Abstract

本发明公开了脑胶质瘤原代培养及其PDX模型构建。PDX模型高度保真并保留了原代脑胶质瘤的组织病理学、遗传学特征以及肿瘤异质性,是目前为止最接近临床研究的相关肿瘤模型,这种模拟人脑胶质瘤特异性的模型对肿瘤临床前期评估、治疗和预后具有重要的转化意义,有望为脑胶质瘤患者个体化治疗带来新突破。

脑胶质瘤原代细胞培养及PDX模型的建立
技术领域
本发明属于生物医药领域,尤其是涉及一种脑胶质瘤原代细胞培养及脑胶质瘤PDX标准化模型库。
背景技术
传统的体内药敏测试方法是将鼠源或人源细胞在体外筛选,经过传代培养,建立稳定细胞株,然后移植到小鼠体内建立肿瘤模型,这种模型的建立在肿瘤研究中的应用已有半个世纪,但是这种模型存在一定的缺陷,因为连续传代的肿瘤细胞株适应了外界培养皿的环境,丢失了肿瘤异质性,因此不能客观反应原代肿瘤的情况。PDX模型是一种直接将病人的新鲜肿瘤细胞或组织通过原位或者是异位(多为皮下或肾包膜)等方式移植到免疫缺陷小鼠上,依靠小鼠提供的环境生长的一种动物模型。其最重要的优势就是可以高度保留患者肿瘤的组织学特性,遗传学特征以及肿瘤特异性,同时也包含了原代肿瘤的微环境,包括细胞外基质,免疫细胞以及肿瘤微环境因子。人源肿瘤异种移植模型(patient-derived xenograft model,PDX模型)是目前为止最接近临床研究的相关肿瘤模型,这种模拟人肿瘤特异性的模型对肿瘤临床前期评估、治疗和预后具有重要的转化意义,有望为肿瘤患者个体化治疗带来新突破。
大量的研究表明,PDX模型高度保真并保留了原代肿瘤的组织病理学、遗传学特征以及肿瘤异质性。此外,全面的基因表达谱数据分析表明PDX肿瘤在很大程度上保留了原代肿瘤中大多数的关键基因与信号通路活性。前期研究较多的PDX模型是在乳腺癌、胃癌、结肠癌等,而对脑胶质瘤的肿瘤组织原代培养及PDX模型研究较少。本研究以脑胶质瘤组织样本体外培养,并结合构建PDX模型,为临床药效评估、个性化治疗提供指导意见。
发明内容
本发明体外培养了原代脑胶质瘤组织,并将组织移植入裸鼠体内,构建PDX模型。脑胶质瘤(脑胶质细胞瘤)是由于大脑和脊髓胶质细胞癌变所产生的、最常见的原发性颅脑肿瘤。约占颅内肿瘤的46%。综合发病年龄高峰在30-40岁,或10-20岁。大脑半球发生的胶质瘤约占全部胶质瘤的51.4%,以星形细胞瘤为最多,其次是胶质细胞瘤和少枝胶质细胞瘤,脑室系统也是胶质瘤较多的发生部位,占胶质瘤总数的23 .9%,主要为管膜瘤,髓母细胞瘤,星形细胞瘤,小脑胶质瘤占胶质瘤总数的13%,主要为星形细胞瘤。新鲜脑胶质瘤样本,剪碎成1 mm3左右组织块,以0.25%、1% 型胶原酶等体积混合消化30 min,过200目细胞筛网,1500 rpm离心10 min,完全培养基重悬,培养出状态较好的原代脑胶质瘤细胞,同时将新鲜脑胶质瘤组织移植于裸鼠皮下,构建PDX模型。从而为脑胶质瘤相关的抗癌药物开发提供一个方便稳定的平台。

实验材料与方法
1.细胞培养
脑胶质瘤来源于病人组织,低温条件下快速剪碎,以0.25%、1% 型胶原酶等体积混合消化30 min,过200目细胞筛网,1500 rpm离心10 min,1640培养基(含10 %肽牛血清、100U/ml青霉素、100 ug/ml链霉素)重悬,置于37°C、5 % CO2饱和湿度培养箱中培养,每1-2天更换培养基一次。当细胞生长到覆盖瓶底壁80 %时,用0.25 %胰蛋白酶消化贴壁细胞,传代后继续放置于培养箱中。
2.PDX模型构建
新鲜脑胶质瘤组织,低温条件下快速剪碎,以0.25%、1% 型胶原酶等体积混合消化30min,过200目细胞筛网,1500 rpm离心10 min,PBS重悬,细胞计数,106/ml,皮下移植0.1 ml于裸鼠双侧腋下,每天观察成瘤情况。当肿瘤大达到400-600 mm3左右,取出肿瘤,剪碎,移植到裸鼠体内。
