Abstract
本发明属于中药制剂领域,具体涉及一种甘草泻心汤固体制剂的制备工艺,主要包括以下步骤:煎煮、过滤、离心、浓缩、干燥、制粒。本发明解决了常规生产工艺存在的无法保证产品与传统工艺获得的产品在内在质量上的一致性的问题,通过使用HPLC多组分指纹图谱检测方法对产品质量进行全面监控,且制备过程较为简单,以传统煎煮工艺为基础,进行现代化改进,但没有醇沉和高温干燥等过程,避免了药物成分发生改变,最大程度上保证了与传统工艺获得的产品在质量上和化学成分上的一致性。
Description
技术领域
本发明属于中药制剂领域,具体涉及一种甘草泻心汤固体制剂的制备工艺。
背景技术
甘草泻心汤出自张仲景的《伤寒论》,是治疗狐惑病和虚痞的名方,是张仲景五大泻心汤之一,由炙甘草、黄芩、干姜、大枣、半夏、黄连等6味药物组成。甘草泻心汤是治疗脾胃之方,因胃虚不能调理上下,故出现上火之口腔溃疡,下寒之大便溏泻,中焦之脾胃痞满,所以服用此方皆可得到治疗。
现代药理研究表明,甘草泻心汤具有调节胃黏液分泌、抗溃疡性结肠炎、护肝、增强机体免疫机能和提高抗缺氧能力等药理作用,现代临床主要用于治疗白塞病、口腔溃疡、反流性食管炎、溃疡性结肠炎、胃肠神经官能症等疾病。
《伤寒论》中甘草泻心汤的组成为:甘草四两(炙),黄芩三两,干姜三两,大枣十二枚(擘),半夏半升(洗),黄连一两。制法为:上六味,以水一斗,煮取六升,去滓,再煎取三升,温服一升,日三服。
目前,中成药大生产中普遍采用的是水提取或醇提取、浓缩后醇沉、高温烘干、粉碎、成型等工艺流程,这种常规的生产工艺具有设备简单、成本低且技术成熟等优势,在实际产业化中深受欢迎。然而,这种生产工艺存在两个明显的缺陷,一是醇沉过程可能产生某些化学反应,二是高温干燥过程会破坏某些热不稳定成分。这两个缺陷均可能会使获得的中药制剂在大生产过程中物质基础发生变化,导致临床疗效上的差异。因此,沿用目前的常规中成药提取工艺对经典名方进行产业化可能存在成药缺陷性,即无法保证大生产与传统工艺获得的产品在内在质量上的一致性。
中国专利申请CN 102895620 A公开了一种甘草泻心汤整合型新剂型制备技术及其生产方法,该发明由甘草15份、黄芩9份、黄连6份、党参12份、半夏9份、干姜9份、大枣4份组成,通过超音速气流粉碎、醇水提取、超声波粉碎提取、水煎浓缩、超音速喷雾干燥、纳米研磨、高压乳匀、纳米粒制备等步骤制得。该发明运用纳米载体联用技术和中西医结合理论研发“纳米药物整合型新剂型”,产品具有靶向分布广、缓控释性能强等特点,但该发明存在制备过程复杂,所得药物中的化学成分较多,与传统处方相差较大,可能会导致临床上使用该药物的疗效出现差异的缺点。
发明内容
为解决现有技术中存在的缺陷和不足,本发明的主要目的在于提供一种甘草泻心汤固体制剂的制备工艺,所述制备工艺方法简单,尊重传统的用药习惯和煎煮工艺,用现代化设备进行提取,并利用HPLC多组分指纹图谱检测方法监控大生产过程中每一步骤前后的化学成分,保证了提取的制剂与传统工艺所得汤剂在化学成分上的一致性。此外,还具有携带方便、稳定性好及服用方便等优点。
为了达到上述目的,本发明通过以下技术方案实现:
一种甘草泻心汤固体制剂的制备工艺,包括以下步骤:煎煮、过滤、离心、浓缩、干燥、制粒。
优选地,所述制备工艺的具体步骤如下:
(1)将炙甘草、黄芩、干姜、姜半夏、黄连、大枣按照重量比为4:3:3:3:1:3的比例混合均匀,加入体积为药材重量5~10倍的水,加热煮沸后用小火保持微沸0.8~1.5h;
(2)微沸提取后用100~300目筛过滤,滤液加热煮沸,用小火保持微沸0.5~1h;
(3)微沸后的提取液通过高速离心除去杂质和大分子物质,得到澄清的提取液;
(4)将步骤(3)得到的提取液通过减压低温浓缩,浓缩到相对密度为1.01~1.20,备用;
(5)将步骤(4)获得的浓缩液加入辅料或不加辅料,混匀,通过喷雾干燥快速干燥成粉末;
(6)将步骤(5)获得的粉末,通过湿法制粒、干法制粒或沸腾制粒制备成颗粒剂;
(7)对获得的颗粒剂进行整体质量控制,通过HPLC多组分指纹图谱检测方法对产品质量进行全面监控。
优选地,所述的高速离心为碟片离心或管式离心,其中碟片离心转速为8000~12000r/min;管式离心转速为8000~25000r/min。
优选地,所述的辅料为可溶性淀粉、麦芽糊精和蔗糖的一种或几种。
优选地,所述的喷雾干燥的进风口温度为130~180℃,出风口温度为60~85℃,料液处理量为8~200kg/h,其中气流式喷雾干燥气流量为0.5~3.0m3/min;离心式喷雾干燥离心速率8000~20000r/min,喷雾盘直径为50~350mm。
优选地,所述的沸腾制粒进料温度为25~50℃,进风口温度为60~155℃,出风口温度35~75℃,料液处理量10~100kg/h。
优选地,所述HPLC多组分指纹图谱的色谱条件如下:使用Phenomenon C18色谱柱(250mm×4.6mm,5μm),柱温为室温,流速为1mL/min,检测波长为230nm,进样量10μL,流动相及梯度洗脱条件如下表所示:

优选地,所述指纹图谱要求样品指纹图谱与对照指纹图谱相比较,相似度在0.90以上。
与现有技术相比,本发明具有以下有益效果:
1、本发明制备的甘草泻心汤固体制剂携带便捷,服用方便,便于储存,有利于市场推广;
2、本发明的煎煮工艺参数以传统煎煮工艺参数为基础,进行了优化和改进,并取得了更加优异的提取效果;
3、本发明的浓缩除杂过程在低温条件下采用物理方式除去杂质,不会引起受热分解、化学成分变化等物质基础的改变;干燥过程采用瞬间干燥技术,不会使物质成分因高温而不稳定;
4、本发明在标准汤剂制备完成后,建立以标准汤剂为参照的整体质量控制模式,采用指纹图谱技术全面监控标准汤剂中整体的化学物质成分组成,将建立的多组分控制指标用于产品的稳定性研究以保证成药的市场化。

具体实施方式
实施例1
一种甘草泻心汤固体制剂的制备工艺,包括以下步骤:
(1)选取按国家标准检验合格的炙甘草3.7kg、黄芩2.8kg、干姜2.8kg、姜半夏2.8kg、黄连0.9kg、大枣2.8kg;
(2)将药材放入提取罐中,加入120L水,加热煮沸,并保持沸腾1h后停止加热;
(3)将得到的提取液用150目的筛网进行过滤,滤液继续加热煮沸,并保持沸腾40min后停止加热;
(4)将药液放冷后,用管式离心机(10000r/min)离心,得澄清提取液(取样检测1)。
(5)将提取液减压(≤0.08MPa)低温(≤65℃)浓缩,浓缩至相对密度为1.04g/mL,停止加热浓缩(取样检测2)。
(6)将药液进行喷雾干燥,喷雾干燥参数为:进风口温度160℃,出风口温度65℃,进药速度8kg/h,气流量为1.6m3/min,获得干燥粉末,备用(取样检测3)。
(7)将制备得到的粉末粉末用沸腾制粒机制粒,进料温度为37℃,进风口温度为110℃,出风口温度60℃,料液处理量10kg/h,制粒后的颗粒剂即为甘草泻心汤颗粒(取样检测4)。
对比例1
一种甘草泻心汤整合型新剂型制备技术及其生产方法,包括以下步骤:
(1)将甘草15份、黄芩9份、黄连6份、党参12份、半夏9份、干姜9份、大枣4份进行预处理,称量后置入粉碎机粉碎至80-200目,然后采用超音速气流粉碎至1-10μm,得不同中药微米粉,备用。
(2)取60%的甘草、黄芩、黄连、党参微米粉用6-12倍量的30%~90%醇水溶液混合浸泡1-2天,回流提取3次,提取水溶性、醇溶性、酯溶性三种有效药物成份,回收酒精,得提取物备用。
(3)取60%的半夏、干姜、大枣微米粉,采用超声波分散萃取设备分别进行分散提取,超声功率为:800~3000W、超声频率:28~120KHz、粉碎提取时间为10-100min,得提取物备用。
(4)将(2)、(3)所得中药提取物,采用纳米球磨机研磨4~16h,得粒径分布为50~1000nm的纳米粉,备用。
(5)取40%的各中药微米粉,用水浸泡2h后用中药煎药机水煎煮3次,煎液合并过滤,滤液浓缩,得中药浸膏或水煎浓缩液以备用。
(6)在GMP10000级洁净室里将水煎浓缩液、纳米脂质体、纳米乳、微乳按4:2:2:2比例进行混合;
