Abstract
本发明公开了一种小分子多肽TAT‑PSYN及其在制备预防或治疗帕金森综合症药物中的应用。通过合成TAT蛋白转导结构域和可以结合并抑制突触核蛋白(alpha‑synuclein)的蛋白多肽的融合蛋白TAT‑pSyn,利用TAT携带pSyn蛋白多肽经由血液运输透过血脑屏障,并被神经元摄取。将该多肽运用到Alpha‑synuclein(A53T)转基因帕金森小鼠模型中,可有效发挥其阻断突触核蛋白(alpha‑synuclein)与死亡相关蛋白激酶1(DAPK1)结合的作用,抑制α‑synuclein磷酸化和异常聚集引发的多巴胺神经元和神经投射的丢失,降低帕金森模型的运动障碍,为临床治疗帕金森综合症的药物提供了分子靶标。

背景技术
1817年英国医生James Parkinson首先对帕金森综合症进行了详细的描述,其临床表现主要包括静止性震颤、运动迟缓、肌强直和姿势步态障碍,同时患者可伴有抑郁、便秘和睡眠障碍等非运动症状。帕金森病突出的病理改变是中脑黑质多巴胺(dopamine,DA)能神经元的变性死亡、纹状体DA含量显著性减少以及黑质残存神经元胞质内出现嗜酸性包涵体,即路易小体(Lewy body)。路易小体(Lewy body)是以帕金森病为代表的路易体病患者脑内的特征性标志物,存在于细胞浆中,中央呈致密核心,周围有细丝状晕圈。显微镜下为圆形粉红色均质状结构,其前体被称为Pale body(灰体),而alpha-synuclein是Lewybody及其前体Pale body的主要成分。帕金森病患者10%的中脑黑质残存的多巴胺能神经元中含有Lewy body,是帕金森综合症主要的病理特征,也是其诊断的重要依据。大部分帕金森症患者是散发性的由多种因素造成的。已有研究表明alpha-synuclein 功能的障碍是大部分帕金森症患者发病的原因。研究人员认为有可能是调节蛋白正常功能的磷酸化修饰过程受到了影响。
帕金森综合症是第二大神经退行性疾病。在2015年,全球有620万帕金森患者,并且有117400患者死亡,然而目前针对帕金森综合症的治疗手段十分有限,现有的药物治疗和手术治疗都不能有效地根治,故多数患者只能对症治疗。迄今为止,全球针对帕金森综合症所开发的小分子化合物和多项临床试验均以失败告终。因此,探究新的治疗方法,对抗帕金森综合症引发的运动障碍,减少神经元的死亡极其重要。
TAT为细胞穿膜肽(cell penetrating peptides),是一种可以穿透细胞膜和核膜的新型高效运输载体。TAT能够携带多肽、蛋白质以及DNA分子等通过受体主动转运进入细胞质和细胞核,从而使携带的分子发挥相应的生物效应。目前体内外试验均显示HIV-TAT可以穿过包括神经细胞在内的所有组织细胞,且无明显毒副作用。TAT的转运效率高,可通过血液运输,并且可以透过血脑屏障进入神经元和胶质细胞,而所携带的蛋白质或多肽可保持其原有的生物活性,发挥其相应的生物学作用。基于申请人的近期研究以及TAT技术,拟合成一种具有预防和治疗帕金森综合症作用的膜渗透小分子多肽。

本发明设计了这种小分子多肽TAT-pSyn。基于申请人的近期研究以及TAT 技术,申请人合成了由alpha-synuclein的一段氨基酸序列(Tyr-Glu-Met-Pro-Ser- Glu-Glu-Gly-Tyr-Gln-Asp,YEMPSEEGYQD)和TAT穿膜肽(Tyr-Gly-Arg-Lys-Lys- Arg-Arg-Gln-Arg-Arg-Arg,YGRKKRRQRRR)组成的膜渗透小分子多肽TAT- pSyn,将其运用到转基因帕金森综合症小鼠模型中,有效发挥其阻断死亡相关蛋白激酶1(DAPK1)与突触核蛋白的结合并且将其引导入溶酶体降解的作用,从而达到抑制alpha-synuclein下游引发的多巴胺神经元丢失,降低帕金森综合症后的运动障碍,为进一步开发临床治疗帕金森综合症的药物提供了分子靶标。
