Abstract
本发明提供了一种缓解视疲劳的保健品,以丹参提取物、木瓜提取物、蓝莓提取物、决明子提取物、枸杞子提取物、黄芪提取物、淀粉制备而成。标志性成分及含量:每100g含丹参酮ⅡA不得少于60mg,花青素不得少于120mg。其制备方法是:将丹参、木瓜、蓝莓、决明子、枸杞子、黄芪分别加水煎煮、滤过、合并提取液,制成浸膏,干燥。加入淀粉,混合,制粒、整粒、灌装、包装。本发明质量稳定可靠,安全有效,成本较低,便于存储和服用。
Description
技术领域
本发明涉及中药领域,具体涉及一种缓解视疲劳的保健品及其制备方法。
背景技术
视力疲劳导致的眼疲劳、眼干涩 、异物感、眼皮沉重感、视物模糊、畏光流泪、眼胀痛及眼部充血等,严重者还可出现头痛、头昏、恶心、精神萎靡、注意力不集中、记忆力下降、食欲不振、以及颈肩腰背酸痛和指关节麻木等全身症候群能够使人心情烦躁,生活质量降低,长期而导致整体健康状况下降。是影响人们心理,生理,健康的重要因素。目前,市场上能够缓解视力疲劳的保健食品并不常见,而且多是化学药物合成,中药材为原料的提取物缓解视疲劳并不多见。常见的剂型有颗粒剂、胶囊剂、片剂、口服液、茶剂等。已有的缓解视力疲劳的中药保健品中药材利用率不高,组方简单,疗效不够明确,可测有效成分含量较低,缺乏严格的质量标准和规范的质量控制体系。
发明内容
本发明的目的是提供一种能够提高视力、缓解视力疲劳功效的保健品及其制备方法。本发明的药物以蓝莓,丹参为主药,蓝莓富含花青素,具有活化视网膜功效,可以强化视力,防止眼球疲劳。丹参有活血化瘀之功效,其中中富含丹参酮ⅡA,具有修复视神经,缓解视神经疲劳的效果。木瓜:包含各种酶元素,维生素(维他命)及矿物质,而最丰富的维生素是维生素A、复合维生素B、维生素C及维生素E。而有研究表明:维生素C和E可排除人体内不正常堆积的氧化物,避免组织破坏。可预防早发性白内障和夜间视力不良和干眼症等病症,对于视网膜具有不可缺乏的保护作用。枸杞子:是补肾之剂,性味平和,滋阴养血,益睛明目。古代文献中早已记载对老年视力衰退有很好的作用。决明子:又称“还瞳子”,含有多种维生素和丰富的氨基酸、肪、碳水化合物等,具有清肝火、祛风湿、益肾明目等功能,常饮决明子茶,可使血压正常,大便通畅,老眼不花。临床实验证明,决明子茶可以清肝明目 、防止视力模糊。决明子可防治夜盲症等。黄芪有补气、养血、益中功效能够使诸药有运行的气血之本。本发明的药物通过各类药物的相互照应,调动了药物的效能,增强了本发明的药物治缓解视力疲劳的作用
本发明的一种缓解视疲劳的保健品包括以下重量的原料:一日配方原料量约2.9g,即丹参提取物1g、木瓜提取物0.5g、蓝莓提取物0.4g、决明子提取物0.4g、枸杞子提取物0.3g、黄芪提取物0.2 g,加入适量辅料制成胶囊剂,其标志性成分及含量:每100g含丹参酮ⅡA不得少于60mg,花青素不得少于120mg。主要有效成分清楚,含量高且相对稳定,质量可以控制。
本发明的一种缓解视疲劳的保健品的制备方法的技术方案是:提供一种缓解视疲劳的保健品-其标志性成分及含量:每100g含丹参酮ⅡA不得少于60mg,花青素不得少于120mg。优选的是每100g含丹参酮ⅡA为60mg-140mg,花青素为120mg-230 mg。
上述缓解视疲劳的保健品的制备方法,包括:a、将丹参、木瓜、蓝莓、决明子、枸杞子、黄芪分别加水煎煮、滤过、合并提取液,制成浸膏,干燥。b、加入淀粉,混合。c、制粒、整粒。d、灌装、包装。
所述的缓解视疲劳的保健品的制备方法可以直接在缓解视疲劳的保健品以及药物中应用。加上食品或是药品辅料或是载体,制备成口服剂。优选剂型包括:片剂、胶囊、或口服液。
本发明缓解视疲劳的保健品有效成分明确,质量稳定可控,安全有效,成本低廉。

