Abstract
本发明涉及CRISPR‑Cas9高效敲入嵌合抗原受体基因到T细胞特定基因组位点的方法及其应用。本发明具体涉及一种将CAR基因精确敲入到T细胞目的基因组DNA的方法,所述方法包括a)采用基因编辑物质剪切目的基因组DNA,其中所述目的基因组DNA位于基因启动子区后,起始密码子和其所处的外显子3端之间;b)将CAR基因克隆至修复模版载体后导入T细胞,细胞以同源重组方式修复被剪切的目的基因组DNA,从而将CAR基因嵌入,其中所述修复模版载体为腺相关病毒载体。该方法可应用于各种CAR‑T细胞制备。
Description
技术领域
本发明涉及细胞治疗领域,特别涉及CRISPR-Cas9高效敲入嵌合抗原受体基因到T细胞特定基因组位点的方法及其应用。
背景技术
CAR-T治疗
传统肿瘤治疗药物包括化疗药、靶向药,虽然它们在一定程度上提高了癌症患者的生存期,但同时也带来了严重的副作用,极大地降低了病人的生存质量。更不幸的是,大部分患者在接受这些传统治疗后仍旧会复发,而一旦复发,无药可救是大概率事件。
近年来,随着免疫治疗的发展,免疫检验点抑制剂药物(如:CTLA-4、 PD-1/PD-L1抗体)的出现,彻底改变了肿瘤治疗的方式。但这类药物在不同癌症病人中的有效率只有20%-40%,仍然有大部分的癌症患者等待新的有效治疗方式的出现。
嵌合抗原受体T细胞(CAR-T细胞)治疗技术是通过体外转导一个识别癌细胞的人工基因于人体T细胞上,使人T细胞具有特异杀伤肿瘤细胞的能力。CAR-T细胞治疗属于过继性免疫疗法(adoptive cell transfer,ACT),它利用人体免疫细胞来对抗肿瘤,被称为“活体药物”。这种ACT疗法在近年已应用于临床实验当中,特别是在儿童白血病方面的有效探索,为研究者以及医生开辟了一条治疗肿瘤的新路径。2017年末,两款CAR-T药物获得美国FDA的许可上市,然而,这种ACT疗法仍旧处于其早期阶段。它与传统药物相比具有如下创新点:
首先,精确靶向肿瘤,且副作用可逆。CAR-T细胞治疗相比化疗药、靶向药具有更特异的肿瘤细胞杀伤能力。在输入病人体内后,虽然短时间内会产生细胞因子效应,但该副反应可控,病人在获得完全缓解的情况下基本上能和健康人一样生活。
其次,持续缓解时间长。CAR-T细胞部分属于记忆T细胞,在输入病人体内后能长期存在于病人身体中,时刻监视身体中是否出现肿瘤细胞,一旦有新的肿瘤细胞出现就会立刻将其杀灭。
CAR转导方式
本领域通常涉及以下CAR转导方式:
1)慢病毒或者逆转录病毒
慢病毒或逆转录病毒可用于包装3kb以下的目的DNA序列,其优势在于:a.较高的感染效率,通常转染后阳性率可以达到50%左右;b.病毒分泌到培养基中较多,浓缩纯化相对容易;c.T淋巴细胞感染病毒后能保持一个良好的细胞状态,存活和扩增不易受到影响,有利于后续回输给病人。其不足之处在于:a.随机插入细胞基因组中,容易导致产生的CAR-T 细胞耗竭及潜在的癌变风险;b.感染T细胞后,残留的病毒有强免疫原性。
2)转座子
转座子优势在于简单的生产工艺流程,无病毒残留的风险。但由于编码转座子的载体为质粒DNA,其在电穿孔入T细胞后有细胞毒性作用,不利于产生的CAR-T细胞扩增。同样,转座子介导也是随机性地将CAR 基因插入到T细胞基因组中。
3)mRNA
mRNA编码的CAR基因转导T细胞依赖于电穿孔,其优势在于只短暂地表达目的蛋白于T细胞中(大约表达7天),对于前期不确定是否存在严重副作用的CAR-T细胞,这种方法能够相对安全的进行人体实验。但其缺点也很明显,就是不能持久的表达,作用时间相对有限。
CRISPR-Cas9技术
