Abstract
本发明公布了一种基于胞外诱捕网原理制成的新型肿瘤细胞疫苗及其制备方法,在肿瘤细胞表面形成胞外诱捕网(Extracellular traps)结构的CpG寡聚脱氧核苷酸网,肿瘤细胞胞外诱捕网的主要成分为CpG寡聚脱氧核苷酸和组蛋白,使得肿瘤细胞成为胞外诱捕网化肿瘤细胞,经灭活即制成安全的肿瘤细胞疫苗。将本发明提供的肿瘤细胞疫苗接种于肿瘤模型小鼠,可以诱导肿瘤细胞产生有效的免疫反应,达到有效治疗肿瘤的作用,具有良好的临床转化前景。在实际应用中,可针对特定的肿瘤细胞制备成特异的胞外诱捕网化肿瘤细胞疫苗。本发明为转化应用于人类肿瘤的治疗应用提供了可行性及技术支持。

本发明涉及肿瘤疫苗及其制备技术,尤其涉及一种基于细胞诱捕网原理制成的新型肿瘤细胞疫苗及其制备方法。该肿瘤疫苗的主要特征为在肿瘤细胞表面包被细胞诱捕网的主要成分CpG寡聚脱氧核苷酸(CpG ODN,一种DNA)和组蛋白,将这种肿瘤细胞灭活后制备成为具有治疗肿瘤作用的新型疫苗。
背景技术
肿瘤全细胞疫苗是一种新的肿瘤疫苗模式。但是,由于肿瘤细胞是自身细胞,免疫系统对自身的肿瘤细胞未能引起有效的免疫反应,因此不能直接利用自身肿瘤细胞制备成疫苗。国内外已经有学者将肿瘤细胞进行改造或修饰,利用基因重组技术将可以增强免疫细胞活性和功能的细胞因子(比如粒细胞-巨噬细胞集落刺激因子、成纤维细胞激活蛋白、白细胞介素-2等一种或多种复合)转入肿瘤细胞来增强免疫反应。尽管已经有肿瘤细胞疫苗进入临床试验,但效果并不理想,仍然需要发明更好的技术或策略来制备新的肿瘤细胞疫苗。
胞外诱捕网(Extracellular Traps)的概念起源于中性粒细胞诱捕网(Neutrophil extracellular traps)。这种现象最早在中性粒细胞中发现,近来由于在单核细胞、巨噬细胞、嗜酸(碱)性粒细胞、NK细胞等免疫细胞中均有发现,并不只限于中性粒细胞,因此用“胞外诱捕网”来统称这一种现象。所谓胞外诱捕网,指的是处于死亡前状态的免疫细胞(上述提到的细胞种类)释放的一种纤维网状结构,主要成分为细胞核内物质,包括浓缩的DNA、组蛋白、颗粒蛋白等。这种网状结构能够将病原微生物甚至某些自身病变细胞包裹住,促使免疫细胞吞噬清除所包裹的病原微生物和/或细胞,在吞噬清除过程中也处理和递呈相应的抗原,进而激活和提高所吞噬微生物和/或细胞的特异性免疫反应。近来研究还发现,自身免疫性疾病病人标志性抗核抗体的产生和胞外诱捕网相关。但是,目前尚未出现利用胞外诱捕网机理或成分有效制备肿瘤细胞疫苗的技术方案。
发明内容
本发明的目的在于:提供一种基于胞外诱捕网原理制成的新型肿瘤细胞疫苗及其制备方法,该新型疫苗利用细胞诱捕网的主要成分DNA(本发明选用具有免疫佐剂功能的CpG ODN)和组蛋白,将它们包被于肿瘤细胞的表面,使肿瘤细胞表面形成一个胞外诱捕网(Extracellular traps)类似结构;本发明将CpG ODN和组蛋白包被于肿瘤细胞表面后,将这种肿瘤细胞称为“胞外诱捕网化肿瘤细胞。具体实施表明,将本发明的胞外诱捕网化肿瘤细胞作为疫苗接种于肿瘤模型小鼠皮下(也可通过血管、肌肉和腹腔内注射等途径注射),可以诱导肿瘤细胞产生有效的免疫反应,达到有效治疗肿瘤的作用,具有良好的临床转化前景。
