Abstract
Description
(a)細胞核抽出試薬(Solarbio)を用いてリンパ腫細胞E.G7−OVA(ATCC CELL BANK)由来の細胞核を抽出する。
E.G7−OVA細胞を10%ウシ胎児血清タンパク質(Gibco社)、1%ペニシリン−ストレプトマイシン(Gibco社)、0.4mg/mLG418(Gibco社)および0.05mMβ−メルカプトエタノール(Gibco社)を含む高グルコース1640培地(Gibco社)に培養する。1:5の継代比率で2日ごとに継代する。遠心分離し、対数増殖期の腫瘍細胞を収集し、PBS(Gibco社)で再懸濁させ、2回洗浄する。細胞核提取キット(Solarbio社)におけるLysis bufferを用いて細胞を沈殿して再懸濁させた後、細胞密度は約2×107/mLである。その後、1mlあたりの細胞懸濁液に20μL Reagent A(Solarbio社)を加え、ピペットで繰り返して吹きかけて均一に混合し、200メッシュの篩に掛けて綿状物を除去する。篩掛けた後の懸濁液700gを5minで遠心分離し、下層の沈殿物は細胞核である。
(a)細胞核抽出試薬(Solarbio)を用いて黒色腫細胞Muc−B16(ATCC CELL BANK)由来の細胞核を抽出する。
Muc1−B16細胞を接着培養し、完全培地は、ウシ胎児血清タンパク質(Gibco社)、1%ペニシリン−ストレプトマイシン(Gibco社)、1μg/mLピューロマイシン(Gibco社)を含む1640培地(Gibco社,HEPESを含まない)である。1:4の比率で2〜3日ごとに継代する。対数期の細胞を選択し、遠心分離して収集し、PBS(Gibco社)で再懸濁させ、2回洗浄する。2×107個の細胞を1mL細胞核抽出キット(Solarbio社)におけるLysis bufferで再懸濁させ、その後、1mlあたりの細胞懸濁液に20μL Reagent A(Solarbio社)を加え、1mLピペットで10回吹きかけ、細胞懸濁液が透明になった後、直ちに10mL PBS(Gibco社)を加えて溶解液を希釈し、1400r/minで5min遠心分離し、下層の沈殿物を再懸濁させ、篩掛けして綿状物を除去し、分散性が良好なMuc1−B16細胞核懸濁液を得る。

4T1細胞を接着培養し、完全培地は、10%ウシ胎児血清タンパク質(Gibco社)、1%ペニシリン−ストレプトマイシン(Gibco社)を含む1640培地(Gibco社,HEPESを含まない)である。1:4の比率で2〜3日ごとに継代する。対数期の細胞を選択し、遠心分離して収集し、PBS(Gibco社)で再懸濁させ、2回洗浄する。2×107個の細胞を1mL細胞核抽出キット(Solarbio社)におけるLysis bufferで再懸濁させ、その後、1mlあたりの細胞懸濁液に20μL Reagent A(Solarbio社)を加え、1mLピペットで10回吹きかけ、細胞懸濁液が透明になった後、直ちに10mL PBS(Gibco社)を加えて溶解液を希釈し、1400r/minで5min遠心分離し、下層の沈殿物を再懸濁させ、篩掛けして綿状物を除去し、分散性が良好な4T1細胞核懸濁液を得る。

A375細胞を接着培養し、完全培地は、ウシ胎児血清タンパク質(Gibco社)、1%ペニシリン−ストレプトマイシン(Gibco社)を含む的DMEM培地(Gibco社,HEPESを含まない)である。1:4の比率で2〜3日ごとに継代する。対数期の細胞を選択し、遠心分離して収集し、PBS(Gibco社)で再懸濁させ、2回洗浄する。2×107個の細胞を1mL細胞核抽出キット(Solarbio社)におけるLysis bufferで再懸濁させ、その後、1mlあたりの細胞懸濁液に20μL Reagent A(Solarbio社)を加え、1mLピペットで10回吹きかけ、細胞懸濁液が透明になった後、直ちに10mL PBS(Gibco社)を加えて溶解液を希釈し、1400r/minで5min遠心分離し、下層の沈殿物を再懸濁させ、篩掛けして綿状物を除去し、分散性が良好なA375細胞核懸濁液を得る。
