Abstract

本发明公开了一种调节肠道菌群和防治肥胖的组合物及其应用,组合物包括低聚半乳糖、抗性糊精、绿茶提取物、葡萄籽提取物、左旋肉碱、果蔬粉、蘑菇粉和燕麦。本发明的组合物热量低、适用于各种人群长期服用的能调节肠道菌群和防治肥胖的组合物,具有显著的减肥效果,且能有效强化肠道功能,并具有一定的保健作用。本发明的组合物可用于防治便秘和肥胖的保健品、药物或食品。

技术领域
本发明涉及保健品领域,尤其涉及一种调节肠道菌群和防治肥胖的组合物及其应用。
背景技术
随着现代的生活节奏逐渐加快,压力增大以及物质条件的丰富,肥胖正越来越成为困扰现代人的重要健康问题,肥胖越来越呈现年轻化的趋势。肥胖是二型糖尿病、心血管疾病等的重要风险因素之一,同时肥胖也会严重影响个人形象,造成相应的社会、心理方面的问题,然而目前对于肥胖的机制认识依然不够清晰,除了遗传因素与社会环境因素之外,目前的研究表明,肠道菌群与肥胖之间存在着密切的关系。
人体肠道内大约存在着1014个微生物,每个人的菌群构成均有所不同,其构成与生活习惯,饮食和宿主的基因密切相关。厌氧菌如拟杆菌属、真杆菌属、双歧杆菌属、梭菌属、消化链球菌属和奇异菌属占了消化道菌群的主要部分,而兼性厌氧菌如乳酸杆菌,肠球菌,链球菌,肠杆菌等只占了很小的一部分。肠道菌群与宿主构成微生态系统,参与营养物质吸收、免疫调节、能量代谢等生命活动,构成肠道黏膜的保护屏障,而肠道菌群的失调与肥胖、免疫低下、炎性肠病、2型糖尿病、肿瘤、抑郁症的发生发展有着密切联系,随着检测技术的发展,肠道微生物与宿主健康和疾病的关系成为研究的热点。
目前的研究结果表明,肥胖人群的肠道菌群厚壁菌门与拟杆菌门的比值升高,而某些有益菌如Akk菌,普氏栖粪杆菌、乳酸杆菌、双歧杆菌等丰度下降,同时肠道菌群与肥胖相关的激素如脂联素、瘦素以及血清代谢物存在着相关性。将诱导肥胖小鼠的菌群移植到正常小鼠体内可诱导正常小鼠肥胖,将肥胖人群的菌群移植到正常小鼠体内可诱导正常小鼠肥胖。因此通过开发产品用于调控肠道菌群结合其他调控成分共同抑制肥是非常有必要和有前景的。
发明内容
本发明要解决的技术问题是克服现有技术的不足,提供一种热量低、适用于各种人群长期服用、对人体无害的能调节肠道菌群和防治肥胖的组合物,该组合物具有显著的减肥效果,且能有效强化肠道功能,并具有一定的保健作用。
为解决上述技术问题,本发明采用以下技术方案:
调节肠道菌群和防治肥胖的组合物,所述组合物包括低聚半乳糖、抗性糊精、绿茶提取物、葡萄籽提取物、左旋肉碱、果蔬粉、蘑菇粉和燕麦、菊粉和聚葡萄糖。
上述的调节肠道菌群和防治肥胖的组合物,优选地,所述组合物中,按质量比计,低聚半乳糖∶抗性糊精∶绿茶提取物∶葡萄籽提取物∶左旋肉碱∶果蔬粉∶蘑菇粉∶燕麦:菊粉:多聚葡萄糖为1.5~5∶2~4∶0.04~0.1∶0.04~0.1∶0.02~0.04∶1~3∶1~3∶5~7∶1.2~2.4∶1.4~2.8。
上述的调节肠道菌群和防治肥胖的组合物,优选地,按质量百分含量计,所述组合物中包括低聚半乳糖7.5~25%、抗性糊精10~20%、绿茶提取物0.2~0.5%,葡萄籽提取物0.2~0.5%、左旋肉碱0.1~0.2%、果蔬粉5~15%、蘑菇粉5~15%、燕麦25~35%、菊粉6~12%和聚葡萄糖7~14%。
上述的调节肠道菌群和防治肥胖的组合物,优选地,按质量百分含量计,所述组合物中包括低聚半乳糖15%、抗性糊精15%、绿茶提取物0.25%、葡萄籽提取物0.25%、左旋肉碱0.15%、果蔬粉10%、蘑菇粉10%、燕麦30%、菊粉8%和聚葡萄糖11.35%。
上述的调节肠道菌群和防治肥胖的组合物,优选地,所述组合物中还含有保健品或药物可接受的添加剂。
上述的调节肠道菌群和防治肥胖的组合物,优选地,所述组合物为片剂、丸剂、胶囊、颗粒剂或粉剂。
作为一个总的发明构思,本发明还公开了前述的组合物在防治便秘和肥胖的保健品、药物或食品中的应用。
上述的应用,优选地,所述食品为牛奶、酸奶或茶叶。
与现有技术相比,本发明的优点在于:
本发明经过大量研究,得到一种合适的成分和配比的有效调节肠道菌群和防治肥胖的组合物,组合物受试者在连续服用该组合物2个月后,肠道内的乳酸杆菌、双歧杆菌、普氏栖粪杆菌和阿克曼菌丰度显著增高,服用4个月后,肠道内的这几种细菌丰度仍维持相对稳定;普氏栖粪杆菌可以调节肠道免疫系统、氧化应激和结肠上皮细胞的代谢,还可以发酵产生丁酸,使肠道的酸碱度降低,有效地促进肠道内正常菌群(如双歧杆菌等)生长,阿克曼菌誉为减肥菌。组合物中的各成分有机配合,同时实现了提高肠道中乳酸杆菌、双歧杆菌、普氏栖粪杆菌和阿克曼菌这4种菌中多种菌的丰度、促进肠道蠕动、促进脂肪燃烧、减少热量摄入,多重作用多管齐下,具有显著的减肥作用,并且还能强化肠道功能,还具有一定的抗氧化和其他保健效果,肥胖试用者在连续服用组合物2月后体重降低了4~7.5kg,连续服用4月后体重降低6~12kg,减肥效果显著,该组合物热量含量低,对身体无害,适合长期服用。