具体实施方式
下面结合具体实施例对本发明做进一步的详细说明,以下实施例是对本发明的解释,本发明并不局限于以下实施例。
将丹参、木瓜、蓝莓、决明子、枸杞子、黄芪六种原材料分别经过挑拣水洗备用;将原料投入提取罐中,第一次按重量加入10倍水,浸泡0.5小时,提取1.5小时;第二次按重量加8倍水,提取1小时;第三次按重量加6倍水,提取1小时。200目滤过,合并滤液。水为纯化水,应符合GB/T17324—2003的要求。浓缩,将滤液减压浓缩(-0.08MPa,70℃)成浸膏(d=1.10—1.15, 65℃);喷雾干燥;粉碎过80目筛;按丹参、木瓜、蓝莓、决明子、枸杞子、黄芪按照药物质量标准分别检验;检验合格后在洁净区内包装。
含量测定方法:
丹参酮ⅡA的测定:
按照高效液相色谱法(中华人民共和国药典2010版一部VID)进行测定。
仪器与试药:
岛津LC-20AT高效液相色谱仪(SPD-20A检测器,LC-20A泵,LC-20B泵);依利特色谱柱(Hypersil ODS2 5μm,尺寸4.6mm×250mm),DK-98-Ⅱ电热恒温水浴锅(天津市泰斯特仪器有限公司);T-214电子分析天平(北京赛多利斯仪器有限公司);
甲醇(一级色谱纯 Honeywell Burdick & Jackson );水为重蒸馏水;丹参酮ⅡA对照品(批号:110766-200619,由中国食品药品检定研究院提供)。
色谱条件:
以十八烷基硅烷键合硅胶为填充剂;以甲醇-水(75:25)为流动相;进样量:10µL;流速:1ml/min;检测波长为270nm。理论板数按丹参酮ⅡA峰计算应不低于2000。
对照品溶液的制备:
取对照品适量,精密称定,置棕色量瓶中,加甲醇制成每1ml含丹参酮ⅡA15μg的溶液,作为对照品溶液。
供试品溶液的制备:
丹参因含有对热不稳定成分,本研究对其提取时间进行了优选,选定甲醇为溶剂,分别加热回流45min、60min、75min,经试验研究,加入甲醇50ml,加热回流60min,可较充分地提取本品中丹参酮ⅡA,使测定结果更准确合理,且该方法成熟,重现性好,故确定为供试品的制备方法。即:取本品内容物约0.5g,精密称定,置具塞锥形瓶中,精密加入甲醇50ml,称定重量,加热回流1小时,放冷,再称定重量,用甲醇补足减失的重量,摇匀,滤过,取续滤液,即得。
空白实验:
按配料比例称取除丹参提取物以外的其它原、辅料,按供试品制备方法制成缺丹参提取物的阴性样品,同法制成阴性样品溶液;再按上述色谱条件,吸取供试品溶液、对照品溶液、阴性样品溶液各10μl,分别注入液相色谱仪中,测定,记录色谱图,分别见图1、图2、图3。结果供试品色谱中,在与对照品峰相同保留时间处,有相同的色谱峰,而阴性样品无干扰峰,如图1所示。
精密度试验:
精密吸取对照品溶液10μl,按上述色谱条件重复进样6次,记录峰面积,结果如表1所示,丹参酮ⅡA的RSD为0.80%,表明仪器的精密度良好。

稳定性试验:
精密吸取供试品溶液(批号:20140226),分别于0、2、4、6、8、10小时进样10μl,记录峰面积,结果如表2所示,测得丹参酮ⅡA的RSD为0.72% (n=6),表明供试品溶液至少在10小时内基本稳定。

重复性试验:
称取同一批样品(批号:201402264)约0.5g,共6份,精密称定,按供试品溶液的制备方法,制备6份供试品溶液,依法进行测定,结果见表3所示,丹参酮ⅡA平均含量为:142.12mg/100g,RSD=1.01%(n=6),表明本方法重复性好。

样品测定:
精密称取三批中试样品(批号:20140701、20140702、20170703),按上述制备项下方法,制备供试品溶液,分别精密吸取对照品溶液与供试品溶液各10μl,按上述色谱条件测定,以外标一点法计算样品中丹参酮ⅡA的含量,三批样品测定结果见表4。

