Abstract
阿托帕沙溴酸作为新型JAK‑STAT3信号通路抑制剂的用途,可用于抑制STAT3活性和STAT3下游基因表达来控制细胞增殖;对细胞因子诱导的STAT3活化和JAKs自磷酸化具有抑制作用;计算机分子模拟可结合于STAT3的SH2结构域,与JAK激酶JH1结构域有很高亲和力;可水平抑制肿瘤细胞生长。阿托帕沙溴酸作为新型JAK‑STAT3信号通路抑制剂的研究方法,包括如下步骤:细胞培养;蛋白提取;Western‑Blot;RNA提取;RT‑PCR及荧光定量PCR;荧光素酶活性分析;细胞活性分析;流式细胞术检测细胞周期;流式细胞术检测细胞凋亡;裸鼠移植瘤实验;计算机分子动力学模拟对接分析;数据计算及处理。本发明为进一步细胞周期检测和后续新型小分子药物研发奠定了基础。

技术领域
本发明属于医药应用领域,涉及阿托帕沙溴酸的医药新用途和研究方法,具体地说,是其作为JAK-STAT3信号通路抑制剂的用途和研究方法。
背景技术
JAK-STAT信号通路是细胞生长,激活,分化,凋亡及其功能过程中重要的细胞内信号转导通路。STAT3信号的异常激活可导致肿瘤发生,例如乳腺癌,肺癌。由于其在肿瘤形成中的重要功能,JAK(Janus激酶)和STAT(信号转导及转录激活因子)已成为药物设计的流行目标。尽管针对JAK和STAT的抑制剂已经FDA,EMA或临床试验批准,但也观察到某些缺陷,例如耐药性和副作用。此外,作为激酶抑制剂,这些药物可以与激酶的ATP袋结合,并且ATP袋是高度保守的,因此它可能导致脱靶和不可预测的毒性作用。因此,有必要开发新的JAK或STAT抑制剂。
阿托帕沙溴酸,Atopaxar Hydrobromide(AHB),CAS号:474550-69-1,分子式C29H38N3O5F.HBr,分子量:608.54,又称为E5555 hydrobromide。E5555是一种可逆蛋白酶激活受体(Protease activated receptor,PAR)-1凝血酶受体拮抗剂,可以干扰血小板信号传导,目前尚无明确报道阿托帕沙溴酸的具体作用靶点。
发明内容
基于上述技术背景,本发明提供阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的用途和研究方法。
本发明主要目的是提供阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的用途。通过本研究所建立的基于STAT3信号通路萤火虫荧光素酶报告基因的抗肿瘤药物筛选平台,筛选发现了小分子化合物阿托帕沙溴酸,实验结果发现,其具有明显的STAT3抑制活性,高特异性、低细胞毒性,还抑制STAT3下游基因表达,包括socs3,细胞周期蛋白D1,c-Myc和Bcl-xL。此外,阿托帕沙溴酸经由JAKs-STATs信号通路来调节与细胞生长密切相关的一些下游基因的表达来控制细胞增殖。Western-Blot和MTT测定显示AHB可以剂量和时间依赖性方式抑制STAT3组成型活化的癌细胞活力,即DU145和A549,HepG2,Hela和MDA-MB-231。阿托帕沙溴酸也对细胞因子诱导的STAT3活化和JAKs自磷酸化具有抑制作用。分子模拟对接实验也在分子结构上直观的表明,阿托帕沙溴酸可结合于STAT3的SH2结构域并形成氢键,与JAK激酶JH1结构域具有很高的亲和力并形成氢键。暗示阿托帕沙溴酸是一种信号特异性抑制剂。详细调查表明,一些关键的STAT3下游基因参与细胞周期和细胞凋亡。荧光素酶活性和细胞毒性分析,阿托帕沙溴酸能够抑制荧光素酶活性,因荧光素酶的表达要依靠上游STAT3的转录活性,故荧光素酶活性可以正比显示STAT3的活性,而MTT细胞活性实验分析其具有较低的细胞毒性,因而说明阿托帕沙溴酸可直接作用于STAT3信号启动。
进一步研究发现,阿托帕沙溴酸具有明显的STAT3抑制活性,高特异性、低细胞毒性,可以有效抑制STAT3信号依赖性前列腺癌DU145细胞中持续激活的STAT3活性,对STAT3所调控的下游基因,如Bcl-XL、CyclinD和c-Myc,的表达也有抑制作用,而这些基因在细胞的生长、凋亡和细胞周期调控中有重要的作用。我们的研究结果显示,阿托帕沙溴酸诱导G1期细胞周期停滞,并促进细胞凋亡。RT-PCR方法在RNA水平也验证了这一点。通过Western-Blot分析,阿托帕沙溴酸也对细胞因子诱导的STAT3活化和JAKs自磷酸化具有抑制作用。分子模拟对接实验也在分子结构上直观的表明,阿托帕沙溴酸可结合于STAT3的SH2结构域并形成氢键,与JAK激酶JH1结构域具有很高的亲和力并形成氢键。在文献报道中,IL-6(白细胞介素-6)、IFNα(干扰素α)、IFNγ(干扰素α)诱导的JAK-STAT信号中,也包含了STAT1的活化,实验结果也发现阿托帕沙溴酸具有同样的抑制效应,暗示阿托帕沙溴酸的作用靶点可能在STAT3和STAT1蛋白的共同上游。其中在JAKs激酶过度活化所介导的信号通路中,STAT3是研究最为透彻的。大部分肿瘤组织中STAT1是失活的,且在效应上与STAT3作用相反。