CRISPR(Clusters of Regularly Interspaced Short Palindromic Repeats)技术在2012年由麻省理工学院和加州大学伯克利分校的科学家共同发现,是一种由RNA序列介导的双链DNA内切酶工具。其由两个部分构成,其中之一是100bp左右的sgRNA,用于靶向识别目的双链DNA,另一部分是1369个氨基酸的Cas9蛋白,其可结合sgRNA,并且具有DNA酶活性,人为设计的sgRNA和Cas9蛋白形成复合体后可对目的DNA进行特异性切割,当切割造成错配修复,则导致基因移码而起到敲除目的,当添加一段修复DNA序列,则会进行有目的的编辑。自CRISPR-Cas9技术问世至今,已经在很多个领域得到应用。
基因敲入技术
虽然基因敲除技术取得了长足发展,但是对于大片段基因敲入效率一直不高。基因敲入和基因敲除区别在于是否添加一个修复模版。其过程是:在核酸酶对特定DNA进行剪切后,修复模版根据同源重组原理对“损伤”DNA进行修复,在修复后将特定DNA片段引入特定位点。目前使用的修复模版主要有质粒DNA,双链DNA和单链DNA,在这其中,有研究显示单链DNA效率相对最高。在原代T细胞中,大片段的基因敲入更是难上加难,研究显示,以上不同形式的修复模版做基因敲入,其效率不超过5%,并且质粒DNA和双链DNA都具有不同程度的细胞毒性,并不适合在T细胞中做基因敲入的修复模版。所以原代T细胞的基因敲入仍需要新的有效方法出现。
发明内容
为了解决以上技术问题,本发明首先提供:
本发明的第一个方面,提供一种将CAR基因精确敲入到T细胞目的基因组DNA的方法,所述方法包括一:
a)采用基因编辑物质剪切目的基因组DNA,其中所述目的基因组DNA位于基因启动子区后,起始密码子和其所处的外显子3端之间;
b)将CAR基因克隆至修复模版载体后导入T细胞,细胞以同源重组方式修复被剪切的目的基因组DNA,从而将CAR基因嵌入,其中所述修复模版载体为腺相关病毒载体。
或二:
a)将CAR基因克隆至修复模版载体后导入T细胞;
b)采用基因编辑物质剪切目的基因组DNA,其中所述目的基因组 DNA位于基因启动子区后,起始密码子和其所处的外显子3端之间;
c)细胞以同源重组方式修复被剪切的目的基因组DNA,从而将CAR 基因嵌入,其中所述修复模版载体为腺相关病毒载体。
本发明的第二个方面,提供根据第一个方面所述的方法,其中所述基因编辑物质包括CRISPR-Cas9、ZFN、TALEN和megaTAL。
本发明的第三个方面,提供根据第一个方面所述的方法,其中CAR 基因敲入的目的基因组DNA位点包括TRAC、TRBC、HIF-1、PD-1启动子调控区域。
本发明的第四个方面,提供根据第二个方面所述的方法,其中所述基因编辑物质为CRISPR-Cas9,其中所述Cas9类型包括:SpCas9、SaCas9、 SpCas9-HF、eSpCas9、xCas9和cpf1,优选SpCas9,其氨基酸序列如SEQ ID NO:1所示。
本发明的第五个方面,提供根据第四个方面所述的方法,其中采用(i) SpCas9蛋白和sgRNA、(ii)SpCas9表达质粒和sgRNA表达质粒、和/或(iii) SpCas9mRNA和修饰的sgRNA递送方式剪切目的基因组DNA,其中 sgRNA修饰方式为:5’和3’端1-10个碱基进行(i)2’-O-甲基化修饰;(ii) 2’-O-甲基化和3’硫代磷酸化修饰;或(iii)能够稳定sgRNA的其它修饰。
本发明的第六个方面,提供根据第一个方面所述的方法,其中所述腺相关病毒载体包含左右同源臂和敲入基因。
本发明的第七个方面,提供根据第六个方面所述的方法,其中敲入的基因从5’端到3’端包括:剪切肽基因、CAR基因、Poly A基因,其中剪切肽优选是T2A、2A和IRES,其DNA序列分别如SEQ ID NO:2-4所示; Poly A优选包括BGHpA,其DNA序列如SEQ ID NO:5所示;CAR基因优选包含信号肽DNA、抗体scFv DNA、铰链区DNA、跨膜区DNA、一个或两个共刺激信号区DNA和信号传导区DNA。

