一种用于提取和纸层析分离叶绿体中色素的提 层试剂的制备方法,它由直馏轻质汽油或石油醚20 份,常温下液态的无苯环无氯的酮类2~4份,常温下 液态的无苯环无氯的醇类0.3~0.6份混合而成。本 发明不含氯,不含苯类致癌物,毒性极弱,对常见绿色 植物叶绿体中色素的分离均理想,而且比以往分离出 来的色素种类多一种。只要注意,层析前,烘干或晾 干点样处色素,则成功率100%,而且时间短,3—5 分钟即可。

Description

本发明涉及一种用于提取和分离尤其纸层析分离叶绿体中色素的有机化学组合物-提层试剂的制备方法。
提取和分离叶绿体中的色素,是植物生理学领域中尤其中学生物学科中的一个重要学生实验。以往提取色素时用丙酮或酒精,但对眼、鼻、喉的刺激性太大,实验者易头昏头胀;纸层析分离色素,以往用四氯化碳,或由石油醚(60-90℃)20份,丙酮2份和苯1份混合而成,作为层析液,但或含氯化物,或含苯类致癌物,实验者出现胸闷、久之心律不齐等毒性现象,而且效果不理想:用于部分材料如上海青、萝卜叶、小白菜叶的实验,层析带上叶黄素难以分离;研磨绿叶加入碳酸钙,部分叶绿素分子形成了叶绿酸钙,因此,在层析时,部分叶绿素分子仍然停留在原点处,实验者往往得出错误结论:叶绿素b比叶绿素a多;叶绿体中只有叶绿素a、叶绿素b、叶黄素和胡萝卜素。
本发明的目的是提供一种无明显毒害的,适用范围更广的、效果更好的、同时用于提取和分离叶绿体中色素的提层试剂的制备方法。
本发明提层试剂组合方案为:将直馏轻质汽油或石油醚20份;常温下液态无苯环无氯的酮类2-4份、常温下液态的无苯环无氯的醇类0.3-0.6份,混合而成。再分装入试剂瓶中。
实施例:取60-90℃石油醚400毫升、丙酮(A.R.或C.P.)40毫升及95%或无水乙醇(A.R.或C.P.)6毫升混合于500毫升的棕色试剂瓶中,加内、外盖,静置或摇匀即成提层试剂。使用该试剂层析色素时,应注意烘干或晾干点样处色素后,再层析。
本发明的优点是:不含氯化物,不含苯类致癌物,利于实验者的身体健康,减少环境污染,对常见绿色植物叶绿体中色素的分离均理想,纸层析带上能显出五种色素带,点样处色素分子迁移较彻底。层析时间短(3-5分钟即可),成功率高,只要注意烘干或晾干点样处色素,再层析,则成功率100%。
本发明的产品在长沙县及长沙市二十多所高中生物“提取和分离叶绿体中的色素”,反映效果明显优于过去的实验,而且速度快,师生身体无不良反映。

Claims (4)

1、一种用于提取和分离叶绿体中色素的提层试剂的制备方法,其特征在于将三种原料按体积比配方为:直馏轻质汽油或石油醚20份、酮类2-4份、醇类0.3~0.6份,混合而成。
2、根据权利要求1所述配方,其特征在于酮类优选用常温下液态无苯环无氯的酮类化合物。
3、根据权利要求1所述配方,其特征在于在醇类优选用常温下液态的无苯环无氯的醇类化合物。
4、根据权利要求1所述的配方,其特征在于优选组份为:石油醚(60-90℃)20份,丙酮2.3份,乙酵0.3份,将其混合而成。