本发明将TAT穿膜肽(Tyr-Gly-Arg-Lys-Lys-Arg-Arg-Gln-Arg-Arg-Arg,YGRKKRRQRRR)与alpha-pSynuclein的一段氨基酸序列(Tyr-Glu-Met-Pro- Ser-Glu-Glu-Gly-Tyr-Gln-Asp,YEMPSEEGYQD)连接,得到具有生物学活性的 TAT-pSyn小分子多肽。利用TAT携带pSyn多肽,经血液运输透过血脑屏障,并被大脑神经元摄取从而发挥其阻断alpha-synnuclein异常聚集和磷酸化的生物学功能。
本发明提供的这种小分子多肽TAT-pSyn能用于制备预防或治疗帕金森综合症的药物。将这种小分子多肽TAT-pSyn给予帕金森模型小鼠尾静脉注射,发现其可以有效减低帕金森综合症后alpha-synuclein的磷酸化和异常聚集,抑制多巴胺神经元的丢失,改善相应的行为学表型。
本专利申请的发明人发现,帕金森综合症后,突触核蛋白(alpha- synnuclien)的Ser129位点被死亡相关蛋白激酶1(DAPK1)磷酸化,并介导下游神经元死亡(坏死与凋亡)。阻断死亡相关蛋白激酶1(DAPK1)与突触核蛋白(alpha-synnuclien)的相互作用,能有效降低帕金森综合症后神经元死亡。针对这一发现,申请人将TAT穿膜肽(Tyr-Gly-Arg-Lys-Lys-Arg-Arg-Gln- Arg-Arg-Arg,YGRKKRRQRRR)与突触核蛋白(alpha-synnuclien)的一段氨基酸序列(Tyr-Glu-Met-Pro-Ser-Glu-Glu-Gly-Tyr-Gln-Asp,YEMPSEEGYQD)连接,获得具有生物学活性的TAT-pSyn。通过小鼠尾静脉注射,可使TAT-pSyn 多肽进入血液并穿过血脑屏障被大脑神经元摄取,从而发挥其生物学效应。

本发明提供的小分子多肽TAT-pSyn在制备预防或治疗帕金森综合症药物中的应用,其应用过程如下:
TAT-pSyn在帕金森综合症动物模型中的应用:
Alpha-synuclein(A53T)(Jax:004479)转基因小鼠模型是公认的帕金森综合症动物模型。该转基因小鼠表达人源性的A53T突变的突触核蛋白(alpha- synuclein,全长140氨基酸),使用小鼠朊蛋白启动子。在八个月的时候,一些纯合子小鼠逐渐地表现出严重的运动障碍。表型显现出来平均是14-15个月。表现有皮肤松弛,体重下降和先于运动障碍的运动迟缓,部分肢体麻痹,震颤以及无法站立。对8-12个月的突变小鼠的免疫组织化学分析,发现在脊髓,脑干,小脑和纹状体中突触核蛋白内含物聚集。突触核蛋白的恶性聚集与运动障碍的发展是同步的。对12个月的模型小鼠给予尾静脉注射10mg/kg(每三天一次(1mg/kg),为期一个月)的TAT-pSyn溶液,在相应的时间后进行动物行为学实验和染色。疲劳转棒和旷场实验结果表明,注射TAT-pSyn的小鼠的平衡协调能力以及运动能力相较于对照组TAT-s-pSyn或者生理盐水Vehicle组有显著改善。蛋白质免疫印迹和filter trap的结果显示给予TAT-pSyn的小鼠的alpha- synuclein和磷酸化的alpha-synuclen的蛋白水平降低。TH染色结果显示给予 TAT-pSyn的小鼠的TH阳性细胞数量比对照组TAT-s-pSyn或者生理盐水 Vehicle组多。此外,以上结果均证明,TAT-pSyn对帕金森综合症具有确切的治疗作用。
与现有的技术相比,本发明具有以下特点:本发明所设计的小分子多肽 TAT-pSyn合成纯度高,可溶性好,适用于静脉注射,无毒副作用,在转化生产和临床应用上均具有优势。
本发明公开了一种小分子多肽TAT-PSYN及其在制备防治帕金森综合症药物中的应用,通过合成TAT蛋白转导结构域和可以结合并抑制突触核蛋白 (alpha-synuclein)的蛋白多肽的融合蛋白TAT-pSyn,利用TAT携带pSyn蛋白多肽经由血液运输透过血脑屏障,并被神经元摄取。将该多肽运用到Alpha- synuclein(A53T)转基因帕金森小鼠模型中,可有效发挥其阻断突触核蛋白 (alpha-synuclein)与死亡相关蛋白激酶1(DAPK1)结合的作用,抑制α- synuclein磷酸化和异常聚集引发的多巴胺神经元和神经投射的丢失,降低帕金森模型的运动障碍,为临床治疗帕金森综合症的药物提供了分子靶标。本发明运用人工合成TAT蛋白转导结构域与结合突触核蛋白(alpha-synuclein)的融合蛋白多肽,意在采用静脉注射的方式治疗帕金森综合症,减少帕金森综合症引起的神经元丢失,改善帕金森综合症后运动障碍。本发明公开的TAT-pSyn重组蛋白多肽中,TAT作为一种高效的转导蛋白,可携带多肽经血液运输透过血脑屏障而被神经元摄取,可以转化应用于帕金森综合症等神经系统疾病中,具有实际操作的可行性。