更进一步的实验结果也发现阿托帕沙溴酸可以抑制细胞因子诱导的JAKs家族成员的自磷酸化。所以可以初步推定阿托帕沙溴酸应该是直接与JAKs相互结合,影响JAK激酶对STAT3的激活。由于JAKs成员都是只有JH1结构域负责底物催化,且与JAK激酶JH1结构域具有很高的亲和力并形成氢键,所以我们可以通过体外激酶实验检测阿托帕沙溴酸对JAKs家族激酶结构域的有效作用关系。可以纯化STAT3蛋白和构建JAKs-JH1过表达稳转细胞株。后续构建荷瘤小鼠进行的体内动物实验,也进一步验证阿托帕沙溴酸的抗肿瘤活性。体内结果还证实了在5mg/kg/天和10mg/kg/天阿托帕沙溴酸处理后其抑制作用。此外,阿托帕沙溴酸治疗显示小鼠体重平稳稳定,证明阿托帕沙溴酸安全,在一定程度上无副作用。
分子模拟对接实验也在分子结构上直观的表明,阿托帕沙溴酸可结合于STAT3的SH2结构域并形成氢键,与JAK激酶JH1结构域具有很高的亲和力并形成氢键。后续可以根据两者之间的分子结构关系,进一步优化阿托帕沙溴酸的分子结构,使其抑制效果更强,细胞毒性更小,为肿瘤相关疾病的治疗提供候选物。总之,我们发现阿托帕沙溴酸是一种新型抑制剂,可用于临床治疗对JAK-STATs信号转导成瘾的肿瘤。
为该靶标开发的JAK抑制剂可用于筛选血液系统疾病,肿瘤,类风湿性关节炎和牛皮癣。自从20世纪90年代早期发现第一种JAK激酶以来,直到2012年,第一种JAK激酶抑制剂Tofacitinib才被批准用于治疗类风湿性关节炎(RA)。目前,FDA和EMA批准的JAK抑制剂包括Ruxolitinib,Tofacitinib,和Baricitinib。根据JAK抑制剂的结构,第一代JAK抑制剂的母体核结构在ADP中含有相似的腺嘌呤结构,其特征在于同时抑制多种JAK亚型激酶的能力。因此,通常存在第一代JAK抑制剂中选择性低的问题。尽管Tofacitinib被认为是RA治疗的一流药物,因为它对JAK3的特异性是JAK1和JAK2的5-100倍,它最终被证明是一个泛-Jaks抑制剂在激酶测定中。通过优化母核,第二代JAK抑制剂特异性靶向不同的JAK亚型。在不同的临床试验中有超过20种第二代JAK抑制剂,包括Filgotinib,Upadacitinib和Solcitinib选择性抑制JAK1:decernotinib和PF06651600选择性抑制JAK3:BMS986165,NDI021232,NDI031407,PF06700841和SAR20347选择性地抑制TYK2。它们的特征在于针对不同亚型设计的选择性JAK抑制剂。当JAK抑制剂的开发刚刚开始时,基于对JAK不同生物学功能的理解,研究人员预测了JAK抑制剂的一些特征。随着JAK抑制剂的大规模临床试验,一些原始预测已经得到证实,然而,在任何情况下,类似于Ruxolitinib的阿托帕沙溴酸,作为JAK1和JAK2的特异性抑制剂,可以像Ruxolitinib一样用于治疗原发性骨髓纤维化,骨髓纤维化患者和特发性血小板减少症患者的真正红细胞增多症。
本发明的另一个目的是提供阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的研究方法,具体包括如下步骤:
S1:细胞培养:hTERT-BJ、HELA、HepG2、A549和SKA等细胞培养于DMEM高糖培养基,DU145和MDA-MB-231等细胞培养于RPMI-1640培养基。
S2:蛋白提取:将培养皿中细胞刮下后收集好,离心后吸走上清使用裂解液裂解细胞,并转移到预冷的离心管中离心、取上清,进行BCA蛋白定量,样品加入5×蛋白上样缓冲液处理,随后进行Western-Blot分析。
S3:Western-Blot分析:配制8%~10%SDS-PAGE凝胶,电泳,然后将凝胶浸泡至预冷的转膜缓冲液中,甲醇处理PVDF膜,进行三明治转膜。取出转印好的PVDF膜,蛋白面向上,室温封闭;一抗配制4℃孵育过夜,TBST清洗5次,每次5min;二抗,TBST清洗PVDF膜4次,每次10min。加ECL显影曝光。
