Abstract
本发明的课题在于提供具有自然免疫活化作用的新型物质。本发明涉及含有具有自然免疫活化作用的多糖类、含有该多糖类作为有效成分的自然免疫活化剂、以及含有该多糖类的饮食品,其中,所述多糖类的特征在于,作为构成糖,含有25~50摩尔份的半乳糖醛酸、15~50摩尔份的半乳糖、0~7摩尔份的葡萄糖、0~30摩尔份的阿拉伯糖、0~6摩尔份的木糖和3~15摩尔份的鼠李糖,作为主链,含有具有α‑1,4键合的半乳糖醛酸的聚半乳糖醛酸链。

技术领域
本发明涉及具有自然免疫活化作用的多糖类、以及含有该多糖类的自然免疫活化剂或饮食品。
背景技术
人等高等脊椎动物具有自然免疫和获得免疫这两种免疫机制(免疫体系),通过两种免疫机制适当地作用,进行对感染源的防御。另一方面,昆虫类等其余的众多生物不具有获得免疫机制,仅以自然免疫机制进行对感染源的防御。
自然免疫机制是生物共同具有的感染防御机制,是非特异性的,因此具有反应快速、对大量感染源有效地发挥功能的特征。对于人等高等脊椎动物而言,从感染初期的抵抗性、癌症及生活习惯病的预防、组织修复等的观点出发,可以认为,相比于对感染源特异性的感染免疫,非特异性的自然免疫显示出更为重要的位置。
因此,自然免疫是在传染病初期的机体防御机制的重要的宿主因子,可以认为自然免疫的活化对于传染病的预防和治疗有效。另外,近年来,对抗癌症的药物开发的重要性也不断提高,以来自蘑菇类的多糖类具有的抗肿瘤活性为代表的自然免疫活化能力受到关注。
目前为止,本发明的发明人等发现了,如果在蚕的肌肉标本中注射来自真菌的β-葡聚糖或来自细菌的肽聚糖等自然免疫活化物质,则作为昆虫细胞因子的麻痹肽被诱导、自然免疫得以活化的同时,会发生肌肉收缩(专利文献1、非专利文献1~3)。另外,建立了以体长因该肌肉的收缩而发生变化为指标的、简便的自然免疫活化物质的探索体系,在目前为止从绿茶等提取的多糖类中发现了自然免疫活化能力(非专利文献4、5)。
经验上已知蔬菜对于摄取维持人类健康所必须的养分而言很重要,而另一方面,作为传统的药草利用也在世界范围内进行,被认为存在含有自然免疫活化物质的可能性,但迄今为止还未能明确地知晓被视作具有活化自然免疫功能的作用的物质。
目前为止,本发明的发明人发现了西兰花的提取物显示出自然免疫活化作用(专利文献1),但具体西兰花中的何种成分直接参与自然免疫活化作用到目前为止并未查明。因此,当然也没有查明该成分(化合物)的哪个部位(化学结构)可能具有自然免疫活化作用。
因此,不仅仅是上述西兰花的提取物,将来自生物的提取成分(化合物)的化学键分解等而仅取出有效部位(化学结构)作为自然免疫活化剂的尝试也几乎没有。换言之,几乎没有以来自生物的提取成分(化合物)为原料来制备(合成)具有对自然免疫活化有效的化学结构的自然免疫活化剂的做法。

发明内容
发明要解决的问题
本发明是鉴于上述背景技术而完成的,其课题在于提供具有自然免疫活化作用的新型物质。
解决问题的方法
本发明的发明人为了解决上述问题,经过反复深入研究的结果,使用本发明的发明人确立的蚕肌肉收缩活性测定法,从西兰花提取物纯化自然免疫活化物质并进行了结构解析。此外,对于其活性所必须的结构也进行了研究。
其结果,通过纯化西兰花的提取物,发现了在迄今为止结构未知的多糖类中存在具有自然免疫活化作用的物质。
此外发现,作为上述多糖类的构成成分的聚半乳糖醛酸链对于自然免疫活化作用发挥着重要作用,从而完成了本发明。