以下结合附图和具体实施方法对本方法做进一步说明。在实施方案中涉及的所有细胞培养,蛋白质免疫印迹,TH染色,尾静脉注射以及行为学检测的所有操作方法,均为本领域研究人员所熟知。本发明中未详细阐述的请参见文献 (Lei Pei,You Shang,HuijuanJin,Shan Wang,Na Wei,Honglin Yan,Yan Wu, Chengye Yao,Xiaoxi Wang,Ling-QiangZhu,and Youming Lu,DAPK1-p53 Interaction Converges Necrotic and ApoptpticPathways of Ischemic Neuronal Death, The Journal of Neuroscience,May7,2014-34(19)-6546-6556)的材料与方法部分。
实施例1
TAT-pSyn的人工合成
TAT-pSyn的序列如SEQ ID NO.1所示,由江苏强耀生物科技有限公司人工合成,合成报告如下所示,色谱如图1所示。

合成的TAT-pSyn多肽的纯度为95.79%,每支1mg,为白色粉末状,完全可溶于水,密封避光保存于-20℃。使用前,用注射用生理盐水按指定浓度进行稀释配置,现配现用。
TAT-pSyn的对照为TAT-scramble-pSyn(TAT-s-pSyn),其序列如下:

实施例2
TAT-pSyn阻断DAPK1与alpha-synuclein结合,抑制alpha-synuclein的过度磷酸化和异常聚集。
体外培养神经纤维瘤母细胞(N2a),转染真核表达alpha-synuclein的质粒,在共转染表达DAPK1质粒的细胞上分别给予0uM,5uM,25uM,50uM的TAT-pSyn 多肽或对照TAT-s-pSyn多肽或vehicle孵育。90分钟后提取细胞蛋白。结果显示:给予25uM的TAT-pSyn后,alpha-synuclein和磷酸化的alpha-synuclein的蛋白量都减少了(图2)。提示TAT-pSyn阻断了DAPK1与alpha-synuclein的相互作用,从而抑制alpha-synuclein的过度磷酸化和异常聚集。
实施例3
TAT-pSyn在帕金森综合症的转基因小鼠模型中的应用
(1)帕金森综合症小鼠模型的建立
从美国Jackson Lab购买了编号004479的Alpha-synuclein(A53T)的转基因小鼠进行繁殖,该转基因小鼠表达人源性的A53T突变的突触核蛋白(alpha- synuclein,全长140氨基酸),使用小鼠朊蛋白启动子。在八个月的时候,一些纯合子小鼠逐渐地表现出严重的运动障碍。表型显现出来平均是14-15个月。表现有皮肤松弛,体重下降和先于运动障碍的运动迟缓,部分肢体麻痹,震颤以及无法站立。对8-12个月的突变小鼠的免疫组织化学分析,发现在脊髓,脑干,小脑和纹状体中突触核蛋白内含物聚集。突触核蛋白的恶性聚集与运动障碍的发展是同步的。我们使用12月龄的小鼠作为实验用模型小鼠。
(2)TAT-pSyn静脉注射
12月龄的Alpha-synuclein(A53T)模型小鼠经小鼠尾静脉注射10mg/kg多肽TAT-pSyn和对照多肽TAT-s-PSYN或注射用生理盐水(vehicle),每三天一次 (1mg/kg),为期一个月。多肽浓度为1mg/ml,用注射用生理盐水溶解。静脉注射完成后,进行疲劳转棒实验和旷场实验检测小鼠的平衡协调能力和运动能力,并进行相应的免疫组织化学染色。以上述的实验结果证明TAT-pSyn对帕金森综合症的治疗作用。
(3)TAT-pSyn治疗效果的评估
