Description

Traditional Chinese medicine composition with moisturizing and hydrating effects and preparation method and application thereof
Technical Field
The invention belongs to the field of cosmetics, and relates to a traditional Chinese medicine composition with moisturizing and hydrating effects, and a preparation method and application thereof.
Background
The stratum corneum of the epidermis of the skin is an important barrier for maintaining the health and stability of the skin, the water content of the healthy stratum corneum of the skin is kept above 10%, and the stratum corneum is directly dry and rough when the stratum corneum lacks water, and various skin problems such as thickening, desquamation, chapping, inflammation, pruritus, fine lines and the like are caused along with the skin. However, the dry and cold climate in autumn and winter is one of the important factors causing skin dryness, and besides, the influence of the working environment on the skin is not negligible, for example, modern urban office workers generally work in air-conditioned rooms for a long time and are easy to suffer from 'air-conditioning dessicator'. Therefore, dry and rough skin becomes a big skin problem which generally troubles people's life. According to statistics of an England latest product database, in Asian skin care products in recent years, cosmetics declared as moisturizing and moisturizing are ranked first, and it is seen that the first skin requirement of consumers is still moisturizing and moisturizing.
Moisturizers in cosmetics are classified into synthetic moisturizers and natural moisturizers according to their source. Polyols are the most widely used traditional synthetic moisturizers in cosmetics, such as glycerin, propylene glycol, sorbitol and the like, and these moisturizers can absorb moisture from the surrounding environment to keep the skin with a certain amount of moisture, but have good moisturizing effect only under the condition of high relative humidity, and can absorb moisture from the skin to make the skin drier in cold and dry environments with low relative humidity. In addition, the synthetic moisturizer can cause adverse reactions such as allergy, irritation and the like to the skin to a certain extent. The natural humectant is a raw material which is extracted from natural materials, has a good moisturizing effect and contains other substances with nutritive value to the skin, is popular with consumers, and has the characteristics of extraction from natural materials, easier skin absorption, reduction of skin irritation and the like, and is more popular with the environment-friendly concept of people's worried green and nature returning than a chemically synthesized humectant.
CN108938929A discloses a traditional Chinese medicine composition with moisturizing and anti-aging effects, a preparation method and an application thereof, wherein the traditional Chinese medicine composition is prepared from the following raw materials in percentage by mass: 15-40% of rose, 10-30% of dendrobium officinale, 10-25% of echinacea purpurea, 10-25% of aloe, 5-15% of tremella and 5-15% of seaweed.
CN103239383A discloses a traditional Chinese medicine composition for whitening and moisturizing, which is composed of the following raw materials in parts by weight: 5-10 parts of Chinese herbaceous peony, 8-15 parts of lotus, 3-10 parts of ganoderma sinensis, 4-15 parts of almond, 2-7 parts of motherwort, 3-12 parts of centella, 5-16 parts of silk, 7-18 parts of green bamboo, 2-6 parts of olive leaf, 5-15 parts of seaweed, 2-5 parts of oat, 3-9 parts of cactus, 1-5 parts of pearl, 3-12 parts of angelica, 7-15 parts of pawpaw, 4-16 parts of sophora flavescens and 5-10 parts of sanguisorba.
Therefore, the development of a traditional Chinese medicine composition which can deeply replenish water, can keep moisture for a long time and has strong oxidation resistance is the focus of research in the field of cosmetics at present.