即,本发明提供一种具有自然免疫活化作用的多糖类,其特征在于,作为构成糖,含有25~50摩尔份的半乳糖醛酸、15~50摩尔份的半乳糖、0~7摩尔份的葡萄糖、0~30摩尔份的阿拉伯糖、0~6摩尔份的木糖、及3~15摩尔份的鼠李糖;作为主链,含有具有α-1,4键合的半乳糖醛酸的聚半乳糖醛酸链。
另外,还提供一种自然免疫活化剂,其含有上述多糖类作为有效成分。
另外,提供一种饮食品,其含有上述多糖类。
发明的效果
按照本发明,可以提供具有新型化学结构的具有自然免疫活化作用的多糖类。
另外,可以提供含有该多糖类作为有效成分的自然免疫活化剂、含有该多糖类的具有自然免疫活化作用的饮食品。
另外,本发明的多糖类是极为安全且无副作用的物质。并且,既容易制成各种剂形,也容易添加到饮食品中,因此作为安全的自然免疫活化剂或功能性食品,能够极为有效地活化自然免疫。

发明的具体实施方式
以下将对本发明进行说明,本发明不限于以下的具体实施方式,可以在本发明的技术范围内任意地变形。
<多糖类>
本发明的多糖类具有自然免疫活化作用,其特征在于,
作为构成糖,含有25~50摩尔份的半乳糖醛酸、15~50摩尔份的半乳糖、0~7摩尔份的葡萄糖、0~30摩尔份的阿拉伯糖、0~6摩尔份的木糖、及3~15摩尔份的鼠李糖,
作为主链,含有具有α-1,4键合的半乳糖醛酸的聚半乳糖醛酸链。
本发明的多糖类必须含有25~50摩尔份的半乳糖醛酸、15~50摩尔份的半乳糖、0~7摩尔份的葡萄糖、0~30摩尔份的阿拉伯糖、0~6摩尔份的木糖及3~15摩尔份的鼠李糖作为构成糖。
优选含有25~45摩尔份的半乳糖醛酸、20~50摩尔份的半乳糖、1~7摩尔份的葡萄糖、0~20摩尔份的阿拉伯糖、0~6摩尔份的木糖、及4~14摩尔份的鼠李糖作为构成糖。
更优选含有25~40摩尔份的半乳糖醛酸、30~50摩尔份的半乳糖、3~7摩尔份的葡萄糖、0~10摩尔份的阿拉伯糖、0~5摩尔份的木糖、及5~13摩尔份的鼠李糖作为构成糖。
特别优选含有30~40摩尔份的半乳糖醛酸、35~45摩尔份的半乳糖、5~6摩尔份的葡萄糖、0~5摩尔份的阿拉伯糖、0~5摩尔份的木糖、及6~12摩尔份的鼠李糖作为构成糖。
本发明的多糖类中,在不损害本发明的效果的范围内,也可以含有上述单糖以外的单糖。
本发明的多糖类含有具有α-1,4键合的半乳糖醛酸的聚半乳糖醛酸链作为主链。聚半乳糖醛酸链中,在不损害本发明的效果的范围内,也可以含有半乳糖醛酸以外的单糖。
作为该“半乳糖醛酸以外的单糖”,可以列举例如鼠李糖等。
另外,就该“聚半乳糖醛酸链”中的半乳糖醛酸(单元)的键合比例而言,相对于该“聚半乳糖醛酸链”整体,优选为20摩尔%~99摩尔%,更优选为30摩尔%~97摩尔%,特别优选为40摩尔%~95摩尔%。
如果作为主链的“聚半乳糖醛酸链”中的半乳糖醛酸(单元)的键合比例过少,则上述多糖类的自然免疫活化作用有时会降低。另一方面,如果过多,则难以得到上述本发明的多糖类。
本发明的多糖类中不存在半乳糖醛酸甲酯,由此与果胶不同,为具有新型结构的多糖类。此外,由于是从天然物质中提取出具有自然免疫活化作用的化学结构而得到了“本发明的多糖类”,因此是新型物质。
本发明的多糖类是通过纯化西兰花提取物而被发现的。西兰花提取物由多种成分构成。从其成分中鉴定出具有自然免疫活化作用的成分并不容易。
