技术领域
本发明涉及个人清洁组合物,其在个人护理应用(例如局部皮肤护理,最优选的包括女性私密卫生产品的清洁)中是特别有用的,该组合物表现出明显的抗微生物益处,但对皮肤温和且无刺激性,并提供防止感染益处的靶向保护,同时维持健康的外阴微生物组的平衡。
背景技术
在污垢清除和卫生维持方面,清洗是生活中对清洁的基本要求之一。
已知每日使用含有某些活性剂的个人卫生组合物会引起皮肤刺激和干燥,特别是在女性身体的敏感/私密区域。这既限制了它们的使用频率,而且经常需要使用矫正性皮肤保湿剂。
不适当的卫生可能会导致女性的气味和外阴阴道感染和问题。此外,微生物平衡的维持在整个外阴阴道健康和防止病原体获得立足点方面起着关键作用。除了刺激性抗菌剂之外,干扰外阴阴道区域的天然pH水平的因素可能会对保护免受感染所需的天然外阴阴道微生物组的组成构成负面影响。适当的私密卫生对女性健康至关重要。英国皇家妇产科学院(RCOG)指南和大多数医生不推荐使用高pH值的肥皂,因为这类肥皂可能导致干燥或其他皮肤问题。
目前市场上的产品能够提供适当水平的抗菌作用的能力有限,而其他产品为私密区提供过高的杀菌水平,并且不提供皮肤平衡/保湿。某些产品使用诸如三氯生、氯己定等杀菌剂,或其他抗菌剂/其他刺激性化学物质来杀死细菌。市场上需要一种女性私密卫生制剂,其提供抗菌作用,同时维持皮肤天然菌群的健康平衡,对皮肤不干燥,提供保湿益处,并且适合日常使用,同时为私密区提供保护。
健康的阴道中乳杆菌(Lactobacillus)占优势,其是一种产生乳酸的非孢子性革兰氏阳性杆菌,产生酸性环境(pH 3-4)。阴道pH与总乳酸浓度相关,因为阴道粘膜也是乳酸的丰富来源,乳酸是雌激素调节的厌氧葡萄糖代谢的副产物。正常的阴道菌群、酸性阴道pH和阴道分泌物都是防止外阴阴道感染的先天防御机制的组成部分。需要强调阴道乳酸的重要性,因为它与阴道健康相关,其抑制与细菌性阴道病相关细菌的生长,并可能在局部免疫防御中发挥作用。
由于与内部清洗/冲洗相关的风险,外部女性洗液被认为更适合于私密健康,特别是含有乳酸的那些外部女性洗液,其具有增加皮肤平衡的酸性pH,并可以作为阴道感染或服用抗生素的女性的辅助疗法。
本发明的目的在于消除或减轻上述的某些缺点。
发明内容
根据本发明的第一方面,提供了一种个人清洗组合物,其提供针对真菌、革兰氏阳性细菌和革兰氏阴性细菌的有效抗微生物益处,并且还提供包括对皮肤的温和清洁,包括保护皮肤的共生微生物群落(microflora),该组合物包含:
a)0.1重量%~10重量%的甜菜碱表面活性剂,

该组合物优选用作女性私密卫生产品,但也可广泛用于个人护理或个人清洁,包括液体洗手液、沐浴露、温和洗发剂或儿童和婴儿沐浴露。
本发明特别配制用于例如外阴或私密区的外部皮肤和粘膜,以提供温和的清洁、新鲜、抗菌保护以保持健康平衡(特别是私密区)。
它具有四个主要优点:(1)温和清洁(2)皮肤保湿(3)对暂居生物体温和但有效的抗微生物作用和(4)共生微生物平衡。这种温和清洁与皮肤保湿的实现是有意义的。此外,这些目标是在最佳皮肤pH下实现的。此外,已经发现本发明的组合物不破坏或损害天然皮肤菌群并促进使用者皮肤上的平衡菌群,并且进一步稳定微生物组。

泡沫的密度优选小于500克/升,更优选小于400克/升,更优选小于300克/升,最优选小于200克/升。
本发明第二方面的泡沫可以由使用者制备,例如,在手动混合过程中。作为另一种选择,本发明第二方面的泡沫可以使用发泡分配器制备,如US 6840408中所述的发泡分配器,将其内容通过参考并入。