S4:RNA提取:细胞培养后弃培养基,用PBS清洗后,加TRIZOL试剂,摇匀,消化后依次收集至EP管中,加氯仿,轻摇。静置,离心。取上清无色水相到新的EP管中,加异丙醇,静置,离心。弃上清,用75%乙醇洗涤,离心。去上清,吸干液体,干燥至管底透明。加入DEPC处理水20~30μl,55~60℃水浴10min溶解总RNA,测OD值。
S5:RT-PCR及荧光定量PCR:根据罗氏逆转录试剂盒进行反转录,取1μg总RNA进行反应。
S6:荧光素酶活性分析:SKA细胞铺于96孔板(100μl)中,待细胞贴壁后,更换含有阿托帕沙溴酸梯度浓度的培养基处理,然后于每孔加入荧光素酶底物,测定荧光素酶的活性。
S7:细胞活性分析(MTT法):将一定数量的细胞铺于96孔板(100μl),然后加不同药物浓度,24h或者48h或者72h后测定,加入20μl 5mg/ml MTT溶液,培养箱培养3~4h,吸走孔内液体,加入DMSO完全溶解结晶,摇床轻晃,490nm测定吸光值。
S8:流式细胞术检测细胞周期:培养细胞于10%FBS的DMEM培养基中,生长至80%汇合度,取样品,用预冷的PBS洗涤。在预冷的70%乙醇中固定、孵育。用PBS洗涤,离心,弃去上清液。用核糖核酸酶处理细胞,加入Rnase储存液和PI储存液,室温避光染色,流式细胞术测定各组细胞周期的百分率。
S9:流式细胞术检测细胞凋亡:培养DU145细胞至70%左右汇合度,不同阿托帕沙溴酸梯度浓度处理。贴壁细胞和培养基中的飘浮细胞均低速离心收集,使用预冷的PBS清洗后,再分别加入PBS将细胞重悬。将细胞悬液,碘化丙啶和Annexin V-FITC混合到一起,避光反应。再分别加入Annexin V结合缓冲液,静置反应后尽快流式细胞仪检测。
S10:裸鼠移植瘤实验:将人源A549细胞瘤块种植于BALB/c裸鼠的左侧腋下,3天后开始给药,将它们随机分为空白对照组、阳性Gefitinib灌胃组、腹腔注射阿托帕沙溴酸低浓度组、以及腹腔注射阿托帕沙溴酸高浓度组,每天给药,定期用游标卡尺测量瘤体的长径和短径,并称量记录小鼠体重,观察记录小鼠异常的生理状况。给药若干天后处死,解剖剥离移植瘤,拍照,称重。利用公式V(mm3)=0.5×a(长径)×b(短径)2计算瘤体体积,绘制A549移植瘤生长曲线。
S11:计算机分子动力学模拟对接分析:JAK1和JAK2的结构来源蛋白质数据库,提取相应的激酶结构域[140]。从PUBCHEM查到化合物阿托帕沙溴酸的3D结构,采用Avogadro软件进行结构优化。AutoDockTools软件分析分子结构,划定网格盒子并将此区域作为分子对接的口袋。PDB2PQR程序用于PH=7时的质子化[141]。采用NAMD程序对添加氢进行优化。对接盒位于JH1域(Janus同源结构域1)中。
S12:数据计算及处理:实验均经过重复验证,与对照组数据的比值表示荧光素酶活性和细胞活性用。数据之间的显著性使用单因素方差分析比较;使用SPSS19软件计算IC50(EC50)计算,用±SD表示标准差。使用Origin 8做柱状图及折线图。
进一步的,S1中DMEM高糖培养基和RPMI-1640培养基中包含双抗青霉素链霉素和10%胎牛血清FBS。
进一步的,S1中细胞培养在37℃,5%CO2条件下进行。

具体实施方式
为了便于理解,下面结合附图,通过实施例,对本发明技术方案作进一步具体描述:
一种阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的研究方法,具体包括如下步骤:
S1:细胞培养:hTERT-BJ、HELA、HepG2、A549和SKA(基于STAT3的抗肿瘤药物高通量筛选平台稳转株,本实验室已构建成功的细胞株)等细胞培养于DMEM高糖培养基,DU145和MDA-MB-231等细胞培养于RPMI-1640培养基,在37℃,5%CO2条件下培养。DMEM高糖培养基和RPMI-1640培养基包含双抗青霉素链霉素和10%胎牛血清FBS。
S2:蛋白提取:100mm细胞培养皿中将细胞培养至80%的汇合度,用4℃预冷的PBS清洗3次,随后用刮棒刮下细胞后收集好,2,500g离心1min,小心吸走上清使用配制的裂解液(50mM HEPES、pH 7.0、250mM NaCl、0.1%NP-40、10%甘油、1mM PMSF、1mM DTT、1×RocheCocktail)4℃,30min内裂解细胞,并全部转移到预冷的离心管中,超声30s,4℃下12,000×rpm离心10min,取上清。进行BCA蛋白定量,样品加入5×蛋白上样缓冲液在100℃下处理8min,随后进行Western-Blot分析。