具体实施方式
以下将结合说明书附图和具体实施例对本发明做进一步详细说明。
实施例
本实施例中,选取23名肥胖人群,编号为1-23,按照30g/天的服用量连续服用组合物4个月,要求餐前服用,组合物的成分如下:按质量百分含量计,含低聚半乳糖(15%)、抗性糊精(15%)、绿茶提取物(0.25%)、葡萄籽提取物(0.25%)、左旋肉碱(0.15%)、果蔬粉(10%)、蘑菇粉(10%)和燕麦(30%),菊粉8%,聚葡萄糖11.35%。
低聚半乳糖,抗性糊精,菊粉:具有降脂和促进肠道有益菌增殖的作用;绿茶提取物:富含绿茶多酚,抗氧化,促进脂肪燃烧;葡萄籽提取物:富含葡萄多酚,抗氧化,促进Akk菌的生长;左旋肉碱:促进脂肪的燃烧,促进减肥;果蔬粉、燕麦:富含膳食纤维及维生素,促进肠道蠕动,减少热量摄入,促进益生菌的生长;蘑菇粉:富含壳聚糖,促进益生菌生长,减少热量摄入,聚葡萄糖:为大分子聚合糖,促进肠道蠕动及有益菌增殖。
在未服用组合物前采取1次粪便,服用过程中,每服用2个月采取一次粪便,采用16S高通量测序检测肠道菌群的变化,同时测量其体重与计算BMI,得到的数据如下表1-4所示。
组合物干预效果验证:
本组合物服用后有饱腹感,会降低食欲,图1为肥胖人员在服用组合前,服用组合物2月和4月之后,对应的体重和BMI值变化,可见在服用组合物之后,肥胖人员的体重和BMI值均明显降低,表明在持续服用组合物具有较显著的减肥效果。
表1为23名便秘人员在未服用组合前,服用2个月和4个月后,所取粪便中测得的乳酸杆菌、双歧杆菌、普氏栖粪杆菌和阿克曼菌丰度,从表1中可看出,服用组合物2个月之后,所取粪便中的四种细菌中的多种丰度显著增高,证实产品对肠道菌群有显著的有益调节作用,在服用4个月之后,与第2个月相比,这几种益生菌的丰度依然维持在一个较高的水平。

