Abstract
本发明公开了一种奇亚籽在制备抑制肠道蠕动的药品或食品中的应用。本发明能有效抑制肠道蠕动,延长药物在肠道内的停留时间,从而可增加药物的治疗效果。本发明同时研究奇亚籽用于代餐减肥产品中的新的作用机制与功能,明确奇亚籽发挥减肥效果的作用过程,为奇亚籽在代餐领域深加工提供指引,也为奇亚籽在代餐减肥中的作用机制提供新的导向。研究发现,奇亚籽发挥减肥效果,除依靠其高泡发,高膨胀,高膳食纤维,来增加饱腹感,进而减少食物的摄入,还能通过抑制肠道的蠕动,延缓食物的排出,来维持饱腹感,起到减肥的作用。

本发明涉及医药技术领域,具体涉及奇亚籽在制备抑制肠道蠕动的药品或食品中的应用。
背景技术
奇亚籽(Chia Seed)是唇形科鼠尾草属薄荷类植物芡欧鼠尾草(SalviaHispanica L)的种子, 2014年我国卫生计生委将其列为新食品原料,正式批准进入我国市场。近年研究表明,奇亚籽富含脂肪酸、蛋白质、矿物质、维生素、多酚和膳食纤维等多种活性物质,在抗氧化、降血脂、改善心血管等方面发挥着重要作用[1]。王志强[2]等人研究发现奇亚籽中含有丰富的矿物质元素,其中P、K、Ca、Mg元素含量较高,奇亚籽中含有丰富的Ca,含量是一般谷物类物质数倍,微量元素Cu也比一般谷物类高。奇亚籽中也富含脂肪酸,其中不饱和脂肪酸总量约占88.79%,其中α-亚麻酸含量最高为59.35%[3]。高浓度的n-3脂肪酸能有效减低冠心病、高血压、Ⅱ型糖尿病、自身免疫性疾病和癌症患病的风险[4]。奇亚籽的蛋白质含量高于其他传统作物如大麦、玉米、燕麦、小麦、大米等[5]。奇亚籽蛋白包括4种成分,其中球蛋白是主要成分(占52%),白蛋白、谷蛋白和醇溶蛋白比例相当,具有良好的持水性和起泡性[6]。奇亚籽中的可溶性膳食纤维可以溶解于水中,吸水膨胀后形成一种胶状物质,浸泡后食用可以起到代餐减肥、持续供能、润肠通便的作用[7],不会引起任何不良过敏反应以及抗营养和毒性作用,受到越来越多人们的关注,被广泛运用于各类代餐减肥产品。

