背景技术
癌症是严重威胁人类生命健康的恶性疾病,每年会带走大量人的生命,给家庭、社会、国家造成了沉重的负担。由于癌症早期常无特殊症状,甚至毫无症状,目前常规的诊断方法诊断结果基本都为癌症中晚期,错过了最佳的治疗时机,因此癌症的死亡率非常高。长期以来,放化疗是癌症治疗的主要手段之一,然而,放疗和化疗在杀伤肿瘤细胞的同时,也将正常细胞和免疫细胞一同杀灭,导致患者免疫功能受损,免疫力低下,给患者的身体造成了极大的损害。
世界卫生组织在其公布的《癌症报告》中指出,1/3的肿瘤是可以预防的;1/3的肿瘤是可以通过早期发现、早期诊断、早期治疗而治愈的;1/3的肿瘤可以通过治疗减轻痛苦、延长生命,提高患者的生活质量。随着分子生物学和现代医学的发展,目前针对癌症早筛陆陆续续出现了一些新的技术方法,如循环肿瘤DNA(ctDNA)检测、循环肿瘤细胞(CTC)检测、DNA甲基化检测等。在现代分子生物学技术、精准医疗技术的支撑下,对癌症进行早筛,,尽可能较早发现癌症,进行及时的有效治疗如手术切除、放化疗、靶向治疗、中医中药治疗,在这些治疗的过程中,采用配方食物进行辅助治疗,能够提高治疗效果、减少毒副作用,延长治疗时间,具有重要的临床意义。
中医认为药食同源。合理健康的膳食不仅对正常人体健康有重要的意义,而且在恶性肿瘤病人治疗中也起着辅助的作用。CN107744137A公开了一种癌症患者专用食品及其制作方法,是由富硒菇粉,螺旋藻,桦褐孔菌提取物,金银花提取物,葡萄糖酸钾,葡萄糖酸钙和葡萄糖酸锌加工而成,该发明通过各种原料的有机配比,能够显著改善免疫功能,然而其抗癌效果未知。CN104116064B公开了一种具有预防癌症功效的草药料酒,由原金刚藤、桑黄、天荞麦、九里香、香蕈、白花蛇舌草、红曲、糯米和净泉水酿制而成,长期食用可以起到强化免疫力,诱导癌细胞自行死亡,抑制癌细胞的增殖及转移,减轻化疗和放疗的副作用,缓解癌症患者特有的疼痛,阻止溃疡、息肉、良性肿瘤等恶变为癌症,预防、避免癌症的复发。然而,受料酒食用方法的限制,该发明存在食用不方便,受众面较小的缺点。
发明内容
为解决现有技术中,癌症辅助治疗食品存在效果不理想,食用不方便的问题,本发明的目的之一是提供一种用于癌症的辅助治疗食品组合物。
本发明的目的之二是提供上述用于癌症的辅助治疗食品组合物的制备方法。
为实现上述目的,本发明采用以下技术方案:
一种用于癌症的辅助治疗食品组合物,原料包含以下重量份计的组分:
灵芝孢子粉 5~45份,
山药 10~20份,
葛根 5~25份,
魔芋 5~15份。
优选地,原料还包含以下重量份计的组分:
木糖醇 1~5份,
食盐 0.1~0.5份。
优选地,所述灵芝孢子粉为富硒灵芝孢子粉。
进一步优选地,所述富硒灵芝孢子粉的硒含量为3000~5000ug/kg。
优选地,所述山药的蛋白质含量≥55%,多糖含量≥10g/kg。
优选地,所述魔芋的魔芋多糖含量≥65%。
上述食品组合物的制备方法,步骤包括:按配比称取各原料组分,将山药、葛根和魔芋洗净干燥后粉碎、过筛,得原料粉,将原料粉与灵芝孢子粉混合均匀。
优选地,所述原料粉的目数为60~80目。
灵芝,别名仙草、神芝或芝草,为担子菌纲多孔菌科真菌赤芝或紫芝的干燥子实体,其性味甘平,具有扶正固本、滋补强壮等功效,药用价值高,在我国、朝鲜半岛、日本及东南亚等国家均有着广泛的应用。灵芝在生长成熟期,会从菌褶中弹射出极其微小的卵形生殖细胞,称为灵芝孢子或灵芝孢子粉。近年来,研究者对其进行了大量研究,发现灵芝孢子粉包含多种活性成分,如多糖、三萜、氨基酸、生物碱、甾醇和无机元素等,并从基因、蛋白或代谢通路等不同层次确证了抗肿瘤、抗癫痫、增强免疫、调节血糖血脂等一系列药理作用。目前,已有相应药品进入市场,并取得了较好的临床疗效。从细胞结构上看,灵芝孢子具有双层壁结构,由几丁质和葡萄糖构成,质地较为坚韧,占比达50%~70%。几丁质为广泛存在于自然界的一种含氮多糖类生物性高分子,研究表明具有抗肿瘤、抗炎和抗菌等作用。
