本发明提供一种冷水虹鳟来源的杀鲑气单胞菌,其保藏编号为CCTCC NO:M2018849。本发明所提供的杀鲑气单胞菌用于制备疫苗。本发明所提供的杀鲑气单胞菌是从发病的冷水虹鳟中筛选获得的,该菌株相比于目前杀鲑气单胞菌产生了变异,用于制备疫苗,从而对虹鳟鱼提供更完善的免疫保护。

Description

一种冷水虹鳟来源的杀鲑气单胞菌
技术领域
本发明属于病原微生物筛选技术领域,具体涉及一种冷水虹鳟来源的杀鲑气单胞菌。
背景技术
虹鳟鱼是属于鲑科的一种冷水性鱼类,是目前最广泛的世界性养殖的淡水鱼类之一,由于虹鳟鱼最适合进行人工集约化养殖,具有高产、高效、市场广阔等特点,所以虹鳟鱼养殖发展迅速,养殖面积范围及养殖产量在逐年增长。
但随着养殖规模的扩大,养殖密度的逐渐增大,在冷水虹鳟的养殖过程中出现了诸多病害问题,而近年来在虹鳟养殖过程中细菌性疾病的发生比较频繁,给养殖户造成了很大的经济损失。常用的孔雀石绿、呋喃唑酮、氯霉素等有效控制细菌疾病的药物,由于会产生残留,已经被国家禁止使用。
疫苗的使用可提高养殖鱼类特异性免疫水平,使鱼体产生抵抗特定病原微生物感染的免疫力,不污染环境,无药物残留等问题,是今后鱼类疾病防控的主流发展方向。
但在微生物疫苗制备领域中,已正式上市的疫苗所使用的抗原菌株为已多年流行的标准菌株,但随着时间推移和环境变化,致病菌株会由于免疫导致的选择压力发生变异,使原有疫苗免疫产生的抗体无法针对变异菌株提供完全的免疫保护,临床应用效果不佳。另一个可能因素是当两个或以上的病菌同时侵袭一个宿主细胞时,基因片段之间相互作用,在转录复制过程中发生交换、交叉或重排,引起基因的变异。而突变株有一定的几率使已有的疫苗的免疫效果降低或完全没有效果。因此,筛选变异菌株就成为疫苗制备领域的研究热点之一。
发明内容
本发明的目的在于提供一种冷水虹鳟来源的杀鲑气单胞菌,能够用于制备灭活疫苗,从而弥补现有技术的不足。
本发明所提供的冷水虹鳟来源的杀鲑气单胞菌杀日本鲑亚种RB0506(Aeromonassalmonicida subsp.masoucida RB0506),于2018年12月03日保藏在中国武汉、武汉大学的中国典型培养物保藏中心,保藏编号为CCTCC NO:M2018849;
本发明所提供的杀鲑气单胞菌在制备疫苗中的应用。
所述的疫苗为灭活疫苗。
本发明所提供的杀鲑气单胞菌是从发病的冷水虹鳟中筛选获得的,该菌株相比于目前杀鲑气单胞菌产生了变异,用于制备疫苗,从而对虹鳟鱼提供更完善的免疫保护。
具体实施方式
下面对本发明进行详细说明。
实施例1:杀鲑气单胞菌菌株的分离及其毒性分析
1)患病虹鳟取自山东日照养鱼厂,但发病虹鳟鱼之前已进行了单胞菌疫苗注射免疫。发病的病鱼平均体重为(100±5)g,平均体长(17±2)cm,主要症状为体表溃烂、腹鳍基部充血发红、肛门突出、肠道充血。
2)取症状典型的濒死病鱼,无菌条件下从鳍基部、肠道等病灶处挑取部分组织,划线于MH琼脂平板上,28℃培养24h,挑取形态一致的优势菌落进行纯化培养,直至获得纯培养菌。
3)将分离获得各菌株纯培养物,在MH琼脂平板上进行扩大培养,28℃培养24h后,用0.9%无菌生理盐水洗脱菌落,获得菌悬液,利用比浊法将菌株的细菌浓度定为1×107CFU/mL,用于注射感染。
4)实验用健康虹鳟每20尾1组,大小规格与患病虹鳟相同,并设生理盐水对照组。养殖水温为17±1℃,全天充气,每天换水50%。暂养7d后,腹腔注射0.2mL不同菌株的菌悬液,对照组注射同体积的0.9%无菌生理盐水。定时观察记录,取濒死鱼的病灶及内脏器官进行细菌再分离,所分离优势菌株再次感染虹鳟,观察结果。
