本发明提供用于抑制柑橘黄龙病菌(Candidatus Liberibacter)感染或黄龙病(Huanglongbing disease)的组合物和方法。更具体地说,本发明涉及植物防御诱导剂组合物和化合物,以及其用于改善植物和作物管理的某些方面的用途,包括治疗植物疾病、改善植物抵御疾病的能力、减少疾病症状、治疗HLB疾病、最小化由植物疾病引起的作物产量降低、改善作物产率以及提高作物质量。具体地说,可以使用化合物和组合物控制和治疗植物疾病,例如黄龙病(HLB),以及缓解植物中HLB和柑橘黄龙病菌感染的疾病症状。预期本发明适用于作物植物,例如柑橘树。
背景技术
黄龙病(HLB),通常称为柑橘“绿化”病,是由树木和其他作物物种的部分系统性细菌感染引起的,引起叶子变色并且降低果实产量。在佛罗里达(Florida),侵袭性HLB疾病的传播造成对柑橘产业的主要威胁,柑橘产业由于这种感染引起的损失已达到每年数百万美元。因为昆虫载体已到达德克萨斯(Texas)和加利福尼亚(California),疾病在这些州爆发也只是时间问题。
认为HLB与由三种韧皮杆菌物种引起的感染相关:对于在亚洲的疾病,柑橘黄龙病菌亚洲种(Candidatus Liberibacter asiaticus;Las),对于在非洲的疾病,柑橘黄龙病菌非洲种(Candidatus Liberibacter africanus;Laf),并且对于在美洲的疾病,柑橘黄龙病菌美洲种(Candidatus Liberibacter americanus;Lam)。统称为柑橘黄龙病菌属。
HLB的当前管理策略是以化学方式控制木虱并且搜寻和移除受感染的树木。其功效有限并且没有常规手段可显示对疾病的持续和有效抑制。
受HLB感染的树木的果实非常苦。果实在其成熟时的颜色甚至仍然保持绿色并且其尺寸较小。患有HLB疾病的树木将落叶,并且不久便会枯萎和死亡。频繁的昆虫控制和树木移除造成的高成本将最终导致柑橘园没有盈利,其引起对园艺和农业的严重损害。并且大规模施用杀虫剂将破坏生态系统并且污染环境。
因此,需要有效治疗HLB疾病。
发明内容
提供用于改善植物(包括重要的作物植物,如柑橘)的健康和抗病性的技术和组合物。所述方法还提供对受疾病影响或对疾病敏感的植物中的植物疾病的治疗和控制。可对植物施用本文中描述的根据本发明的实施例的微生物类组合物以治疗植物疾病,如HLB;改善抗病性;改善抵御疾病的能力;减少疾病症状;最小化由植物疾病引起的作物产量降低;改善柑橘中的作物产率和作物质量;以及提高柑橘中的果汁含量和果汁质量。因此,可以使用微生物类组合物为健康植物和患病植物带来益处。所描述的方法还涉及对植物施用组合物,包括通过注射或土壤浇灌方法施用于植物周围的土壤、施用于植物表面,如喷雾到植物或植物的一部分上,如通过叶面喷雾,或注射到植物中,如通过树干注射。本发明涵盖的植物包括任何植物,尤其作物植物,但也包括观赏类植物和柑橘属植物。
在一个方面中,本发明涉及生物杀虫剂治疗柑橘绿化病领域。更具体地说,本发明涉及发现赭曲霉(Aspergillus ochraceus)、绿木霉菌(Trichoderma virens)或哈茨木霉(Trichoderma harzianum)的菌株,其在将发酵的完全细胞培养液、滤液、上清液或提取物施用于植物、植物部分和/或果实时可以抑制柑橘黄龙病菌属的生长。

如本文中所定义,“完全细胞培养液”是指含有细胞、孢子和/或培养基的液体培养物。如果细菌或真菌在盘上生长,那么可以在水或其它液体或完全培养物中收集细胞。
术语“上清液”是指当在培养液中生长或从琼脂盘在另一液体中收集的细胞通过离心、沉降或所属领域中已知的其它手段被移除时剩余的液体。
如本文中所定义,“滤液”是指来自完全培养液培养物的已经穿过膜的液体。形成滤液的非限制性方式是使一或多种微生物发酵以产生完全细胞培养液,接着使培养液穿过膜,其中滤液是无细胞的。

本文中所披露的真菌可以使用所属领域中已知的方法在营养物培养基中培育。有机体可以在允许细胞生长的条件下,通过振荡烧瓶培养、在合适的培养基中进行的在实验室或工业发酵器中的小规模或大规模发酵(包括(但不限于)连续、分批、分批进料或固态发酵)来培养。培养可以在包含碳和氮气来源以及无机盐的合适的营养物培养基中,使用所属领域中已知的程序进行。合适的培养基可以从商业来源获得或可以根据已公开的论文制备。