実施例1における蛍光染料で染色されていない雑種細胞を培養し、腫瘍細胞核を加え、4日後、雑種細胞の培養液上清を収集し、300gで10分間遠心分離し、残った細胞沈殿を除去し、上清を取り出して引き続き2000gで10分間遠心分離し、死細胞沈殿を除去し、上清を取り出して引き続き10000gで30分間遠心分離し、細胞破片沈殿を除去し、最後に上清を取り出し、120000gで2時間遠心分離し、上清を捨て、沈殿物をPBS(Gibco社)で再懸濁させ、エクソソーム懸濁液を得る。
超遠心分離により抽出された新鮮な実施例5のエクソソームと等量の4%のパラホルムアルデヒド溶液とを混合し、つまり、エクソソームを2%のパラホルムアルデヒド溶液に懸濁させ、30min固定する。その後、10μLエクソソーム懸濁液を密封膜に滴下し、メッキ層の面が液滴に接触するように銅メッシュを液滴上に被覆し、室温下で銅メッシュ膜で10min吸着する。その後、ピンセットで銅メッシュを慎重に取り出し、濾紙で銅メッシュに残った液滴を除去し、事前に密封膜に滴下したPBS液滴上で2回洗浄する(5分間/回)。その後、銅メッシュを取り外し、濾紙で銅メッシュに残った液体を除去する。その後、銅メッシュを50μLの酢酸ウラニル液滴上に移し、30s対比染色した後、銅メッシュを取り外し、濾紙で銅メッシュ上に残った酢酸ウラニル液体を吸い取って乾燥させ、銅メッシュのメッキ層面を上に向け、空気中で10min乾燥させる。その後、120kV生物学的透過型電子顕微鏡下観察し、結果を図2に示す。超遠心分離で抽出されたエクソソーム原液をPBSで500−2000倍希釈し、注射器で1mLエクソソーム希釈液をナノ粒子トラッキングアナライザーの試料注入口に注入し、システムで検出して分析し、希釈液中のエクソソームの数を計算し、エクソソーム粒度分布図(図3)を得る。Wes全自動タンパク質発現分析システムによりエクソソーム内の各タンパク質を検出する。まず、ビオチン化した標準サンプルladderを調製し、タンパク質濃度が同じであるエクソソームと1/5体積のFluorescent Master Mixとを均一に混合し、サンプルを調製する。Ladderとサンプルを95℃で5min加熱し、タンパク質を変性させる。その後、それぞれマイクロプレート内の対応するサンプルウェルに加え、マイクロプレートの一次抗体ウェルにCD9、CD86、MHC IおよびMHC IIの一次抗体希釈液を加え、下の行のウェルに異なる一次抗体の由来源に対応するHRP−二次抗体希釈液を加え、最も下の行のウェルにLuminol−Peroxide混合液を加える。その後、装置にキャピラリーインジェクターを取り付け、マイクロプレートをインジェクターの下に置き、装置を起動して検出分析を実行し、実験結果を図4に示す。
実施例2の雑種細胞に終濃度1μg/mLのリポ多糖(Sigma社)で刺激し、4日後、雑種細胞の培養液の上清を収集し、300gで10分間遠心分離し、残りの細胞沈殿を除去し、上清を取り出し、引き続き2000gで10分間遠心分離し、死細胞沈殿を除去し、上清を取り出して引き続き10000gで30分間遠心分離し、細胞破片沈殿を除去し、最後に上清を取り出し、120000gで2時間遠心分離し、上清を除去し、沈殿をPBS(Gibco社)で再懸濁させ、本発明のエクソソーム懸濁液を得る。