硒是人体必需的微量元素。硒参与合成人体内多种含硒酶和含硒蛋白。其中谷胱甘肽过氧化物酶,在生物体内催化氢过氧化物或脂质过氧化物转变为水或各种醇类,消除自由基对生物膜的攻击,保护生物膜免受氧化损伤,硒参与构成碘化甲状腺胺酸脱碘酶。在体内硒和维生素E协同,能够保护细胞膜,防止不饱和脂肪酸的氧化。微量硒具有防癌作用及保护肝脏的作用。硒能提高人体免疫,促进淋巴细胞的增殖及抗体和免疫球蛋白的合成。硒对结肠癌、皮肤癌、肝癌、乳腺癌等多种癌症具有明显的抑制和防护的作用,其在机体内的中间代谢产物甲基烯醇具有较强的抗癌活性。硒与维生素E、大蒜素、亚油酸、锗、锌等营养素具有协同抗氧化的功效,增加抗氧化活性。同时,硒具有减轻和缓解重金属毒性的作用。
硒水平,尤其是硒蛋白的不足与许多人类的疾病相关,这些疾病包括癌症、糖尿病、心血管和免疫系统疾病。利用植物的生长特性,将人工施加的无机硒或某些有机硒吸收利用,在生物体内自然转化成为特有的硒形态,随同植物提供的食物、药物供给人们食用,不但达到补充硒的保健作用,而且结合原植物的功效成份,发挥更强的、甚至新的功效。
山药有“神仙之食”的美名。山药最大的特点是能防止心血管系统脂肪沉淀,保持血管的弹性,防止动脉粥样硬化过早发生,减少皮下脂肪沉积。山药块茎富含多糖,可刺激和调节人类免疫系统,因此常作增强免疫能力的保健药品使用。中医认为山药具有健脾、补肺、固肾、益精等多种功效。并且对肺虚咳嗽、脾虚泄泻、肾虚遗精、带下及小便频繁等症,都有一定的疗补作用。山药健脾、除湿、补气、益肺、固肾、益精的功效,山药含有可溶性纤维,能推迟胃内食物的排空,控制饭后血糖升高。还能助消化、降血糖。用于糖尿病脾虚泄泻,小便频数。控制进食欲作为高营养食品,山药中含有大量淀粉及蛋白质、B族维生素、维生素C、维生素E、葡萄糖、粗蛋白氨基酸、胆汁碱、尿囊素等。其中重要的营养成分薯蓣皂,是合成女性荷尔蒙的先驱物质,有滋阴补阳、增强新陈代谢的功效,而新鲜块茎中含有的多糖蛋白成分的粘液质、消化酵素等,可预防心血管脂肪沉积,有助于胃肠的消化吸收。
葛根为豆科植物野葛的干燥根,秋、冬二季采挖,其对癌症化疗引起的呕吐等副作用有极好的疗效并能同时起到辅助抑制肿瘤的效果。葛根中含有异黄酮、大豆素、大豆甙、葛根素、葛根素-7-木糖甙等物质,味甘辛,性凉,具有解肌退热、透疹、生津止渴、升阳止泻、降三高、美容、改善睡眠、调节情绪等功效。葛根为豆科植物野葛根,归脾、胃经。《神农本草经》记载葛根主治消渴,身大热,呕吐等,《药性论》曰葛根能治天行上气呕逆,开胃下食。
魔芋为天南星科植物,是我国民间传统的药食两用的植物。民间将魔芋还用于治疗脑瘤、甲状腺癌、淋巴肉瘤等。据《中药大辞典》记载:魔芋性温味辛,内服化痰散积,行瘀消肿,治疗痰咳、积滞、疟疾、经闭、肿瘤、糖尿病,还可以健胃、消饱胀、利尿和护发。外用可解毒消肿、治疗颈淋巴结结核、丹毒、跌打损伤、烫火伤和蛇咬伤等。近代我国各地方中药志及中草药书籍均把魔芋收入,研究发现,魔芋中含有60%左右的甘露聚糖(KGM),这种物质具有强大的膨胀力和吸水性,超过任何一种植物的粘韧度,KGM溶于水后会形成一种十分粘稠的溶胶,是魔芋中的主要成分,使得人食用后不在胃中消化,能有效地吸附胆固醇和胆汁酸,并能抑制肠道对胆固醇和胆汁酸的吸收,因此具有降脂、降血糖、降血压、开胃、通便、防癌等功能。同时它可填充胃肠,消除饥饿感,尤其所含热量微乎其微,故可控制体重,可以以调节营养平衡。
木糖醇是在自然界中分布范围很广,广泛存在于各种水果、蔬菜、谷类之中,但含量很低。商品木糖醇是将玉米芯、甘蔗渣等农业作物进行深加工而制得的,是一种天然、健康的甜味剂,用于矫味。
本发明的有益效果
1、本发明提供了一种用于癌症的辅助治疗食品组合物,该食品组合物营养丰富,富含多种药食同源中草药,搭配合理,食用方便,使用温水冲服即可;
