Description

Herbal additive for promoting sheep growth and preparation and use methods thereof
Technical Field
The invention relates to the technical field of sheep feed additives, in particular to a herbal medicine additive for promoting sheep growth and a preparation method and a use method thereof.
Background
The Liangshan state possesses rich characteristic Yi-nationality herbal medicine (Yi-nationality medicine for short) resources, and in the big background of a project [1] listed in the development center of the national feed industry development strategy in the last ten years, the novel, pollution-free and multifunctional sheep-using Yi-nationality medicine feed additive is developed aiming at the unique Yi-nationality medicine resources of the Liangshan state and the vigorously developed sheep breeding industry according to the unique Yi-nationality medicine theory, so that the development of the green sheep breeding industry with the Liangshan state characteristics is promoted, safe and pollution-free sheep products are produced, and the ecological environment of the Liangshan state is protected.
At present, the research of using Yi medicine as animal feed additive at home and abroad is still blank. In Liangshan, more than 1000 kinds of long-term and unique Yi medicine resources are widely used for preventing and treating human diseases, and many Yi medicines have the characteristics of homology and common use with food and have double functions of nutrition and medicines, thereby providing valuable resources for developing natural livestock and poultry feed additives with the characteristics of Liangshan.
The Yiyi medicine with Liangshan characteristics belongs to a natural medicine as the traditional Chinese medicine, and has the outstanding advantages of low cost, wide source, simple and convenient processing, safety, effectiveness and the like when being used as a feed additive. According to incomplete statistics, more than 200 Chinese herbal medicines in more than 1000 Chinese herbal medicines clinically used by veterinarians at present can be used as feed additive raw materials [2], and have various effects of improving the quality of livestock and poultry products, improving the quality of feeds, preventing diseases, protecting health and the like.
The research of the traditional Chinese medicines in the livestock breeding aspect can be used for reference by developing the Liangshan special Yi-nationality medicine into the sheep feed additive. According to reports, the Sudan [3] and the like are added with compound traditional Chinese medicine superfine powder feed consisting of radix codonopsitis, angelica, astragalus, hawthorn, malt, honeysuckle and common andrographis herb with 0.5 percent of basic daily ration in the feed for fattening pigs to feed the fattening pigs, so that the effects of improving daily gain, disease resistance and total oxidation resistance of serum and reducing the feed-weight ratio and the content of serum Malondialdehyde (MDA) can be achieved. The Ma Mingzhi [4] adds 20g of traditional Chinese medicine powder such as astragalus root, uniflower swisscentaury root, angelica, cowherb seed, beautiful sweetgum fruit, ricepaperplant pith and the like into daily ration of the milk goat in the lactation peak period, so that the milk yield of the goat is remarkably improved, and the daily income increase can reach 36 yuan. The result of adding superfine powder of astragalus root, licorice, selfheal, reed rhizome, gypsum, mint and other Chinese medicinal materials in different dosages into daily ration of laying hens by Sun-Qi Ying and other [5], shows that the addition amount of 1.0% can obviously improve the content of heat shock protein 70 in serum of laying hens, is 63.3% higher than that of blank groups, reduces liver and muscle damage of laying hens in heat stress, and enhances the heat stress resistance of organisms. The results of 0.25%, 0.5% and 1% of Chinese herbal medicine ultrafine powder agents are respectively added into basic daily ration of 21-day-old weaned piglets in Zhao hongmei and the like [6], show that compared with antibiotics, the Chinese herbal medicines in each group have no obvious difference on the influence of biochemical indexes of blood of the piglets, but can obviously improve the levels of thyroxine T3 and T4 of the piglets, and the formula can promote the growth of the piglets. Zhang Shi Chang et al [7] adopts crude extracts of 3 Chinese medicines of barberry root, astragalus root and hawthorn fruit to be added into the daily ration of weaned pigs according to the dosage of 0.125 percent and 0.25 percent, has the trend of improving the weight and daily gain of the piglets, and can obviously reduce the feed conversion ratio and the diarrhea rate; the high-dose traditional Chinese medicine crude extract can improve GSH-PX by 30.4%, can also improve the level of serum superoxide dismutase (SOD), reduces the content of MDA, and improves spleen index, which indicates that the traditional Chinese medicine additive can improve the antioxidation function of piglets and promote the development of immune organs. The Chinese medicines of epimedium, broomrape, dodder, wolfberry fruit, morinda root and astragalus root etc. 8 are added into the daily ration of ram according to the dose of 10g/d, thus obviously increasing the content of reproductive hormone of ram's plasma follicle stimulating hormone, testosterone and luteinizing hormone etc. and finally improving the production performance. Guo Jianying et al [9] adding superfine powder of compound traditional Chinese medicine raw astragalus, motherwort, angelica, dandelion, tuckahoe, hawthorn, purslane, rhizoma atractylodis, raw liquorice, dried orange peel and the like into daily ration of lactating sows at the dosage of 7.5g/kg and 15g/kg can obviously improve the rosette rate of red blood cells E-C3bR of sows, the CD4+/CD8+ of lymphocytes and improve the contents of serum immunoglobulin, IL-2, IL-10, TNF-alpha and IFN-gamma, and shows that the adding of the superfine powder of the compound traditional Chinese medicine into the feed of the lactating sows can improve the immunologic function of