疲劳转棒实验:在检测前的连续5天进行训练,在每天的同样时间训练10 分钟,具体操作如下:第一天,小鼠放置在4转/分钟的转棒上待5分钟,接下来将每30秒增加1转匀加速至15转/分钟,最后保持1分钟;第二天,小鼠放置在4转/分钟的转棒上待1.5分钟,接下来将每30秒增加1转匀加速至20转/ 分钟,最后保持10分钟;第三,四,五天则统计在由4转/分钟线性加速到32转 /分钟的各5分钟内小鼠从开始到由转棒掉下的时间。最后测试时与最后三天的训练方法一样进行。从放置小鼠到小鼠落下的时间间隔记作保留时间,反应动物的四肢力量及运动协调能力。小鼠滞留时间越长,说明其运动能力越强。
旷场实验:旷场实验可以检测啮齿类动物的运动能力,焦虑水平和自发探索情况。我们将实验小鼠置于旷场实验系统(50cmx50cmx40cm)检测10分钟,统计小鼠的运动距离,运动速度和运动时间比。运动距离越长,速度越快,运动时间所占百分比越多说明其运动能力越强。
蛋白质免疫印迹(Western Blot)和filter trap:蛋白免疫印迹是是将电泳分离后的细胞或组织总蛋白质从凝胶转移到固相支持物NC膜或PVDF膜上,然后用特异性抗体检测某特定抗原的一种蛋白质检测技术,通过分析着色的位置和着色深度获得特定蛋白质在所分析的细胞或组织中表达情况的信息,是检测蛋白质特性、表达与分布的一种最常用的方法。我们提取了实验小鼠的中脑黑质组织总蛋白进行蛋白质免疫印迹,从而分析alpha-synuclein和磷酸化alpha-synuclein的蛋白水平变化。filter trap是一种分析总蛋白中特定蛋白聚集程度的免疫印迹方法,无需进行凝聚电泳,其余与蛋白质免疫印迹类似。可以用该方法分析组织蛋白中 alpha-synuclein的聚集程度。
络氨酸羟化酶(Tyrosine hydroxylase,TH)染色:TH染色是一种免疫组织化学染色,使用TH(络氨酸羟化酶)抗体可以标记多巴胺神经元的数量。络氨酸羟化酶是负责催化氨基酸L-酪氨酸转变为二羟基苯丙氨酸(多巴)的酶,多巴是多巴胺的一个前体,因此TH染色阳性的细胞是多巴胺神经元。我们将实验小鼠经心脏灌流后取出脑组织,接着固定脱水进行冰冻切片,将相应连续切片进行 TH染色。之后观察和统计TH阳性细胞数量。
12月龄的Alpha-synuclein(A53T)模型小鼠经小鼠尾静脉注射10mg/kg TAT-pSyn溶液或者对照组TAT-s-pSyn溶液,或生理盐水vehicle组。疲劳转棒结果显示,给予TAT-pSyn后Alpha-synuclein(A53T)小鼠在不同转速停留在转棒上的时间比未给多肽处理的帕金森模型小鼠要显著延长,平衡协调能力与正常小鼠没有差异(图3)。说明TAT-pSy注射后能防止Alpha-synuclein(A53T)模型小鼠的平衡协调能力障碍。旷场实验结果显示,给予TAT-pSyn后Alpha-synuclein (A53T)小鼠的运动距离,运动速度和运动时间百分比均比未给多肽处理的帕金森模型小鼠显著改善,运动能力与正常小鼠没有差异(图4)。说明TAT-pSy注射后能防止Alpha-synuclein(A53T)模型小鼠的运动能力障碍。小鼠中脑黑质组织的免疫蛋白印迹的结果显示,给予TAT-pSyn后Alpha-synuclein(A53T)小鼠的alpha-synuclein和磷酸化alpha-synuclein的蛋白水平均显著低于未给多肽处理的帕金森模型小鼠,与正常小鼠的蛋白没有显著差异,filter trap的结果也一致 (图5)。说明TAT-pSy注射后能减少Alpha-synuclein(A53T)模型小鼠的alpha- synuclein的过度磷酸化和异常聚集。TH染色结果显示,未给予多肽处理的帕金森模型小鼠的多巴胺神经元数量减少了,而给予TAT-pSyn的Alpha-synuclein (A53T)小鼠的多巴胺神经元数量与正常12月龄对照组小鼠没有差异(图6)。说明TAT-pSy注射后能防止Alpha-synuclein(A53T)模型小鼠大脑多巴胺神经的丢失。
上述组织学与行为学的实验结果均显示,注射TAT-pSyn多肽后,能显著减少帕金森综合症小鼠的多巴胺神经元的死亡,改善小鼠的运动平衡能力和运动能力,是开发帕金森综合症临床治疗药物的潜在靶点。