S3:Western-Blot分析:配制8%~10%SDS-PAGE凝胶,80V,30min,120V 1h电泳,完成后将聚丙烯酰胺凝胶浸泡至预冷的转膜缓冲液中,甲醇处理PVDF膜25s,进行三明治转膜,设定恒压90V 2h。配制含5%脱脂奶粉的TBST,小心取出转印好的PVDF膜,蛋白面向上,室温封闭1~2h;一抗按说明书比例配制4℃孵育过夜,TBST清洗5次,每次5min;二抗,TBST清洗PVDF膜4次,每次10min。加ECL显影曝光。
S4:RNA提取:6孔板细胞培养至90~100%,弃培养基,用PBS洗两次后,每孔加TRIZOL试剂(Gibco公司)1ml,摇匀,超净台内消化3~5min(待液体变粘稠,细胞脱壁)。依次收集至DEPC处理过的1.5ml EP管中,加新开的氯仿0.2ml,轻摇15s。室温静置2~3min,12,000rpm离心(15min,4℃)。取上清无色水相到新的DEPC处理过的EP管中,加0.5ml新开的异丙醇,室温静置10min,12,000rpm离心(10min,4℃)。观察总RNA在管底的白色沉淀,弃去上清,用DEPC水新配制的75%乙醇1.0ml洗涤,4℃温度下7,500rpm离心5min。去上清,吸干液体,室温干燥5~10min至管底透明。加入DEPC处理水20~30μl,55~60℃水浴10min溶解总RNA,测OD值。
S5:RT-PCR及荧光定量PCR:根据罗氏逆转录试剂盒进行反转录,取1μg总RNA进行反应。PCR引物如下[137-138]:

S6:荧光素酶活性分析:SKA细胞铺于96孔板中,每孔0.8×104个细胞,100μl体系,待细胞贴壁12h以后,更换含有阿托帕沙溴酸梯度浓度(0.5μM、1μM、2.5μM、5μM、7.5μM、10μM、12.5μM、15μM、20μM)的新鲜培养基处理24h,处理24h后直接于每孔加入20μl Steady-Glo稳定型荧光素酶底物,避光放置5min,测定荧光素酶的活性。
S7:细胞活性分析(MTT法):将一定数量的细胞铺于96孔板(100μl),12h后加不同药物浓度,24h或者48h或者72h后测定,加入20μl 5mg/ml MTT溶液,培养箱培养3~4h,吸走孔内液体,加入100μl DMSO完全溶解结晶,摇床轻晃5min,490nm测定吸光值。
S8:流式细胞术检测细胞周期:培养细胞于10%FBS的DMEM培养基中,生长至80%汇合度,每个样品收集约106~107个细胞,用预冷的PBS洗涤两次。在预冷的70%乙醇中固定,在涡旋搅拌的同时向细胞沉淀中逐滴加入乙醇,最大限度避免细胞聚集,在恒定涡流下固定细胞,然后在4℃下孵育1h。用PBS洗涤2次,850g离心30s,小心弃去上清液。用核糖核酸酶处理细胞,加入50μl Rnase储存液(100μg/mL),200μl PI储存液(50μg/mL),室温避光染色30min,流式细胞术测定各组细胞周期的百分率。
S9:流式细胞术检测细胞凋亡:培养DU145细胞至70%左右汇合度,不同阿托帕沙溴酸梯度浓度(7.5μM、12.5μM)处理24h。贴壁细胞和培养基中的飘浮细胞均低速离心收集,使用预冷的PBS清洗两遍后,再分别加入500μLPBS将细胞重悬。将100μL细胞悬液,、4μL碘化丙啶和4μL Annexin V-FITC混合到一起,避光反应15min。再分别加入400μL Annexin V结合缓冲液,静置反应5min。随后尽快流式细胞仪检测。
S10:裸鼠移植瘤实验:以组织插块法将人源A549细胞瘤块种植于BALB/c裸鼠的左侧腋下,接种瘤块大小2×2mm,3天后开始给药,将它们随机分成四组,每组六只小鼠。分为空白对照组、阳性Gefitinib(50mg/kg/day)灌胃组,腹腔注射阿托帕沙溴酸(5mg/kg/day)低浓度组、以及腹腔注射阿托帕沙溴酸(10mg/kg/day)高浓度组,每天给药并持续22天,每隔3天用游标卡尺测量瘤体的长径和短径,并记录称小鼠体重,观察记录小鼠异常的生理状况。给药22天后处死,解剖剥离移植瘤,拍照,称重。利用公式V(mm3)=0.5×a(长径)×b(短径)2计算瘤体体积,绘制A549移植瘤生长曲线。

对上述研究结果进行试验验证:
实验1:验证阿托帕沙溴酸STAT3磷酸化有显著的抑制作用。