表2为对表1中的数据进行整理得到,通过对干预前、干预2个月和干预4个月对应四种有益菌的丰度进行对比,通过秩和检验检验它们之间的差异是否具有统计学意义,一般认为P值小于或等于0.05,则对比的两组差异是显著的,可见干预2个月、干预4个月与干预前相比,这4种菌的丰度均呈增加趋势。平均值为每种菌丰度的平均值统计数据,从表2可得出在干预2个月和4个月后有益菌的丰度与干预前相比,显著增加,干预2个月和4个月相比,各细菌丰度维持在相对稳定的状态。

表3为31名肥胖人员在服用组合物前、服用2个月和4个月后测得的体重及BMI值,从表3中可看出,服用2个月之后,受试者的体重和BMI显著降低,服用4月后受试者体重和BMI进一步降低,表明该组合物的效果是持续起作用的。

表4为对表3的数据进行进一步整理得到,通过对23名肥胖人员干预前、干预2个月、干预4个月的体重进行对比,通过秩和检验检验它们之间的差异是否具有统计学意义,一般认为P值小于或等于0.05,则对比的两组差异是显著的,可见干预2个月、干预4个月与干预前相比,所有受试人员的体重和BMI均降低。平均值为体重与BMI的平均值统计数据,从表4中可得出,干预2个月和4个月体重与BMI干预前相比,显著降低,2个月和4个月相比,体重进一步降低,表明其作用是持续的。

本组合物热量含量低,适合各个人群的长期服用,本品可与加入牛奶、酸奶或茶叶等中作为食品添加剂使用,也可用于保健品或药品。

1.调节肠道菌群和防治肥胖的组合物,其特征在于,所述组合物包括低聚半乳糖、抗性糊精、绿茶提取物、葡萄籽提取物、左旋肉碱、果蔬粉、蘑菇粉和燕麦、菊粉和聚葡萄糖。
2.根据权利要求1所述的调节肠道菌群和防治肥胖的组合物,其特征在于,所述组合物中,按质量比计,低聚半乳糖∶抗性糊精∶绿茶提取物∶葡萄籽提取物∶左旋肉碱∶果蔬粉∶蘑菇粉∶燕麦:菊粉:多聚葡萄糖为1.5~5∶2~4∶0.04~0.1∶0.04~0.1∶0.02~0.04∶1~3∶1~3∶5~7∶1.2~2.4∶1.4~2.8。
3.根据权利要求2所述的调节肠道菌群和防治肥胖的组合物,其特征在于,按质量百分含量计,所述组合物中包括低聚半乳糖7.5~25%、抗性糊精10~20%、绿茶提取物0.2~0.5%,葡萄籽提取物0.2~0.5%、左旋肉碱0.1~0.2%、果蔬粉5~15%、蘑菇粉5~15%、燕麦25~35%、菊粉6~12%和聚葡萄糖7~14%。
4.根据权利要求2所述的调节肠道菌群和防治肥胖的组合物,其特征在于,按质量百分含量计,所述组合物中包括低聚半乳糖15%、抗性糊精15%、绿茶提取物0.25%、葡萄籽提取物0.25%、左旋肉碱0.15%、果蔬粉10%、蘑菇粉10%、燕麦30%、菊粉8%和聚葡萄糖11.35%。
5.根据权利要求1~4任意一项所述的调节肠道菌群和防治肥胖的组合物,其特征在于,所述组合物中还含有保健品或药物可接受的添加剂。
6.根据权利要求1~4任意一项所述的调节肠道菌群和防治肥胖的组合物,其特征在于,所述组合物为片剂、丸剂、胶囊、颗粒剂或粉剂。