具体实施方式
定义
“免疫检验点抑制剂”涉及目前肿瘤免疫治疗药物最主要方面,通过抑制肿瘤细胞的免疫逃逸,调动自身免疫系统功能消除肿瘤。
“CTLA-4”,即细胞毒性T淋巴细胞相关蛋白4(Cytotoxic T-lymphocyteassociated protein 4),是最早获批的免疫检验点抑制剂靶点。
“PD-1/PD-L1”,:即程序性细胞死亡蛋白1(programmed cell death 1)/PD-1蛋白的配体,是最著名的两个免疫检验点抑制剂靶点。
“过继性免疫疗法”是指将供体的淋巴细胞转移给受体,增强其细胞免疫功能。过继性细胞免疫可分为特异性和非特异性两类,前者是用已知抗原致敏的淋巴细胞注入受体后使其获得对该抗原的细胞免疫能力;后者是用未经特殊抗原致敏的正常人淋巴细胞注入受体后使其获得对多种抗原的细胞免疫能力。
“记忆T细胞”是一种人体免疫细胞。人体免疫的第三道防线中分为两部分,一是体液免疫、二是细胞免疫。T细胞分裂和分化后,会分别形成记忆T细胞和效应T细胞,而记忆T细胞则会在下一次抗原入侵时再次将记忆中的杀毒方法再次调动出来,再次破坏靶细胞即受病菌或病毒感染的细胞,释放出靶细胞中的抗原。
“转座子”是一类在很多后生动物中(包括线虫、昆虫和人)发现的可移动的遗传因子。一段基因可以从原位上单独复制或断裂下来,环化后插入另一位点,并对其后的基因起调控作用,此过程称转座。这段序列称为跳跃基因或转座子,可分为插入序列(Is因子),转座(Tn),转座噬菌体。
“CRISPR”是一种细菌免疫系统,今年来被科学家改造为最热门的基因编辑工具。
“基因敲入”是指目的基因位置引进特定的突变或外源基因。
在一个方面,本发明涉及一种将CAR基因精确敲入到T细胞目的基因组DNA的方法,所述方法包括:
a)采用基因编辑物质剪切目的基因组DNA,其中所述目的基因组 DNA位于基因启动子区后,起始密码子和其所处的外显子3端之间;
b)将CAR基因克隆至修复模版载体后导入T细胞,细胞以同源重组方式修复被剪切的目的基因组DNA,从而将CAR基因嵌入,其中所述修复模版载体为腺相关病毒载体。
在一个方面,本发明涉及一种将CAR基因精确敲入到T细胞目的基因组DNA的方法,所述方法包括:
a)将CAR基因克隆至修复模版载体后导入T细胞;
b)采用基因编辑物质剪切目的基因组DNA,其中所述目的基因组 DNA位于基因启动子区后,起始密码子和其所处的外显子3端之间;
c)细胞以同源重组方式修复被剪切的目的基因组DNA,从而将CAR 基因嵌入,其中所述修复模版载体为腺相关病毒载体。
在一些实施方案中,其中CAR敲入的基因组DNA位点包括TRAC、 TRBC、HIF-1、PD-1启动子调控区域。
在一些实施方案中,采用ZFN、TALEN、CRISPR-Cas9或megaTAL 等基因编辑的方法剪切目的基因组基因。将上述方法递送至T细胞的方式包括质粒载体、慢病毒载体、逆转录病毒载体、腺相关病毒载体、mRNA、 RNA蛋白复合体等,其中质粒载体、mRNA、RNA蛋白复合体等方式需采用电穿孔、脂质体或者其他转染物质递送。在一些实施方案中,所述基因编辑方法为CRISPR-Cas9,其中Cas9类型包括:SpCas9、SaCas9、 SpCas9-HF、eSpCas9、xCas9和cpf1,优选SpCas9。在一些实施方案中,采用Cas9mRNA和修饰的sgRNA剪切目的基因组DNA。在一些实施方案中,采用(i)SpCas9蛋白和sgRNA、(ii)SpCas9表达质粒和sgRNA表达质粒、和/或(iii)SpCas9mRNA和修饰的sgRNA递送方式剪切目的基因组 DNA。在一些实施方案中,修饰的sgRNA头尾1至10个(例如,1、2、3、 4、5、6、7、8、9或10个)碱基用甲基化和/或硫代磷酸化修饰。在一些实施方案中,所述sgRNA头尾三个碱基分别用甲基化修饰。