実施例3の雑種細胞に終濃度1μg/mLのリポ多糖(Sigma社)で刺激し、4日後、雑種細胞の培養液の上清を収集し、300gで10分間遠心分離し、残りの細胞沈殿を除去し、上清を取り出し、引き続き2000gで10分間遠心分離し、死細胞沈殿を除去し、上清を取り出して引き続き10000gで30分間遠心分離し、細胞破片沈殿を除去し、最後に上清を取り出し、120000gで2時間遠心分離し、上清を除去し、沈殿をPBS(Gibco社)で再懸濁させ、本発明のエクソソーム懸濁液を得る。
実施例4の雑種細胞に終濃度1μg/mLのリポ多糖(Sigma社)で刺激し、4日後、雑種細胞の培養液の上清を収集し、300gで10分間遠心分離し、残りの細胞沈殿を除去し、上清を取り出し、引き続き2000gで10分間遠心分離し、死細胞沈殿を除去し、上清を取り出して引き続き10000gで30分間遠心分離し、細胞破片沈殿を除去し、最後に上清を取り出し、120000gで2時間遠心分離し、上清を除去し、沈殿をPBS(Gibco社)で再懸濁させ、本発明の本発明のヒトエクソソーム(h−nc−lps−exo)懸濁液を得る。
超遠心分離により抽出された新鮮な実施例9のヒトエクソソームと等量の4%のパラホルムアルデヒド溶液とを混合し、つまり、エクソソームを2%のパラホルムアルデヒド溶液に懸濁させ、30min固定する。その後、10μLエクソソーム懸濁液を密封膜に滴下し、メッキ層の面が液滴に接触するように銅メッシュを液滴上に被覆し、室温下で銅メッシュ膜で10min吸着する。その後、ピンセットで銅メッシュを慎重に取り出し、濾紙で銅メッシュに残った液滴を除去し、事前に密封膜に滴下したPBS液滴上で2回洗浄する(5分間/回)。その後、銅メッシュを取り外し、濾紙で銅メッシュに残った液体を除去する。その後、銅メッシュを50μLの酢酸ウラニル液滴上に移し、30s対比染色した後、銅メッシュを取り外し、濾紙で銅メッシュ上に残った酢酸ウラニル液体を吸い取って乾燥させ、銅メッシュのメッキ層面を上に向け、空気中で10min乾燥させる。その後、120kV生物学的透過型電子顕微鏡下観察し、結果を図5に示す。
1μM近赤外蛍光親油性染料DiR(北京泛博生物化学有限公司)を用いて100μLエクソソームを標識し、37℃で1hインキュベートし、その後、実施例5で調製された本発明のエクソソームをゲル濾過カラム(Thermo Scientific社)の上側に滴下し、700gで2min遠心分離したところ、遊離染料はゲルカラムの内部に遮断され、下層液体はDiR標識雑種エクソソーム溶液である。3.7×109個のDiR−エクソソームを皮下注射によりC57BL/6マウス(維通利華実験動物技術有限公司)の背部に注射し、次いで、96時間内の異なる時点でマウスを麻酔し、小動物ライブイメージングシステムにより750nmでDiR−雑種エクソソームのマウス体内における分布状況を観察し、異なる時点の蛍光信号のリンパ節および腫瘍部位での変化状況を分析する。実験結果を図7および図8に示す。また、48h後にマウスを解剖し、それぞれ鼠径部および腋下のリンパ節、腫瘍、心臓、肝臓、脾臓、肺臓、腎臓を取り出し、小動物ライブイメージングシステムにより蛍光信号を検出し、実験結果を図9に示す。