在干燥粉末的情况下,活性成分与可湿润粉末类似地配制,但添加如矿物油等粘着剂而非湿润剂。举例来说,根据由莱达古玛(Nandakumar)等人(2001)所描述的方法,在无菌条件下混合一千克纯化滑石粉末(灭菌12小时)、15g碳酸钙以及10g羧甲基纤维素。以1:2.5比率(悬浮液比干燥混合物)混合一或多种活性成分,并且阴干产物以将水分含量降低到20-35%。

组合物还可以选自多种配制物类型,包括可尘化粉末(DP)、可溶性粉末(SP)、水溶性颗粒(SG)、水分散性颗粒(WG)、可湿润粉末(WP)、颗粒(GR)(缓慢或快速释放)、可溶性浓缩物(SL)、油可混溶的液体(OL)、超低容量液体(UL)、可乳化浓缩物(EC)、可分散浓缩物(DC)、乳液(水包油(EW)和油包水(EO)两种)、微乳液(ME)、悬浮液浓缩物(SC)、基于油的悬浮液浓缩物(OD)、气雾剂、成雾/烟雾配制物、胶囊悬浮液(CS)以及种子处理剂配制物。在任何实例中所选择的配制物类型取决于所设想的具体目的以及真菌的物理、化学和生物学特性。
组合物还可以呈液体、凝胶或固体形式。可以通过使固体载剂悬浮于一或多种活性成分的溶液中,并且在温和条件下干燥所述悬浮液(如在室温下蒸发或在65℃或更低的温度下真空蒸发)来制备固体组合物。组合物可以包含用凝胶囊封的活性成分。这类凝胶囊封的物质可以通过以下方式制备:混合胶凝形成剂(例如明胶、纤维素或木质素)与活的或灭活的真菌的培养物或悬浮液或真菌培养物或悬浮液的无细胞滤液或细胞溶离份,或喷雾或冷冻干燥的培养物、细胞或细胞溶离份或在本发明的方法中使用的抗菌化合物的溶液中;并且诱导试剂的凝胶形成。
组合物可以另外包含用于以下目的的表面活性剂:乳化、分散、湿润、扩散、整合、控制崩解、稳活性成分以及改善流动性或抑制衰退。在具体实施例中,表面活性剂是无植物毒性的非离子型表面活性剂,其优选属于EPA清单4B(以引用的方式并入本文中)。在另一个具体实施例中,非离子型表面活性剂是聚氧乙烯(20)单月桂酸酯。表面活性剂的浓度可以在全部配制物的0.1%-35%之间的范围内,优选范围是5-25%。通过组合物的性质和试剂促进本发明的组合物分散的能力来确定分散剂和乳化剂的选择,如非离子型、阴离子型、两性和阳离子型分散剂和乳化剂,以及所使用的量。

如上所述,上述组合物和物质可以用于调节植物、其种子、根部、果实、叶子、茎干、块茎中的柑橘黄龙病菌属感染量,并且具体地说,抑制和/或防止所述柑橘黄龙病菌属感染,具体地说,降低所述植物中所述柑橘黄龙病菌属感染的传播率和/或传播程度。并且,植物包括(但不限于)果实(例如草莓、蓝莓、黑莓、桃子和其它核果类果实)、植物(例如番茄、笋瓜、胡椒、茄子、马铃薯、胡萝卜)或谷物作物(例如大豆、小麦、大米、玉米、高粱)、树木、花、观赏类植物、灌木(例如棉花、玫瑰)、鳞基植物(例如洋葱、大蒜)或藤本植物(例如葡萄藤)、草皮、块茎(例如马铃薯、胡萝卜、甜菜)。或者,所述组合物可以用于调节植物中柑橘黄龙病菌属感染量并且具体地说,防止或抑制所述柑橘黄龙病菌属感染和/或降低所述植物中所述疾病感染的传播率和/或传播程度。并且,植物包括(但不限于)果实(例如草莓)、植物(例如番茄、笋瓜、胡椒、茄子)或谷物作物(例如大豆、小麦、大米、玉米)、树木、花、观赏类植物、灌木(例如棉花、玫瑰)、鳞基植物(例如洋葱、大蒜)或藤本植物(例如葡萄藤)。