本发明是通过数次采用道义上的问题少且简便的蚕肌肉收缩活性测定法分别测定上述“西兰花提取物的多种成分”的自然免疫活化作用才得以完成的。进一步,由该成分分解而提取出化学结构(单元),同样地数次采用蚕肌肉收缩活性测定法从大量化学结构(单元)中发现了具有自然免疫活化作用的化学结构(单元),进而完成了本发明。
本发明的多糖类可以是通过纯化、分解等由天然物质得到的源自天然物质的多糖类、以天然物质为原料进行化学修饰而得到的多糖类等,也可以是完全由合成得到的多糖类。
<自然免疫活化剂>
本发明的自然免疫活化剂的特征在于,含有上述多糖类作为有效成分。
本发明的自然免疫活化剂中作为有效成分而包含的多糖类既可以是天然的多糖类,也可以是合成的多糖类。
本发明的自然免疫活化剂中的作为有效成分的多糖类相对于自然免疫活化剂整体的含量没有特别限制,可以根据目的而适当确定。将自然免疫活化剂整体设为100质量份时,以多糖类的合计量计,优选为0.01~100质量份的含量,更优选为0.1~99质量份,特别优选为1~95质量份,进一步优选为10~90质量份的含量。
本发明的自然免疫活化剂中除了作为有效成分的上述多糖类以外,还可以含有“其他成分”。
作为该“其他成分”,没有特别限制,在不损害本发明的效果的范围,可以根据目的适当选择,可以列举例如药学上可接受的载体等。
作为这样的载体,没有特别限制,例如可以根据后述的剂型等适当选择。另外,作为该自然免疫活化剂中的该“其他成分”的含量,也没有特别限制,可以根据目的适当选择。
作为本发明的自然免疫活化剂的剂型,没有特别限制,例如可以根据如后所述所希望的给药方法适当选择。
具体可以列举例如口服固形剂(片剂、包衣片剂、颗粒剂、散剂、胶囊剂等)、口服液剂(内服液剂、糖浆剂、酏剂等)、注射剂(溶剂、悬浮剂等)、软膏剂、贴剂、凝胶剂、霜剂、外用散剂、喷雾剂、吸入散布剂等。
作为上述口服固形剂,例如可以在上述有效成分中加入赋形剂、并进一步根据需要加入结合剂、崩解剂、润滑剂、着色剂、矫味/矫臭剂等添加剂,根据常规方法制造。
作为上述赋形剂,可以列举例如乳糖、白糖、氯化钠、葡萄糖、淀粉、碳酸钙、高岭土、微晶纤维素、硅酸等。
作为上述结合剂,可以列举例如水、乙醇、丙醇、单糖浆、葡萄糖液、淀粉液、明胶液、羧甲基纤维素、羟丙基纤维素、羟丙基淀粉、甲基纤维素、乙基纤维素、虫胶、磷酸钙、聚乙烯吡咯烷酮等。
作为上述崩解剂,可以列举例如干燥淀粉、海藻酸钠、琼脂粉、碳酸氢钠、碳酸钙、十二烷基硫酸钠、硬脂酸单甘油酯、乳糖等。
作为上述润滑剂,可以列举例如精制滑石、硬脂酸盐、硼砂、聚乙二醇等。

作为上述贴剂,例如可以在公知的支撑体上利用常规方法涂布作为上述软膏剂的霜剂、凝胶剂、糊剂等来制造。作为上述支撑体,可以列举例如由棉、短纤维、化学纤维制成的织布、无纺布、软质氯乙烯、聚乙烯、聚氨酯等的膜、发泡片等。
本发明的自然免疫活化剂例如可以通过在需要使自然免疫机制活化的个体(例如,需要健康维持或疲劳恢复的个体;需要预防或治疗癌症或生活习惯病的个体;被细菌、真菌、病毒等感染的个体等)给药来使用。
作为本发明的自然免疫活化剂的给药对象动物,没有特别限制,可以列举例如:人;小鼠;大鼠;猴;马;牛、猪、羊、鸡等家畜;狗、猫等宠物等。
另外,作为上述自然免疫活化剂的给药方法,没有特别限制,例如可以根据上述自然免疫活化剂的剂型等适当选择,可以列举口服给药、腹腔内给药、向血液中注射、向肠内注入等。
