Abstract
一种高效抗氧化的富硒植物酵素原液,主要由以下重量份原料组成:富硒甘蓝或其提取物2‑4份,灰树花粉或其提取物3‑5份,紫云英花粉或其提取物2‑4份,蓝莓提取物或其提取物1‑3份。其优点是:富硒甘蓝和灰树花、紫云英花粉等的组合,不仅可以起到直接的抗氧化作用,还有助于体内产生内源性抗氧化酶GSH‑Px,协同发挥抗氧化作用;原料均为天然可食用植物或食用菌来源,安全性有保障。

技术领域
本发明涉及天然保健品及护肤品技术领域,具体地说是一种高效抗氧化的富硒植物酵素原液及其制备和应用。
背景技术
正常状态下,人体内自由基的代谢维持动态平衡,不止有生成,也有有效的清除机制,体内防御氧化的体系主要有抗氧化酶体系,通过谷胱甘肽过氧化物酶(GSH-Px),超氧化物歧化酶(SOD)等起到清除自由基,维持人体的氧化还原稳态;还有一些非酶类物质也具有清除自由基的能力,被称为抗氧化剂,例如维生素C、维生素E、多酚类、黄酮类、多糖类、微量元素硒等,其中,硒元素是GSH-Px的活性中心的必要组成部分,可分部在全身多个器官,产生有效的抗氧化,保护细胞和组织器官的作用。
由于人体自身的衰老变化,以及空气污染、生活压力、睡眠不足、精神紧张等内外因素的影响,导致人体自由基过量累积,不能通过自身抗氧化体系有效的清除,则可引起蛋白质、核酸变性,细胞脂质过氧化损伤,诱发慢性病变,促进机体的衰老进程。并且,皮肤的衰老,皱纹的产生和色素沉淀也在过量自由基的堆积后,加速了进程。因此清除体内无用的代谢产物,减少自由基的堆积是目前延缓衰老,使人体内部器官和皮肤保持较好状态的有效方法。
而传统的抗氧化剂主要是化学制品,对人体可能有毒副作用,制备有效且安全的纯天然抗氧化制品是目前的必然趋势。
发明内容
本发明的目的是为解决上述技术缺陷,提出一种高效抗氧化的富硒植物酵素原液及其制备和应用,具体是:将富硒植物多肽和其它天然抗氧化植物进行复配,并用益生菌发酵,制备出一种高效抗氧化的富硒植物酵素原液,以利用富硒植物酵素原液中的硒元素和天然植物酵素中的多酚,多糖类等协同作用,发挥强效的抗氧化、清除自由基的作用。
本发明第一方面保护一种高效抗氧化的富硒植物酵素原液,主要由以下重量份原料组成:
富硒甘蓝或其提取物2-4份,灰树花粉或其提取物3-5份,紫云英花粉或其提取物2-4份,蓝莓提取物或其提取物1-3份。
优选为,富硒甘蓝或其提取物3-4份,灰树花粉或其提取物4-5份,紫云英花粉或其提取物2-3份,蓝莓提取物或其提取物1-2份;
优选地,一种高效抗氧化的富硒植物酵素原液,还包括刺梨粉或其提取物1-3份、红枣粉或其提取物1-2份。
本发明第二方面保护第一方面所述富硒植物酵素原液的制备方法:
S1、制备原料水提液:各原料按比例混合均匀、过筛,再将过筛后的原料与水按照1:10的质量比混合,90℃浸泡60min后,用高速组织捣碎机打浆以获得原料浆备用;采用纤维素酶和果胶酶对原料浆进行酶解,酶添加量500U/g,酶解液过滤后即得原料水提液;
优选地,步骤S1中,纤维素酶和果胶酶的酶活比为1.5:1;
步骤S1中,酶解条件为:55℃温度下酶解150min,酶解结束后90℃灭酶15min;
步骤S1中,酶解液经100目筛网过滤。
S2、杀菌:原料水提液分装后经80℃杀菌20min;
S3、益生菌发酵:将益生菌进行菌种活化,制备种子培养液进行接种发酵,接种量10%;
优选地,步骤S3中,益生菌由保加利亚乳杆菌、嗜热链球菌、嗜酸乳杆菌构成,且保加利亚乳杆菌、嗜热链球菌、嗜酸乳杆菌的菌种比例为1:1:1;
步骤S3中,发酵温度为37℃,静置发酵7天。
S4、发酵液过滤:发酵结束,发酵液经3000r/min、2~8℃条件下离心15min,再经40-60μm滤膜过滤,即得富硒酵素原液。
步骤S4中,优选为4℃条件下离心。