已发现其凝胶或泡沫形式的组合物具有出色的奶油状泡沫状态,并且出人意料地具有不麻烦的冲洗能力(用水除去)。
最优选的用途是作为女性(密切)清洗组合物。
当用作女性清洗制剂时,已经发现该组合物提供/最大/最佳抗菌益处,此外还是柔和的,并且经临床测试为温和的。该洗去型制剂在临床上也被证明具有保湿作用,有助于防止干燥,并且不会伤害私密天然微生物菌群。
本发明利用天然的、生物相似的活性乳酸并且在适当的pH值下为身体的这个区域提供温和的抗菌作用。本发明通过维持最佳的皮肤pH值,湿润的皮肤和平衡的阴道菌群而进一步为私密区提供平衡。
已经发现它可以防止关键的私密卫生问题(由暂居细菌引起的感染、B链球菌、气味、刺激、瘙痒、炎症、疼痛、不适)。对B组链球菌的保护对孕妇特别重要,因为它会导致尿路感染、上生殖道感染、产后子宫内膜炎、肺炎和产后脓毒症。
因此,根据本发明的第三方面,提供了用于为局部表面,特别是皮肤表面提供杀菌益处的方法,所述方法包括以下步骤:使其上已知或怀疑存在一种或多种不期望的病原体,优选细菌的局部表面与本发明的第一或第二方面的组合物接触。
本发明的组合物提供了如抗菌剂(即抗微生物活性)、清洁、调理和保湿以及安全性和功效,易于储存和使用以及美学性质等性质的最佳组合。
本发明的组合物/方法提供了减少不期望的微生物(特别是减少革兰氏阳性和革兰氏阴性微生物)作为主要技术益处,同时提供包括皮肤调理和/或皮肤清洁的第二益处。能够通过可以包含在本发明的制剂或组合物中的一种或多种可选组分的存在来提供进一步的辅助益处。
本发明第一方面的具体实施方式
甜菜碱表面活性剂的存在量更优选为1重量%~3重量%,最优选为约1.3重量%。
烷基甜菜碱是已知的表面活性剂,其主要通过胺化合物的羧烷基化(优选羧甲基化)产生。典型的实例是己基甲基胺、己基二甲基胺、辛基二甲基胺、癸基二甲基胺、十二烷基甲基胺、十二烷基二甲基胺、十二烷基乙基甲基胺、C12/14椰油烷基二甲基胺、十四烷基二甲基胺、十六烷基二甲基胺、硬脂基二甲基胺、硬脂基乙基甲基胺、油基二甲基胺、C16/18牛油烷基二甲基胺及其技术混合物的羧甲基化产物。代表酰氨基胺的羧烷基化产物的烷基酰氨基甜菜碱也是合适的。典型的实例是含有6-22个碳原子的脂肪酸(即己酸、辛酸、癸酸、月桂酸、肉豆蔻酸、棕榈酸、棕榈油酸、硬脂酸、异硬脂酸、油酸、反油酸、岩芹酸、亚油酸、亚麻酸、桐酸、花生酸、鳕油酸、二十二酸和芥酸及其技术混合物)和与氯乙酸钠缩合的N,N-二甲基氨基乙基胺、N,N-二甲基氨基丙基胺、N,N-二乙基氨基乙基胺和N,N-二乙基氨基丙基胺的反应产物。
最优选的是甜菜碱包含椰油酰胺丙基甜菜碱。
更优选的是乳酸的存在量为0.2重量%~8重量%,更优选为0.3重量%~7重量%,更优选为0.4重量%~5重量%,并且最优选为2重量%~5重量%,例如,约2.25重量%。

更优选的是甜菜碱表面活性剂的存在量为0.1重量%~2重量%,最优选为约0.6重量%。

最优选的是甜菜碱包含椰油酰胺丙基甜菜碱。
更优选的是乳酸的存在量为0.2重量%~5重量%的乳酸,更优选为0.3重量%~5重量%,更优选为0.4重量%~5重量%,并且最优选为2重量%~5重量%,例如约2.75重量%。
更优选的是C2-C6多元醇的存在量为2重量%~18重量%,更优选为6重量%~17重量%,更优选为8重量%~16重量%,并且最优选为10重量%~15重量%。
最优选的是C2-C6多元醇包含甘油。
具体实施方式