本发明的原理:目前研究发现,自身免疫性疾病病人标志性抗核抗体的产生和胞外诱捕网相关,说明胞外诱捕网可以诱导免疫系统产生针对所包裹细胞的特异性免疫反应。Toll样受体(TLRs)是先天和后天免疫的桥梁。CpG寡聚脱氧核苷酸(CpG ODN)是一种特殊的DNA和疫苗佐剂,也是Toll样受体9(TLR9)的配体,在病原体感染阶段,它能被树突状细胞(DCs)等抗原呈递细胞上的TLR9识别,促进DCs的成熟;成熟的DCs通过分泌INF-α等细胞因子促进CD8+T细胞、NK和Th1细胞的免疫反应。因此,CpG ODN作为免疫佐剂已经应用于临床和多种开发中的疫苗。肿瘤表面表达某些免疫检查点分子(比如PD-L1、CD47),它们能被免疫细胞上的分子(比如PD-1和SIRPα)识别并促使免疫细胞对肿瘤细胞失去反应。本发明基于胞外诱捕网的原理,利用肿瘤细胞表面和CpG ODN均带有负电荷的特性,提出一种将DNA(CpG ODN)和组蛋白包被于肿瘤细胞表面,在肿瘤细胞表面形成一个胞外诱捕网结构的CpG ODN网,制备成胞外诱捕网化肿瘤细胞,再将这种胞外诱捕网化肿瘤细胞灭活后即可制备成为具有治疗肿瘤作用的新型疫苗。上述疫苗由CpG ODN、组蛋白和肿瘤细胞三种成分组成。通过这种设计,免疫细胞就有可能通过以下两种途径诱导产生肿瘤细胞特异的免疫反应:(1)肿瘤细胞表面的CpG ODN是一种危险信号,可以通过TLR9信号通路诱导特异的免疫反应;(2)包被肿瘤细胞的CpG ODN网有可能覆盖住细胞表面的PD-L1、CD47等检查点分子,从而阻断PD-1/PD-L1和CD47/SIRPα的信号通路,也可促进免疫反应。将这种胞外诱捕网化肿瘤细胞灭活后作为肿瘤疫苗注射肿瘤模型小鼠,能诱导免疫系统产生肿瘤细胞特异的免疫反应,具有很好的治疗肿瘤作用。本发明通过“胞外诱捕网化肿瘤细胞”来制备肿瘤疫苗,这是一种新的制备肿瘤疫苗的技术方法,具有临床转化应用的前景。
本发明提供的技术方案是:
一种基于胞外诱捕网制备的新型肿瘤细胞疫苗,疫苗的成分包括DNA(如CpG寡聚脱氧核苷酸(CpG ODN))、组蛋白和肿瘤细胞;
其中的DNA和组蛋白是构成细胞诱捕网的主要成分;具体实施时,DNA选用CpGODN;CpG ODN和组蛋白包被于肿瘤细胞表面,成为胞外诱捕网化肿瘤细胞;具体地,肿瘤细胞膜骨架和CpG ODN主要成分均有磷酸,肿瘤细胞表面和CpG ODN均带有负电荷;组蛋白含大量碱性的精氨酸和赖氨酸而带有正电荷;通过组蛋白的桥梁作用,带负电荷的CpG ODN联结到同样带负电荷的肿瘤细胞表面,由此成为本发明提供的胞外诱捕网化肿瘤细胞;
其中的肿瘤细胞可以从肿瘤患者手术后(离体)的肿瘤组织进行分离提取,从而制成肿瘤患者个体化的肿瘤细胞疫苗。
本发明将细胞诱捕网的主要成分DNA(CpG ODN)和组蛋白包被于肿瘤细胞表面,图1为制备胞外诱捕网化肿瘤细胞的主要原理和方法步骤。如图1所示,肿瘤细胞表面和CpGODN都带有负电荷;组蛋白则带有正电荷。通过组蛋白的桥梁作用,将带负电荷的CpG ODN联结到同样带负电荷的肿瘤细胞表面,即成为本发明提供的胞外诱捕网化肿瘤细胞。
本发明提供了上述基于胞外诱捕网原理制成的新型肿瘤细胞疫苗及其制备方法,包括以下步骤:
1)培养肿瘤细胞,达到需要的细胞数量后收集对数生长期的细胞,计数后用磷酸盐缓冲溶液(PBS溶液)将肿瘤细胞数量调整至1×106个细胞/100μL。也可以用生理盐水等和人体体液渗透压相同或相近的溶液替换PBS溶液。