実施例5で調製された本発明のエクソソームを実施例11の方法により蛍光標識し、DiR標識エクソソームを得る。3.7×109個のDiR−エクソソームを皮下注射によりC57BL/6マウス(維通利華実験動物技術有限公司)の背部に注射し、48hの時点でマウスを安楽死させ、リンパ節を取り出して研磨して単細胞懸濁液を得る。その後、それぞれ0.25μgのPacific Blue標識anti−F4/80、0.25μgのAlex Fluro700標識anti−CD11b、0.25μgのAlex Fluro 488標識anti−CD11c、0.25μgのBV605標識anti−MHC II、0.25μgのBV510標識anti−CD3、0.25μgのAPC標識anti−CD4、0.25μgのPE標識anti−CD8蛍光抗体(いずれもebioscience社)で単細胞懸濁液を暗所(4℃)で25分間標識する。500μL Stain buffer(ebioscience社)で細胞を2回洗浄した後、細胞を再懸濁させ、フローサイトメーターにより検出し、DiR信号が陽性である細胞集団におけるマクロファージ(F4/80+CD11b+)、樹状細胞(CD11c+MHC II+)、CD4 T細胞(CD3+CD4+)、CD8 T細胞(CD3+CD8+)の4種類の細胞タイプが占める比率を分析し、各サンプルについては500000個の細胞を収集して分析し、結果を図10に示す。
カルボキシフルオレセインジアセテートスクシンイミジルエステル(CFSE)蛍光標識方法によりT細胞の増殖状況を調査する。基本的な原理は以下のとおりである。CFSEは、生細胞を蛍光標識することができる細胞染色剤である。CFSEは、細胞に入った後、不可逆的に細胞内のアミノ基に結合して細胞タンパク質にカップリングし、細胞分裂増殖の過程において、CFSE標識蛍光は2つの娘細胞に均等に分配されるため、その蛍光強度は母細胞の半分である。フローサイトメーターにより488nmの励起光で分析し、リンパ球の増殖状況を正確に検出することができる。
実施例9のヒトエクソソームとCFSE染色ヒトT細胞とを共培養し、ヒトT細胞増殖効果を検出する。具体的な実験手順は以下のとおりである。1、ヒト末梢血T細胞染色:健康ヒト末梢血を採取し、ヒトリンパ球分離液を用いて末梢血からリンパ球を分離し、さらにPan T選別磁気ビーズ(Miltenyi社)を用いて説明書に従って操作し、ヒトT細胞の分離を行い、その後、0.5μMのCFSE(Life Technologies社)作業濃度で細胞を7min(37℃)染色し、4倍体積の氷ウシ胎児血清で染色を停止させ、氷浴で5min処理した後、遠心分離する。新鮮な培地で細胞を再懸濁させ、2回洗浄する。2、96ウェルプレート前処理:96ウェルプレートを事前に1μg/mLのヒトCD3抗体溶液(ebioscience社)で前処理し、4℃で一晩静置し、翌日にCD3抗体溶液を吸って取り除き、PBS(Gibco社)で2回洗浄した後、保存する。3.エクソソームとT細胞の共培養:CFSE−T細胞を96ウェルプレートに接種し、それぞれ異なる用量のヒト雑種エクソソームを加えて共培養し、一定の時間後、細胞を収集する。4、染色およびフローサイトメトリー:SuperBright600標識anti−CD3、APC標識anti−CD4、AF700標識anti−CD8抗体(いずれもebioscience社)を用いてヒトT細胞を染色し、フローサイトメーターによりCD3+CD8+T細胞、CD3+CD4+T細胞の分裂増殖状況を検出する。各サンプルについては、10000個の細胞を収集して分析する。結果を図14に示す。