另外,作为上述自然免疫活化剂的给药量,没有特别限制,可以根据作为给药对象的个体的年龄、体重、所希望的效果的程度等适当选择,例如就对成人的1天的给药量而言,以作为有效成分的上述多糖类整体的量计,优选为1mg~30g,更优选为10mg~10g,特别优选为100mg~3g。
另外,作为上述自然免疫活化剂的给药时期,也没有特别限制,可以根据目的适当选择,例如可以预防性地给药,也可以治疗性地给药。
<饮食品>
本发明的饮食品的特征在于,含有上述多糖类或上述本发明的自然免疫活化剂。
本发明的饮食品具有自然免疫活化作用。
含有上述多糖类或上述自然免疫活化剂的饮食品(以下,有时简记为“本发明的饮食品”)中的多糖类或自然免疫活化剂的含量没有特别限制,可以根据目的或饮食品的形态(种类)适当选择,在将饮食品整体设为100质量份时,以上述自然免疫活化剂的合计量计,优选以0.001~100质量份含有,更优选为0.01~100质量份,特别优选为0.1~100质量份的含量。
另外,可以单独使用多糖类或自然免疫活化剂中的任何1种,也可以组合使用2种以上。在组合使用2种以上的情况下,上述饮食品中的各个物质的含量比没有特别限制、可以根据目的适当选择。
本发明的饮食品除了上述多糖类或上述本发明的自然免疫活化剂以外,还可以含有“其他成分”。
作为具有这样的自然免疫活化作用的本发明的饮食品中的上述“其他成分”,没有特别限制,可以在不损害本发明的效果的范围内根据目的适当选择,可以列举例如各种食品原料等。另外,“其他成分”的含量没有特别限制,可以根据目的适当选择。
作为上述饮食品的种类,没有特别限制,可以根据目的适当选择,可以列举例如果冻、糖果、巧克力、饼干等点心类;绿茶、红茶、咖啡、冷饮等适口饮料;发酵乳、酸奶、冰激凌等乳制品;蔬菜饮料、果实饮料、果酱类等蔬果加工品;汤等液体食品;面包类、面类等谷物加工品;各种调味料等。
作为这些饮食品的制造方法,没有特别限制,例如可以根据通常的各种饮食品的制造方法适当制造。
另外,上述饮食品例如可以制成片剂、颗粒剂、胶囊剂等口服固形剂或内服液剂、糖浆剂等口服液剂制造。上述口服固形剂、口服液剂的制造方法没有特别限制,可以根据目的适当选择,例如可根据上述药剂的口服固形剂、口服液剂的制造方法制造。
上述饮食品作为以自然免疫机制的活化为目的的功能性食品、健康食品等被认为是特别有用的。
在饮食品的制造中使用本发明的多糖类或自然免疫活化剂的情况下,制造方法可以利用本领域技术人员公知的方法进行。只要是本领域技术人员,就可以将下述工序适当组合来制作目标的饮食品,所述工序包括:将本发明的多糖类与其他成分混合的工序、成形工序、灭菌工序、发酵工序、烧制工序、干燥工序、冷却工序、造粒工序、包装工序等。
实施例
以下,列举实施例、比较例、试验例更加具体地说明本发明,本发明只要不超出其主旨,就不限定于这些实施例等。
<热水提取>
切好各种蔬菜后,加入水进行密蒸处理(121℃、20分钟)。自然冷却后离心(8000rpm、10分钟),将上清液作为“热水提取物”。
<蚕肌肉收缩活性测定法>
将样品溶解于缓冲液后,向蚕的肌肉标本注射100μL,测定肌肉的收缩。在用注射前后的肌肉标本的长度之差除以注射前的长度得到的值(比活性或C值(Contractionvalue,收缩值))显示为0.15以上的情况下,判定为具有自然免疫活化能力。