5)感染实验结果显示,仅有菌株RB0506显示出对健康虹鳟具有较高的毒力,可引起90%虹鳟死亡,且死亡虹鳟出现与患病虹鳟相同的症状。因此,证实菌株RB0506为引起虹鳟皮肤溃疡的病原。且在感染患病的虹鳟体内可再次分离到该菌株。
菌株RB0506于2018年12月03日保藏在中国武汉、武汉大学的中国典型培养物保藏中心,保藏编号为CCTCC NO:M2018849。
6)从GenBank下载已知杀鲑气单胞菌的封闭带毒素(ZOT)、OmpW基因和气溶素Aerolysin基因的序列,经过alignment分析比对后,根据保守序列设计用于扩增Primer5.0软件设计封闭带毒素(ZOT)、OmpW基因和气溶素Aerolysin基因的扩增引物,采用RT-PCR方法扩增各基因节段序列,并用BLASTn分析各基因编码情况。与GenBank上已发表的三个抗原基因进行核苷酸和氨基酸的同源性比对。
结果表明,三种抗原蛋白都存在5-10个氨基的差异。分子生物学分析结果表明新分离的杀鲑气单胞菌RB0506菌株与已报到的杀鲑气单胞菌有着明显区别,主要表现为各基因片段核苷酸之间的点突变和基因重组。
其中RB0506菌株ZOT基因的氨基酸序列为SEQ ID NO:1,OmpW基因的序列为SEQ IDNO:2,气溶素Aerolysin基因的氨基酸序列为SEQ ID NO:3。
实施例2:使用杀鲑气单胞菌制备灭活疫苗
将MH肉汤中过夜培养的杀鲑气单胞菌RB0506菌株室温6000rpm离心10min,细菌沉淀用生理盐水(0.9%NaCl溶液)重悬,再离心收集沉淀,重复2次,最后用生理盐水重悬并调整细菌浓度至1.5×109个/mL,分装于50mL离心管内,每管30mL,利用0.1%的甲醛溶液在28℃条件下对杀鲑气单胞菌灭活5h制成灭活疫苗抗原悬液。
取19ml上述灭活疫苗抗原悬液,加入1ml灭菌Tween-80搅拌混匀,制备成乳化疫苗的水相;而后用疫苗用白油38.4ml,1.6ml Span-80,0.6g硬脂酸铝相混合,加热使硬脂酸铝完全溶化,灭菌制备成乳化疫苗的油相;使用分散机10000rpm转速,将40ml的油相缓慢加入到20ml水相中,搅拌3分钟形成乳白色油包水性油乳疫苗。
实施例3:疫苗的效果检测
按照步骤2制备疫苗的步骤,将杀鲑气单胞菌CICC 23564株和杀鲑气单胞菌E11I4株(CGMCC1.16015)作为抗原制备成疫苗,
将3种灭活疫苗浓度调整至OD600为0.1±0.01,利用腹腔注射的方法分别免疫健康虹鳟,每尾100μL,对照组注射同体积的无菌生理盐水,每组免疫20尾,每组做4个重复。养殖水温为17±1℃,全天充气,每天换水50%;免疫后40d用三种杀单胞菌菌株进行攻毒,每尾腹腔注射浓度为5.0×108CFU/mL的菌液100μL;每天定时观察并记录各实验组鱼体死亡数量,计算3种灭活疫苗对杀鲑气单胞菌感染的相对免疫保护率。
结果表明,攻毒后注射生理盐水组的鱼体在攻毒后第2天便开始出现死亡,并且鱼体在接下来的一周内出现大量死亡,直至攻毒后第10天死亡率达到96.7%;攻毒受感的鱼体表现出腹鳍基部充血发红、肛门突出、肌肉出血、肠道出血等杀鲑气单胞菌感染的典型临床症状,并能从受感死亡鱼体中再次分离到该菌株。而3种疫苗免疫组鱼体的死亡率显著低于生理盐水对照组。但实施例2中使用RB0506菌株制备的灭活疫苗的免疫组,在RB0506菌株攻毒组的死亡率显著低于另外两株杀鲑气单胞菌制备的疫苗(表1)。

上述结果表明本发明所筛选获得的杀鲑气单胞菌相比于已报到的杀鲑气单胞菌发生了变异,从而在感染保护上与已有细菌株在感染保护上有着显著的差异。