出乎意料的是,已经发现本发明的组合物实现令人满意的清洁和杀菌,同时避免了由这些含硫化合物(特别是诸如月桂基醚硫酸钠(SLES)等含硫表面活性剂)的刺激引起的问题。SLES是一种便宜且非常有效的发泡剂。还显示SLES会导致皮肤刺激性。考虑到本发明主要优选私密用途,这是特别重要的。对于这种优选用途,明显有利的是避免了刺激。
更详细的是,已经观察到,当根据ASTM E2315-03“使用时间杀灭程序评估抗微生物活性的标准指南”的标准化测试方案进行测试时,组合物的特征可以在于表现出诸如大肠杆菌等各种微生物/细菌的至少1个log10的减少。
抗微生物测试方案:
使用根据ASTM E2315-03“使用时间杀灭程序评估抗微生物活性的标准指南”的测试方案来评估针对革兰氏阳性(金黄色葡萄球菌,Staphylococcus aureus)(ATCC 6538)、革兰氏阴性(大肠杆菌,Escherichia coli)(ATCC 10536)细菌和酵母白色念珠菌(Candidaalbicans)(ATCC 10231)的抗微生物功效。根据该方案,首先,通过在胰蛋白酶氯化钠中悬浮制备激发培养物(18小时~24小时),其在室温下平衡至20℃~22℃。在所述测试当天,在室温(20℃~22℃)下,在无菌容器(例如试管)中将5ml组合物产品样品与4ml ml标准硬水(约300ppm CaCO3)混合。随后加入1ml测试培养物,其导致50%v/v稀释。然后将试管涡旋5秒,并使其保持接触60+/-5秒,之后立即取出1ml等分试样并加入到含有9ml中和剂的另一管中。中和允许发生5分钟,然后将使用胰蛋白酶氯化钠的系列十倍稀释液铺平板,并在36±1℃温育24小时~48小时。所用的接种物也被连续稀释,铺平板并在36±1℃温育24小时~48小时。温育后,对激发生物体的存活集落形成单位(CFU)计数,并且从一个或多个重复样品确定每个测试制剂的log10降低值,在多个重复样品的情况下,报告平均结果。
pH

粘度
本发明第一方面的组合物是粘性的并且如使用Brookfield粘度计,3型心轴在12rpm下测量的,在室温下表现出至少500cps的粘度。在这些条件下测量,本发明第一方面的组合物优选表现出在至少约500cps范围内的粘度。更优选的是本发明的局部组合物表现出约1000cps~约10,000cps,还更优选约1500cps~7000cps,并且特别优选约1500cps~4500cps的粘度。

实施例
实施例1-基本配方和抗微生物性能
该表显示了基础凝胶沐浴露配方(E1)的总表面活性剂、乳酸和盐浓度变化对微生物性能的影响。当根据ASTM E2315-03“使用时间杀死程序评估抗微生物活性的标准指南”的标准化测试方案进行测试时,通过配方(passing formula)被定义为实现同时针对金黄色葡萄球菌和大肠杆菌的至少1个对数的杀灭。鉴于其在私密区的相关性,针对白色念珠菌的1个对数的杀灭也是优选的。
数据表明,基于实施例E2,总表面活性剂体系减少5%导致较低的大肠杆菌性能。在实施例E4中,通过将乳酸降低至1.75%,微生物效果也受到负面影响。实施例E3证明了通过用氯化钙代替氯化钠保留了阳性效果。实施例E5证明了通过用50%乳酸代替88%乳酸保留了阳性效果。实际上,注意到白色念珠菌杀死量增加0.5log。怀疑这种益处可能来自这样的事实,即乳酸的更稀溶液更富含乳酸的单体形式,这比聚合物形式更有效。

实施例2-皮肤刺激或致敏研究
将凝胶配方E5置于临床研究中以确定外部接触人类受试者皮肤后的皮肤刺激性和致敏潜力。
