我国医学对痔疮的病因认识较为深入,《素问》云:“风客淫气,精乃亡,邪伤肝也,顾而饱食,筋膜横解,肠僻乃痔”;《诸病源候论》记载“房事不慎”可损伤气血筋脉,“渗漏肠间,冲发下部”可致“诸痔”;劳伤筋脉则生热,热积于血,血得热则流散,渗入大肠,故大便血也;《医宗金鉴》云:“痔疮形名亦多般,不外风湿燥热源”,故而概之,以脏腑虚弱为本,感受风湿燥热邪气所致;痔的发生多因经脉壁薄弱,兼饮食不节,多食辛辣醇酒厚味,燥热内生,下迫大肠,乃久坐久蹲,负重远行,便秘怒责,妇女生育过多,致血性不畅,血热淤积,血热相搏,气血纵横,经脉交错,结滞不散,从而形成痔疮。
目前,治疗痔疮的方法有很多,主要采用手术和非手术治疗,因为手术治疗具有痛苦大,安全系数低,费用高,伤口愈合时间长等缺点,大多数患者不愿接受,而中医采用口服药和外用膏剂,大多存在治疗周期长、治疗效果不明显、难以根治;如专利CN201510554768.8公开一种痔疮外用组合物,采用络石藤、鱼腥草、苦豆子、甘草、陈皮、天蓬草、马齿苋等多种中药制备而成,但存在药材繁多,加大原料获取难度,提高了药物成本,不利于药物的推广,并且其治疗效果不理想,治疗周期长。
发明内容
为了解决现有技术中存在的上述技术问题,本发明提供了一种治疗痔疮的中药组合物,原料以重量份计为:柴草20-40份、血余炭20-40份、仙鹤草20-40份、无花果20-40份、马齿苋20-40份、连翘20-40份、野菊花20-40份、蒲公英20-40份。
优选地,所述原料以重量份计为:柴草30份、血余炭30份、仙鹤草30份、无花果30份、马齿苋30份、连翘30份、野菊花30份、蒲公英30份。
上述治疗痔疮的中药组合物的制备方法,包括以下制备步骤:
(1)按照配方量取各中药,备用;(2)取柴草、仙鹤草、马齿苋、蒲公英、无花果,混合后粉碎,加入闪式提取器内,加入其质量8-12倍的60-80%乙醇,处理60-100s,固液分离后,回收乙醇,浓缩成稠膏状;(3)取连翘和野菊花,混合后粉碎,按照料液比为1:(8-12)比例加水,进行匀浆处理,加入0.05-0.1%wt的复合酶,于35-45℃条件下处理30-60min,得到的酶解产物加入到闪式提取器内,处理60-100s,固液分离后,浓缩成稠膏状;(4)将步骤(2)和步骤(3)获得的产物混合后即可。
所述复合酶为纤维素酶、果胶酶和木瓜蛋白酶按照3:1:1的质量比组成。
上述方法制备的中药组合物可制成口服制剂和外用制剂,其中,口服制剂包括颗粒剂、胶囊剂、散剂、片剂中任一种,外用制剂包括膏剂、涂剂、栓剂中任一种;
上述中药组合物在治疗肛肠疾病中的应用,本发明药物用于内痔、外痔和混合痔的治疗外,对肛痿、肛门脓肿、肛周湿疹、肛周尖锐湿疣等肛肠疾病具有良好的治疗效果。
有益效果
①本发明以传统的辩证施治为原则,结合临床实践进行科学配伍,并反复确定各种中药的最佳使用剂量,对内痔、外痔以及混合痔具有良好的治疗效果,配方中柴草可透表泄热,疏气解郁,散火;血余炭、仙鹤草可收敛止血、活血化瘀、补虚止痢;无花果、马齿苋、蒲公英可清热解毒、散血消肿;连翘可清热解毒,消肿散结,野菊花可清热解毒、消肿止痛、疏风平肝;全方配伍合理,攻补互施,标本兼治。
②本发明通过将柴草、仙鹤草、马齿苋、蒲公英、无花果经过粉碎处理后,以质量分数为60-80%乙醇作为溶剂,结合闪式提取参数的控制,进而有效的萃取出药材中的有效成分;另外,通过对连翘和野菊花进行酶解,并结合复合酶成分的调整,有效的促使细胞壁破裂,充分溶出连翘和野菊花中的成分,再配合闪式提取工艺,进而使得有效成分的提取效率得到显著的提升,尤其是与柴草、仙鹤草、马齿苋、蒲公英、无花果的提取产物混合使用时,进而提升药物对痔疮的治疗效果,此外,对肛痿、肛门脓肿、肛周湿疹、肛周湿疣等也具有良好的治疗作用。