(7)采用2010年版《中国药典》一部制剂部分软胶囊剂标准,不添加辅料、防腐剂,采用全自动软胶囊灌装机分装软胶囊,制成软胶囊剂;
(8)对软胶囊剂进行数粒、装瓶、封口、粘贴标贴、打码、装箱等内外包装,最后按生产批次统一瞬间灭菌,并开具灭菌报告单。
由实施例1和对比例1可知,对比例1的制备过程中有醇提取过程,可能会在提取过程中产生某些化学反应,导致临床上使用时的疗效出现差异,且该制备方法较为繁琐,工艺流程较本发明更复杂一些。本发明制备过程较为简单,以传统煎煮工艺为基础,进行现代化改进,但没有醇沉和高温干燥等过程,避免了药物成分发生改变,最大程度上保证了与传统工艺获得的产品在质量上和化学成分上的一致性。
实施例2
甘草泻心汤指纹图谱的建立,包括以下步骤:
1、中药标准汤剂的制备
(1)在市场上购买15批以上的代表性的炙甘草、黄芩、干姜、姜半夏、黄连和大枣,每批处方量为:炙甘草62.4g,黄芩46.8g,干姜46.8g,姜半夏46.8g,黄连15.6g,大枣46.8g;
(2)将以上药材置于药煲中,加入2000mL水,加热煮沸,并用小火保持微沸60min;微沸提取结束后,过滤,滤液继续加热煮沸,并用小火保持微沸40min,即得15批甘草泻心汤标准汤剂。
2、指纹图谱的建立
(1)供试品溶液的制备
取本品的标准汤剂,稀释2倍后,用0.45μm水相滤膜过滤即得标准汤剂的供试品溶液(1mL含药材0.22g);生产过程中的浓缩液、中间体、粉末均分别用水定容至1mL含药材0.22g的溶液,用0.45μm水相滤膜过滤即得以上样品的供试品溶液。
(2)色谱条件的选择
方法一:Phenomenon C18色谱柱(250mm×4.6mm,5μm),柱温为室温,流速为1mL/min,检测波长:230、254、350nm,进样量10μL,流动相及梯度洗脱条件如下表所示。

得到的色谱图见附图1。
结果:色谱峰在30~40min、50~55min较密集,个别色谱峰之间分离度不好,230nm下的色谱信息较丰富。
方法二:Phenomenon C18色谱柱(250mm×4.6mm,5μm),柱温为室温,流速为1mL/min,检测波长:230nm,进样量10μL,流动相及梯度洗脱条件如下表所示。
得到的色谱图见附图2。
结果:色谱峰分离度较好,色谱信息较丰富。
方法三:Phenomenon C18色谱柱(250mm×4.6mm,5μm),柱温为室温,流速为1mL/min,检测波长:230nm,进样量10μL,流动相及梯度洗脱条件如下表所示。

结果:个别色谱峰之间有重叠。

结果:个别色谱峰之间有重叠,35~40min色谱峰较密集。
方法五:Phenomenon C18色谱柱(250mm×4.6mm,5μm),柱温为室温,流速为1mL/min,检测波长:230nm,进样量10μL,流动相及梯度洗脱条件如下表所示。

结果:52min左右的色谱峰之间有重叠。
(3)指纹图谱色谱条件确定
通过上述实验,确认指纹图谱方法为方法二,即:Phenomenon C18色谱柱(250mm×4.6mm,5μm),柱温为室温,流速为1mL/min,检测波长:230nm,进样量10μL,流动相及梯度洗脱条件如下表所示。

