本发明涉及食品及保健品加工领域,具体涉及一种综合利用甘薯的加工方法,本加工方法利用有机溶剂或其水溶液对甘薯干粉进行提取,避免了传统加工方法中产生大量淀粉母液,进而产生废水的问题;利用A溶剂提取的甘薯功能提取物制备的甘薯复合颗粒具有抗肿瘤作用,该甘薯功能提取物与大豆异黄酮共同制备的甘薯复合颗粒的抗癌作用远优于大豆异黄酮的抗肿瘤作用;加工过程中产生的甘薯残渣的蛋白质、赖氨酸和淀粉相对含量比原甘薯原料高,是作为甘薯食品和饲料的优质原料。

本发明涉及食品及保健品加工领域,具体涉及一种综合利用甘薯的加工方法。
背景技术
甘薯(Ipomoea batatas Lam.)具有产量高,适应性广的特点,其薯块和藤叶富含碳水化合物、维生素、矿物质、食用纤维等营养物质,不仅是我国的重要粮食和工业原料作物,而且也是仅次于玉米的重要饲料作物。据FAO统计,我国自1990年以来,甘薯年均种植总面积为6.0×106hm2,年均总产量为113.6×106t,分别占全球的65%和86%,均居世界首位。然而我国甘薯总产量的50%用于饲料,15%用于直接鲜食,10%用于加工,6%-8%用于留种,其余接近20%在运输、贮藏等环节中腐烂掉了。甘薯加工主要就是提取甘薯淀粉,生产1吨甘薯淀粉,要排放10-20吨淀粉母液,而甘薯“淀粉母液”COD高达1万以上,对环境造成了严重的污染。
专利CN105054079A公开了一种甘薯加工利用的方法,具体是包括原料加工、淀粉精制、分离膳食纤维和分离功能提取物,其中,淀粉精制阶段将产生大量“淀粉母液”,后续处理过程产生大量废水,大大增加了处理废水的成本。
甘薯淀粉母液的再利用,主要是以下几个方面:作为原料直接用于微生物培养,如蜡样芽孢杆菌、木霉菌、苏云金芽孢杆菌等;甘薯蛋白提取、多酚氧化酶、β-淀粉酶和储藏蛋白提取。ZL2012100523560公开利用甘薯淀粉母液提取工艺提取富含甘薯蛋白的提取物,专利ZL2013101948990公开利用甘薯淀粉母液提取工艺提取富含多酚氧化酶、β-淀粉酶和储藏蛋白的提取物等。但无论哪一种再利用方式,都不能完全解决淀粉母液携带的大量废水,而这些废水同样将大量增加企业的环保成本。
发明内容
为解决上述问题本发明提供一种综合利用甘薯的加工方法,其在加工过程中能够提取具有抗癌作用的保健品原料,同时实现对甘薯全部利用,且无废水产生。
一种综合利用甘薯的加工方法,包括以下步骤:
(1)将洗净的新鲜甘薯切片、干燥、粉碎过筛的甘薯干粉;
(2)利用A溶剂提取甘薯干粉中的功能成分得甘薯提取液和甘薯残渣;
(3)回收甘薯提取液中的A溶剂得甘薯功能提取物;
(4)将甘薯功能提取物用作原料制备甘薯复合颗粒,甘薯残渣用作原料制备甘薯食品或饲料。
所述A溶剂是有机溶剂或有机溶剂水溶液,所述有机溶剂是甲醇、乙醇、丙醇、丙酮或乙酸乙酯。
上述加工方法中不会产生大量的淀粉母液,有机溶剂或其水溶液提取时,有机溶剂和水均会被回收且可重复使用,有效地降低了甘薯加工的成本;上述甘薯残渣的蛋白质、赖氨酸和淀粉相对含量比原甘薯原料高,是作为甘薯食品和饲料的优质原料。
优选的,步骤(1)中,干燥是指烘干、晒干或冻干。
优选的,步骤(1)中,粉碎过筛是粉碎过10目及以上的筛。