将约8×103SKA细胞铺于96孔板(100μl培养基/孔)。待细胞贴壁12h以后,更换含有阿托帕沙溴酸梯度浓度(0.5μM、1μM、2.5μM、5μM、7.5μM、10μM、12.5μM、15μM、20μM)的新鲜培养基处理24h,测定荧光值和相对细胞存活率(MTT法)。对照使用DMSO,误差棒表示±SD(标准差)。基于STAT3驱动的荧光素酶报告系统[142]共筛选出16081个类药物分子,分别来自FDA批准的药物、临床试验化合物、生物活性化合物和天然产物化学物质。与其他化合物相比,阿托帕沙溴酸是从对STAT3驱动的报告基因表达有较强抑制作用的天然产物文库中选择的,如图1(A)所示,说明从天然产物文库中筛选出阿托帕沙溴酸,它具有明显的STAT3信号抑制作用,其倍数变化大于平均值。然后我们利用萤火虫荧光素酶活性分析方法及细胞活性检测分析方法,验证并评估阿托帕沙溴酸对STAT3信号抑制特异性和细胞毒性。利用本实验室已成熟的含STAT3启动元件并带有荧光素酶报告基因的稳定细胞株检测阿托帕沙溴酸对STAT3信号的抑制,发现阿托帕沙溴酸从0.5μM到20μM处理24h后,对荧光素酶活性有浓度依赖效应,并使用细胞活性检测相同浓度下细胞生长的相对抑制率,计算得出IC50=9.96μM;EC50=4.90μM。可见阿托帕沙溴酸对A549细胞信号的STAT3抑制作用远大于细胞毒性作用,说明阿托帕沙溴酸具有良好的信号特异性,如图1(B)所示,说明阿托帕沙溴酸以剂量依赖性方式阻断STAT3信号传导(如图1所示)。
实验2:验证阿托帕沙溴酸对多种肿瘤细胞生长具有抑制作用。
将hTERT-BJ、DU145、A549、MDA-MB-231、HepG2和Hela细胞以细胞数目约8×103细胞每孔铺于96孔板中。待12h后细胞贴壁,更换含有7.5μM阿托帕沙溴酸的新鲜培养基,每种细胞每个时间段3个复孔,分别于24h、48h及72h后以MTT实验测定细胞相对存活率。误差棒表示±SD,空白处理使用DMSO。
STAT3信号的异常激活在肿瘤发生中起关键作用。我们进一步比较了它对几种常见肿瘤细胞的抑制作用,包括前列腺癌细胞DU145、肺癌细胞A549、乳腺癌MDA-MB-231、肝癌HepG2、宫颈癌Hela细胞以及正常人成纤维细胞hTERT-BJ,结果表明,使用7.5μM阿托帕沙溴酸处理24h、48h和72h后,结果显示阿托帕沙溴酸对肿瘤细胞具有较好的抑制作用,随处理时间的延长,差异逐步明显(如图2所示)。
实验3:验证阿托帕沙溴酸抑制DU145细胞中持续活化的STAT3。
将细胞铺于100mm培养皿培养12h,更换含有阿托帕沙溴酸(10μM)的新鲜1640完全培养基处理DU145细胞2h,收集蛋白,做Western-blot检测,发现阿托帕沙溴酸对DU145细胞中STAT3信号具有显著的抑制作用。
组成型活化的STAT3信号传导在肿瘤细胞中是常见的,例如乳腺癌、肺癌和前列腺癌等。根据文献报道,人前列腺癌细胞DU145对STAT3信号具有生长依赖性,是STAT3超活化细胞。我们将通过阿托帕沙溴酸对这类细胞的作用来检测阿托帕沙溴酸的信号抑制效应(如图3所示)。
实验4:验证阿托帕沙溴酸可下调STAT3下游基因Bcl-XL、CyclinD、c-Myc的表达。
DU145细胞铺于100mm细胞培养皿培养12h,更换含有5μM阿托帕沙溴酸的新鲜完全培养基,处理24h,提取总蛋白,进行Western-Blot分析。
在许多STAT3组成型活化的癌细胞中,阻断STAT3信号通过下调STAT3下游基因(包括CyclinD1,Bcl-xl,c-Myc)来阻止细胞生长,阻滞细胞周期并增加细胞凋亡。根据STAT3启动的下游目的基因的类型,我们检测了与细胞存活相关的基因的表达活性,如c-Myc、Bcl-xL和CyclinD1,阿托帕沙溴酸处理DU145细胞24h后,c-Myc、Bcl-xL和CyclinD1的表达量显著下降(如图4所示)。
实验5:流式细胞术细胞周期分析,验证阿托帕沙溴酸可将肿瘤细胞周期阻滞至G1与G2期,并促进肿瘤细胞的凋亡。
DU145细胞培养至80%的汇合度,更换含有阿托帕沙溴酸(5μM)的新鲜培养基(DMEM,10%FBS),处理24h,通过PI染色,FACS流式细胞仪分析。使用DMSO作为NC对照。
众所周知,细胞周期由一些检查点蛋白精确调节,例如CyclinD1。为了通过诱导细胞周期停滞来确定阿托帕沙溴酸是否影响细胞活力,我们在阿托帕沙溴酸处理24h后用PI染色DU145细胞,然后通过流式细胞术做了进一步更直观的展示,阿托帕沙溴酸(10μM)处理DU145细胞24h,通过PI染色,FACS流式细胞仪分析。使用DMSO作为NC对照(Vehicle),结果发现阿托帕沙溴酸可将细胞周期阻滞至G1及G2期(如图5所示)。
实验6:流式细胞术细胞凋亡分析。验证阿托帕沙溴酸可促进肿瘤细胞的凋亡。
将DU145细胞铺于100mm的细胞培养皿中,培养至70%的汇合度,分别更换含有DMSO、阿托帕沙溴酸梯度浓度(7.5μM、12.5μM)来处理细胞24h。通过碘化丙啶(propidium)和Annexin V-FITC,避光染色。流式细胞仪检测。