在一些实施方案中,在步骤b)中,将CAR基因克隆至修复模版载体中,CAR融合基因两端DNA序列与敲入的目的区域同源,修复模版为腺相关病毒载体,腺相关病毒需在目的DNA剪切后2到5小时内完成。在一些实施方案中,所述腺相关病毒载体包含左右同源臂和敲入的基因。在一些实施方案中,所述腺相关病毒载体的血清型包括1、2、3、4、5、6、 7、8、9、DJ。
在一些实施方案中,所述方法还包括从人血中分离PBMC并激活T细胞的步骤。可选择分离纯化T淋巴细胞或者不分离。T细胞激活可以采用如下方法:单独的抗CD3抗体包被、抗CD3抗体/抗CD28抗体包被、直接添加单独的抗CD3抗体、直接添加抗CD3抗体/抗CD28抗体、抗 CD3抗体/抗CD28抗体磁珠激活。激活时间为2到3天,若采用抗CD3 抗体/抗CD28抗体磁珠激活,需去除磁珠。
在一些实施方案中,所述方法还包括T细胞培养扩增。培养采用1640 培养基+10%血清或者其它专用于T细胞培养的无血清培养基,如 X-VIVO-15。放培养瓶、培养皿或者培养袋中培养,每隔1天换一次液,保证T细胞密度在1×106个/ml左右。
在一些实施方案中,敲入的基因从5’端到3’端包括:剪切肽基因、CAR 基因、PolyA,其中剪切肽基因包括P2A、T2A和IRES序列,Poly A包括BGHpA。在一些实施方案中,CAR基因优选包含信号肽DNA、抗体 scFv DNA、铰链区DNA、跨膜区DNA、一个或两个共刺激信号区DNA 和信号传导区DNA。
在一些实施方案中,CAR的跨膜区选自以下蛋白:CD3、CD4、CD5、 CD8、CD9、CD16、CD22、CD27、CD28、CD33、CD37、CD45、CD64、 CD80、CD86、CD134、CD137、CD152、CD154和PD1。在一些实施方案中,CAR的跨膜结构选自CD8,其氨基酸序列如SEQ ID NO:6所示。
在一些实施方案中,CAR的铰链结构选自以下蛋白:IgG1、IgG4、IgD 和CD8所示。在一些实施方案中,CAR的铰链结构选自CD8,其氨基酸序列如SEQ ID NO:7所示。

本发明的有益技术效果在于:
1.更精确的转导方式,可以任意精确插入指定的基因组位置。
本发明利用了基因编辑和同源重组修复双重介导,可以准确将CAR 基因插入到指定位置。特别地,不依赖外源启动子表达CAR基因,即插入的CAR基因依赖T细胞天然启动子调控。
2.更高效、更稳定的转导方式
本发明所采用的CAR基因敲入技术效率超过50%,比现有的慢病毒和逆转录病毒效率都高。另外慢病毒和逆转录病毒所用的启动子容易受到 T细胞甲基化的影响,长时间会失活而使CAR-T细胞失效,而我们所采用的CAR基因敲入技术可以将CAR基因插入到天然启动子下游并受调控,而很多天然启动子并不会被甲基化。
3.更安全
慢病毒、逆转录病毒、转座子技术介导的CAR基因导入是一种随机插入的方式,可能引起T细胞的癌变而严重影响移植安全。
4.更多变的设计
精确敲入技术可以使CAR基因插入的方式更加多变,如:插入TRAC 和TRBC基因组位点可以在插入CAR基因的同时破环TCR基因而做成通用型CAR-T细胞、插入PD-1基因组位点受PD-1基因启动子控制可以起到更好的疗效和特异性、插入HIF-1基因组位点受其启动子调控可以做成低氧诱导的CAR-T细胞而更特异的应用于实体瘤中。
5.更温和的转导方式
之前的基因敲入方法采用质粒为修复模版,在电穿孔后对细胞的毒性较大,不利于细胞的扩增繁殖,而本发明在敲入过程中采用了腺相关病毒,其对细胞更加温和。
实施例1:PBMC提取