优选的,步骤(2)中,A溶剂与甘薯干粉的体积比为1~10︰1;优选的,步骤(2)中,A溶剂与甘薯干粉的体积比为2~4︰1。
优选的,步骤(4)中,甘薯功能提取物的质量占甘薯复合颗粒的质量的1~90%。
优选的,步骤(4)中,甘薯复合颗粒还包括大豆异黄酮和辅料,大豆异黄酮的质量占甘薯复合颗粒的质量的1~90%,余量为辅料。
优选的,步骤(4)中,甘薯功能提取物占甘薯复合颗粒的质量的20~80%,大豆异黄酮的质量占甘薯复合颗粒的质量的20~80%,余量为辅料。
优选的,步骤(4)中,甘薯功能提取物占甘薯复合颗粒的质量的20~50%,大豆异黄酮的质量占甘薯复合颗粒的质量的20~50%,余量为辅料。
上述加工方法制备的甘薯复合颗粒属于本发明的保护范围。
上述加工方法制备的甘薯复合颗粒在制备治疗和/预防肿瘤的药物/保健品中的应用也属于本发明的保护范围。
本发明的有益效果在于:本加工方法利用有机溶剂或其水溶剂液甘薯干粉进行提取,避免了传统加工方法中产生大量淀粉母液,进而产生废水的问题;利用有机溶剂或其水溶液提取的甘薯功能提取物制备的甘薯复合颗粒具有抗肿瘤作用,该甘薯功能提取物与大豆异黄酮共同制备的甘薯复合颗粒的抗癌作用远优于大豆异黄酮的抗肿瘤作用;加工过程中产生的甘薯残渣的蛋白质、赖氨酸和淀粉相对含量比原甘薯原料高,是作为甘薯食品和饲料的优质原料。
具体实施方式
实施例1甘薯功能提取物的制备
将100公斤甘薯洗净,晒干,粉碎过10目,用100升甲醇(1倍体积)浸泡24小时,过滤;残渣回收溶剂后,作为饲料;提取液回收甲醇后,得到甘薯功能提取物。甘薯残渣蛋白质相对含量比甘薯原料提高4%,赖氨酸含量增加8%,淀粉含量提高2%,是优质饲料;提取物水分含量10%以下,是预防肿瘤的优质保健品。
实施例2甘薯功能提取物的制备
将100公斤甘薯洗净,烘干,粉碎过500目,用500升乙醇(5倍体积)浸泡0.1小时,过滤;残渣再用500升乙醇(5倍体积)浸泡10小时;过滤,回收溶剂后,作为饲料;提取液回收乙醇后,得到甘薯功能提取物。甘薯残渣蛋白质相对含量比甘薯原料提高3%,赖氨酸含量增加6%,淀粉含量提高2%,是优质饲料;提取物水分含量10%以下,是预防肿瘤的优质保健品。
实施例3甘薯功能提取物的制备
将100公斤甘薯洗净,烘干,粉碎过200目,用100升丙醇(1倍体积)浸泡1小时,过滤;残渣再用100升乙醇(1倍体积)浸泡2小时;过滤,回收溶剂后,作为饲料;提取液回收丙醇后,得到甘薯功能提取物。甘薯残渣蛋白质相对含量比甘薯原料提高1%,赖氨酸含量增加5%,淀粉含量提高1%,是优质饲料;提取物水分含量10%以下,是预防肿瘤的优质保健品。
实施例4甘薯功能提取物的制备
将100公斤甘薯洗净,烘干,粉碎过100目,用200升丙酮(2倍体积)浸泡1小时,过滤;残渣再用200升丙酮(2倍体积)浸泡1小时;过滤,回收溶剂后,作为饲料;提取液回收丙酮后,得到甘薯功能提取物。甘薯残渣蛋白质相对含量比甘薯原料提高1%,赖氨酸含量增加6%,淀粉含量提高1%,是优质饲料;提取物水分含量10%以下,是预防肿瘤的优质保健品。
实施例5甘薯功能提取物的制备
