Abstract
本发明涉及一种精血补片提取液及其应用,所述精血补片提取液又精血补片提取而成,精血补片由以下重量份的组分制备而成:生晒参100份,红参100份,制何首乌200份,紫河车200份,五味子200份和陈皮120份;所述精血补片提取液在提高T细胞免疫活性中的应用。本发明的优点在于:本发明精血补片提取液在250ug/mL浓度时对于T细胞的增值有接近于两倍的刺激作用。
Description
技术领域
本发明属于生物医药领域,特别涉及一种精血补片提取液及其应用。
背景技术
《中华人民共和国卫生部药品标准》(中药成方制剂)第六册中具有用于神经衰弱,精神萎靡,头晕目眩心悸失眠的药物精血补片,该复方制剂由中药材生晒参100g,红参100g,制何首乌200g,紫河车200g,五味子200g,陈皮120g组成,上述原料共制成4600片。生晒参是方中主要药味,为五加科植物人参Panax ginseng C.A.Mey.的干燥根及根茎,具有大补元气,复脉固脱,补皮益肺,生津,安神之功效。红参也是方中主要药味,为五加科植物人参Panax ginseng C.A.Mey.的栽培品经蒸制后的干燥根及根茎,具有大补元气,复脉固脱,益气摄血之功效。制何首乌为萝科植物何首乌Polygonum multiflorum Thunb.的干燥块根的炮制加工品,具有补肝肾,益精血,乌须发,强筋骨之功效。紫河车为健康人的干燥太胖,具有温肾补精,益气养血之功效。五味子为木兰科植物北五味子Schisandra chinensis(Turcz.)Baill.的干燥成熟果实,具有收敛固涩,益气生津,补肾宁心之功效。陈皮为芸香科植物橘Citrus reticulta Blanco 及其栽培变种的干燥成熟果皮,具有理气健脾,燥湿化痰之功效。
关于对精血补片的提取,以及在免疫细胞中的应用,经检索,目前还没有报道类似的专利文献。
发明内容
本发明要解决的技术问题是提供一种精血补片提取液及其应用,
以挖掘精血补片新的应用。
为解决上述技术问题,本发明的技术方案为:一种精血补片提取液,其创新点在于:所述精血补片由以下重量份的组分制备而成:生晒参100份,红参100份,制何首乌200份,紫河车200份,五味子200份和陈皮120份;所述精血补片提取液是通过以下步骤制备而成:
步骤1:精确称量精血补片9.2g置于一50mL的离心管中,然后向离心管中加入30mL的磷酸缓冲盐溶液,并振荡离心管混匀;
步骤2:步骤1中精血补片与磷酸缓冲盐溶液混匀后,冰上超声粉碎,超声功率为45~80KHZ,每次超声处理6S停9S,共30min,共超声处理至少两次;
步骤3:冰上超声处理后,将混匀的精血补片与磷酸缓冲盐溶液在10000rpm转速下离心30min,然后取上清液20mL,下沉淀物于65~75℃下烘干;
步骤4:将步骤3中的上清液依次经0.22µm过滤膜和0.1µm过滤膜过滤,得到精血补片提取液,备用。
一种上述的精血补片提取液的应用,其创新点在于:所述精血补片提取液在提高T细胞免疫活性中的应用。
本发明的优点在于:
本发明精血补片提取液及其应用,通过对精血补片进行提取,并通过提取液进行免疫细胞株增殖试验,进而发现本发明精血补片提取液在250ug/mL浓度时对于T细胞的增值有接近于两倍的刺激作用。

具体实施方式
下面的实施例可以使本专业的技术人员更全面地理解本发明,但并不因此将本发明限制在所述的实施例范围之中。
实施例
本实施例精血补片提取液,该精血补片由以下重量份的组分制备而成:生晒参100份,红参100份,制何首乌200份,紫河车200份,五味子200份和陈皮120份;所述精血补片提取液是通过以下步骤制备而成:
步骤1:精确称量精血补片9.2g置于一50mL的离心管中,然后向离心管中加入30mL的磷酸缓冲盐溶液,并振荡离心管混匀;
步骤2:步骤1中精血补片与磷酸缓冲盐溶液混匀后,冰上超声粉碎,超声功率为45~80KHZ,每次超声处理6S停9S,共30min,共超声处理至少两次;
步骤3:冰上超声处理后,将混匀的精血补片与磷酸缓冲盐溶液在10000rpm转速下离心30min,然后取上清液20mL,下沉淀物于65~75℃下烘干;
步骤4:将步骤3中的上清液依次经0.22µm过滤膜和0.1µm过滤膜过滤,得到精血补片提取液,备用。
对精血补片提取液进行免疫细胞株增殖试验,包括如下步骤:
步骤1:使用RPMI1640(10%FBS、1%双抗)完全培养基梯度稀释制备含水提取液1000、500、250 、100 、20 ug/mL;
步骤2:取对数期生长良好的Raw264.7(巨噬细胞)、CTLL-2(T细胞)、NALM-6(B细胞)重悬计数;将密度调至5*10^4/mL,100ul/孔接种于96孔板,每孔加入100ul制备好的水提物完全培养基,每个浓度三个复孔,并设置0浓度对照孔;
步骤3:48h后,加入细胞活性检测试剂CCK8,20ul/孔,37℃培养4小时;
步骤4:酶标仪读取OD450nm 读数,其结果见下表1和图1。

由表1和图1可以看出,精血补片提取液在250ug/mL浓度时对于T细胞的增值有接近于两倍的刺激作用,进一步提高浓度没有发现有进一步增殖作用现象。在同等浓度范围内,JH01对于巨噬细胞和B细胞的刺激作用则不明显。

1.一种精血补片提取液,其特征在于:所述精血补片由以下重量份的组分制备而成:生晒参100份,红参100份,制何首乌200份,紫河车200份,五味子200份和陈皮120份;所述精血补片提取液是通过以下步骤制备而成:
步骤1:精确称量精血补片9.2g置于一50mL的离心管中,然后向离心管中加入30mL的磷酸缓冲盐溶液,并振荡离心管混匀;
步骤2:步骤1中精血补片与磷酸缓冲盐溶液混匀后,冰上超声粉碎,超声功率为45~80KHZ,每次超声处理6S停9S,共30min,共超声处理至少两次;
步骤3:冰上超声处理后,将混匀的精血补片与磷酸缓冲盐溶液在10000rpm转速下离心30min,然后取上清液20mL,下沉淀物于65~75℃下烘干;
步骤4:将步骤3中的上清液依次经0.22µm过滤膜和0.1µm过滤膜过滤,得到精血补片提取液,备用。
2.一种权利要求1所述的精血补片提取液的应用,其特征在于:所述精血补片提取液在提高T细胞免疫活性中的应用。