(4)指纹图谱的建立
取15批次甘草泻心汤标准汤剂,按指纹图谱色谱条件进行高效液相色谱检测,得到的图谱通过中药色谱指纹图谱相似度评价系统进行分析,得到甘草泻心汤的对照指纹图谱,见附图6。
(5)指纹图谱中色谱峰的定位
选取黄芩苷对照品溶液,按指纹图谱色谱条件进行高效液相色谱检测,对甘草泻心汤对照指纹图谱中的黄芩苷进行定位,结果如附图7所示。其中S为黄芩苷色谱峰,S1(1)为黄芩苷对照品色谱图,S2(1)为甘草泻心汤对照指纹图谱。
实施例3
甘草泻心汤颗粒制备过程整体质量控制,包括以下步骤:
按照甘草泻心汤指纹图谱方法,对产品制备过程中离心后(实施例1中取样检测1),浓缩后(实施例1中取样检测2),干燥后(实施例1中取样检测3),制粒后(实施例1中取样检测4)得到的样品进行高效液相色谱分析,得到的色谱图见附图8。
与甘草泻心汤对照指纹图谱进行相似度分析,相似度均在0.90以上。说明各生产环节均能保证甘草泻心汤的内在质量基本不发生变化,制备的甘草泻心汤固体制剂基本不改变汤剂的物质基础。由此可以证明,本发明制备工艺得到的甘草泻心汤颗粒剂与传统工艺获得的产品相比,能在最大程度上保证质量上和化学成分上的一致性。
上述实施例仅示例性说明本发明的原理及其性能,不应视为对本发明的限制,本发明的保护范围应当以权利要求所限定的范围为准。对于本技术领域的普通技术人员而言,皆可在不违背本发明的精神及范畴下,对上述实施例进行修饰或改变。因此,举凡所属技术领域中具有通知常识者在未脱离本发明所揭示的精神与技术思想下所完成的一切等效修饰或改变,仍应由本发明的权利要求所涵盖。
Claims (5)
1.一种甘草泻心汤固体制剂的制备工艺,包括以下步骤:煎煮、过滤、离心、浓缩、干燥、制粒,所述制备工艺的具体步骤如下:
A、将炙甘草、黄芩、干姜、姜半夏、黄连、大枣按照重量比为4:3:3:3:1:3的比例混合均匀,加入体积为药材重量5~10倍的水,加热煮沸后用小火保持微沸0.8~1.5h;
B、微沸提取后用100~300目筛过滤,滤液加热煮沸,用小火保持微沸0.5~1h;
C、微沸后的提取液通过高速离心除去杂质和大分子物质,得到澄清的提取液;
D、将步骤C得到的提取液通过减压低温浓缩,浓缩到相对密度为1.01~1.20,备用;
E、将步骤D获得的浓缩液加入辅料或不加辅料,混匀,通过喷雾干燥快速干燥成粉末;
F、将步骤E获得的粉末,通过湿法制粒、干法制粒或沸腾制粒制备成颗粒剂;
G、对获得的颗粒剂进行整体质量控制,通过HPLC多组分指纹图谱检测方法对产品质量进行全面监控;
所述HPLC多组分指纹图谱的色谱条件如下:使用Phenomenon C18色谱柱(250mm×4.6mm,5μm),柱温为室温,流速为1mL/min,检测波长为230nm,进样量10μL,流动相及梯度洗脱条件如下表所示:
2.根据权利要求1所述的一种甘草泻心汤固体制剂的制备工艺,其特征在于,所述的高速离心为碟片离心或管式离心。
3.根据权利要求1所述的一种甘草泻心汤固体制剂的制备工艺,其特征在于,所述的辅料为可溶性淀粉、麦芽糊精和蔗糖的一种或几种。
4.根据权利要求1所述的一种甘草泻心汤固体制剂的制备工艺,其特征在于,所述的喷雾干燥的进风口温度为130~180℃,出风口温度为60~85℃,料液处理量为8~200kg/h,其中气流式喷雾干燥气流量为0.5~3.0m3/min。
5.根据权利要求1所述的一种甘草泻心汤固体制剂的制备工艺,其特征在于,所述的沸腾制粒进料温度为25~50℃,进风口温度为60~155℃,出风口温度35~75℃,料液处理量10~100kg/h。