将100公斤甘薯洗净,烘干,粉碎过50目,用200升乙酸乙酯(2倍体积)浸泡1小时,过滤;残渣再用200升乙酸乙酯(2倍体积)浸泡1小时;过滤,回收溶剂后,作为饲料;提取液回收乙酸乙酯后,得到甘薯功能提取物。甘薯残渣蛋白质相对含量比甘薯原料提高1%,赖氨酸含量增加5%,淀粉含量提高1%,是优质饲料;提取物水分含量10%以下,是预防肿瘤的优质保健品。
实施例5甘薯功能提取的制备
将100公斤甘薯洗净,烘干,粉碎过20目,用200升乙醇-水(90:10)浸泡2小时,过滤;残渣再用200升乙醇-水(90:10)浸泡2小时;过滤,回收溶剂后,作为饲料;提取液回收乙醇溶液后,得到甘薯功能提取物。甘薯残渣蛋白质相对含量比甘薯原料提高3%,赖氨酸含量增加7%,淀粉含量提高3%,是优质饲料;提取物水分含量10%以下,是预防肿瘤的优质保健品。
实施例6甘薯功能提取物的制备
将100公斤甘薯洗净,烘干,粉碎过20目,用200升乙醇-水(10:90)浸泡2小时,过滤;残渣再用200升乙醇-水(10:90)浸泡2小时;过滤,回收溶剂后,作为饲料;提取液回收乙醇溶液后,得到甘薯功能提取物。甘薯残渣蛋白质相对含量比甘薯原料提高2%,赖氨酸含量增加6%,淀粉含量提高1%,是优质饲料;提取物水分含量10%以下,是预防肿瘤的优质保健品。
实施例7甘薯复合颗粒的制备
取甘薯功能提取物90g、大豆异黄酮1g、糊精8.9g、甜蜜素0.1g,混合均匀,按照一般制粒工艺制粒,60℃下烘干,分装即可。大豆异黄酮为购买的食品级大豆提取物,异黄酮含量为99%。该产品具有较好的预防肿瘤效果。
实施例8甘薯复合颗粒的制备
取甘薯功能提取物1g、大豆异黄酮90g、糊精5g、蔗糖4g,混合均匀,按照一般制粒工艺制粒,60℃下烘干,装胶囊即可。大豆异黄酮为购买的食品级大豆提取物,异黄酮含量为1%。该产品具有较好的预防肿瘤效果。
实施例9甘薯复合颗粒的制备
取甘薯功能提取物20g、大豆异黄酮50g、糊精25g、白糖5g,混合均匀,按照一般制粒工艺制粒,60℃下烘干,分装即可。大豆异黄酮为购买的食品级大豆提取物,异黄酮含量为20%。该产品具有较好的预防肿瘤效果。
实施例10甘薯复合颗粒的制备
取甘薯功能提取物50g、大豆异黄酮20g、糊精25g、白糖5g,混合均匀,按照一般制粒工艺制粒,60℃下烘干,分装即可。大豆异黄酮为购买的食品级大豆提取物,异黄酮含量为80%。该产品具有较好的预防肿瘤效果。
实施例11甘薯复合颗粒的制备
取甘薯功能提取物45g、大豆异黄酮45g、糊精9g、甜蜜素1g,混合均匀,按照一般制粒工艺制粒,60℃下烘干,分装即可。大豆异黄酮为购买的食品级大豆提取物,异黄酮含量为50%。该产品具有较好的预防肿瘤效果。
对照实施例1甘薯全粉
甘薯洗净,干燥,粉碎过100目。作为对比原料。
对照实施例2甘薯全提取物的制备
将10L甘薯淀粉母液干燥,减压浓缩至固型物含量20%,喷雾干燥,得到甘薯全提取物。
预防肿瘤效果试验
1.试验材料
甘薯功能提取物,实施例1-6任一实施例制备的甘薯功能提取物;
大豆异黄酮:为购买的产品,异黄酮含量50%;
甘薯复合颗粒,实施例7-11任一实施例制备;
MCF-7肿瘤细胞,选择生长对数期的MCF-7肿瘤细胞,用0.25%胰蛋白酶液消化1~2min,倒出消化液,加入PBS洗涤,反复吹打,将细胞悬液移至离心管中,1300rpm离心5min,弃上清,加入DMEM完全培养基,调整细胞浓度为1×108个/ml,0.4%台盼蓝检测活细胞>95%,备用。