③本发明中药组合物临床有效率达到80%以上,具有治疗周期短、毒性低、治愈率高、不易复发、成本低的特点。
具体实施方式
下面结核具体的实施方式来对本发明的技术方案做进一步的限定,但要求保护的范围不仅局限于所作的描述。
实施例1
一种治疗痔疮的中药组合物,原料以重量份计为:柴草20g、血余炭20g、仙鹤草20g、无花果20g、马齿苋20g、连翘20g、野菊花20g、蒲公英20g;
制备方法:取各原料药,加入8-12倍量重量倍数的体积百分数为70-90%的乙醇,回流提取2-3h,滤过,药渣再加入6倍量重量倍数同等浓度的乙醇,回流提取1-2h,滤过,合并两次醇提取液,减压回收乙醇,并浓缩至相对密度为1.30-1.35的稠膏,加入甘油,制成膏剂,即得。
实施例2
一种治疗痔疮的中药组合物,原料以重量份计为:柴草40g、血余炭40g、仙鹤草40g、无花果40g、马齿苋40g、连翘40g、野菊花40g、蒲公英40g;
制备方法:取各原料药,加入8-12倍量重量倍数的水煎煮2-3次,合并煎煮液,用纱布过滤,加入甘油,制成涂剂。
实施例3
一种治疗痔疮的中药组合物,原料以重量份计为:柴草30g、血余炭30g、仙鹤草30g、无花果30g、马齿苋30g、连翘30g、野菊花30g、蒲公英30g;
制备方法:将上述原料分别粉碎成粒径为10-100微米的粉末,混合后加入液体石蜡搅拌均匀,备用;将40型混合脂肪酸甘油酯和甘油明胶熔化后,加入到前述的物料中混合均匀,然后将其倾入栓模中,室温冷却至凝固后,即得栓剂。
实施例4
一种治疗痔疮的中药组合物,原料以重量份计为:柴草25g、血余炭25g、仙鹤草25g、无花果25g、马齿苋25g、连翘30g、野菊花30g、蒲公英25g。
上述治疗痔疮的中药组合物的制备方法,包括以下制备步骤:
(1)按照配方量取各中药,备用;(2)取柴草、仙鹤草、马齿苋、蒲公英、无花果,混合后粉碎,加入闪式提取器内,加入其质量8倍的60%乙醇,处理60s,固液分离后,回收乙醇,浓缩成稠膏状;(3)取连翘和野菊花,混合后粉碎,按照料液比为1:8比例加水,进行匀浆处理,加入0.05%wt的复合酶,于35℃条件下处理30min,得到的酶解产物加入到闪式提取器内,处理60s,固液分离后,浓缩成稠膏状;所述复合酶为纤维素酶、果胶酶和木瓜蛋白酶按照3:1:1的质量比组成;(4)将步骤(2)和步骤(3)获得的产物混合后,喷雾干燥,粉碎,过80目筛,制成颗粒。
实施例5
一种治疗痔疮的中药组合物,原料以重量份计为:柴草35g、血余炭35g、仙鹤草30g、无花果30g、马齿苋30g、连翘25g、野菊花35g、蒲公英35g。
上述治疗痔疮的中药组合物的制备方法,包括以下制备步骤:
(1)按照配方量取各中药,备用;(2)取柴草、仙鹤草、马齿苋、蒲公英、无花果,混合后粉碎,加入闪式提取器内,加入其质量12倍的80%乙醇,处理100s,固液分离后,回收乙醇,浓缩成稠膏状;(3)取连翘和野菊花,混合后粉碎,按照料液比为1:12比例加水,进行匀浆处理,加入0.1%wt的复合酶,于45℃条件下处理60min,得到的酶解产物加入到闪式提取器内,处理100s,固液分离后,浓缩成稠膏状;所述复合酶为纤维素酶、果胶酶和木瓜蛋白酶按照3:1:1的质量比组成;(4)将步骤(2)和步骤(3)获得的产物混合后,喷雾干燥,粉碎,过120目筛,制成散剂。