招募健康志愿者(不便透露信息),无感冒发烧症状。由专业医务人员在人肘正中静脉处取血100ml到BD抗凝血管。采完血后,血液与等量的 PBS缓冲液(含2%的胎牛血清)混合。取PBMC分离管Sepmate-50,小心加入15ml的Ficoll缓冲液,再加入血液PBS的混合液,每管小心加入 30ml左右。1200g离心10分钟后迅速将上清倒入新50ml管中,200g离心8分钟,弃上清,加入10ml PBS缓冲液重悬沉淀,再弃上清后加10ml PBS缓冲液重悬,离心弃上清后10ml上清PBS缓冲液重悬所有沉淀。对重悬细胞计数,取10μl混悬液加入10μl 0.1%的台盼蓝混匀,上机计细胞数和存活率,结果显示于表1中。

实施例2:T细胞激活
取上述PBMC细胞1×107个,用6ml VIVO-15培养基重悬,用抗-CD3/ 抗-CD28抗体磁珠激活。取抗-CD3/抗-CD28抗体磁珠(Life Technology) 6×106个,用PBS缓冲液(含2mM的EDTA和1%的胎牛血清)重悬,加入磁极中静置2分钟后小心弃去上清。重复4次上述过程。取洗后磁珠,磁珠加入PBMC细胞,混匀,放入37度培养3天。3天后取出磁珠,首先将T细胞用移液枪重悬多次。将细胞悬液置于磁极中,静置两分钟后,弃去管壁上的磁珠。重新计数,计数结果如表2所示。

实施例3:腺相关病毒载体构建
本发明中CAR基因敲入TRAC基因位点采用了CRISPR-Cas9技术结合腺相关病毒载体转导的方法。首先用CRISPR-Cas9对TRAC基因位点产生一个切口,进而利用腺相关病毒递送的CAR基因模版对缺口进行同源重组修复,最终将CAR基因敲入目的位点。示意图如图2。
(1)该腺相关病毒载体包含了两个部分:
a.左右同源臂。左右同源臂用于识别目的DNA,并进行重组交换,为了使敲入基因能正确表达,本发明将敲入基因“登陆”到TRAC基因外显子1的5’端区域。根据碱基序列的长短,分别设计了以下几对同源臂:
左右分别为1170bp的同源臂,左臂序列如SEQ ID NO:12,右臂序列如SEQ ID NO:13;
左右分别为600bp的同源臂,左臂序列如SEQ ID NO:14,右臂序列如SEQ ID NO:15;
左右分别为300bp的同源臂,左臂序列如SEQ ID NO:16,右臂序列如SEQ ID NO:17;
b.敲入的基因。敲入基因依次融合了P2A DNA、抗-CD19scFv DNA、 CD8铰链区DNA、CD8跨膜区DNA、CD137共刺激信号区DNA、CD3ζ信号传导区DNA和BGHpA序列,如图3所示。其DNA序列如SEQ ID NO: 18。
其中P2A序列如SEQ ID NO:3;CD8铰链区DNA序列如SEQ ID NO: 7;CD8跨膜区DNA序列如SEQ ID NO:6;CD137共刺激信号区DNA序列如SEQ ID NO:8;CD3ζ信号传导区DNA序列如SEQ ID NO:10;BGHpA DNA序列如SEQ ID NO:5。
在合成重组臂和敲入基因DNA后,将其克隆至腺相关病毒载体中,腺相关病毒载体采用pAAV-MCS质粒,使用时将ITR中间的碱基全部用 Not1酶切删除,重组臂和敲入基因连接克隆至pAAV载体两个ITR序列中间。
实施例4:腺相关病毒包装、纯化和滴度测定
(1)包装:293T细胞(购自ATCC)在转染前一天在225cm2培养皿中长满,按1:3传代,每个培养皿培养基为40ml,每个腺相关病毒包装5 个225cm2培养皿。转染按照Lipo3000的厂家步骤做,先配制转染体系(针对1个225cm2培养皿),如表3,

混匀体系1和2,静置5分钟后将二者混匀,再静置10分钟。小心加入293T细胞中。6小时后换新鲜培养基。72小时后分别收取培养基和细胞。