<DEAE-纤维素柱吸附级分的纯化>
对西兰花的热水提取物进行乙醇沉淀。热水提取方法按照专利文献1记载的方法。将所得到的沉淀溶解于Milli Q水,对Milli Q水进行透析后,进行了冷冻干燥。将其供于DEAE-纤维素(DEAE-cellulose)柱色谱,对各级分进行了基于苯酚-硫酸法的还原糖量的测定。收集糖质的洗脱的峰,进行透析后,进行了冷冻干燥。
<DEAE-纤维素柱吸附级分的结构解析>
[[1.单糖组成分析]]
利用三氟乙酸使DEAE-纤维素柱吸附级分水解,对水解产物进行氨基苯甲酸乙酯标记,进行利用ODS柱的HPLC分析。对于将作为标准单糖的L-阿拉伯糖(Ara)、L-岩藻糖(Fuc)、D-半乳糖(Gal)、D-葡萄糖(Glc)、D-甘露糖(Man)、L-鼠李糖(Rha)、D-核糖(Rib)、D-木糖(Xyl)、D-半乳糖醛酸(GalA)、D-葡萄糖醛酸(GlcA)、N-乙酰基-D-半乳糖胺(GalNAc)、N-乙酰基-D-葡糖胺(GlcNAc)、N-乙酰基-D-甘露糖胺(ManNAc)混合得到的试样,进行同样的处理,并进行了保留时间的比较。
[[2.NMR分析]]
将27mg的DEAE-纤维素柱吸附级分于40℃溶解于重水(D2O),进行了1H、13C NMR和DQF-COSY、HMQC、HMBC、TOCSY分析。
[[3.甲基化分析]]
对DEAE-纤维素柱吸附级分进行甲基化后,利用三氟乙酸进行了水解。将其乙酰化后,制成糖醇乙酸酯,利用GCMS进行了分析。
<DEAE-纤维素柱吸附级分的选择性分解>
[[1.基于草酸水解的阿拉伯糖的选择性分解]]
对DEAE-纤维素柱吸附级分进行草酸水解,中和后,进行了透析、冷冻干燥。
[[2.基于果胶酶的聚半乳糖醛酸链的分解]]
对DEAE-纤维素柱吸附级分进行了果胶酶(Sigma公司)处理。通过加热使酶失活后,供给至Bio-gel P4凝胶过滤柱色谱,对各级分进行了基于苯酚-硫酸法的还原糖量的测定。Bio-gel P4凝胶过滤柱使用1150nm×15mmφ的柱,并使用0.2M乙酸作为洗脱液。收集糖质的洗脱的峰,进行了减压干燥。
实施例1
[包含自然免疫活化物质的蔬菜的探索]
针对17种蔬菜(西兰花、卷心菜、胡萝卜、青椒、罗汉果、菠菜、大蒜、南瓜、姜、圣女果、白萝卜、豆芽、香菜、黄瓜、茄子、葱、白菜)的热水提取物,进行了使用蚕肌肉收缩活性测定法的自然免疫活化能力的比较。

表1是对17种蔬菜的热水提取物使用蚕肌肉收缩活性测定法得到的比活性(C值)的结果。在比活性(C值)显示为0.15以上的情况下,判定为具有自然免疫活化能力。由表1的结果确认到西兰花的热水提取物具有强的自然免疫活化能力。
实施例2
[来自西兰花的自然免疫活化物质的纯化]
以蚕肌肉收缩活性为指标从西兰花的热水提取物纯化了自然免疫活化物质。表2是各纯化阶段的比活性(C值)的结果。

对热水提取物进行乙醇沉淀的结果,由于沉淀物中活性被回收(表2),因此表明活性物质为多糖类。接着进行了DEAE-纤维素柱色谱,结果发现,根据NaCl的梯度而洗脱出均匀的糖质的峰,在该级分中确认到了比活性的上升(表2)。
接着,针对该级分研究了用量反应性。将其结果示于图1。图1中,纵轴表示比活性(C值),横轴表示DEAE-纤维素吸附级分(μg)。