在不同浓度(7.5μM和12.5μM)的阿托帕沙溴酸处理24h后,用膜联蛋白V-FITC和PI双重染色DU145细胞,使用DMSO作为NC对照(Vehicle),结果显示阿托帕沙溴酸确实诱导细胞凋亡(如图6所示)。
实验7:验证阿托帕沙溴酸可抑制细胞因子IL-6、IFNα和IFNγ诱导的STAT3和STAT1的活化。
将Hela细胞和A549细胞铺于100mm培养皿,12h以后,加入10μM阿托帕沙溴酸预处理30min,加20ng/ml IL-6刺激第二个30min,进行Western-Blot分析,发现阿托帕沙溴酸可有效抑制外源细胞因子IL-6诱导的STAT3活化(如图7所示)。
实验8:验证阿托帕沙溴酸抑制细胞因子IL-6诱导的STAT3下游基因socs3的mRNA的活性。
A549细胞和DU145细胞铺于100mm培养皿,12h以后,加入10μM阿托帕沙溴酸预处理1h,加20ng/ml IL-6刺激另外3h,提取总RNA,进行RT-PCR分析。
细胞因子IL-6调控的STAT3最直接的下游基因中,受STAT3活性的严格调控的分子中,socs3是最典型的,其可以通过RT-PCR检测socs3mRNA的变化,发现阿托帕沙溴酸能有效抑制A549细胞和DU145细胞中IL-6诱导的socs3mRNA增加(如图8所示),在A549细胞中这种抑制高达50%以上。
实验9:验证阿托帕沙溴酸可抑制细胞因子IFNα和IFNγ诱导的STAT3活化。
将A549细胞铺于100mm培养皿12h,加入10μM阿托帕沙溴酸处理30min,再加5,000U/ml IFNα或1,500U/ml IFNγ处理另一个30min。提取总蛋白,进行Western-Blot分析,发现阿托帕沙溴酸也能有效抑制外源细胞因子IFNα、IFNγ诱导的STAT3活化(图9所示)。
实验10:进一步检测STAT1的磷酸化活性。
A549细胞铺于100mm培养皿,12h以后,加入10μM阿托帕沙溴酸预处理30min,加20ng/ml IL-6刺激第二个30min,进行Western-Blot分析。结果发现阿托帕沙溴酸对STAT1的活化也有抑制作用(图10所示)。
实验11:验证阿托帕沙溴酸IFNα诱导的mRNA水平STAT1下游基因irf-1的抑制作用。
将A549细胞培养至汇合度80%,加入10μM阿托帕沙溴酸预处理1h,加20ng/ml IL-6刺激另外3h,提取总RNA,进行RT-PCR分析,发现STAT1下游基因irf mRNA的表达和实验10是一致的结果(图11所示),揭示阿托帕沙溴酸的作用靶点可能在STAT3和STAT1蛋白的共同上游。
实验12:验证阿托帕沙溴酸对JAKs激酶家族中其他成员的影响。
A549细胞培养至汇合度80%,加入10μM阿托帕沙溴酸处理30min,再加5,000U/mlIFNα或1,500U/ml IFNγ处理另一个30min。提取总蛋白,进行Western-Blot分析。结果显示阿托帕沙溴酸对外源细胞因子IL-6、IFNα和IFNγ诱导的诱导的JAK1及JAK2激酶具有抑制作用,尤其对JAK1的磷酸化抑制作用明显(图12所示)。
实验13:验证阿托帕沙溴酸可结合于STAT3的SH2结构域。
为了更直观的验证阿托帕沙溴酸对JAK-STAT3的抑制作用,我们进行了分子对接实验(如图13)。通过LeDock软件选取STAT3SH2结构域与阿托帕沙溴酸进行分子对接,蛋白数据库获取STAT3β的晶体结构。STAT3为绿色;阿托帕沙溴酸为红色;氢键为黄色。利用LeDock软件通过分子对接实验得到了阿托帕沙溴酸与STAT3(PDB ID:1BG1)激酶的SH2结构域的亲和力为:-5.8kcal/mol。此外,阿托帕沙溴酸与STAT3的Ser613、Ser636和Arg595氨基酸之间形成3对氢键,表明理论上其与STAT3有很强的亲和力。
实验14:验证阿托帕沙溴酸与JAK激酶JH1结构域具有很高的亲和力。
通过LeDock软件选取JAK激酶JH1结构域与阿托帕沙溴酸进行分子对接。JAK激酶JH1结构域为绿色;阿托帕沙溴酸为红色;氢键为黄色。我们进一步利用利用LeDock软件分析了阿托帕沙溴酸与JAK激酶JH1结构域之间的亲和力,以及形成氢键的情况。结果发现,阿托帕沙溴酸与JAK1(PDB ID:4I5C)激酶的JH1结构域的亲和力为:-9.24kcal/mol,与JAK1的Asp1003、Arg1007氨基酸之间形成两对氢键(如图14A);阿托帕沙溴酸与JAK2(PDB ID:5CF5)激酶的JH1结构域的亲和力为:-8.41kcal/mol,与JAK2的Asp939氨基酸之间形成1对氢键,与Ser936氨基酸之间形成2对氢键(如图14B);阿托帕沙溴酸与TYK2(PDB ID:4GJ2)激酶的JH1结构域亲和力为:-8.55kcal/mol,与TYK2的Arg901、V981氨基酸之间形成2对氢键(如图14C),从而使得阿托帕沙溴酸与JAK激酶JH1结构域的亲和力大大增加。