(2)纯化:首先用高速离心机将培养基用50000g离心2小时,离心后去除培养基上清,加入1ml PBS重悬所有病毒沉淀,放置于4度冰箱备用。然后将收取的细胞用10ml PBS重悬,反复在液氮和37度水浴锅冻融 4次,加入上清病毒后,2000rpm 4度离心30分钟去除细胞碎片。取上清,加入全能核酸酶后37度处理30分钟。按一定的比例配制不同浓度的碘克沙醇,分别为:60%、40%、25%、15%。取一13.2ml PP超速离心管,逐层加入60%层2ml、40%层2ml、25%层2ml、15%层2ml,最后小心加入样品3ml。250000g超速离心3小时,离心完毕后,用注射器在40%和60%交界层吸取2ml,然后加入PBS 18ml稀释并用0.45um滤器过滤,去除污染。最后将过滤的病毒用100KDa超滤管离心,弃掉离下的液体,重新加入PBS进行离心,最后得到约200μl的病毒量。
(3)滴度测定:0.5μl病毒液检测(稀释40倍),第一步先用DNA酶消化,体系如表4。

混匀后放置于37度水浴锅反应10分钟,再在75度水浴锅放置10分钟终止反应。第二步用蛋白酶K消化腺相关病毒外壳,配制反应体系如表 5(进一步稀释了5倍),

最后根据Ct值结果计算腺相关病毒滴度,先用标准品制作散点图(如图4所示)。
根据散点图公式得出pAAV-CAR-TRAC腺相关病毒滴度为10的N次方,N=-0.1548×7.87+9.2876=8.069,即117307019.4vg/μl。
加上DNase消化和蛋白酶K反应时稀释的200倍,最终的滴度应该是:
117307019.4×1000×200=2.34614×1013vg/μl。
实施例5:CRISPR-Cas9与腺相关病毒介导靶向CD19CAR基因敲入TRAC基因位点
T细胞用抗-CD3/抗-CD28抗体磁珠激活三天后,将磁珠去除,并对细胞进行计数,得到细胞密度为:1.9×106个/ml,存活率96.5%,共约5ml 细胞体积,取其中2.73ml细胞悬液置于离心管中进行离心,转速为200g, 5分钟。离心后完全去除培养基,并用Lonza电穿孔缓冲液进行重悬,然后加入SpCas9mRNA 5μg和设计合成的sgRNA 1μg至T细胞悬液,混匀后加入电转杯中用Lonza 4D程序E0-115程序电击,后放置于37度培养箱15分钟后加入5ml预热的细胞培养基中,其中sgRNA序列如如SEQ ID NO:19,其中该sgRNA 5’和3’端各三个碱基同时进行2’-O-甲基化和3’硫代磷酸化修饰,该修饰sgRNA由Sythego公司合成。
电穿孔后2小时,将5ml电击的T细胞加入上述腺相关病毒5×1012VG,使MOI值为106左右。16小时后将感染腺相关病毒的两种细胞200g离心 5分钟,完全去除培养基和病毒后加入5ml新的培养基重悬,37度继续培养。在培养过程中,保持细胞密度在1×106个/ml,培养基中重组IL-2细胞因子浓度为10ng/ml。
实施例6:流式检测靶向CD19CAR基因敲入TRAC基因位点效率
腺相关病毒感染后继续培养4天,取2×105个相应细胞做流式细胞分析,具体步骤为:取相应细胞加入1.5ml离心管中,用PBS+1%胎牛血清缓冲液洗2遍,完全弃去上清,加入100μl缓冲液重悬细胞后加入FITC- 抗-小鼠-Fab流式抗体1μl,混匀后室温避光静置15分钟,加入缓冲液洗2 遍后上机检测,结果如图5所示,可以看出CAR基因敲入效率超过60%,比普通的慢病毒或者逆转录病毒转导效率都高。
实施例7:制备的靶向CD19CAR-T细胞体外对肿瘤细胞的杀伤实验
首先将T细胞、CAR-T细胞与Raji细胞按照5:1混合,其中T细胞数量为2.5×105个/孔,Raji细胞数为5×104个/孔,每个样品做两个副孔,体积为180μl/孔,在培养基1640+1%胎牛血清中37℃孵育4小时。在检测信号前,加入10×裂解缓冲液20μl至Raji细胞最大释放孔,200g离心5 分钟,吸取50μl培养基加入96孔板中,再加入50μl/孔的LDH底物,室温反应20分钟后,酶标仪检测492nm波长激发光信号,结果如表9。