本发明第四方面保护第一方面所述富硒植物酵素原液在保健食品方面的应用。
富硒植物是天然抗氧化剂的很好的载体,其中含有的多糖类抗氧化剂不仅可以清除自由基,抑制细胞脂质过氧化,并且可以促进人体内产生抗氧化酶,比合成抗氧化剂更具优势。富硒甘蓝中的硒元素含量高达1200μg/g,其中的植物有机硒化合物如植物硒蛋白、硒多糖可以有效促进人体内源性含硒酶GSH-Px等的合成,发挥抗氧化、抑制慢性炎症病变的作用。
灰树花是一种富含蛋白质、膳食纤维、多种矿物质元素的食品,享有“食用菌王子”和“华北人参”的美誉。灰树花中含有灰树花多糖等活性物质,有研究者从灰树花中分离提纯出1种脂肪酸和3种麦角淄醇化合物,并对它们进行了体外抗氧化试验,结果显示它们均有抗氧化和抑制环氧合酶的作用。灰树花多糖对受辐射的小鼠有一定的防护作用,可以提高小鼠的存活率。
紫云英花粉的蛋白质含量可达到21.8%,维生素E,维生素C等抗氧化活性的维生素也有较高含量,其中还有多种游离氨基酸,可以被人体高效吸收利用。紫云英花粉性甘平,有益于消化系统病变及食欲不佳者,可促进消化吸收和溃疡的愈合。
蓝莓含有丰富的蓝莓花色苷,蓝莓花色苷具有抗氧化、抗癌等生物活性。有研究发现其抗脂质过氧化的能力强于抗坏血酸,还原能力和清除超氧阴离子自由基的能力略低于抗坏血酸。
刺梨中富含多种维生素、氨基酸及其他重要的生物活性物质,其中维生素C和芦丁的含量居蔬菜水果之首。并且刺梨中的氨基酸有18种。刺梨具有调节机体免疫功能,延缓衰老、解毒等功能。
红枣属于药食同源植物,早在《名医别录》、《本草纲目》等书中就有记载,红枣味甘性温,久食有补气养血,益脾胃、润肤养颜等功效。
本发明一种高效抗氧化的富硒植物酵素原液及其制备和应用,其优点在于:
(1)通过利用富硒甘蓝和灰树花、紫云英花粉等的组合,不仅可以起到直接的抗氧化作用,还有助于体内产生内源性抗氧化酶GSH-Px,协同发挥抗氧化作用;
(2)本发明利用独特的益生菌组合发酵,有助于将大分子蛋白质分解成小分子氨基酸,同时将大分子植物有机硒分解为小分子硒代氨基酸或者其衍生物,将硒的促进体内抗氧化酶合成的作用和其他原料中的维生素、花色苷等活性物质的清除自由基的作用相结合,有助于皮肤和其他各器官对抗氧化成分的有效利用,以提高清除自由基的效果;
(3)本发明所述原料均为天然可食用植物或食用菌来源,安全性有保障。
具体实施方式
下面结合具体实施案例,进一步阐述本发明。这些实施案例仅用于说明本发明而不用于限制本发明的范围。下列实施案例中未注明具体条件的实验方法,通常按照常规条件,或按照制造厂商所建议的条件。

