンパク質受容体の再生を10mM HClの溶液(5mL、40mL/分)の注入によって行った。さらに、対照抗体、正常マウスIgGを、111nMの濃度で両表面に注入した。図16(時間に対するRUのグラフ)は、曲線Aにおいて、HSA表面への抗HSAの結合;曲線Bにおいて、BSA表面への抗HSAの結合;曲線Cにおいて、HSA表面への抗マウスIgGの結合;および、曲線Dにおいて、BSA表面へのマウスIgGの結合;の結果を示す。

- ZがSまたはSO2である、請求項6に記載のポリマー。
- Rが、電子求引基に共役した不飽和基、芳香族基、複素環式芳香族基および求電子基の1つを含んで成る、前記請求項のいずれか1つに記載のポリマー。
- 電子求引基が、低級アルキルオキシカルボニル、ニトリル、ニトロ、低級アルキルスルホニルおよびトリフルオロメチルの1つまたは複数を含んで成る、請求項8に記載のポリマー。
- 芳香族基が、任意に置換されたフェニルを含んで成り、該置換されたフェニルにおいて、ニトロ、トリフルオロメチル、ニトリル、低級アルキルオキシカルボニルまたは他の電子求引基から選択される3個までの置換基が存在してよい、請求項8に記載のポリマー。
- 複素環式芳香族基が、フェニル環または付加的5または6員芳香族複素環の残基に任意に縮合した5または6員環を含んで成り、前記芳香族複素環または付加的芳香族複素環が、1個または2個の低級アルキル、フェニル、=O、=S、トリフルオロメチル、ニトロまたはニトリル基によって任意に置換されていてよい、請求項8に記載のポリマー。
- 成分−Z−Rが、芳香族チオール、複素環式芳香族チオールまたはそれらのチオン互変異性体から誘導される、前記請求項のいずれか1つに記載のポリマー。
- 成分−Z−Rが、下記から成る群から誘導される、請求項12に記載のポリマー:
イミダゾール;ピロリジン−2−チオン;1,3−イミダゾリジン−2−チオン;1,2,4−トリアゾリン−3(5)−チオン;1,2,3,4−テトラゾリン−5−チオン;
2,3−ジフェニル−2,3−デヒドロテトラゾリウム−5−チオン;N(1)−メチル−4−メルカプトピペリジン;チオモルフィリン−2−チオン;チオカプロラクタム;ピリジン−2−チオン;ピリミジン−2−チオン;2−チオウラシル;2,4−ジチオウラシル;2−チオシトシン;キノキサゾリン−2,3−ジチオン;1,3−チアゾリン−2−チオン;1,3−チアゾリジン−2−チオン;1,3−チアゾリジン−2−チオン−5−オン;1,3,4−チアジアゾリン−2,5−ジチオン;1,2−オキサゾリジン−2−チオン;ベンズ−1,3−オキサゾリン−2−チオン;1,3,4−オキサジアゾリン−2−チオン、および硫黄がセレンまたはテルルによって置き換えられた類似体。 - Rが2−ピリジル基である、前記請求項のいずれか1つに記載のポリマー。