在某些实施例中,本发明中药组合物还可制成片剂或胶囊剂。
实施例6
一种治疗痔疮的中药组合物,原料以重量份计为:柴草30g、血余炭30g、仙鹤草30g、无花果30g、马齿苋30g、连翘30g、野菊花30g、蒲公英30g。
上述治疗痔疮的中药组合物的制备方法,包括以下制备步骤:
(1)按照配方量取各中药,备用;(2)取柴草、仙鹤草、马齿苋、蒲公英、无花果,混合后粉碎,加入闪式提取器内,加入其质量10倍的75%乙醇,处理90s,固液分离后,回收乙醇,浓缩成稠膏状;(3)取连翘和野菊花,混合后粉碎,按照料液比为1:10比例加水,进行匀浆处理,加入0.08%wt的复合酶,于37℃条件下处理45min,得到的酶解产物加入到闪式提取器内,处理90s,固液分离后,浓缩成稠膏状;所述复合酶为纤维素酶、果胶酶和木瓜蛋白酶按照3:1:1的质量比组成;(4)将步骤(2)和步骤(3)获得的产物混合,喷雾干燥,粉碎,过100目筛,按照1:2重量配比加入85%蔗糖溶液,混合均匀制成糖浆剂。
实施例7
一种治疗痔疮的中药组合物,原料以重量份计为:柴草30g、血余炭30g、仙鹤草30g、无花果30g、马齿苋30g、连翘30g、野菊花30g、蒲公英30g。
上述治疗痔疮的中药组合物的制备方法,包括以下制备步骤:
(2)按照配方量取各中药,备用;(2)取柴草、仙鹤草、马齿苋、蒲公英、无花果,混合后粉碎,加入闪式提取器内,加入其质量10倍的75%乙醇,处理90s,固液分离后,回收乙醇,浓缩成稠膏状;(3)取连翘和野菊花,混合后粉碎,按照料液比为1:10比例加水,进行匀浆处理,加入0.08%wt的复合酶,于37℃条件下处理45min,得到的酶解产物加入到闪式提取器内,处理90s,固液分离后,浓缩成稠膏状;所述复合酶为纤维素酶、果胶酶和木瓜蛋白酶按照3:1:1的质量比组成;(4)将步骤(2)和步骤(3)获得的产物混合后,制成膏剂。
试验例1
试验选取贵州某肛肠医院从2014-1017收治的116例患者,均符合痔疮的诊断标准,其中男性68例,女性48例,年龄为30-70岁,临床症状主要包括大便出血、疼痛、直肠坠痛、肿物脱出、肛门分泌物外流等,临床诊断内痔42例,外痔26例,混合痔48例;将这116例患者随机分为4组,其中试验组使用本发明实施例1-实施例3制备的外用制剂,对照组采用专利CN201510554768.8制备的外用制剂;每组每日将药物涂抹在患处1次,连续使用10d,结果如下表1:
临床诊断标准:治愈:临床症状消失,痔核消失或全部萎缩;好转:临床症状减轻,痔核缩小或部分萎缩;无效:症状和体征均无变化;

由表1中试验结果可知,采用本发明实施例1-实施例3方案制备药物对痔疮患者的治疗效果明显优于对照组,其中以实施例3方案制备的药物效果最佳,总有效率达到84.4%。
此外,本发明还针对该中药组合物的制备工艺进行研究,采用实施例1-3实施例的原料组成,按照本发明方案的制备方法制备成膏剂,将67名患者随机分为A/B/C组,将制备的3种药物分别每日将药物涂抹在患处1次,连续使用10d,其中A组例数为23,治愈20例,好转3例,无效0,治愈率为87.0%;B组例数18,治愈16例,好转2例,无效1例,治愈率为88.9%;C组例数26,治愈24例,好转2例,无效0,治愈率为92.3;三组的总有效率均为100%,由此可见本发明制备工艺对于药效的提升是十分显著的。