実施例9のヒトエクソソームとヒトT細胞とを共培養し、増殖後のヒトT細胞とA375腫瘍細胞とを共培養し、LDHキット(碧云天公司)により腫瘍細胞の殺傷効果を調査する。具体的な実験手順は以下のとおりである。1、ヒト末梢血T細胞分離:健康ヒト末梢血を採取し、ヒトリンパ球分離液を用いて末梢血からリンパ球を分離し、さらにPan T選別磁気ビーズ(Miltenyi社)を用いて説明書に従って操作し、ヒトT細胞の分離を行う。2、96ウェルプレート前処理:96ウェルプレートを事前に1μg/mLのヒトCD3抗体溶液(ebioscience社)で前処理し、4℃で一晩静置し、翌日にCD3抗体溶液を吸って取り除き、PBS(Gibco社)で2回洗浄した後、保存する。3.エクソソームとT細胞の共培養:T細胞を96ウェルプレートに接種し、それぞれ異なる用量のヒト雑種エクソソームを加えて共培養し、一定の時間後、T細胞を収集する。4、T細胞とA375細胞の共培養:T細胞とA375細胞をそれぞれ2.5:1、5:1、15:1の比率(E:T)で混合し、新しい96ウェルプレートに接種して共培養し、24h後にCCK8キットを用い、説明書に従って操作し、マイクロプレートリーダーにより450nmでのOD値を測定し、腫瘍細胞の殺傷率を計算し、結果を図15に示す。
(a)E.G7リンパ腫モデルの構築:週齢6〜8の雄C57BL/6マウス(維通利華実験動物技術有限公司)を選択し、その左側の腋窩部位に5×105個のE.G7リンパ腫細胞(ATCC CELL BANK)を皮下注射し、E.G7リンパ腫モデルを構築する。
まず、週齢6〜8の雄C57BL/6マウス(維通利華実験動物技術有限公司)を選択し、0日目にその左側の腋窩部位に5×105個のE.G7リンパ腫細胞(ATCC CELL BANK)を皮下注射し、E.G7リンパ腫モデルを構築する。7日目に腫瘍体積が近いマウスを選択し、ランダムに群を分ける。腫瘍内注射により実施例5の雑種エクソソーム(nc−lps−exo)を体積が同じマウスE.G7腫瘍の内部に注入し、腫瘍内部の各免疫細胞の浸潤状況を調査し、対照群PBS群、処理されていないマクロファージエクソソーム群(pbs−exo)、リポ多糖で刺激されていない実施例5のエクソソーム群(nc−exo)、リポ多糖のみで刺激されたマクロファージエクソソーム群(lps−exo)を作る。隔日に注射し、毎回の注射量は3.7×109個であり、合計3回注射する。14日目にマウスを安楽死させ、腫瘍組織を取り出し、研磨、赤血球破砕、洗浄した後、単細胞懸濁液を得る。その後、単細胞懸濁液を染色し、異なる免疫細胞が占める比率を分析する。具体的な計画は以下のとおりである。
ヒト腫瘍細胞、M2ヒトマクロファージおよびヒトT細胞で3D細胞球を調製し、人体腫瘍微小環境に対してインビトロシミュレーションを行い、本発明のエクソソームを加えて刺激し、3D細胞球におけるM1/M2型マクロファージ比率およびTreg比率を測定し、顕微鏡イメージングにより3D細胞球のサイズを観察する。具体的なステップは以下の通りである。
10mLのdMEM不完全培地に0.15gアガロースを加え、1hかけて100℃に加熱し、アガロースを融解させる。溶液が冷却して完全に凝固していないときに、調製されたアガロースを96ウェルプレートに入れ、室温に冷却し、凹型の細胞培養基質を形成する。
3D細胞球を軽く吸い出した後、4%パラホルムアルデヒドを固定して一晩静置し、その後、Hoechst(Solarbio社)を用いて細胞核染色を行い、室温で15min静置した後、細胞球をウェルプレートに移し、蛍光イメージングを行い、実験結果を図22に示す。
週齢6−8の雄C57BL/6マウス(維通利華実験動物技術有限公司)を選択し、0日目にリンパ腫細胞E.G7−OVA腫瘍細胞(5×105)を接種し、5日目に、実施例5で調製された雑種エクソソームワクチン製剤を用いて皮下免疫を行い、エクソソームの単回接種量は3.7×109個である。さらに、二次免疫を受けた二次免疫群(2nc−lps−exo)を作り、8日目に強化ワクチン接種を行う。また、対照群PBS群、処理されていないマクロファージエクソソーム群(pbs−exo)、リポ多糖で刺激されたいない実施例5の雑種細胞のエクソソーム群(nc−exo)、リポ多糖のみで刺激されたマクロファージエクソソーム群(lps−exo)を作り、対照群エクソソームの注射量は3.7×109個である。その後、マウスの担癌体積および生存状況を記録し、結果を図23および図24に示す。