- スペーサXが、1個または複数の、低級アルキルオキシ、ハロ、オキソ、トリフルオロメチル、ニトリル、または−Y−Z−R成分の形成および使用を妨げない他の基によって置換されていなくてもよくまたは置換されていてもよいアルキレン基またはフェニル基を含んで成る、前記請求項のいずれか1つに記載のポリマー。
- スペーサXが、側鎖をポリマーの残部に結合させる結合成分を含んで成り、該結合成分が、−O−、−O−CO−、−O−CO−O−、−NH−、低級アルキル置換−NH−、−O−CO−NH−、および低級アルキルN置換−O−CO−NH−から成る群から選択される、前記請求項のいずれか1つに記載のポリマー。
- スペーサ基が、式−A−B−で示され、式中、Bは請求項16に記載の非置換または置換アルキレンまたはフェニル基であり、Aは請求項17に記載の結合成分である、前記請求項のいずれか1つに記載のポリマー。
- スペーサが、成分Bを含んで成り、該成分が、酸素原子によって任意に中断された低級アルキレン基、カルボキシル基またはカルボキシルオキシ基である、請求項17または19に記載のポリマー。
- スペーサXが、成分Bを含んで成り、該成分が、直鎖アルキレニル基−(CH2)n−であり、式中、nが1〜4、好ましくは2である、請求項20に記載のポリマー。
- Xが−CO−NH−CH2−CH2であり、YがSであり、ZがSであり、Rが2−ピリジルである、前記請求項のいずれか1つに記載のポリマー。
- 親水性である、前記請求項のいずれか1つに記載のポリマー。
- 中性である、前記請求項のいずれか1つに記載のポリマー。
- −X−Y−Z−R側鎖が、下記から成る群から選択される分子に結合している、前記請求項のいずれか1つに記載のポリマー:
デキストラン、ヒアルロン酸、セファロース、アガロース、ニトロセルロース、ポリビニルアルコール、部分加水分解ポリビニルアセテートまたはポリメチルメタクリレート、カルボキシメチルセルロース、およびカルボキシメチルデキストラン。 - −X−Y−Z−R側鎖が、糖モノマー単位から誘導される分子に結合している、前記請求項のいずれか1つに記載のポリマー。
- −X−Y−Z−R側鎖に加えて、ポリマーが、式−X−Y−Z−R1で示される側鎖も含んで成り、式中、X、YおよびZは請求項1に定義された通りであり、R1は特異的結合対の構成員である、前記請求項のいずれか1つに記載のポリマー。
- 側鎖の−Z−R基の少なくともいくつかが置換され、ポリマーが−X−Y−側鎖を介して基材に共有結合するような、前記請求項のいずれか1つに記載のポリマーと反応した基材。
- 基材の少なくとも一部が、請求項1〜27のいずれか1つに記載のポリマーで被覆され、該ポリマーが、−X−Y−側鎖を介して基材に共有結合し、側鎖の−Z−R基の少なくともいくつかが置換されていない、請求項28に記載の基材。
- 金属表面を含んで成る、請求項28または29に記載の基材。
- 金属表面が、金、銀、白金、パラジウム、ニッケル、クロム、チタン、銅およびそれらの任意の合金を含んで成る、請求項30に記載の基材。
- 基材がバイオセンサの一部を構成する、請求項28〜31のいずれか1つに記載の基材。

- 請求項28〜35のいずれか1つに記載の基材を含んで成るバイオセンサ。
- SPRバイオセンサおよび音響バイオセンサから成る群から選択される、請求項36に記載のバイオセンサ。
- 基材に成分を間接的に結合させる方法であって、基材を、請求項1〜27のいずれか1つに記載のポリマーと反応させる工程を含んで成り、該ポリマーが、基材に間接的に結合すべき成分を含んで成る側鎖を有する方法。
- 下記の工程を含んで成る、基材に成分を間接的に結合させる方法:
基材を、請求項1〜27のいずれか1つに記載のポリマーと反応させる工程であって、該反応が、ポリマーの側鎖から全てではなくいくつかの−Z−R基を置換し、それによって、ポリマーが−X−Y基によって基材に結合する工程;および
基材に間接的に結合すべき成分を含んで成る試薬に、該結合ポリマーを接触させ、それによって、該成分をポリマーに結合させる工程。 - 試薬が、結合ポリマー上に存在する非置換−Z−R基と反応する、請求項39に記載の方法。
- 基材に間接的に結合すべき成分を付加する試薬との接触前に、結合ポリマーを、化学物質によってさらに修飾する、請求項39に記載の方法。
- ポリマー上に存在するヒドロキシ、カルボキシ、エポキシまたはアミノ基を使用して成分を結合させる、請求項41に記載の方法。
- 成分が、特異的結合対の構成員である、請求項38〜42のいずれか1つに記載の方法。