该研究在104名健康受试者中进行,使用半封闭性贴片,将含有测试产品(稀释至1%)的半封闭贴片应用在上背部(在肩胛骨之间)标记的皮肤部位,以在3周诱导阶段中进行9个24小时暴露期。每次应用后约48小时评估皮肤部位的红斑、水肿和其他刺激征兆。在诱导期后大约2周的休息期后,将半封闭性产品激发贴片应用于背部先前未经处理的部位24小时。在贴片应用后24小时、48小时和72小时评估激发部位的红斑、水肿和其他刺激征兆以确定致敏潜力。
未观察到可见的皮肤反应。

实施例3-本发明的女性凝胶与竞争产品的比较
Kora是一种含有乳酸/洋甘菊抗微生物复合物的私密卫生凝胶产品。但是,当测试时,Kora产品中的低剂量乳酸不能提供足够的抗微生物功效来促进私密女性区域的充分卫生。参见下面的结果。
另外,发现Kora产品(类似于许多商业的私密卫生产品)具有非常低水平的甘油,与根据本发明的凝胶相比,其可能不提供足够的保湿益处。
本发明的制剂证明了有效的抗微生物益处的关键是具有高量乳酸协同作用的三种表面活性剂体系。
实施例4-私密泡沫清洁剂
私密泡沫清洗剂是另一种广受欢迎的女性产品。在泡沫配方中再次使用乳酸作为抗微生物活性物质。与先前的凝胶实施例相比,所述制剂含有较低量的总表面活性剂,并且还含有较高水平的乳酸以帮助平衡该制剂的微生物效果。这些制剂是水稀释的,可以使用泡沫泵包装或挤压泡沫分配器。
当根据ASTM E2315-03“使用时间杀死程序评估抗微生物活性的标准指南”的标准化测试方案进行测试时,泡沫的通过制剂被定义为同时针对金黄色葡萄球菌和大肠杆菌实现至少1个对数的杀灭。鉴于其在私密区的相关性,针对白色念珠菌的1个对数的杀灭也是优选的。
实施例5-女性卫生清洗剂使用中的耐受性和微生物研究
在pH为5.6的淋浴凝胶产品的7天预研调理期后,34名健康女性受试者使用测试产品(配方E5)每天至少一次洗涤外部私密区28天,在基线(第0天)以及第14天和第28天进行评估。妇科医生进行了对皮肤干燥、红斑、水肿、脱屑、瘙痒(口头询问受试者,视觉评估划伤痕迹)、灼烧(口头询问受试者)、刺痛(口头询问受试者)和私密区的其他临床刺激征兆的耐受性评估。受试者在基线(第0天)和第14天和第28天提供主观耐受性评估。
在基线(第0天)和第14天和第28天,使用皮肤-pH计PH 905探针用Multi ProbeAdaptor系统(MPA系统)(Courage&Khazaka GmbH)进行中-大阴唇的皮肤pH测量。在受试者保持在气候控制条件下(适应22±2℃和35-55%RH)后,使用具有Corneometer探针的多探针适配器(MPA)6,用CM 825(Courage&Khazaka GmbH)进行20分钟的皮肤水合和保湿测量。在基线(第0天)和第14天和第28天使用Corneometer测量腹股沟区域的皮肤水合作用以评估更持久的益处。在研究技术人员施用测试产品之前和之后的两个时间点,在基线(T-0h),T-0清洗后(在测试产品清洗后立即)和施用测试产品清洗后的T-1h使用Corneometer测量上内腿区域的皮肤保湿以评估单次使用益处/短期益处。
在基线(第0天)和第14天和第28天通过液杯擦洗方法从中-大阴唇区域采集微生物样品。按压玻璃量筒(约4cm2面积)以牢固地与皮肤接触,施加足够的压力以防止收集过程中液体泄漏。将1ml 0.1%清洁剂溶液(缓冲至pH 7.5-8.0的含有0.1%(v/v)Triton X-100(全强度清洗液)的75mM磷酸盐缓冲液)移液到围绕约3.0cm2面积的无菌玻璃量筒中,使用钝的无菌玻璃棒(擦洗装置)尽可能均匀地小心摩擦皮肤表面,施加中等恒压1分钟。将样品在96℃煮沸10分钟以进行灭菌,包装在干冰中并运送用于微生物多样性分析。
