Pharmacological toxicity test
I. Acute toxicity test: acute toxicity experiments using mice show that: the antibacterial peptide and Chinese herbal medicine compound preparation for preventing and treating pig diarrhea prepared in the embodiment 2 of the invention is orally taken by mice, under the dose of 488.8g crude drugs/kg, the slight activity of the mice is reduced after administration, the mice recover to normal after about 1 hour, no animal dies after continuous observation for 7 days after administration, the general condition, diet, water intake, urination and weight increase of the animals are all normal, and the experimental result shows that: the maximum dose of the compound preparation of the invention fed by oral administration to mice is 488.8g crude drug/kg/d (LD 50>488.8g crude drug/kg). The total daily clinical medication amount of the compound preparation is O.1g crude drugs/kg/d; the medicine has low acute toxicity and safe clinical medication.

And long-term toxicity test: 300 SD rats with good health state are selected, each half of the SD rats are divided into three groups, each group comprises 100 rats, the number of the males and the females in each group is the same, and the groups are respectively a group A, a group B and a group C, wherein the group A is a high-dose group, the group B is a medium-dose group, and the group C is a low-dose group. Group A, administering the antibacterial peptide and Chinese herbal medicine composite preparation for preventing and treating porcine diarrhea, which is prepared in the embodiment 2 of the invention and has the concentration of 18.0g crude drugs/kg, and feeding the composite preparation once a day for 90 days continuously, wherein after 24 hours after the last administration, each group kills 1/2 animals, and each half of the animals, and the rest 1/2 animals are killed after continuously observing for 2 weeks; group B, the antibacterial peptide and Chinese herbal medicine compound preparation for preventing and treating porcine diarrhea, which is prepared in the embodiment 2 of the invention and has the concentration of 6.0g crude drugs/kg, is fed once a day for 90 days continuously, and 24 hours after the last administration, each group kills 1/2 animals, and each half of the animals, and the rest 1/2 animals are killed after continuously observing for 2 weeks; group C the antibacterial peptide and Chinese herbal medicine composite preparation for preventing and treating porcine diarrhea prepared in example 2 of the present invention, which was given at a concentration of 2.Og crude drug/kg, was fed once a day for 90 consecutive days, 24 hours after the last administration, and each group survived killing 1/2 animals, half males and females, and the remaining 1/2 animals survived 2 weeks after observation. Other conditions are the same.
The animals were observed for changes in appearance, general behavior, food intake, and body weight during the test period, and examined for indices of hematology (RBC, HB, reticulocyte, PLT, CT, WBC, and classification) and blood biochemistry (AST, ALT, ALP, Glu, BUN, Crea, TP, T.BIL, ALB, CHOL), urine biochemistry, organ coefficients, histopathology, etc. 90 days after administration and 2 weeks after discontinuation.
The test result shows that: the compound preparation of the invention has good general state in high, medium and low dose animals, and has no abnormal change in appearance physical sign, behavior activity, food intake and weight growth; the three dose groups and the control group are in normal range of hematology examination, blood biochemistry examination and urine biochemistry examination, and have no obvious difference among the groups; no obvious abnormality is found in histopathological examination of main organs in each group, and the indexes are not changed after the medicine is stopped for 2 weeks. The test dosage is 180, 60 and 20 times of the clinical dosage, the continuous administration of the compound preparation for 90 days at high, medium and low dosages, namely 18.0g crude drug/kg, 6.0g crude drug/kg and 2.Og crude drug/kg, has no obvious influence on rats, has no specific toxic target organs and sensitive indexes, and has no delayed toxic reaction even in the recovery period, namely two weeks after stopping the administration, thereby prompting that the compound preparation has higher safety of clinical application dosage.