将C值(Contraction value,收缩值)显示为0.15的活性定义为1unit时,显示出了67unit/mg的比活性(图1)。在本实施例中,将该级分作为纯化级分进一步尝试了分析。
实施例3
[DEAE-纤维素柱吸附级分的结构解析]
为了解明所得DEAE-纤维素柱吸附级分的单糖组成,利用HPLC分析了三氟乙酸水解物。将分析结果示于图2。图2a为对标准样品的HPLC分析的结果,图2b为对DEAE-纤维素柱吸附级分的HPLC分析的结果。
图2中,1表示D-葡萄糖醛酸,2表示D-半乳糖醛酸,3表示D-半乳糖,4表示D-甘露糖,5表示D-葡萄糖,6表示L-阿拉伯糖,7表示D-核糖,8表示N-乙酰基-D-甘露糖胺,9表示D-木糖,10表示N-乙酰基-D-葡糖胺,11表示L-岩藻糖,12表示L-鼠李糖,且13表示N-乙酰基-D-半乳糖胺。
图2的结果,检测到了半乳糖醛酸(GalA)、半乳糖(Gal)、葡萄糖(Glc)、阿拉伯糖(Ara)及鼠李糖(Rha),其摩尔比为GalA:Gal:Glc:Ara:Rha=12.4:4.9:1.0:7.3:1.2。
接着,尝试了基于NMR分析的糖链结构的解析。将结果示于图3。
1DNMR分析的结果,没有检测到蛋白质和脂质的特征信号,显示出了糖质的特征图谱(图3)。
接着,进行各种2DNMR分析,进行检测到的信号的归属,汇总于表3。

作为主要构成成分的GalA(半乳糖醛酸)的信号强度强,能够确定形成了α-1,4键合的聚半乳糖醛酸链。关于含量第二高的Ara(阿拉伯糖),能够归属部分信号,由其化学位移值表明形成了α-1,5键合,但在单糖组成分析中检测到的其他较小糖残基的信号强度弱,无法进行全部信号的归属。
因此,尝试了利用甲基化分析来确定糖键合位置。将结果示于图4。图4中,1表示T-Araf,2表示T-Rhap,3表示3-Araf,4表示5-Araf,5表示T-Glcp,6表示T-Galp,7表示3,5-Araf,8表示2,4-Rhap,9表示4-GalpA,10表示4-Galp,11表示3-Galp,12表示6-Galp,及13表示3,6-Galp。
图4的结果,检测到了13种部分甲基化糖醇乙酸酯的峰。从各峰的保留时间和MS碎片模式确定糖种类和键合位置,汇总于表4。
表4中,例如“T-Araf”表示非还原末端阿拉伯糖,“3-Galp”表示3-键合半乳糖,“3,5-Araf”表示3,5-键合阿拉伯糖。

通过甲基化分析,作为主要构成成分检测到了1→4键合的半乳糖醛酸(GalA)和1→5键合的阿拉伯糖(Ara),支持了NMR分析的结果。另外,关于在单糖组成分析中检测到的其他较小糖残基,也能够确定键合位置。
实施例4
[自然免疫活化能力所需要的DEAE-纤维素柱吸附级分的结构]
为了确定自然免疫活化能力所需要的DEAE-纤维素柱吸附级分的结构,制备选择性分解作为主要构成成分的阿拉伯糖和半乳糖醛酸而得到的结构修饰物,利用蚕肌肉收缩活性测定评价了自然免疫活化能力。将其结果示于表5。

首先,尝试了基于草酸水解的阿拉伯糖(Ara)的选择性分解、除去。单糖组成分析的结果,Ara含量明显减少(表5)。
另外,将上述草酸水解物于70℃溶解于重水(D2O),进行了NMR分析。将结果示于图5。
1DNMR分析的结果,阿拉伯糖的信号消失(图5)。另外,对该样品(草酸水解物)进行蚕肌肉收缩活性测定的结果,自然免疫活化能力丧失(表5)。