2、本发明的食品组合物,可以作为癌症患者的辅助食品,对癌症有一定的辅助治疗作用或治疗作用,长期食用可以有效预防癌症的发生,降低癌症发病风险;
3、本发明组合物的制备工艺简单,质量可控,适合工业化大生产的需求;
4、现有技术中的相关产品中都含有药物,不属于癌症治疗中的食品辅助治疗范围,且长期服用可能不可避免产生毒副反应,本发明的食品组合物,不但可以用于癌症的辅助治疗,而且还可以用于癌症的预防。
具体实施方式
以下是本发明的具体实施例,并结合实施例对本发明的技术方案作进一步的描述,但本发明并不限于这些实施例。
实施例1
一种用于癌症的辅助治疗食品组合物,原料包含以下重量份计的组分:
灵芝孢子粉5份,山药10份,葛根5份,魔芋5份。
实施例2
一种用于癌症的辅助治疗食品组合物,原料包含以下重量份计的组分:
灵芝孢子粉45份,山药20份,葛根25份,魔芋15份。
实施例3
一种用于癌症的辅助治疗食品组合物,原料包含以下重量份计的组分:
灵芝孢子粉25份,山药15份,葛根15份,魔芋10份。
实施例4
用于癌症的辅助治疗食品组合物的制备方法,步骤包括:将山药、葛根和魔芋粉,按配比称取后过60-80目筛,得原料粉,再将原料粉与富硒灵芝孢子粉混合均匀,包装即得。
对比例1
一种用于癌症的辅助治疗食品组合物,原料包含以下重量份计的组分:
灵芝孢子粉2份,山药15份,葛根15份,魔芋10份。
对比例2
一种用于癌症的辅助治疗食品组合物,原料包含以下重量份计的组分:
灵芝孢子粉25份,山药15份,葛根15份。
对比例3
一种用于癌症的辅助治疗食品组合物,原料包含以下重量份计的组分:
灵芝孢子粉25份,山药15份,魔芋10份。
检测例1-动物急慢性毒性实验
预实验显示,本发明食品组合物,毒性很小,无法测出LD50,因此,按照最大的食用量喂食。
1、动物急性毒性试验
药物:通过本发明实施例4的方法,根据实施例1和实施例2配方制备所得食品组合物。
试验动物:昆明种小鼠,体重20±2g,雌雄各半。
方法与结果:
(1)将两种食品组合物各40g,分别用160ml温开水调制成糊状。
(2)将实验小鼠随机分成两组雌雄各半,分别喂食两种食品组合物糊,容积0.2-1.0ml/ 只,灌胃,密切观察,并作记录,继续观察14天;
(3)14天中,小鼠无死亡,行为活泼,摄食量如常,毛色光亮无疏松污,无惊厥嗜唾,眼、口、鼻、耳及肛周无异常分泌物,无断尾烂趾。分别从两组中选择10只解剖后,肉眼所见:心、脑、肝、肺、胃内及胸腺等重要脏器,未见异常改变,表明两种该食品组合物均无急性毒性。
2、动物长期毒性试验
药物:通过本发明实施例4的方法,根据实施例1和实施例2配方制备所得食品组合物。
动物:普通级SD大鼠,体重210g±10g,雌雄各半。
方法与结果:
(1)两种食品组合物的配置:均分为高、中、低三个剂量组,分别为患者食用量的240、 120、60倍,分别用温开水1:1混合成糊状;
(2)将实验鼠随机分成两组,分别连续喂食调制好的食品组合物,连续喂食180天,观察动物全身毒性反应及严重程度,解剖后按操作规程检查各部位,并进行血液学,ALT、BUN 及心、肝、脾、肺、肾、胃等主要脏器的病理学检查;
(3)经过长期喂食,两组大鼠均未出现毒性反应。发育良好。肉眼外观及主要脏器未见异常。外周血象及血清ALT、BUN与对照组比较无病理性改变。病理报告心、肝、脾、肺、肾、胃等均未有意义的改变,因此,认为经病理证实,两种食品组合物对动物无慢性毒性表现。
通过以上两个毒性实验,证明本发明通过本发明实施例4的方法,根据实施例1和实施例2配方制备所得食品组合物是安全的,无毒副作用的,可以被患者食用。
检测例2-小鼠抑瘤试验
本发明食品组合物对移植性小鼠Lewis肺癌抑瘤率影响试验。
1、研究目的
研究本发明食品组合物对腋下种植肺癌Lewis瘤株的小鼠的实体瘤抑制作用。
2、供试品及对照品
供试品:通过本发明实施例4的方法,根据实施例1~3和对比例1~3配方制备所得食品组合物。