Test example 1
Haemolysis test of erythrocytes
The erythrocyte hemolysis test is one of the alternatives of the rabbit eye irritation test (Draize test), and the basic principle is to evaluate the damage of chemicals to eye tissue cells by measuring the dissolution amount and denaturation degree of hemoglobin. The RBC test is used internationally for evaluating eye irritation studies on chemicals such as cosmetics and raw materials.
the Chinese medicinal compositions prepared in examples 1 to 7 and the Chinese medicinal extracts prepared in comparative examples 1 to 9 were subjected to a red blood cell hemolysis test in accordance with the RBC test method and grade standard of the European Alternatives Committee for the validation of methods (ECVAM). ECVAM. ECVAM DB-ALM INVITOX Protoco, Red blood cells test system INVITOX No.37 Ispra. Italy ECVAM,1994 ], ECVAM cosmetic products RBC experimental irritation grading criteria are shown in Table 1:
TABLE 1
the results of experiments on hemolysis of erythrocytes of the herbal compositions prepared in examples 1 to 7 and the herbal extracts prepared in comparative examples 1 to 9 are shown in Table 2, in which HD50 is the sample concentration at which 50% of erythrocytes are hemolyzed, DI is the protein denaturation index, and L/D is the ratio of HD50 to DI.
TABLE 2
According to the test results, the irritation of the traditional Chinese medicine composition prepared in the embodiments 1 to 7 is classified as no irritation, which indicates that the traditional Chinese medicine composition has the characteristics of mildness and no irritation; wherein, the sample concentration HD50 of the traditional Chinese medicine composition prepared in the examples 1-7 when 50% of erythrocytes are hemolyzed is more than 70096mg/L and is far greater than HD50 of the traditional Chinese medicine extract prepared in the comparative examples 1-9, and the protein denaturation index DI is less than 0.55% and is also obviously less than the traditional Chinese medicine extract prepared in the comparative examples 1-9, which indicates that the five traditional Chinese medicine compositions are mutually matched, and the toxic and side effects and the irritation of each traditional Chinese medicine can be obviously reduced after the synergy, and the traditional Chinese medicine composition has scientific and reasonable component collocation.

in vitro weighing method moisturizing performance test research
The principle of the in vitro weighing method for testing the moisture retention performance is as follows: according to the difference of moisture absorption and retention performances of different components, the acting force of different humectant molecules on water molecules is different, and the capacities of absorbing and retaining water are different. The adhesive tape is used for simulating biological materials such as cuticle and the like, the adhesive tape is coated with the humectant and placed for a period of time under the conditions of constant temperature and constant humidity, the mass of the sample placed for different periods of time is weighed, and the moisture retention rate of the sample can be calculated, so that the moisture retention performance of the sample can be evaluated.
The tests were conducted using the conventional high-performance humectant glycerin for cosmetics as a control sample, the Chinese medicinal composition prepared in the above examples 1 to 7 and the medicinal extracts prepared in the comparative examples 1 to 9. The test was as follows:
1. Experimental procedure
The glycerin, the traditional Chinese medicine composition prepared in examples 1 to 7 and the traditional Chinese medicine extract of comparative examples 1 to 9 are respectively weighed and 0.2000g is respectively and uniformly coated on a glass plate which is 5cm by 5cm and is stuck with the microporous ventilating adhesive tape, the glass plate is placed in a dryer with constant temperature and constant humidity, the mass of the glass plate after being placed for 4 hours and 8 hours is respectively weighed, and the moisture retention rate is calculated.
the moisture retention rate calculation formula is as follows:
moisture retention inhibition rate/% (M2-M0)/(M1-M0) × 100%
Wherein M0 is the glass plate mass/g, M1 is the glass plate mass/g after sample loading, and M2 is the glass plate mass/g after being placed in a dryer for several hours.
2. Results of the experiment
The results of the in vitro weighing method for determining the moisture retention rate of the traditional Chinese medicine composition prepared in examples 1 to 7, the traditional Chinese medicine extract prepared in comparative examples 1 to 9 and the glycerin serving as a control sample are shown in the following table 3:
TABLE 3
As can be seen from table 3, the moisture retention rates of glycerin and the Chinese medicinal compositions prepared in examples 1 to 4 were higher than 35% at 4 hours and higher than 34% at 8 hours, and the moisture retention rates were within 1% with time, and the moisture retention effect was stable; the traditional Chinese medicine composition prepared in the embodiments 1 to 4 has higher moisturizing rate than glycerin in 4 hours and 8 hours, which shows that the traditional Chinese medicine composition has superior moisturizing performance to the traditional high-efficiency moisturizing agent glycerin and has excellent moisturizing effect. Meanwhile, the experimental results show that the moisture retention rates of the traditional Chinese medicine extracts prepared in the comparative examples 1-9 for 4h and 8h are far lower than that of the traditional Chinese medicine composition, and the moisture retention effect is not as stable as that of the traditional Chinese medicine composition, which indicates that the components of the traditional Chinese medicine composition are mutually matched to play a synergistic effect so as to achieve the effects of deep water replenishing and lasting moisture retention.