the sows. Gong Wei Zhong, etc. [10] adding the astragalus root, the malt, the common andrographis herb, the codonopsis pilosula, the glossy privet fruit and other compound traditional Chinese medicine superfine powder into daily ration of the chickling according to the dosage of 0.25 percent and 0.5 percent respectively, and adding the traditional Chinese medicine coarse powder into the daily ration of the chickling according to the dosage of 1.0 percent, compared with a blank control group, the weight of the 0.5 percent traditional Chinese medicine superfine powder group and the 1 percent traditional Chinese medicine coarse powder group is obviously improved, the material-meat ratio is reduced, the T-AOC and SOD; tests show that the 0.5 percent Chinese medicinal superfine powder can improve the production performance and the oxidation resistance of broiler chickens, has the same curative effect as 1 percent Chinese medicinal coarse powder, and can obviously reduce the dosage of medicinal materials. Li Chun Yan and so on [11] add 5% of dosage of compound Chinese herbal medicine powder of tuber fleeceflower root, basket fern, hawthorn, malt, dried orange peel, medicated leaven, white peony root, radix astragali, eucommia bark, Chinese yam, radix codonopsitis, pine needle and so on into the daily ration of 3-month-old sheep, can obviously promote the weight gain of sheep, have raising effects on most indicators of red cells and white cells, and have reducing effects on the number of platelets; meanwhile, the Chinese herbal medicine additive can improve the immune function of the organism, and the action effect is positively correlated with the additive amount. Different formulas are selected for the Liu Aixia [12], the traditional Chinese medicine formula is crushed and then added into daily ration to feed the milk goat, and the result shows that the effect of improving the milk yield of the milk goat, improving the milk quality and increasing the economic benefit is best when the additive amount is 1%.
Compared with China, foreign research on natural medicine feed additives is relatively late. In the Sulian, acanthopanax root was used as a feed additive in the 60 th of the 20 th century, and a good effect is achieved. In the 70 s of the 20 th century, Japan extracted effective ingredients from garlic as animal growth promoters. In recent years, much attention has been paid to Chinese herbs and their use as feed additives, especially in the direction of plant extracts, in many countries such as the United states, Canada, Germany, Singapore. In asia, japan and korea are countries where studies on plant extracts were performed earlier. The European and American countries have also carried out large-scale research on their traditional medicinal plants, and have applied for many patents on the basis of the research, and developed many products for human and animal production. The types of plants studied in foreign countries include vegetable, fruit, green tea, onion, garlic, clove, locust tree leaf, juniper, malt, marigold, peach, panda leaf, thistle, coptis, sage, cardamom, bay, extracts or essential oils of herbs/oregano, seaweed, mushroom and others [13-14], but the results are rarely applied in the feed industry. The volatile essential oil additive extracted from oregano is a novel feed additive product which appears in almost twenty years and is applied to certain range in European and American areas, the first product on sale is the norbiddar (ROPADLAR) of Ropafam corporation in Netherlands, more than ten European and American manufacturers produce the same product, most of the products are chemically synthesized, and the effect is different from that of natural extraction.
At present, the research on natural medicine feed additives for ruminants such as sheep is less at home and abroad [15], so that the research on natural medicine feed additives for sheep has important significance.

The administration time of the herbal additive is 11 months to 3 months in the coming year in the south of the Yangtze river, and 11 months to 4 months in the coming year in the north of the Yangtze river.
As shown in figure 1, the addition amount of 0.25-0.8% can improve the weight gain rate of sheep in different degrees within 60 days after the feeding of the herbal additive. Blank (group) was not added, and the low dose was added at 0.25%, the medium dose was added at 0.5% and the high dose was added at 0.8%.
As shown in FIG. 2, the herbal additive of the present invention can promote the secretion of sheep growth hormone and increase its serum concentration. The Growth Hormone (GH) can promote the growth of bones, internal organs and the whole body of animals, promote protein synthesis, influence the metabolism of fat and minerals and play a key role in the growth and development of sheep. The herbal additive of the invention increases the content of sheep serum GH at the administration time of 30 days, and the administration group is significantly different from the blank group at the administration time of 60 days.
As shown in FIG. 3, the herbal additive of the present invention can promote the secretion of sheep I insulin-like growth factor and increase its serum concentration. Insulin-like growth factors (IGFs) are a group of polypeptides with growth-promoting effects, and their secretory cells are widely distributed in tissues such as liver, kidney, lung, heart, brain and intestine of animals. The IGF family includes two types, IGF-I and IGF-II. The production of IGF-I is more dependent on GH, and the growth promoting effect is strong, and is an important growth factor in the young period of sheep. IGF-I synthesized in various tissues plays a role in promoting growth in an autocrine or paracrine manner, and IGF-I synthesized by the liver enters the blood circulation and acts on target cells in an endocrine manner. The IGF-I level in vivo is regulated by GH, and IGF-I also has negative feedback regulation effect on GH secretion. At 30 days and 60 days after the administration of the herbal additive, the herbal additive can obviously improve the IGF-I serum concentration of sheep and has the effect of promoting growth.