接着,利用果胶酶处理尝试了聚半乳糖醛酸的分解。将分解产物利用Bio-gel P4凝胶过滤柱色谱进行分级的结果示于图6。图6中的1~4分别对应于表5中的“Peak1”~“Peak4”。
图6的结果,检测到4个峰。对这些峰分别进行单糖组成分析的结果(表5),由Peak1和2检测到了GalA、Gal、Glc、Ara、Rha和Xyl。由Peak3未检测到了GalA,检测到了Gal、Glc、Ara、Rha和Xyl。由Peak4检测到了经果胶酶处理而游离出的GalA单体。对各峰进行蚕肌肉收缩活性测定的结果(表5),伴随低分子化,自然免疫活化能力降低,由Peak3和Peak4没有检测到活性。
<实施例的总结和考察>
使用蚕肌肉收缩活性测定法对17种蔬菜的热水提取物尝试自然免疫活化物质的探索的结果,确认到了西兰花具有高活性。而且,通过本实施例,由西兰花纯化出了具有自然免疫活化能力的DEAE-纤维素柱吸附级分。
尝试DEAE-纤维素柱吸附级分的结构解析的结果,表明其为包含GalA、Gal、Glc、Ara和Rha的酸性多糖类,作为主要结构,是具有α-1,4键合的聚半乳糖醛酸链作为主链、具有α-1,5键合的阿拉伯聚糖链作为侧链的结构。
根据上述结果,可以认为DEAE-纤维素柱吸附级分为作为植物细胞壁成分的果胶的一种,但NMR分析的结果,一般地几乎检测不到果胶所含的GalA甲酯的信号,这表明是与目前为止有报道的果胶不同的结构。
为了确定自然免疫活化能力所需要的DEAE-纤维素柱吸附级分的结构,将作为主要构成成分的阿拉伯糖和半乳糖醛酸选择性分解的结果,通过利用果胶酶处理将聚半乳糖醛酸链分解,自然免疫活化能力伴随低分子化而降低、消失。
将由实施例的结果预测的、关于本发明的多糖类的结构和该多糖类的处理物的结构的一例的示意图示于图7。本发明的多糖类的结构不限于图7的示意图所示的结构。
目前为止,大量报道了从药草等各种植物提取的果胶具有免疫活化能力。而另一方面,涉及到免疫活性与结构的关系的报道则很少,有报道称果胶所含的半乳聚糖结构对于活性是必须的。
然而,就本实施例中纯化的DEAE-纤维素柱吸附级分而言,由于聚半乳糖醛酸结构对于活性是必须的,因此表明,具有与目前为止有报告的果胶的免疫活化能力不同的作用机制。
工业实用性
根据本发明,可以得到活化自然免疫功能的效果高的多糖类,因此能够广泛利用于各种饮食品、片剂、颗粒、咀嚼片等。另外,本发明的多糖类特别适合作为机体内具有自然免疫机能活化作用的功能性食品、特定保健用食品、具有免疫功能低下问题的病人饮食或护理饮食等的原材料加以利用。此外,也可以作为药物的原材料而得以广泛利用。
Claims (4)
1.一种具有自然免疫活化作用的多糖类,其中,
作为构成糖,含有30~40摩尔份的半乳糖醛酸、35~45摩尔份的半乳糖、5~6摩尔份的葡萄糖、0~5摩尔份的阿拉伯糖、0~5摩尔份的木糖、及6~12摩尔份的鼠李糖,
作为主链,含有具有α-1,4键合的半乳糖醛酸的聚半乳糖醛酸链,
其中所述多糖类是从西兰花获得的,并且
其中所述聚半乳糖醛酸链中所述半乳糖醛酸单元的比例为20摩尔%至99摩尔%。
2.根据权利要求1的多糖类,其是通过柱色谱从西兰花的热水提取物中纯化并用草酸水解获得的。
3.一种自然免疫活化剂,其含有权利要求1或2所述的多糖类作为有效成分。
4.一种饮食品,其含有权利要求1或2所述的多糖类。