具体实施方式
下面通过参考附图描述的实施例是示例性的,仅用于解释本发明,而不能理解为对本发明的限制。
一、原料及设备
原料:奇亚籽,产地美国,购于品牌Viva Naturals
实验设备:高速多功能粉碎机(运邦YB-1000A型)、BL-420N生物信号采集与分析系统 (四川泰盟)、生物张力传感器(四川泰盟,FT-102)、HW200S恒温平滑肌实验系统(四川泰盟),GrapdhPad Prism 6.0
二、实验过程
1、奇亚籽预处理
将奇亚籽经高速粉碎机粉碎,并过40目筛,装罐,密封备用。
2、台氏液的配制和横纹平滑肌实验系统的准备
(1)准确称取氯化钠8.00g、氯化钾0.20g、氯化钙0.20g、碳酸氢钠1.00、磷酸二氢钠0.05g、氯化镁0.10g、葡萄糖1.00g溶解于1000ml蒸馏水中,装于广口瓶中备用。
(2)在恒温平滑肌槽的中心管加入台氏液,外部容器中加装纯净水,启动仪器,使水浴槽温度稳定在38℃。调节供氧导管旋钮,使气泡一个接一个地通过中心管,为台氏液持续供氧。
3、离体小肠标本制作
用木锤猛击家兔头枕部,使其昏迷后,迅速剖开腹腔,以胃幽门与十二指肠交界处为起点,先将肠系膜沿肠缘剪去,再剪取20~30cm肠管。肠段取出后,置于38℃左右台氏液内轻轻漂洗,在肠管外壁用手轻轻挤压以除去肠管内容物。当肠腔内容物洗净后,用38℃左右的乐氏液中浸浴,当肠管出现明显活动时,将其剪成约3cm长的肠段。
4、奇亚籽对肠管生理特性的影响
取出一段长约3~4cm的肠段,用线结扎其两端,迅速将小肠一端的结扎线固定于通气管的挂钩上,另一端固定在张力换能器上。适当调节换能器的高度,使肠段勿牵拉过紧或过松,用生物信号采集处理系统记录肠管收缩曲线,观察正常离体肠管自动节律性收缩运动。滴加美国奇亚籽溶液(1:50)2ml,观察肠管节律性运动变化,更换溶液,用38℃台式液反复冲洗肠管,待肠管恢复稳定后,滴加奇亚籽溶液(1:50)2ml,重复上述操作。
5、消化道平滑肌作用机制
选取活动性较好的两段肠管,分别悬挂在张力换能器上,并使其完全浸没于38℃持续通气的台式液中,调节挂线的张弛程度,使两段肠管均能自主运动,直至肠管收缩曲线稳定。分别滴加乙酰胆碱(1:500)20-40ul,普萘洛尔(1:500)20-40ul,观察肠管运动变化。再次滴加奇亚籽溶液(1:50)2ml,观察并记录肠管运动变化,并做出分析。
6、在体肠管推进率实验
取14只禁食12小时,体重(20±2g)雄性昆明白鼠,随机分为空白组和奇亚籽组,用苦味酸标记。空白组7只,活性炭生理盐水混悬液0.1g/ml,每只0.4ml灌胃;奇亚籽组,奇亚籽生理盐水混悬液0.0g/ml(含活性炭0.02g/ml),每只0.4ml灌胃。给药25min后脱颈椎处死,打开腹腔分离肠系膜,剪取上端至幽门,下端至回盲部的肠管,置于托盘上。轻轻将小肠拉成直线,测量肠管长度作为“小肠总长度”。从幽门至炭末前沿的距离作为“炭末在肠内推进距离”。用公式计算炭末推进百分率。
炭未推进率=炭末在肠内推进距离(cm)/小肠全长(cm)×100%
三、实验结果
1、奇亚籽对家兔离体肠管生理特性作用
由图1、图2-1、图2-2可知,与空白组相比,分别加入奇亚籽后肠管运动频率基本没有明显变化。加入奇亚籽后,肠管运动的峰峰值明显降低(P<0.01)。说明,在相同条件下,奇亚籽能够明显抑制肠管的运动幅度。
2、奇亚籽对家兔离体肠管抑制作用通路
由图3、图4-1、图4-2可知,与空白组相比,当加入乙酰胆碱后,和再加入奇亚籽混合液后,肠管的运动频率没有明显变化,与文献相符[8];当加入乙酰胆碱后,肠管的峰峰值较空白组有明显的增加(P<0.01),再次滴加奇亚籽之后肠管的峰峰值降低与空白组相比有显著差异(P<0.001),两者之间差异明显(P<0.01)。乙酰胆碱为M受体激动剂,可以使肠道平滑肌兴奋,促进肠道平滑肌的收缩[9]。在乙酰胆碱存在的情况下,奇亚籽仍能起到抑制作用,由此推测,奇亚籽不通过M受体通路发挥作用。
由图5、图6-1、图6-2所知,离体肠管加入普萘洛尔后,肠管的运动频率基本不发生变化,在此基础上加入奇亚籽,也没有改变(图5);离体肠管在加入普萘洛尔之后,肠管的峰峰值发生了显著增加(P<0.01),待运动稳定后,再加奇亚籽溶液,肠管运动没有发生明显改变 (图6-1,图6-2)。普萘洛尔能通过阻断肠管上的β受体,兴奋肠管平滑肌,使肠管运动幅度加强[10]。在普萘洛尔阻断β受体的情况下,奇亚籽不能再起作用,由此推测奇亚籽是通过兴奋β受体通路,从而抑制肠管运动。
3、在体肠管推进率实验
由图7可知,奇亚籽组的在体肠道推进率明显低于空白组(P<0.001),再次证明了奇亚籽能明显抑制肠道的运动。
四、关于代餐
现在市场上各类代餐产品中加入奇亚籽,均是通过奇亚籽中含有大量的膳食纤维,其中可溶性膳食纤维占1/3以上,可溶性膳食纤维溶解于水中,吸水膨胀形成一种胶状物质,从而达到高泡发,高膨胀,进而维持长时间的饱腹感,减少热量摄入,达到减肥的目的。没有对奇亚籽其他的的作用机制进行探讨和实验。
本发明实施例的上述实施可以探究奇亚籽用于代餐减肥产品中的新的作用机制与功能,明确奇亚籽发挥减肥效果的作用过程,为奇亚籽在代餐领域深加工提供指引,也为奇亚籽在代餐减肥中的作用机制提供新的导向

研究发现,奇亚籽除了目前已知的高泡发,高膨胀,高膳食纤维,来增加饱腹感,进而减少食物的摄入以外,还能通过抑制肠道的蠕动,延缓食物的消化时间,延长饱腹感,属于新功能的发现,为以后的药物研发和开发应用提供了新的素材。