根据计算,制备的靶向CD19CAR-T细胞的杀伤能力如图6所示。可以看出制备的靶向CD19CAR-T细胞对比T细胞具有明显的杀伤优势,进一步证明靶向CD19CAR敲入成功。
Claims (15)
1.一种将CAR基因精确敲入到T细胞目的基因组DNA的方法,所述方法包括一:
a)采用基因编辑物质剪切目的基因组DNA,其中所述目的基因组DNA位于基因启动子区后,起始密码子和其所处的外显子3端之间;
b)将CAR基因克隆至修复模版载体后导入T细胞,细胞以同源重组方式修复被剪切的目的基因组DNA,从而将CAR基因嵌入,其中所述修复模版载体为腺相关病毒载体,
或二:
a)将CAR基因克隆至修复模版载体后导入T细胞;
b)采用基因编辑物质剪切目的基因组DNA,其中所述目的基因组DNA位于基因启动子区后,起始密码子和其所处的外显子3端之间;
c)细胞以同源重组方式修复被剪切的目的基因组DNA,从而将CAR基因嵌入,其中所述修复模版载体为腺相关病毒载体,
其中所述CAR基因敲入目的基因组DNA位点为TRAC启动子调控区域,并且其中:
1)腺相关病毒载体包含的左右同源臂分别为:i)1170bp的同源臂,其中左臂包含序列SEQ ID NO:12,右臂包含序列SEQ ID NO:13;ii)600bp的同源臂,其中左臂包含序列SEQ IDNO:14,右臂包含序列SEQ ID NO:15;或iii)300bp的同源臂,其中左臂包含序列SEQ ID NO:16,右臂包含序列SEQ ID NO:17;所述CAR基因包括靶向CD19 CAR,其序列如SEQ ID NO:18所示;
2)所述基因敲入到TRAC基因外显子1的5’端区域;和
3)采用Cas9 mRNA和修饰的sgRNA剪切TRAC基因,其中所述修饰的sgRNA如序列SEQ IDNO:19所示,并且所述sgRNA头尾三个碱基用甲基化和/或硫代磷酸化修饰。
2.根据权利要求1所述的方法,其中敲入的基因从5’端到3’端包括:剪切肽基因、CAR基因、Poly A基因。
3.根据权利要求2所述的方法,其中剪切肽是T2A、P2A和IRES,其DNA序列分别如SEQ IDNO:2-4所示。
4.根据权利要求2所述的方法,其中Poly A包括BGHpA,其DNA序列如SEQ ID NO:5所示。
5.根据权利要求2所述的方法,其中CAR基因包含信号肽DNA、抗体scFv DNA、铰链区DNA、跨膜区DNA、一个或两个共刺激信号区DNA和信号传导区DNA。
6.根据权利要求5所述的方法,其中CAR的跨膜结构选自CD8,其氨基酸序列如SEQ IDNO:6所示。
7.根据权利要求5所述的方法,其中CAR的铰链结构选自CD8,其氨基酸序列如SEQ IDNO:7所示。
8.根据权利要求5所述的方法,其中CAR的共刺激信号选自CD137,其氨基酸序列如SEQID NO:8所示。
9.根据权利要求5所述的方法,其中CAR的信号传导区选自CD3ζ,其氨基酸序列如SEQID NO:10所示。
10.根据权利要求5所述的方法,其中CAR信号肽选自CD8,其氨基酸序列如SEQ ID NO:11所示。
11.根据权利要求1所述的方法,其中所述腺相关病毒载体的血清型包括1、2、3、4、5、6、7、8、9、DJ。
12.根据权利要求1-11中任一项所述的方法制备的CAR-T细胞在制备药物中的用途,其中所述药物用于治疗白血病、实体肿瘤或免疫疾病。
13.根据权利要求12所述的用途,所述免疫疾病为艾滋病。
14.根据权利要求1-11中任一项所述的方法制备的通用型CAR-T细胞在制备药物中的用途,其中CAR基因敲入TRAC基因组位点,并且其中所述药物用于治疗白血病、实体肿瘤或免疫疾病。
15.根据权利要求14所述的用途,所述免疫疾病为艾滋病。