S2、杀菌:原料水提液分装后经80℃杀菌20min;
S3、益生菌发酵:将益生菌(由保加利亚乳杆菌、嗜热链球菌、嗜酸乳杆菌构成,且保加利亚乳杆菌、嗜热链球菌、嗜酸乳杆菌的菌种比例为1:1:1)进行菌种活化,制备种子培养液,并将种子培养液接种至步骤S2获得的原料水提液中,接种量10%,发酵温度为37℃,静置发酵7天;
S4、发酵液过滤:发酵结束,发酵液经3000r/min、4℃条件下离心15min,再经40-60μm滤膜过滤,即得富硒酵素原液。
本实施例中制得产品每1ml硒含量为50微克(检测方法:按GB5009.93规定方法检测)。
使用方法:按照营养学会对硒的每日推荐服用量加入食品、保健食品中使用,或以1%-5%的比例加入护肤品中使用。

富硒植物酵素原液在清除自由基方面的应用
对实施例1获得产品的自由基清除效率进行试验验证。
1试验材料和方法:
1.1试验待测样品
富硒植物酵素原液、维生素C
1.2试验试剂
DPPH、无水乙醇、水杨酸、盐酸、硫酸亚铁、过氧化氢、ABTS、过硫酸钾、超纯水、磷酸盐缓冲液。
1.3试验方法
1.3.1待测样品的配制方法
富硒植物酵素原液用超纯水分别稀释0倍、2倍、4倍和8倍,以构成浓度不同的四组富硒植物酵素原液,测得四组富硒植物酵素原液的浓度分别为100%、50%、25%、12.5%。维生素C用超纯水配制成0.2mg/ml。
1.3.1 DPPH自由基清除率的测定
将1mL的待测样品溶液和1mL的0.1mmol/L的DPPH乙醇溶液均匀混合,在室温下避光静置30min,随后在517nm处用无水乙醇作参比溶液测定其吸光值A1。同时将1mL无水乙醇和1mL浓度为0.1mmol/L的DPPH溶液混合液的吸光值A0作为空白值,1mL的待测样品溶液和1mL无水乙醇混合液的吸光值A2作为背景值。DPPH自由基清除率的计算公式如下:

式中:
A0是1mL 0.1mmol/L DPPH溶液与1mL无水乙醇混合的吸光值;
A1是1mL的0.1mmol/L的DPPH乙醇溶液与1mL的待测样品溶液混合的吸光值;
A2是1mL的乙醇溶液与1mL的待测样品溶液混合的吸光值。
1.3.2羟基自由基清除率的测定
将1mL浓度为9mM的FeSO4、1mL浓度为9mM的水杨酸乙醇溶液,加入到1mL的样品溶液中,再加入1mL浓度为8.8mM的H2O2,于37℃水浴锅中恒温反应60min,在510nm的条件下以超纯水作为参比溶液测定反应体系的吸光值A3。同时将1mL浓度为9mM的FeSO4、1mL浓度为9mM的水杨酸乙醇溶液、1mL浓度为8.8mM的H2O2和1mL超纯水混合液的吸光值A4作为空白值,将1mL浓度为9mM的FeSO4、1mL浓度为9mM的水杨酸乙醇溶液、1mL的样品溶液和1mL超纯水混合液的吸光值A5作为背景值。羟基自由基清除率的计算公式如下:
羟自由基清除率(%)=[1-(A3-A5)/A4]×100
公式中:
A4为超纯水代替样品的空白吸光值;
A3为待测样品的吸光值;
A5为超纯水代替H2O2的背景吸光值。
1.3.3 ABTS自由基清除率的测定
将浓度为7mmol/L的ABTS溶液和2.45mmol/L的K2S2O8溶液等体积混合均匀,混合液在室温避光条件下静置12-16h,以磷酸盐缓冲液(0.2M,pH7.4)稀释,使其在734nm时吸光度达到0.70±0.02,形成ABTS+测定液。取4.0mL的ABTS+测定液,加入100μL样品溶液,准确振荡10s,测定反应体系在734nm处的吸光值A6。将磷酸盐缓冲液与ABTS+测定液混合溶液的吸光值A7作为空白值,磷酸盐缓冲液与样品混合溶液的吸光值A8作为背景值。ABTS自由基清除率的计算公式如下:
ABTS自由基清除率(%)=[1-(A6-A8)/A7]×100
式中:
A7为磷酸盐缓冲液与ABTS+测定液混合溶液的吸光值;
A6为待测样品液与ABTS+测定液混合溶液的吸光值;
A8为磷酸盐缓冲液与样品混合溶液的吸光值。
2试验结果:
2.1:富硒植物酵素原液清除DPPH自由基的能力
由表1可见,富硒植物酵素原液的DPPH自由基清除能力随着其浓度的增加而增强,当富硒植物酵素原液浓度从25%增加到50%时,DPPH自由基清除能力增加迅速,富硒植物酵素原液的浓度达到100%时,其清除DPPH自由基的清除率与维生素C溶液效果相当。