试验例2
试验选用实施例7制备的药物为实验组,对照组A采用同实施例7相同的技术方案,区别在于原料中不添加连翘;对照组B采用同实施例7相同的技术方案,区别在于原料中不添加野菊花;试验选取贵州某肛肠医院从2013-2014年收治的84例患者,将制备的3种药物每日涂抹在患处1次,每日3次,连续使用10d,试验结果如下表2:

可见,本发明药材中连翘和野菊花的加入对该组合物的治疗效果具有十分重要的,单独加入连翘和单独加入野菊花对痔疮患者的治愈效果并不明显,而同时加入这两种成分后,使得治愈率达到96.2%,治疗效果突出。
此外,本申请人针对实施例4、实施例5方案制备的药物对于肛痿、肛门脓肿、肛周湿疹、肛周尖锐湿疣等肛肠疾病的治疗效果观察,实施例4方案制备的药物针对42例肛痿患者的治愈率达到78..6%,对27例肛门湿疹的治愈率达到81.4%,对22例肛周湿疹的治愈率达到77.3%,对19例肛周尖锐湿疣的治愈率达到73.7%,因此,本发明药物对于肛痿、肛门脓肿、肛周湿疹、肛周尖锐湿疣等具有良好的治疗效果。
试验例3
毒性试验:选用昆明种小白鼠50只,雌雄各半,体重为18-24g,随机分为5组,每组10只,服用本发明制备的就颗粒剂,采用口服灌胃法进行给药,5组给药的药量分别相当于原药材为30g/kg、25g/kg、15g/kg、5g/kg及生理盐水对照组,每组重复实验两次,观察给药后1周内动物死亡情况,结果动物无一死亡,进食、行为活动、体重正常;肉眼部检心、肝、脾、肺、肾等重要脏器,结果均无异常改变,进行解剖并作各脏器组织学检查,均未见异常病理学改变,未见对小鼠产生毒性反应。
在此有必要指出的是,以上实施例和试验例仅限于对本发明的技术方案做进一步的阐述和理解,不能理解为对本发明的技术方案做进一步的限定,本领域技术人员作出的非突出实质性特征和显著进步的发明创造,仍然属于本发明的保护范畴。
Claims (5)
1.一种治疗痔疮的中药组合物,其特征在于,原料以重量份计为:柴胡20-40份、血余炭20-40份、仙鹤草20-40份、无花果20-40份、马齿苋20-40份、连翘20-40份、野菊花20-40份、蒲公英20-40份;
所述治疗痔疮的中药组合物的制备方法,包括以下制备步骤:(1)按照配方量取各中药,备用;(2)取柴胡、仙鹤草、马齿苋、蒲公英、无花果,混合后粉碎,加入闪式提取器内,加入其质量8-12倍的60-80%乙醇,处理60-100s,固液分离后,回收乙醇,浓缩成稠膏状,(3)取连翘和野菊花,混合后粉碎,按照料液比为1:(8-12)比例加水,进行匀浆处理,加入0.05-0.1%wt的复合酶,于35-45℃条件下处理30-60min,得到的酶解产物加入到闪式提取器内,处理60-100s,固液分离后,浓缩成稠膏状;(4)将步骤(2)和步骤(3)获得的产物混合后即可。
2.如权利要求1所述治疗痔疮的中药组合物,其特征在于,所述原料以重量份计为:柴胡30份、血余炭30份、仙鹤草30份、无花果30份、马齿苋30份、连翘30份、野菊花30份、蒲公英30份。
3.如权利要求1所述治疗痔疮的中药组合物,其特征在于,所述复合酶为纤维素酶、果胶酶和木瓜蛋白酶按照3:1:1的质量比组成。
4.如权利要求1或2所述治疗痔疮的中药组合物,其特征在于,该中药组合物可制成口服制剂和外用制剂。
5.如权利要求4所述治疗痔疮的中药组合物,其特征在于,所述口服制剂包括颗粒剂、胶囊剂、散剂、片剂中任一种,外用制剂包括膏剂、涂剂、栓剂中任一种。