实验15:验证阿托帕沙溴酸可抑制小鼠体内肿瘤细胞的生长。
为了确认阿托帕沙溴酸是否能抑制体内肿瘤细胞的生长,建立了Bal b/c裸鼠A549人肺癌模型。将1×107个A549细胞皮下注射到Bal b/c小鼠中两周。然后解剖肿瘤并切成8mm3的小块,将这些瘤块植入Bal b/c小鼠体内。每4天测量肿瘤体积。50mg/kg/天吉非替尼作为阳性对照,0.9%盐水作为阴性对照。药物治疗3周后,结果显示,在5mg/kg/天和10mg/kg/天的组中阿托帕沙溴酸(AHB)抑制肿瘤体积(图15A所示)。阿托帕沙溴酸处理后小鼠体重没有显着变化,但吉非替尼处理后小鼠体重下降(图15B所示)。在5mg/kg/天和10mg/kg/天的组中,阿托帕沙溴酸分别使肿瘤重量抑制18%和26%(图15C所示)。肿瘤图片也证实了阿托帕沙溴酸的抑制作用(图3-15D)。
本发明研究方法所用试剂及耗材:毫克级阿托帕沙溴酸(纯度>95%)由陶素公司提供;荧光素酶底物Steady-Glo购自Promega公司;DMEM(高糖)、RPMI-1640及胎牛血清购自Hyclone Thermo公司;RNA反转录试剂盒购自Invitrogen公司;96孔板购自Corning公司;IL-6、IFNα和IFNγ购自Peprotech公司;一抗磷酸化STAT3Tyr705、Tyr1022/1023JAK1、Tyr1007/1008JAK2、Tyr1054/1055Tyk2、总蛋白STAT3、JAK1、JAK2和Tyk2,以及GAPDH和α-Tubulin的抗体均购自Cell Signaling Technology公司;二抗购自爱必信;PVDF膜、ECL显影液购自Millipore公司;Fugene 6试剂购自Roche公司;二抗(包括HRP羊抗鼠,HRP羊抗兔)购自爱必信;BCA蛋白定量试剂盒购自Thermo公司;Cyber Green PCR mix试剂盒,磷酸酶抑制剂和蛋白酶抑制剂Cocktail购自Roche公司;Steady-Glo荧光素酶底物(Promega公司);TRIZOL试剂(Gibco公司);5×蛋白上样缓冲液、氯仿、异丙醇、柠檬酸三钠、DEPC、Tris-HCl、Tween-20、NaCl、KCl、MgCl2、NP-40、BSA、甘油、PMSF、DTT、EDTA、EGTA等常规化学试剂,上海生工或者麦克林提供;细胞培养皿及常规实验耗材离心管等由上海生工提供。
Claims (9)
1.阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的用途。
2.根据权利要求1所述的阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的用途,其特征在于,阿托帕沙溴酸可用于特异性抑制STAT3活性。
3.根据权利要求1所述的阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的用途,其特征在于,阿托帕沙溴酸可用于抑制STAT3下游基因Bcl-XL、CyclinD1、c-Myc的表达来控制细胞增殖,具有长时程效应。
4.根据权利要求1所述的阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的用途,其特征在于,阿托帕沙溴酸对细胞因子IL-6、IFNα和IFNγ诱导的STAT3活化、可抑制细胞因子IL-6诱导的STAT1活化以及JAKs家族成员的自磷酸化具有抑制作用。
5.根据权利要求1所述的阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的用途,其特征在于,阿托帕沙溴酸可以剂量和时间依赖性方式抑制STAT3组成型活化的癌细胞活力。
6.根据权利要求1所述的阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的用途,其特征在于,阿托帕沙溴酸可结合于STAT3的SH2结构域并形成氢键,与JAK激酶JH1结构域具有很高的亲和力并形成氢键,是一种信号特异性抑制剂。
7.根据权利要求1所述的阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的用途,其特征在于,阿托帕沙溴酸可直接作用于STAT3信号启动,从而抑制荧光素酶活性。