2)用PBS将CpG ODN与组蛋白按质量比1:0.5~10混匀,然后加入肿瘤细胞(按CpGODN1μg加入103~104个肿瘤细胞的比例),在室温下摇动10~20分钟。
3)离心去除上清,即得到胞外诱捕网化肿瘤细胞;
可用PBS溶液清洗沉淀的胞外诱捕网化肿瘤细胞二次后,再次用PBS溶液将胞外诱捕网化肿瘤细胞数量调整至适当浓度(如1×106个细胞/100μL),调整的浓度大小可视注射疫苗所需要的浓度而定。
4)将制备得到的胞外诱捕网化肿瘤细胞放置于X线辐照仪(具体实施时采用RadSource RS2000)中,按辐射强度50~100Gy对胞外诱捕网化肿瘤细胞进行灭活,使胞外诱捕网化肿瘤细胞当作疫苗注射后不再继续生长、增殖和转移,从而达到安全的目的。
具体实施时,将灭活的胞外诱捕网化肿瘤细胞作为疫苗给小鼠注射,每只小鼠每次注射3~5×105个胞外诱捕网化肿瘤细胞(注射时调整到需要的浓度,最好将胞外诱捕网化肿瘤细胞溶于80~100μL PBS中),注射途径可为皮下、肌肉、血管、腹腔内等等。
较佳地,步骤2)具体为:先将组蛋白溶于PBS溶液,然后用1M氢氧化钠溶液将其调整至PH值为7~9,使溶于PBS溶液中的组蛋白变得澄清透明后再加入CpG ODN和肿瘤细胞,整个过程最好在4℃环境并在摇床(摇动幅度不宜过大)上进行,使用的PBS温度最好也为4℃。
较佳地,步骤2)具体应注意:组蛋白和CpG ODN的量要保证适当的比例(按质量宜为1:0.3~0.7),使得CpG ODN与组蛋白结合后组蛋白还保留部分正电荷用于与肿瘤细胞上的负电荷结合。具体实施时,可将组蛋白溶于PBS溶液,然后用1M氢氧化钠溶液将其调整至PH值为7~9,使溶于PBS溶液中的组蛋白变得澄清透明。此后按组蛋白1mg混合103~104个肿瘤细胞的比例先和肿瘤细胞混合10~20分钟,然后才加进CpG ODN。
较佳地,步骤(3)在清洗肿瘤细胞时:离心力不宜过大,以500~800g为宜,PBS清洗过程动作也不宜过大,轻轻转动离心管3~5周就可。
较佳地,步骤(4)具体应注意:在进行X线辐射灭活肿瘤细胞时,最好用PBS将肿瘤细胞数量调整至注射需要的浓度,本发明一般将肿瘤细胞数量浓度调至3~5×105个/100μL(取上述浓度为1×106个细胞/100μL的胞外诱捕网化肿瘤细胞溶液50μL再加PBS 50μL),给小鼠注射时每只小鼠注射100μL时所注射的胞外诱捕网化肿瘤细胞数量则为5×106个,其它数量可依最终注射的细胞数量进行适当调整,最好最终注射体积为80~100μL。
本发明的有益效果是:
本发明通过胞外诱捕网化肿瘤细胞制备肿瘤细胞疫苗的技术是一种创新的制备肿瘤疫苗的方法,具有临床转化应用的前景。具体实施表明,将本发明的胞外诱捕网化肿瘤细胞经灭活后作为疫苗接种于肿瘤模型小鼠的皮下或通过血管、肌肉和腹腔内注射等途径注射,可以诱导肿瘤细胞产生有效的免疫反应,达到有效治疗肿瘤的作用,具有良好的临床转化前景。在实际应用中,可针对特定的肿瘤细胞(如从肿瘤患者肿瘤组织中获得肿瘤细胞后),用本发明的方法制备成相应肿瘤细胞(如患者肿瘤细胞)特异的胞外诱捕网化肿瘤细胞疫苗。本发明为转化应用于人类肿瘤的治疗应用提供了可行性及技术支持。

具体实施方式