Research on influence of human skin moisture content
the water content of the human skin stratum corneum is measured by adopting a capacitance method, the difference of dielectric constants of water and other substances is obvious, the measured capacitance value of the skin is different according to the difference of the water content of the skin stratum corneum, and the parameter can represent the water content of the skin.
The test was performed using the herbal composition prepared in examples 1-7 and the herbal extract prepared in comparative examples 1-9. The test was as follows:
3 experimental areas of 3 × 3cm2 size were respectively drawn on the inner sides of the left and right forearms of the testee, the left arm was used as a test area of the herbal composition extract, the herbal compositions prepared in examples 1 to 7 and the herbal extracts prepared in comparative examples 1 to 9 were respectively subjected to the test and uniformly spread on the corresponding test areas, the corresponding symmetric area of the right arm was used as a blank control, the moisture content of the skin before the sample was spread on the test area and the blank area of the skin moisture tester and after the sample was spread for 2 hours, and the evaluation criteria of table 4 were performed. Wherein, the change value of the skin moisture content in 2h is the skin moisture content in the test area after the sample is smeared for 2 h-the skin moisture content in the test area before smearing, and the specific test results are shown in table 5.
TABLE 4
TABLE 5
from the test results, the traditional Chinese medicine compositions prepared in the embodiments 1 to 7 of the present invention have a change value of more than 25% in water content for 2 hours, and have excellent moisture retention effect grade, which indicates that the traditional Chinese medicine composition of the present invention can effectively increase the moisture content of skin, and has excellent and lasting moisturizing effect; the change value of the water content of the traditional Chinese medicine extract prepared in the comparative examples 1-9 in 2h is obviously less than 25%, the grade of the moisturizing effect is good or general, and the traditional Chinese medicine extract is not the traditional Chinese medicine composition prepared in the examples 1-7, so that the traditional Chinese medicine composition can achieve the matching effect of monarch, minister, assistant and guide, has the synergistic effect, obviously enhances the moisturizing effect, and is reasonable in formula collocation of the traditional Chinese medicines.
test example 4
study of DPPH radical scavenging ability
The ability to scavenge DPPH free radicals reflects to some extent the antioxidant capacity of the material. The greater the free radical clearance rate, the stronger the antioxidant capacity and the stronger the anti-aging capacity. Therefore, the anti-aging effect of the traditional Chinese medicine composition can be judged by researching the capability of the traditional Chinese medicine composition for eliminating DPPH free radicals.
The tests were carried out using the Chinese medicinal compositions prepared in examples 1 to 7 and comparative examples 1 to 9. The test was as follows:
Adding 2mL of DPPH test solution with the mass concentration of 47.7mg/L and a certain amount of sample to be tested into a test tube, supplementing the total volume to 3mL by absolute ethyl alcohol, shaking up, reacting for 30min in a dark place, and measuring the absorbance at the wavelength of 517nm by using a 1cm cuvette and marking As; adding 2mL of absolute ethyl alcohol and a sample to be detected with a corresponding volume into the test tube, supplementing the total volume to 3mL by the absolute ethyl alcohol, and recording the measured absorbance as Ar; 2mL of DPPH test solution and 1mL of absolute ethanol were added to the tube, and the absorbance measured was recorded as A0.