8.阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的研究方法,其特征在于,包括如下步骤:S1:细胞培养;S2:蛋白提取;S3:Western-Blot分析;S4:RNA提取;S5:RT-PCR及荧光定量PCR;S6:荧光素酶活性分析;S7:细胞活性分析(MTT法);S8:流式细胞术检测细胞周期;S9:流式细胞术检测细胞凋亡;S10:裸鼠移植瘤实验;S11:计算机分子动力学模拟对接分析;S12:数据计算及处理。
9.根据权利要求8所述的阿托帕沙溴酸作为新型JAK-STAT3信号通路抑制剂的研究方法,其特征在于,S1-S12具体操作如下:
S1:细胞培养:hTERT-BJ、HELA、HepG2、A549和SKA细胞培养于DMEM高糖培养基,DU145和MDA-MB-231细胞培养于RPMI-1640培养基;
S2:蛋白提取:将培养皿中细胞刮下后收集好,离心后吸走上清使用裂解液裂解细胞,并转移到预冷的离心管中离心、取上清,进行BCA蛋白定量,样品加入5×蛋白上样缓冲液处理,随后进行Western-Blot分析;
S3:Western-Blot分析:配制8%~10%SDS-PAGE凝胶,电泳,然后将凝胶浸泡至预冷的转膜缓冲液中,甲醇处理PVDF膜,进行三明治转膜,取出转印好的PVDF膜,蛋白面向上,室温封闭,一抗配制4℃孵育过夜,TBST清洗5次,每次5min,二抗,TBST清洗PVDF膜4次,每次10min,加ECL显影曝光;
S4:RNA提取:细胞培养后弃培养基,用PBS清洗后,加TRIZOL试剂,摇匀,消化后依次收集至EP管中,加氯仿,轻摇,静置,离心,取上清无色水相到新的EP管中,加异丙醇,静置,离心,弃上清,用75%乙醇洗涤,离心,去上清,吸干液体,干燥至管底透明,加入DEPC处理水20~30μl,55~60℃水浴10min溶解总RNA,测OD值;
S5:RT-PCR及荧光定量PCR:根据罗氏逆转录试剂盒进行反转录,取1μg总RNA进行反应;
S6:荧光素酶活性分析:SKA细胞铺于96孔板(100μl)中,待细胞贴壁后,更换含有阿托帕沙溴酸梯度浓度的培养基处理,然后于每孔加入荧光素酶底物,测定荧光素酶的活性;
S7:细胞活性分析(MTT法):将一定数量的细胞铺于96孔板(100μl),然后加不同药物浓度,24h或者48h或者72h后测定,加入20μl 5mg/mlMTT溶液,培养箱培养3~4h,吸走孔内液体,加入DMSO完全溶解结晶,摇床轻晃,490nm测定吸光值;
S8:流式细胞术检测细胞周期:培养细胞于10%FBS的DMEM培养基中,生长至80%汇合度,取样品,用预冷的PBS洗涤,在预冷的70%乙醇中固定、孵育,用PBS洗涤,离心,弃去上清液,用核糖核酸酶处理细胞,加入Rnase储存液和PI储存液,室温避光染色,流式细胞术测定各组细胞周期的百分率;
S9:流式细胞术检测细胞凋亡:培养DU145细胞至70%汇合度,不同阿托帕沙溴酸梯度浓度处理,贴壁细胞和培养基中的飘浮细胞均低速离心收集,使用预冷的PBS清洗后,再分别加入PBS将细胞重悬,将细胞悬液,碘化丙啶和Annexin V-FITC混合到一起,避光反应,再分别加入Annexin V结合缓冲液,静置反应后尽快流式细胞仪检测;
S10:裸鼠移植瘤实验:将人源A549细胞瘤块种植于BALB/c裸鼠的左侧腋下,3天后开始给药,将它们随机分为空白对照组、阳性Gefitinib灌胃组、腹腔注射阿托帕沙溴酸低浓度组、以及腹腔注射阿托帕沙溴酸高浓度组,每天给药,定期用游标卡尺测量瘤体的长径和短径,并称量记录小鼠体重,观察记录小鼠异常的生理状况,给药若干天后处死,解剖剥离移植瘤,拍照,称重,利用公式V(mm3)=0.5×a(长径)×b(短径)2计算瘤体体积,绘制A549移植瘤生长曲线;
S11:计算机分子动力学模拟对接分析:JAK1和JAK2的结构来源蛋白质数据库,提取相应的激酶结构域,从PUBCHEM查到化合物阿托帕沙溴酸的3D结构,采用Avogadro软件进行结构优化,AutoDockTools软件分析分子结构,划定网格盒子并将此区域作为分子对接的口袋,PDB2PQR程序用于PH=7时的质子化,采用NAMD程序对添加氢进行优化,对接盒位于JH1域中;
S12:数据计算及处理:实验均经过重复验证,与对照组数据的比值表示荧光素酶活性和细胞活性用,数据之间的显著性使用单因素方差分析比较,使用SPSS19软件计算IC50(EC50)计算,用±SD表示标准差,使用Origin 8做柱状图及折线图。