在研究中纳入36名(36)受试者。两名(2)受试者退出,剩下三十四(34)名受试者完成研究,他们的微生物组数据被纳入分析。受试者年龄在19岁至55岁之间(平均值36.61,标准差11.28)。12名受试者(33.3%)年龄在18岁至29岁之间;13名(36.1%)受试者年龄在30岁至44岁之间,11名(30.6%)受试者年龄在45岁至55岁之间。
基于5分量表的皮肤刺激评分(平均总分),0=无,0.5=非常轻微,1=轻微,2=中等,3=严重,从第0天的0.01增加至第14天的0.12和第28天的0.13。受试者在第14天和第28天的基线总刺激性评分的平均值分别为0.10和0.12,满足可接受的耐受性标准。使用28天后,妇科医生作出的总体耐受评级为:“测试产品对大多数受试者耐受性良好”。
第0天外部私密区的皮肤pH(总群体)为5.88。在第14天,pH值略微下降至5.87,在第28天时下降至5.85。受试者从基线pH变化的平均值在第14天为-0.01,在第28天为-0.03。分析显示在时间点之间没有显著差异;D-0和D-14之间的p值为0.992,D-0和D-28之间的p值为0.883,并且D-14和D-28之间的p值为0.933;其全部远高于95%置信水平的0.05。统计分析证实,随着产品的使用,外部(外阴)区域的皮肤pH值随时间变化无显著差异(总群体)。
在腹股沟区域测量的皮肤水合作用的平均Corneometer得分在第0天为39.14。在第14天,其增加至45.97,在第28天增加至48.86。在研究的28天期间皮肤水合作用增加。分析显示在D-14和D-28(p=<0.0001)在皮肤水合作用上具有统计学显著增加(p<0.05)。
对于测试产品,通过平均Corneometer得分的上内腿区域测量的皮肤保湿在T-1小时(p=<0.0001)时与基线相比具有统计学上显著增加(34.43比29.98)。
为了进行微生物多样性分析,基于NCBI数据库对16s rRNA序列进行扩增,测序和OTU(操作分类单位)分配到每个序列。
从两个角度考察了多样性。首先,总体丰富度(即微生物组内存在的不同生物的数量)表示为操作分类单位(OTU)的数量,并且使用Chao1丰富度估计量来量化。
其次,整体多样性(由丰富度和均匀度决定,不同类群之间丰度的分布)表示为Shannon多样性。
使用方差分析(ANOVA)筛查群组差异的多样性度量。结果(如下所示)表明测试的凝胶洗涤制剂在针对外部私密皮肤微生物组的细菌或真菌评估的任何时间点对物种丰富度和多样性没有显著影响(p>0.05)。事实上,显示其可以稳定私密皮肤的微生物组。

结论:在4周的时间内(28天),女性在外部生殖器区域每天使用至少一次时,凝胶清洗剂显示可接受的耐受性,没有显著改变外部外阴区域的皮肤pH并提供了显著的皮肤水合作用和保湿益处。此外,外部外阴区域的微生物组分析显示该配方对细菌或真菌的物种丰富度和多样性没有显著影响,对微生物组具有稳定作用。
Claims (24)
1.一种个人清洗组合物,其针对真菌、革兰氏阳性细菌和革兰氏阴性细菌提供有效的抗微生物益处,所述组合物包含:
a)0.1重量%~10重量%的甜菜碱表面活性剂,
b)0.1重量%~5.0重量%的乳酸,
c)0.1重量%~20重量%的C2-C6多元醇,
d)0重量%~10重量%的式RO(CH2CH2O)kCH2COO-M+的烷基聚乙氧基羧酸盐,其中,R是C8-C22烷基,k是0~20的整数,并且M是可溶性成盐阳离子,