上述实验证明本发明的缓解视力的保健品成分明确,质量稳定可靠,制备过程可以重复,含量稳定。
实施例1
一种缓解视疲劳的保健品的制备方法,如图4所示:
将丹参、木瓜、蓝莓、决明子、枸杞子、黄芪六种原材料分别投入提取罐中,第一次加入10倍水(分别按对应原材料的重量计),浸泡0.5小时,提取1.5小时;第二次加8倍水,提取1小时;第三次加6倍水,提取1小时。200目滤过,合并滤液。水为纯化水,应符合GB/T17324—2003的要求。浓缩,将滤液减压浓缩(-0.08MPa,70℃)成浸膏(d=1.10—1.15, 65℃);喷雾干燥;粉碎过80目筛;按丹参、木瓜、蓝莓、决明子、枸杞子、黄芪按照药物质量标准分别检验;检验合格后在洁净区内包装。
取丹参提取物105-115重量份、木瓜提取物50-60重量份、蓝莓提取物40-50重量份、决明子提取物40-50重量份、枸杞子提取物30-40重量份、黄芪提取物30-40重量份,合并,然后加入淀粉33-43重量份,混合30分钟,加95%食用酒精,用量约20%,挤压通过 16目筛,50℃干燥约6小时,控制水分≤6.5%,24目筛整粒,备用。取干颗粒,混合约10min,得总混颗粒。灌装与包装。
优选的,制成0.36g/粒,配方中丹参提取物:木瓜提取物:蓝莓提取物:决明子提取物:枸杞子提取物:黄芪提取物:淀粉约为111.1:55.5:44.4:44.4:33.3:33.3:38。即:

实施例2

实施例3
取实施例1或2制备的丹参提取物、木瓜提取物、蓝莓提取物、决明子提取物、枸杞子提取物、黄芪提取物加水1000ml,PH值调为7,搅拌,过滤,灌装,每支15ml,灭菌,用法用量为每次1支,一日三次。
对实施例 1 所得到的药物做试验结果如下:
对150例16-30岁的经常性视力疲劳人群进行了市场应用调查研究,对给予接收调查的人群每日3次,每次3粒;口服。其中总有效率95.5%,满意度达到91%。
对400例16-45岁的经常性视力疲劳人群进行了市场应用调查研究,对给予接收调查的人群每日3次,每次3粒;口服。其中总有效率93%。满意度达到90%。
对506例16-65岁的经常性视力疲劳人群进行了市场应用调查研究,对给予接收调查的人群每日3次,每次3粒;口服。其中总有效率89.6%。满意度达到88.2%。
Claims (9)
1.一种缓解视疲劳的保健品,其特征是:以重量比计,每100g含丹参酮ⅡA不少于60mg,花青素不少于150mg。
2. 根据权利要求1所述的缓解视疲劳的保健品,其特征在于:以重量比计,每100g含丹参酮ⅡA为60mg-140mg,花青素为120mg-230 mg。
3.根据权利要求1所述的一种缓解视疲劳的保健品的制备方法,其特征是,包括以下步骤:a、将丹参、木瓜、蓝莓、决明子、枸杞子、黄芪分别加水煎煮、滤过、合并提取液,制成浸膏,干燥;b、加入淀粉,混合;c、制粒、整粒;d、灌装、包装。
4.根据权利要求3所述的一种缓解视疲劳的保健品的制备方法,其特征是:所述步骤a中的丹参、木瓜、蓝莓、决明子、枸杞子、黄芪分别提取,然后干燥,粉碎过80目筛。
5. 根据权利要求3所述的一种缓解视疲劳的保健品的制备方法,其特征是:所述步骤b中,混合30分钟,加入酒精为95%食用酒精,用量为20%,挤压通过 16目筛,50℃干燥6小时,控制水分≤6.5%,24目筛整粒。
6. 根据权利要求5所述的一种缓解视疲劳的保健品的制备方法,其特征是:所述步骤b中,取丹参提取物105-115重量份、木瓜提取物50-60重量份、蓝莓提取物40-50重量份、决明子提取物40-50重量份、枸杞子提取物30-40重量份、黄芪提取物30-40重量份,合并,然后加入淀粉33-43重量份,混合30分钟,加95%食用酒精,用量约20%,挤压通过 16目筛,50℃干燥约6小时,控制水分≤6.5%,24目筛整粒。
7.根据权利要求5所述的一种缓解视疲劳的保健品的制备方法,其特征是:所述步骤b中,所述丹参提取物:木瓜提取物:蓝莓提取物:决明子提取物:枸杞子提取物:黄芪提取物:淀粉的重量比为111.1:55.5:44.4:44.4:33.3:33.3:38。
8.根据权利要求2-7任一项所述的一种缓解视疲劳的保健品的制备方法,其特征是:所述丹参、木瓜、蓝莓、决明子、枸杞子、黄芪的提取物,再加上辅料或载体,制备成口服剂。
9.根据权利要求8所述的一种缓解视疲劳的保健品的制备方法,其特征是:所述口服剂是片剂、胶囊或者口服液。