投药方法
所属领域的技术人员了解向植物施用基于微生物的组合物以用于表面施用或吸收的各种方法,并且本发明中涵盖使用这些方法中的任一种。作为非限制性实例,对植物进行投药的方法包括施用于植物的任何部分,通过包含于灌溉水中、通过注射到植物中或注射到植物周围的土壤中、通过使根系统暴露于含有化合物的水性溶液、通过用于水耕或气培系统中、通过在含有诱导剂的培养基中培养个别植物细胞或植物细胞群、通过种子处理剂、通过使用于接枝的柑橘植物的插条暴露于含有化合物的水性溶液、通过施用于根部、茎干或叶子或通过施用于植物内部或所处理的植物的任何部分。涵盖所属领域的技术人员已知的任何手段。一种投药模式包括在植物根部处、植物根部上或植物根部附近施用组合物,或树干注射。
基于微生物的组合物的施用可以在苗圃环境、温室、水耕设备中,或在田野中,或在任何需要处理植物以防止可能的疾病或治疗疾病和其影响的环境中进行,例如在已暴露于或可能变成暴露于HLB或柑橘黄龙病菌感染的植物中。本发明的方法和化合物可以用于治疗由任何柑橘黄龙病菌物种或类型引起的感染并且可以用于改善未受感染的植物中的植物抵抗力。因此,在本发明的情形下,任何有需要的植物包括任何和所有需要改良健康和活力、生长和产率或抵抗疾病的能力的植物。本发明涵盖柑橘或其它对疾病(如HLB或由柑橘黄龙病菌物种引起的感染)敏感的植物,无论目前受到感染或具有潜在感染危险。

对水耕或培养基的施用可以如下进行,但也可以使用所属领域中已知的任何方法。可以将含有某一活性成分剂量的化合物的溶液或媒剂以适用于植物生长和发育的最终浓度添加到水耕或培养基中。浓度和体积可以视植物而变化。
对根部的施用可以通过在实验室、苗圃或水耕环境中将根部结构浸泡于溶液或媒剂中,或通过如上文所描述对根部周围的土壤进行土壤注射或土壤浇灌来进行。根部结构的浸泡可以如下进行,但也可以使用所属领域中已知的任何方法。可以通过使用根部给料器以0.5到1加仑/树木将含有某一活性成分剂量的化合物的溶液或媒剂施用于根部。浓度、体积和持续时间可以视植物而变化。
对植物的茎干或叶子的施用可以通过对植物的所需区域的喷雾或其它直接施用方法来进行,但也可以使用所属领域中已知的任何方法。对含有某一活性成分剂量的化合物的溶液或媒剂可以用喷雾器施用于茎干或叶子直到径流以确保完全覆盖,并且在生长季节中重复三次或四次。浓度、体积和重复处理可以视植物而变化。
在本发明中,以下表面活性剂可以与上文所提及的微生物发酵物一起使用以促进乳化、分散、溶解和渗透。
非离子型表面活性剂的非限制性实例包括脱水山梨糖醇脂肪酸酯、聚氧亚烷基脱水山梨糖醇脂肪酸酯、聚氧亚烷基脂肪酸酯、甘油脂肪酸酯、聚氧亚烷基甘油脂肪酸酯、聚甘油脂肪酸酯、聚氧亚烷基聚甘油脂肪酸酯、蔗糖脂肪酸酯、树脂酸酯、聚氧亚烷基树脂酸酯、聚氧亚烷基烷基醚、聚氧亚烷基烷基苯基醚、烷基(聚)糖苷和聚氧亚烷基烷基(聚)糖苷。优选地,可以使用不具有氮原子的含有醚基的非离子型表面活性剂和含有酯基的非离子型表面活性剂。
阴离子型表面活性剂的非限制性实例包括羧酸、磺酸、含有硫酸酯基的表面活性剂和含有磷酸酯基的表面活性剂,以及含有羧酸和磷酸酯基的表面活性剂。

实例1
因为柑橘黄龙病菌属不在受感染植物的外部活体外生长,使用新月裂杆菌(Liberibacter crescens)作为所属领域中已知的活体外模型,作为HLB抑制的指示。
在这种分析法中,通过使柑橘黄龙病菌在候选微生物滤液(无细胞)存在下,在不同浓度下生长2天以观察哪些抗菌剂和哪些浓度可以抑制生长来测试每种抗微生物化合物。通过将细菌/抗微生物混合物与PrestoBlue Cell Viability试剂(一种在活细胞存在下从蓝色和无荧光变成红色和显著荧光的荧光团)一起培育来定量生长量。使用荧光微板读数仪检测这种荧光变化并且显示于表1中。

实例2
温室分析法
将使用接枝后五到八个月并且未用系统性杀虫剂处理的年龄为一年的单茎干柑橘树进行分析法。在足够大的盆中提供树木,由此在本分析法持续期间不会变成根部缠绕。将树木保持在无木虱温室中并且在开始主动冲洗时输送。将树木与被柑橘黄龙病菌亚洲种感染的柑橘木虱(Diaphorina citri)一起置放于生长室中直到树木被均匀感染。在植物接种之后收集木虱的子样品以确认昆虫的柑橘黄龙病菌亚洲种感染率。接着,以物理方式移除木虱并且向树木施用杀虫皂,接着移送到无木虱温室中。