本発明の実施例7で調製された雑種細胞のエクソソームとPD−1抗体との併用療法により黒色腫悪性腫瘍の免疫治療試験を行い、方法は以下の通りである。
週齢6−8の雄C57BL/6マウス(維通利華実験動物技術有限公司)を選択し、0日目にMuc1−B16黒色腫細胞(ATCC)を接種し、接種用量は5×105個であり、Muc1−B16黒色腫インサイチュ増殖モデルを構築する。実施例7で調製された雑種エクソソームとPD−1抗体の併用により治療を行い(a−PD1+vac.)、エクソソームの皮下投与量は1.85×109個のエクソソームである。一次免疫時間は腫瘍細胞接種後の5日目、二次免疫の時間は8日目であり、PD−1抗体(BioXell社)の投与は腹腔内投与を採用し、それぞれ腫瘍細胞接種後の3日目、6日目、9日目、12日目に投与し、単回投与量は100μgである。また、ブランク群(pbs)、2倍用量雑種エクソソーム単独治療群(2×vac.)、および2倍用量PD−1抗体単独治療群(2×a−PD1)の3群の対照群を作る。その後、マウスの担癌体積を記録し、結果を図25に示す。
Claims (12)
- 抗原提示細胞が腫瘍細胞核を貪食して形成される雑種細胞であって、
好ましくは、前記抗原提示細胞は、樹状細胞、マクロファージまたはB細胞である、雑種細胞。 - 免疫調節剤処理され、
前記免疫調節剤は、リポ多糖、モノホスホリルリピドAまたは非メチル化シチジル酸グアニル酸モチーフである、請求項1に記載の雑種細胞。 - 請求項1に記載の雑種細胞が分泌するナノ小胞である、エクソソーム。
- 腫瘍細胞の細胞核を抽出し、抗原提示細胞培養液に前記腫瘍細胞核を加え、インキュベートした後、この培養液の上清液を収集し、ディファレンシャル遠心分離し、エクソソームを収集するステップを含む、エクソソームの調製方法。
- 前記抗原提示細胞は、腫瘍細胞核を貪食して雑種細胞を形成する、請求項4に記載の調製方法。
- 前記抗原提示細胞は、樹状細胞、マクロファージまたはB細胞である、請求項4に記載の調製方法。
- 抗原提示細胞培養液に免疫調節剤が添加されており、前記免疫調節剤は、リポ多糖、モノホスホリルリピドAまたは非メチル化シチジル酸グアニル酸モチーフである、請求項4に記載の調製方法。
- 請求項1から7のいずれか1項に記載の雑種細胞またはエクソソーム、および薬学的に許容されるキャリアを含む医薬組成物であって、
前記医薬組成物は抗腫瘍ワクチンである、医薬組成物。 - 抗腫瘍薬の調製における請求項1から8のいずれか1項に記載の雑種細胞またはエクソソームの使用であって、
前記抗腫瘍薬は抗腫瘍ワクチンである、使用。 - 請求項1から9のいずれか1項に記載のエクソソーム、モノクローナル抗体、および薬学的に許容されるキャリアを含む医薬組成物であって、
前記モノク1ローナル抗体は、PD−1抗体、PD−L1抗体またはCTLA−4抗体である、医薬組成物。 - 抗腫瘍薬、または腫瘍細胞もしくは腫瘍に対する細胞毒性細胞応答を誘導する医薬品の調製における請求項1から10のいずれか1項に記載のエクソソームの使用。
- 患者体内で腫瘍細胞または腫瘍に対する細胞毒性細胞応答を誘導する方法であって、
前記患者に有効量の請求項1から11のいずれか1項に記載の雑種細胞、エクソソームまたは医薬組成物を含む、方法。