As shown in FIGS. 4 and 5, thyroxine (tetraiodothyronine T4) and triiodothyronine (T3) were maintained at higher levels, significantly higher than the blank group, within 30-60d after administration of the herbal additive of the present invention. The thyroid hormone has the most obvious effect of promoting growth and development in the first five months after birth of sheep, and mainly promotes the growth and development of bones, brain and reproductive organs. Meanwhile, the thyroid hormone mainly promotes protein synthesis under normal conditions, particularly obviously increases protein synthesis of bones, skeletal muscles, livers and the like, and has important significance on growth and development of sheep in young period. In the absence of thyroid hormone, the pituitary gland produces and secretes GH less and fails to function. T4 and T3 are a group of tyrosine which takes iodine and tyrosine as raw materials to synthesize iodine in thyroid gland cells, wherein T4 accounts for about 90 percent of the total amount, T3 has less secretion amount but high activity which is 5 times of that of T4, about 50 percent of T4 is deiodinated and converted into T3 every day, and therefore the effect of T3 cannot be ignored. The herbal additive of the invention can up-regulate the concentrations of T4 and T3 in sheep serum after administration, which is consistent with the high level expression of 60dGH after administration, and has synergistic growth promoting effect.
The herbal additive disclosed by the invention can promote the secretion of sheep growth hormone, improve the serum concentration of sheep growth hormone, promote the secretion of sheep I-type insulin-like growth factors, improve the serum concentration of sheep I-type insulin-like growth factors, and up-regulate the concentrations of T4 and T3 in sheep serum, so that the growth of sheep is promoted.
As shown in FIG. 6, the herbal additive of the present invention significantly increased the serum interleukin-2 (IL-2) concentration in sheep 15 days after administration. IL-2 is mainly produced by T lymphocyte system, and can promote proliferation and differentiation of T lymphocyte activated by antigen or mitogen, maintain balance of T cell group (Tc, Ts and Th) of organism, and induce production of various cytokines such as IFN-gamma, IL-4, IL-6 and TNF-beta; simultaneously can induce the differentiation and effect functions of a plurality of killer cells such as CTL, NK, LAK and the like, and induce the killer cells to generate cytokines such as IFN-gamma, TNF-alpha and the like; IL-2 can directly act on B cells to promote proliferation, differentiation and immunoglobulin secretion of the B cells; IL-2 can activate macrophages, enhance the function of phagocytosing foreign bodies and presenting antigens. In conclusion, IL-2 can enhance the cellular immunity (enhance T cell function) and humoral immunity (enhance B cell function) of the body, and the herbal additive of the present invention may have mitogen components, and can activate T cells and B cells of sheep within 30 days after administration, thereby enhancing the immune function.
As shown in FIG. 7, the herbal additive of the present invention significantly increased the serum concentration of sheep interleukin-6 (IL-6) at 60 days of administration. IL-6 is a lymphokine produced by activated T cells and fibroblasts, primarily capable of rendering B cell precursors into antibody-producing cells; and the colony stimulating factor can promote the growth and differentiation of original bone marrow-derived cells and enhance the lysis function of natural killer cells. The herbal additive can gradually increase the serum concentration of IL-6 after being administrated for 30 days, and has the enhancement effect on both cell and humoral immunity of sheep, especially the humoral immunity function of sheep.
As shown in FIG. 8, the herbal additive of the present invention significantly increased the concentration of Lysozyme (LZM) in sheep serum 15 days after administration. LZM is an alkaline enzyme capable of hydrolyzing mucopolysaccharide in pathogenic bacteria, and is an important component of the innate immunity function of sheep. LZM is present in body fluids to lyse bacteria primarily by disrupting the beta-1, 4 glycosidic bond between N-acetylmuramic acid and N-acetylglucosamine in the bacterial cell wall, breaking down the cell wall insoluble mucopolysaccharide into soluble glycopeptides, causing the contents of the broken cell wall to escape. Lysozyme can also be directly combined with virus protein with negative charge, and forms double salt with DNA, RNA and apoprotein to inactivate virus. Therefore, LZM has multiple functions such as antibiosis, antiphlogosis, antivirus, etc. The herbal additive of the invention can significantly improve the anti-infective ability of sheep during the administration period.
The herbal additive can improve the concentration of interleukin-2 (IL-2), the serum concentration of interleukin-6 (IL-6) and the Lysozyme (LZM) in sheep serum, thereby enhancing the immunity of sheep.
The foregoing is illustrative of the preferred embodiments of this invention, and it is to be understood that the invention is not limited to the precise form disclosed herein and that various other combinations, modifications, and environments may be resorted to, falling within the scope of the concept as disclosed herein, either as described above or as apparent to those skilled in the relevant art. And that modifications and variations may be effected by those skilled in the art without departing from the spirit and scope of the invention as defined by the appended claims.