2.试验动物
裸小鼠(nude mice):BALB/cnu,4周龄,80只,16-20g/只。
3.试验方法
将80只4周龄BALB/c nu裸鼠单笼饲养,屏障隔离系统平衡3~5天,无菌饲料和水充足。将裸鼠随机分为8组,每组10只,除阴性对照组外,各组裸鼠均接种肿瘤细胞(0.1ml/只),给予实验药物饲养,5g/(只*天),持续30天,30天后,处死老鼠,取下肿瘤块,称重,分析比较每组肿瘤组织平均重量,结果见表1。
4.试验结果
表1、抗肿瘤效果实验
试验结果表明,与阳性对照组相比(*组),甘薯功能提取物和大豆异黄酮均有较好的抗肿瘤效果(达到显著水平),而甘薯全粉和全提取物虽然有一定的抗肿瘤效果,但抗肿瘤效果很差(未达显著水平);甘薯功能提取物与大豆异黄酮复合后,尽管给药剂量相同,其抗肿瘤效果显著提升,达极显著水平,表明二者有明显的协同作用;随着配比的进一步优化,其抗肿瘤效果进一步提高。
与大豆异黄酮和甘薯功能提取物组相比(#组),甘薯复合颗粒抗肿瘤效果显著提升,达显著水平,表明二者有明显的协同作用;随着配比的进一步优化,其抗肿瘤效果进一步提高,达到极显著水平。
此外,从肿瘤发生率看,大豆异黄酮和甘薯工功能提取物均有预防肿瘤的作用,而甘薯全提取物却没有该作用;甘薯复合颗粒预防肿瘤的效果进一步提高,表明甘薯复合颗粒具有预防肿瘤的效果。
Claims (10)
1.一种综合利用甘薯的加工方法,包括以下步骤:
(1)将洗净的新鲜甘薯切片、干燥、粉碎过筛的甘薯干粉;
(2)利用A溶剂提取甘薯干粉中的功能成分得甘薯提取液和甘薯残渣;
(3)回收甘薯提取液中的A溶剂得甘薯功能提取物;
(4)将甘薯功能提取物用作原料制备甘薯复合颗粒,甘薯残渣用作原料制备甘薯食品或饲料;
所述A溶剂是有机溶剂或有机溶剂水溶液,所述有机溶剂是甲醇、乙醇、丙醇、丙酮或乙酸乙酯。
2.如权利要求1所述的加工方法,其特征在于,步骤(1)中,干燥是指烘干或晒干。
3.如权利要求1所述的加工方法,其特征在于,步骤(1)中,粉碎过筛是粉碎过10目及以上的筛。
4.如权利要求1所述的加工方法,其特征在于,步骤(2)中,A溶剂与甘薯干粉的体积比为1~10︰1;优选的,步骤(2)中,A溶剂与甘薯干粉的体积比为2~4︰1。
5.如权利要求1所述的加工方法,其特征在于,步骤(4)中,甘薯功能提取物的质量占甘薯复合颗粒的质量的1~90%。
6.如权利要求5所述的加工方法,其特征在于,步骤(4)中,甘薯复合颗粒还包括大豆异黄酮和辅料,大豆异黄酮的质量占甘薯复合颗粒的质量的1~90%,余量为辅料。
7.如权利要求6所述的加工方法,其特征在于,步骤(4)中,甘薯功能提取物占甘薯复合颗粒的质量的20~80%,大豆异黄酮的质量占甘薯复合颗粒的质量的20~80%,余量为辅料。
8.如权利要求7所述的加工方法,其特征在于,步骤(4)中,甘薯功能提取物占甘薯复合颗粒的质量的20~50%,大豆异黄酮的质量占甘薯复合颗粒的质量的20~50%,余量为辅料。
9.权利要求1-8任一项所述的加工方法制备的甘薯复合颗粒。
10.权利要求1-8任一项所述的加工方法制备的甘薯复合颗粒在制备治疗和/预防肿瘤的药物/保健品中的应用。