下面结合实施例对本发明作进一步的说明,但本发明不限于下述实施例,利用任何细胞诱捕网成分和原理方法制备的肿瘤细胞疫苗都应属于本专利方法的范围。
图1为胞外诱捕网化肿瘤细胞的主要原理和步骤,如图1所示,肿瘤细胞膜骨架和CpG ODN主要成分均有磷酸,肿瘤细胞表面和CpG ODN均带有负电荷;组蛋白含大量碱性的精氨酸和赖氨酸而带有正电荷;将带正电荷的组蛋白和带负电荷的CpG ODN结合后,通过组蛋白的桥梁作用,带负电荷的CpG ODN联结到同样带负电荷的肿瘤细胞表面,由此成为本发明提供的胞外诱捕网化肿瘤细胞,将此细胞灭活后即可作为肿瘤疫苗。
具体地,CpG寡聚脱氧核苷酸和组蛋白包被于肿瘤细胞表面,在肿瘤细胞表面形成一种类似胞外诱捕网的结构;胞外诱捕网是免疫细胞(比如中性粒细胞、巨噬细胞)死亡前形成的一种特殊结构,这些细胞在临死前利用胞内DNA(CpG寡聚脱氧核苷酸是一种特殊形式的DNA)、组蛋白等成分形成一个网状结构,这种网状结构可以将细菌、寄生虫、病毒甚至病变细胞(包括肿瘤细胞)等致病物质(对象)包围,然后促使其它免疫细胞清除这些被包围的物质,进而引起免疫反应。本发明将这种包被了CpG寡聚脱氧核苷酸和组蛋白的肿瘤细胞称为“胞外诱捕网化肿瘤细胞”。
一、培养和收集肿瘤细胞
本发明具体实施例采用小鼠来源的结肠癌CT26细胞,来源于美国模式培养物集存库(ATCC),由发明人实验室保存于液氮环境中。PBS(成分为:磷酸二氢钾0.27g,磷酸氢二钠1.42g,氯化钠8g,氯化钾0.2g,加超纯水至1 L,PH值7.4))由发明人配制并经高温清毒后使用。
培养前,将CT26细胞从液氮罐中取出,迅速放入37℃水浴锅中,反复摇晃至完全溶解,大约需要1~2分钟。此后离心(2000rpm)5分钟,去上清,加入培养液RPMI 1640、胎牛血清、抗生素、生长因子等(具体根据细胞商品说明书中推荐的方法进行),混匀后加入细胞培养瓶中,置于37℃、5%CO2细胞培养箱中培养,然后每隔1~2天更换细胞培养液。待细胞生长到培养瓶80%左右弃去培养液,PBS溶液洗两遍,加入0.25%胰酶消化肿瘤细胞,然后换瓶继续扩增培养(以便达到所需要的细胞数量),待细胞生长到所需要的数量时,再用PBS溶液洗两遍,加入0.25%胰酶消化肿瘤细胞,然后离心(2000rpm)5分钟,去除上清,PBS溶液洗两次并计数,计数后用PBS溶液将肿瘤细胞数量调整至1×106个细胞/100μL,供后续胞外诱捕网化操作使用。
二、制备胞外诱捕网化肿瘤细胞
制备胞外诱捕网化肿瘤细胞所用的CpG ODN购自Invitrogen上海公司,组蛋白购自Sangon Biotech上海公司。
实施例一
(1)将1mg组蛋白倒入PBS 1.8mL中,摇动使组蛋白完全溶解,如果溶液出现混浊不清则用1M氢氧化钠调整至PH值为9,此时溶液澄清透明,此后加进PBS使总体积成为2mL;接着,将CpG ODN 10mg倒入调定好的组蛋白溶液并将其摇匀,此后加入2×107个CT26肿瘤细胞,在室温下摇动20分钟。
(2)离心(3000rpm,5min)去除上清(实为去除掉不和CT26肿瘤细胞结合的组蛋白/CpG ODN复合物),用PBS清洗肿瘤细胞二次后再次用PBS将细胞数量调整至1×105个细胞/100μL,此时的细胞即为胞外诱捕网化的CT26肿瘤细胞。
实施例二:
(1)将10mg组蛋白倒入PBS 1.8mL中,摇动使组蛋白完全溶解,如果溶液出现混浊不清则用1M氢氧化钠调整至PH值为9,此时溶液澄清透明,此后加进PBS使总体积成为2mL;接着加入2×107个CT26肿瘤细胞,在室温下摇动10~20分钟。
(2)离心(3000rpm,5min)去上清(实为去除掉不和肿瘤细胞结合的游离组蛋白),用PBS清洗沉淀的肿瘤细胞二次后溶于PBS 2mL中,此后再将1.5mg CpG ODN加进溶液中,在室温下摇动10~20分钟,使CpG ODN完全与组蛋白上所剩余(因为部分已经和CT26肿瘤细胞表面的负电荷结合)的正电荷结合。
(3)离心去除上清(实为去除掉不和肿瘤细胞结合的游离CpG ODN),用PBS清洗沉淀的肿瘤细胞二次后再次用PBS将细胞数量调整至1×105个细胞/100μL,此时的细胞也为胞外诱捕网化的肿瘤细胞。
三、检测肿瘤细胞是否成功制成为胞外诱捕网化肿瘤细胞
为了证明是否成制备以上胸外诱捕网化肿瘤细胞,在以上制备过程中采用可发出绿色荧光的异硫氰酸荧光素(FITC)标记的CpG。取上述制成的胞外诱捕网化肿瘤细胞100μL放置于共聚焦显微镜(本发明使用奥林巴斯FV100)的专用检测平皿中,然后观察所检测的细胞是否有发出绿色荧光。检测前因为同时用DAPI对细胞核进行染色(染成蓝色),因此观察过程中在同一个视野下同时观察蓝色荧光图像,然后将绿色和蓝色荧光图像重叠,以判断绿色荧光(CpG)和蓝色荧光(细胞核)的关系。进行染色并用共聚焦显微镜进行观察,因为DAPI能够将所有DNA染成蓝色,正功制备的胞外诱捕网化的CT26肿瘤细胞表面上应有绿色荧光,这则绿色荧光应该在细胞核(蓝色荧光)外围。图2A为绿色荧光检测结果,图2B为蓝色荧光检测结果,图2C为二者的重叠,两种信号关系和以上设想结查一致,说明本发明成功制备了胞外诱捕网化CT26肿瘤细胞。
四、灭活胞外诱捕网化肿瘤细胞制成疫苗
(1)将以上制备得到的胞外诱捕网化CT26肿瘤细胞放置于X线辐照仪(具体实施时采用Rad Source RS2000)中,按辐射强度50~100Gy对胞外诱捕网化肿瘤细胞进行灭活,使胞外诱捕网化肿瘤细胞当作疫苗注射后不再继续生长、增殖和转移,从而达到安全的目的。
(2)然后将灭活的胞外诱捕网化CT26肿瘤细胞作为疫苗给小鼠注射,每只小鼠每次注射3~5×105个胞外诱捕网化肿瘤细胞,注射途径可为皮下、肌肉、血管、腹腔内等等。
五、在CT26负荷小鼠中观察胞外诱捕网化肿瘤细胞疫苗的治疗效果
按预防程序进行验证。选用6-8周龄雌性BALB/c小鼠10只,先在背部右上测皮下注射以上灭活的细菌化CT26结肠癌细胞5×105个(溶于100 100μL PBS中,相当于先注射接种疫苗),一周后在背部左下侧皮下注射CT26小鼠结肠癌细胞2×106建立肿瘤模型(相当于生成肿瘤),然后观察小鼠肿瘤的生长状况,在注射肿瘤细胞28天后断颈处死小鼠,取血清检测CT26结肠癌细胞特异性抗体,取脾脏淋巴细胞作为效应细胞对CT26结肠癌细胞进行杀伤实验。结果发现:疫苗治疗组5只小鼠中,4只小鼠肿瘤几乎完全没有生长(附图3A),另外1只小鼠生长的肿瘤明显小于对照组肿瘤(附图3B),疫苗注射组小鼠血清中有CT26结肠癌细胞特异的抗体,同时来源于疫苗治疗组的淋巴细胞对CT26结肠癌细胞也具有明显的杀伤作用。这些结果说明胞外诱捕网化CT26肿瘤细胞疫苗可以有效诱导CT26结肠癌细胞特异的免疫反应,可以有效抑制小鼠结肠癌肿瘤的生长。