2.2:富硒植物酵素原液清除清除羟自由基的能力
由表6可见,富硒植物酵素原液的羟自由基清除能力随着其浓度的增加而增强,富硒植物酵素原液的浓度达到100%时,其清除羟自由基的清除率略高于维生素C溶液。

由表7可见,富硒植物酵素原液的ABTS自由基清除能力随着其浓度的增加而增强,富硒植物酵素原液的浓度达到100%时,其清除ABTS自由基的清除率接近于维生素C溶液。

试验结果表明,富硒植物酵素原液具有较高的抗氧化活性,且当其浓度增加到50%以上时,抗氧化活性更强,浓度为100%时,抗氧化活性与维生素C接近。

Claims (8)
1.一种高效抗氧化的富硒植物酵素原液,其特征在于:主要由以下重量份原料组成:
富硒甘蓝或其提取物2-4份,灰树花粉或其提取物3-5份,紫云英花粉或其提取物2-4份,蓝莓提取物或其提取物1-3份。
2.根据权利要求1所述一种高效抗氧化的富硒植物酵素原液,其特征在于:富硒甘蓝或其提取物3-4份,灰树花粉或其提取物4-5份,紫云英花粉或其提取物2-3份,蓝莓提取物或其提取物1-2份。
3.根据权利要求1所述一种高效抗氧化的富硒植物酵素原液,其特征在于:还包括刺梨粉或其提取物1-3份、红枣粉或其提取物1-2份。
4.制备权利要求1至3任一所述富硒植物酵素原液的方法,其特征在于:包括如下步骤:
S1、制备原料水提液:各原料按比例混合均匀、过筛,再将过筛后的原料与水按照1:10的质量比混合,90℃浸泡60min后,用高速组织捣碎机打浆以获得原料浆备用;采用纤维素酶和果胶酶对原料浆进行酶解,酶添加量500U/g,酶解液过滤后即得原料水提液;
S2、杀菌:原料水提液分装后经80℃杀菌20min;
S3、益生菌发酵:将益生菌进行菌种活化,制备种子培养液进行接种发酵,接种量10%;
S4、发酵液过滤:发酵结束,发酵液于3000r/min、2~8℃条件下离心15min,再经40-60μm滤膜过滤,即得富硒酵素原液。
5.根据权利要求4所述方法,其特征在于:步骤S1中,酶解条件为:55℃温度下酶解150min,酶解结束后90℃灭酶15min。
6.根据权利要求4所述方法,其特征在于:步骤S3中,益生菌由保加利亚乳杆菌、嗜热链球菌、嗜酸乳杆菌构成,且保加利亚乳杆菌、嗜热链球菌、嗜酸乳杆菌的菌种比例为1:1:1。
7.权利要求1至3任一所述富硒植物酵素原液在化妆品方面的应用。
8.权利要求1至3任一所述富硒植物酵素原液在保健食品方面的应用。