The DPPH radical clearance (Y) is calculated as: y (%). 1- (As-Ar)/A0X 100%
The DPPH half-clearance (IC50, μ g/mL) of butanediol, VC standard solution, examples 1-7, and comparative examples 1-9 were determined in this order according to the procedure described above.
TABLE 2 measurement of DPPH clearance for Chinese medicinal composition
In the experiment, the antioxidant effect of the sample is evaluated by taking the DPPH half-clearance rate IC50 as an index, and the smaller the DPPH half-clearance rate IC50 value is, the stronger the clearance capability is, the better the antioxidant effect is. VC is a high-efficiency antioxidant accepted by the cosmetic industry, and the test results show that the DPPH half clearance rate IC50 of the traditional Chinese medicine composition prepared in the examples 1-7 is between 10 and 21 mu g/mL, and the traditional Chinese medicine composition prepared in the examples 1-4 is even close to IC50 of VC, so that the traditional Chinese medicine composition has good DPPH free radical scavenging capability, and the traditional Chinese medicine composition prepared in the invention has obvious effects of scavenging free radicals and resisting oxidation; the semi-clearance rate IC50 of DPPH of the traditional Chinese medicine extracts prepared in comparative examples 1-9 is far greater than that of IC50 of VC, and the clearance rate of DPPH free radicals is not ideal, which shows that the traditional Chinese medicine composition prepared by the invention has obvious synergistic effect on all components, has excellent clearance rate of DPPH free radicals, can effectively reduce active oxygen free radicals of skin, resists skin injury and has good antioxidant effect.

Test example 5
Research on influence of human skin roughness and scale level
The dry skin is often accompanied by raised tiny scaly dandruff in the grain gaps, the skin looks dry and rough, for example, the dandruff is more serious due to excessive dryness of the skin, so that the skin feels increased in frictional resistance and obvious in rough feeling.
in the test, skin detectors such as Visia and the like are adopted to analyze the roughness and the scale level of human skin. The rougher the skin, the larger the roughness parameter; the lower the skin scale level, the lower the scale index value and the less skin scale. The test was as follows:
160 subjects who reported self-report dry skin were selected and divided into 10 groups, and the moisturizing and hydrating essence prepared in application example 2 was applied to the inner side of the left forearm of 10 groups of subjects, and the moisturizing and hydrating essence prepared in application comparative examples 1 to 10 was applied to the inner side of the right forearm of 10 groups of subjects in the same amount, respectively. The roughness and skin scale level of the same part of the inner skin of the forearm of the subject were tested before, 5min and 2h after application, respectively. The results of the human skin roughness test of this test are shown in table 12 and fig. 1, and the results of the skin scale level test are shown in table 13 and fig. 2:
TABLE 12
Watch 13
From the test results, after the application comparative example 1 (the moisturizing and moisturizing essence without the traditional Chinese medicine composition) is used for 5min, the rough and scurf states of the skin are obviously relieved due to instant exogenous moisturizing, but after 2h, the roughness parameter of the skin and the skin scale index are increased along with the evaporation of water in the essence matrix, the skin becomes dry and rough again, and the scurf is prominent. After 5min, the skin roughness parameter is reduced and the skin scale index is obviously reduced, and the skin roughness parameter and the scale index are still kept at a lower level after 2h along with the lapse of time after the application example 2 (the moisturizing essence containing 5% of the traditional Chinese medicine composition prepared in the example 1) is used, so that the effects of effectively smoothing upwarping scales and improving rough skin are realized, the skin is kept moist and smooth, and the moisturizing essence is obviously different from a matrix group.
In addition, after the essence of application example 2 is used for 2 hours, the roughness parameter and the skin scale index of the essence are greatly lower than those of application comparative examples 2-10, which fully shows that the traditional Chinese medicine components of the traditional Chinese medicine composition supplement each other and are synergistic, so that the traditional Chinese medicine composition can effectively moisturize and replenish water, relieve dryness and improve the moistening degree and smoothness of skin for a long time.