以上所描述的仅为本发明的较佳实施例子,不能以此来限定本发明之权利范围,因此依本发明权利要求使用细胞诱捕网原理或成分制成的疫苗仍属于本发明所涵盖的范围。
Claims (9)
1.一种肿瘤细胞疫苗,其特征是:所述肿瘤细胞疫苗的成分包括DNA、组蛋白和肿瘤细胞;
所述DNA为CpG寡聚脱氧核苷酸;
所述肿瘤细胞从任何离体的肿瘤组织进行分离提取;
所述CpG寡聚脱氧核苷酸和组蛋白包被于肿瘤细胞表面,在肿瘤细胞表面形成胞外诱捕网结构的CpG寡聚脱氧核苷酸网,主要成分为CpG寡聚脱氧核苷酸和组蛋白,由此使得肿瘤细胞成为胞外诱捕网化肿瘤细胞;
胞外诱捕网化肿瘤细胞经灭活即为安全的肿瘤细胞疫苗。
2.如权利要求1所述肿瘤细胞疫苗,其特征是,所述肿瘤细胞的细胞膜骨架和CpG寡聚脱氧核苷酸均含有磷酸,肿瘤细胞表面和CpG寡聚脱氧核苷酸均带有负电荷;组蛋白含碱性的精氨酸和赖氨酸,组蛋白带有正电荷;CpG寡聚脱氧核苷酸通过组蛋白联结到肿瘤细胞表面,由此使得CpG寡聚脱氧核苷酸和组蛋白包被于肿瘤细胞表面,成为胞外诱捕网化肿瘤细胞。
3.如权利要求1所述肿瘤细胞疫苗,其特征是,所述肿瘤细胞为结肠癌CT26细胞。
4.一种基于胞外诱捕网制备权利要求1所述肿瘤细胞疫苗的方法,其特征是,在肿瘤细胞表面形成胞外诱捕网结构的CpG寡聚脱氧核苷酸网,使得肿瘤细胞成为胞外诱捕网化肿瘤细胞,经灭活即制成安全的肿瘤细胞疫苗;包括如下步骤:
1)培养肿瘤细胞,收集对数生长期的细胞并计数;然后用生理盐水或磷酸盐缓冲溶液调整肿瘤细胞的数量浓度;
2)用生理盐水或磷酸盐缓冲溶液将CpG寡聚脱氧核苷酸与组蛋白按质量比1:0.5~10混匀,然后按1μg的CpG寡聚脱氧核苷酸与103~104个肿瘤细胞的比例加入肿瘤细胞,在室温下轻摇得到的混合液;
3)将步骤2)得到的混合液进行离心去除上清,通过清洗肿瘤细胞除掉不与肿瘤细胞结合的游离的CpG寡聚脱氧核苷酸,即得到胞外诱捕网化肿瘤细胞;
4)将制备得到的胞外诱捕网化肿瘤细胞进行灭活,使胞外诱捕网化肿瘤细胞当作疫苗注射后不再继续生长、增殖和转移,从而得到安全的肿瘤细胞疫苗。
5.如权利要求4所述的方法,其特征是,所述肿瘤细胞从肿瘤患者的离体肿瘤组织进行分离提取得到,制成的肿瘤细胞疫苗即为肿瘤患者个体化的肿瘤细胞疫苗。
6.如权利要求4所述的方法,其特征是,步骤2)具体包括:
先将组蛋白溶于生理盐水或磷酸盐缓冲溶液;
然后用1M氢氧化钠溶液将其调整至PH值为7~9,使溶液中的组蛋白变得澄清透明后再加入CpG寡聚脱氧核苷酸和肿瘤细胞,整个过程在4℃环境并在摇床上进行;
所述组蛋白和CpG寡聚脱氧核苷酸的量的比例应使得CpG寡聚脱氧核苷酸与组蛋白结合后组蛋白还保留部分正电荷用于与肿瘤细胞上的负电荷结合。
7.如权利要求6所述的方法,其特征是,按1mg组蛋白混合103~104个肿瘤细胞的比例,先将组蛋白和肿瘤细胞混合10~20分钟,然后再加入CpG寡聚脱氧核苷酸。
8.如权利要求4所述的方法,其特征是,步骤3)在进行离心清洗肿瘤细胞时,离心力为500~800g,轻轻转动离心管3~5周。
9.如权利要求4所述的方法,其特征是,步骤4)在进行X线辐射灭活肿瘤细胞制成安全的肿瘤细胞疫苗时,用磷酸盐缓冲溶液将肿瘤细胞数量调整至疫苗注射需要的浓度。