e)0重量%~10重量%的式RO(CH2CH2O)kCH2CONH2的烷基聚乙氧基酰胺,其中,R是C8-C22烷基,k是0~20的整数,
f)>8重量%的烷基醚硫酸盐,
g)水,
h)3.8-4.5的pH,
i)10重量%~20重量%的总表面活性剂,
j)0.1重量%~10重量%的金属盐。
2.如权利要求1所述的组合物,其中,所述烷基聚乙氧基羧酸盐的存在量为2重量%~7重量%,最优选为约4.8重量%。
3.如权利要求1或2所述的组合物,其中,在所述烷基聚乙氧基羧酸盐中,R包含C12烷基;K是11并且M是Na。
4.如权利要求1、2或3所述的组合物,其中,所述甜菜碱表面活性剂的存在量为1重量%~3重量%,最优选为约1.3重量%。
5.如权利要求1~4中任一项所述的组合物,其中,所述甜菜碱包含椰油酰胺丙基甜菜碱。
6.如权利要求1~5中任一项所述的组合物,其中,所述乳酸的存在量为0.2重量%~8重量%,更优选为0.3重量%~7重量%,更优选为0.4重量%~5重量%,并且最优选为2重量%~5重量%,例如约2.25重量%。
7.如权利要求1~5中任一项所述的组合物,其中,所述C2-C6多元醇的存在量为2重量%~18重量%,更优选为6重量%~17重量%,更优选为8重量%~16重量%,并且最优选为10重量%~15重量%。
8.如权利要求1~7中任一项所述的组合物,其中,所述C2-C6多元醇包含甘油。
9.如权利要求1~8中任一项所述的组合物,其中,所述烷基聚乙氧基酰胺包含PEG 4油菜籽酰胺。
10.如权利要求1~9中任一项所述的组合物,其中,所述金属盐的存在量为0.1重量%~5重量%,更优选为0.2重量%~3重量%,并且最优选为约2重量%。
11.如权利要求1~10中任一项所述的组合物,其中,所述金属盐包含碱金属和/或碱性金属卤化物。
12.如权利要求1~11中任一项所述的组合物,其中,所述烷基醚硫酸盐包含月桂基醚硫酸钠。
13.一种泡沫形式的个人清洗组合物,其针对革兰氏阳性细菌和革兰氏阴性细菌提供有效的抗微生物益处,所述组合物包含:
a)0.1重量%~5重量%的甜菜碱表面活性剂,
b)0.1重量%~5重量%的乳酸,
c)0.1重量%~20重量%的C2-C6多元醇,
d)水,
e)3.8-4.5的pH,
f)0.1重量%~5重量%的总表面活性剂。
14.如权利要求13所述的组合物,其中,所述泡沫的密度优选小于500克/升,更优选小于400克/升,更优选小于300克/升,并且最优选小于200克/升。
15.如权利要求13或14所述的组合物,其中,所述甜菜碱表面活性剂的存在量为0.1重量%~2重量%,并且最优选为约0.6重量%。
16.如权利要求13~15中任一项所述的组合物,其中,所述甜菜碱包含椰油酰胺丙基甜菜碱。
17.如权利要求13~16中任一项所述的组合物,其中,所述乳酸的存在量为0.2重量%~5重量%,更优选为0.3重量%~5重量%,更优选为0.4~5重量%,并且最优选为2~5重量%,例如约2.75重量%。
18.如权利要求13~17中任一项所述的组合物,其中,所述C2-C6多元醇的存在量为2重量%~18重量%,更优选为6重量%~17重量%,更优选为8重量%~16重量%,并且最优选为10重量%~15重量%。
19.如权利要求13~18中任一项所述的组合物,其中,所述C2-C6多元醇包含甘油。
20.一种为局部表面,特别是诸如私密皮肤/外表面等皮肤表面提供杀菌益处的方法;所述方法包括以下步骤:使其上已知或怀疑存在一种或多种不期望的病原体,优选细菌的局部表面与权利要求1~19中任一项所述的组合物接触。