Diagnostic criteria for porcine diarrhea
1. The clinical symptoms comprise diarrhea of sick pigs, body temperature rise, mental depression, cold ears and nose, thin nasal discharge, fear of cold and tired lying, appetite reduction or abominability, daily diarrhea for more than 10 times, stinking water-like feces excretion, vomiting, high dehydration of organisms, emaciation and death.
2. And (3) autopsy lesions, such as post-mortem abdominal tympany, intestinal wall relaxation, gastrointestinal mucosa congestion, full stomach content, thin and full gas intestinal content, mesenteric lymph node swelling and juicy cut surface.
Hereinafter, the antibacterial peptide and Chinese herbal medicine composite preparation for preventing and treating porcine diarrhea prepared by the application is referred to as an antibacterial peptide and Chinese herbal medicine composite preparation for short, and the traditional Chinese medicine for preventing and treating porcine diarrhea prepared by the application is referred to as a Chinese herbal medicine prescription for short.
Test of influence of compound preparation of enterobacter peptide and Chinese herbal medicine added into daily ration on production performance and diarrhea of weaned piglets
A single-factor completely random design is adopted, healthy 30-day-old white piglets are selected, 180 piglets are selected, 12 piglets in each group are randomly divided into 15 groups, and the test period is 15 days. The conditions were the same for each group except for the daily dose. The administration of each group is shown in table 1 below.

By comparing table 1 and table 2, the average daily gains of the test 1 group, the test 3 group, the test 4 group, the test 5 group, the test 7 group, the test 9 group and the test 11 group which are fed with the enterobacter peptide and the test 13 group which is only added with the enterobacter peptide, and the test 2 group, the test 6 group, the test 8 group, the test 10 group and the test 12 group which are corresponding to the Chinese herbal medicine formula prepared by the same example, and the test 14 group and the control group of the colistin sulfate are respectively higher than that of the basic ration by 40g, 47g, 20g, 13g, 38g, 27g, 33g, 66g and 86g, and the feed-meat ratio is respectively reduced by 0.417, 0.383, 0.168, 0.111, 0.351, 0.236, 0.351, 0.316, 0.383 and 0.805. Therefore, the influence of the enterobactin peptide and Chinese herbal medicine composite preparation on the production performance and diarrhea of weaned piglets is obviously better than that of the enterobactin peptide, Chinese herbal medicine formula and 10% colistin sulfate which are fed separately, and the effect of the enterobactin peptide and Chinese herbal medicine composite preparation prepared in the embodiment 2 is optimal. Among the Chinese herbal medicine prescription sets, the Chinese herbal medicine prescription set prepared in example 2 is the most effective.
(II) test of influence of composite preparation of enterobacter peptide and Chinese herbal medicine added into daily ration on reproductive performance of sows
40 long white X approximately gram multiparous sows with close mating time, approximately same gestation times, approximately similar weight and good health condition are randomly selected and divided into two groups, each group has 4 repetitions, and each repetition has 5 repetitions. The test is divided into 2 groups, 1 group is a control group, and basic daily ration is fed; the treatment group 2 was a test group, and the pig farm was fed 5000 mg/kg of the enterobacter peptide and Chinese herbal medicine composite preparation prepared in ten examples 2 of the basic ration. Before the test, the sows are driven to a positioning fence for feeding, the pre-feeding period is 7d, and the test is completed from 90 d of sow pregnancy to the end of piglet weaning. The results of the effect of the enterobactin-Chinese herbal medicine compound preparation on the productivity of sows are shown in table 3 below.

(III) application case
160 piglets of the same pig farm, which are aged 6 days and have similar weight, are selected, 10 piglets in each group are randomly divided into 16 groups, and the test period is 5 days. The medicine is applied 1 time in the afternoon on day 5, 13 and twice a day from day 5, 14 until no diarrhea occurs in piglets. The conditions were the same for each group except for the daily dose. The administration and treatment effects of each group are shown in Table 4 below.

Through the experimental results of table 4, the enterobactin-Chinese herbal medicine composite preparation has a remarkable treatment effect on piglet dysentery, and the curative effect is obviously superior to that of the enterobactin-Chinese herbal medicine, norfloxacin and furazolidone which are singly used. The treatment effect of the enterobactin peptide and Chinese herbal medicine composite preparation prepared in the example 2 on piglet dysentery is optimal, and the Chinese herbal medicine formula prepared in the example 2 is optimal in the Chinese herbal medicine formula group.
The foregoing is only a preferred embodiment of the present invention, and it should be noted that, for those skilled in the art, various changes and modifications can be made without departing from the overall concept of the present invention, and these should also be considered as the protection scope of the present invention.