对照品:替加氟,白色颗粒,室温,密闭避光保存。
3、试验动物及饲养方法
试验动物:SPF级KM小鼠80只,雌雄各半,4周龄,22±2g;
动物饲养环境条件
普通环境,20-25℃,日温差≤3℃;湿度40%-70%;动物照度:15-20lx;工作照度:150lx以上;试验开展前及试验结束后对房间进行彻底清洁消毒。每天全部作业结束后清扫、消毒。清洁消毒:笼架每天湿擦消毒,消毒剂为0.1%的新洁尔灭和75%的乙醇溶液,消毒剂每周交替使用。SPF鼠料,辐照饲料,常规饲养,自由采食,自由饮水。
4、试验方法
剂量设计:试验剂量设计见表1。
表1小鼠试验剂量及方式
取出10只小鼠做空白对照,将其余80只小鼠分别接种,然后随机分为8组,每组10只,雌雄各半。这8组分别为模型组、替加氟组、实施例1组、实施例2组、实施例3组、对比例1组、对比例2组、对比例3组。实施例1、2、3和对比例1、2和3组除用替加氟灌胃以外,每日喂食10g食品组合物,每日2次,5g/次,间隔8小时。
试验方法:
小鼠肺癌Lewis瘤株细胞培养于1640培养液中,37℃,5%CO2下常规培养,平均每两天传代一次,至对数生长期时用生理盐水制备成密度约为3.0×107个/ml单细胞悬液,在无菌条件下注射于小鼠腹腔内,接种后7天见小鼠腹腔明显肿大,此时,脱颈处死,放入盛有75%乙醇的烧杯中浸泡2~3分钟,将消毒后的小鼠放入超净工作台中,暴露腹部,用无菌注射器抽取腹水放入无菌试剂瓶内备用。将上述腹水用台盼蓝计数,用生理盐水稀释,
调整细胞数至1.0×107个/ml,接种于小鼠右腋下,每只0.2ml。
动物接种肿瘤后第四天开始给药,每天按表1灌胃给药,给药容积为10ml·kg-1;空白组灌胃等量的生理盐水。连续给药15天。
5、指标检测
体重检测频率:分组前及解剖前称量动物空腹体重;
肿瘤质量:
给药15天后结束试验,放血处死,剖检。称量重量的组织器官:腋下肿瘤;剖检方法:腹腔注射2%戊巴比妥钠(0.3ml·kg-1)麻醉,腹主静脉取血完毕后,剪断腹主动脉放血处死,然后进行常规腋下取瘤。
6、数据统计处理方法
试验数据以表示,采用SPSS11.5统计软件进行数据处理。
7、试验结果
实验结果显示(实验结果见表2):与替加氟组相比,本发明的食品组合物加入后抑瘤率明显提高,并具有显著性差异。其中实施例6所得食品组合物辅助治疗的效果与其它各组相比具有极显著性差异,治疗的效果最好,另外两个实施例组次之,不添加中药粉的实施例7 组和单纯中药粉的中药粉组效果较差。

8、试验结果的分析
从表2可以看出,本发明食品组合物在辅助替加氟对肿瘤进行治疗的时候,有效地阻抑了肿瘤的生长,效果显著。
Claims (8)
1.一种用于癌症的辅助治疗食品组合物,其特征在于,原料包含以下重量份计的组分:
灵芝孢子粉5~45份,
山药10~20份,
葛根5~25份,
魔芋5~15份。
2.根据权利要求1所述用于癌症的辅助治疗食品组合物,其特征在于,原料还包含以下重量份计的组分:
木糖醇1~5份,
食盐0.1~0.5份。
3.根据权利要求1所述用于癌症的辅助治疗食品组合物,其特征在于,所述灵芝孢子粉为富硒灵芝孢子粉。
4.根据权利要求3所述用于癌症的辅助治疗食品组合物,其特征在于,所述富硒灵芝孢子粉的硒含量为3000~5000ug/kg。
5.根据权利要求1所述用于癌症的辅助治疗食品组合物,其特征在于,所述山药的蛋白质含量≥55%,多糖含量≥10g/kg。
6.根据权利要求1所述用于癌症的辅助治疗食品组合物,其特征在于,所述魔芋的魔芋多糖含量≥65%。
7.如权利要求1~6任一项所述食品组合物的制备方法,其特征在于,步骤包括:按配比称取各原料组分,将山药、葛根和魔芋洗净干燥后粉碎、过筛,得原料粉,将原料粉与灵芝孢子粉混合均匀。
8.根据权利要求7所述食品组合物的制备方法,其特征在于,所述原料粉的目数为60~80目。
