Abstract
本发明提出了一种调节血压的新型压片糖果及其制备方法,包括青钱柳15~25重量份、金花茶35~55重量份、辣木叶35~55重量份、蒲公英15~25重量份、葛根85~115重量份,葡萄糖粉10~30重量份、麦芽糊精10~30重量份、柠檬酸2~6重量份、硬脂酸镁10~15重量份、薄荷醇2~6重量份。制备方法包括提取、浓缩、干燥、压片等步骤,最佳提取条件为水体积1.5L,每次提取1h,提取4次。解决了现有技术中调节血压的新型压片糖果活性物质总黄酮含量低、治疗效果不好等技术问题。本发明实验设计合理,制得的成品总黄酮含量高、稳定性好,提高了调节血压的新型压片糖果的质量,简单方便,安全有效,无毒副作用。
Description
技术领域
本发明属于食品、保健品技术领域,尤其涉及一种压片糖果及其制备方法,具体涉及一种调节血压的新型压片糖果及其制备方法。
背景技术
高血压是持续血压过高的疾病,会引起中风、心脏病、血管瘤、肾衰竭等疾病。高血压是一种以动脉压升高为特征,可伴有心脏、血管、脑和肾脏等器官功能性或器质性改变的全身性疾病,分为原发性高血压和继发性高血压,严重影响着人们的健康。
压片糖又称粉糖或片糖,也常被称作汽水糖。它是以精制糖粉为主体,添加奶粉、香料等填充料和淀粉糖浆、糊精、明胶等粘合剂,经制粒压片成型的混合物。随着人们健康意识的增强,各种具有治疗疾病效果的压片糖果出现。但是,目前,关于调节血压的压片糖果产品较少,且存在活性物质含量低、治疗效果不好、稳定性差等缺点,现有技术有待发展完善。
发明内容
本发明提供了一种调节血压的新型压片糖果及其制备方法,解决现有技术中调节血压的压片糖果活性物质含量低、治疗效果不好、稳定性差等技术问题。
为了实现上述目的,本发明采用如下技术方案:
一种调节血压的新型压片糖果,包括青钱柳15~25重量份、金花茶35~55重量份、辣木叶35~55重量份、蒲公英15~25重量份、葛根85~115重量份,葡萄糖粉10~30重量份、麦芽糊精10~30重量份、柠檬酸2~6重量份、硬脂酸镁10~15重量份、薄荷醇2~6重量份。
作为优选,包括青钱柳15~20重量份、金花茶50~55重量份、辣木叶50~55重量份、蒲公英20~25重量份、葛根100~115重量份,葡萄糖粉10~15重量份、麦芽糊精10~15重量份、柠檬酸2~4重量份、硬脂酸镁10~12重量份、薄荷醇2~4重量份。
作为优选,包括青钱柳15重量份、金花茶55重量份、辣木叶55重量份、蒲公英25重量份、葛根115重量份,葡萄糖粉10重量份、麦芽糊精10重量份、柠檬酸2重量份、硬脂酸镁10重量份、薄荷醇2重量份。
本发明还包括一种调节血压的新型压片糖果的制备方法,包括以下步骤:
(1)干浸膏的制备
a提取:称取所述重量份的青钱柳、金花茶、辣木叶、蒲公英、葛根,加1~1.5L水,每次提取时间为1~2h,提取次数为2~4次;测定提取物中的总黄酮含量,动物实验评价提取物的功效;
b浓缩:合并滤液,将滤液进行蒸馏,收集馏分;
c干燥:将收集的馏分分别进行热风干燥、冷冻干燥处理,制成干浸膏;比较两种干燥工艺对干浸膏总黄酮含量的影响,确定最佳干燥工艺;
(2)压片的制备
将干浸膏粉碎后与所述重量份的葡萄糖粉、麦芽糊精、柠檬酸混合,加入水,放入制粒机,筛大小为20~40目;温度设定为55~60℃,干燥至糖粉水分在3%~6%之间;将干燥好的糖粉颗粒过20~40目筛,加入所述重量份的薄荷醇和硬脂酸镁,在V型混合机中混合5~10min后,装袋备用;然后,将糖粉放入进料斗,压片机压力设为5N~20N,压片,制得压片糖果成品。
作为优选,所述提取的水体积为1.5L,所述每次提取时间为1h,所述提取次数为4次。
作为优选,所述浓缩可以采用常压蒸馏或减压蒸馏的方法。
作为优选,所述冷冻干燥工艺为最佳干燥工艺。
作为优选,所述V型混合机中混合时间为5min。
作为优选,所述压片糖果硬度为70N~120N。
作为优选,所述压片糖果硬度为100N。
以上所述原料功效和主治病症如下:
青钱柳:别名:摇钱树、麻柳,青钱李、山麻柳、山化树。青钱柳树皮、叶具有清热消肿,止痛的功效。用于顽癣的治疗。中医临床用于治疗糖尿病,因其有药理作用能明显降低血糖、减脂肪和尿糖。
金花茶:属于山茶科、山茶属,是国家一级保护植物之一。金花茶具有明显的降血糖和尿糖作用,能有效的改善糖尿病“三高”症状,能够增强免疫力、调节血流量,防止动脉粥样硬化,抗菌消炎、清热解毒、通便利尿去湿,增进肝脏代谢、防癌抑制肿瘤生长等。
辣木叶:原产于印度的多年生热带落叶乔木辣木树(又称为鼓槌树)的树叶,可食用。辣木的种子和叶子中含有丰富的营养成分。有降血糖、降脂、降压、抗肿瘤等的功效。
蒲公英:性寒,味苦。入肝、胃经。具有清热解毒,利尿散结的功效。主治:急性乳腺炎,淋巴腺炎,瘰疬,疔毒疮肿,急性结膜炎,感冒发热,急性扁桃体炎,急性支气管炎,胃炎,肝炎,胆囊炎,尿路感染。
葛根:性凉,味甘、辛。具有解表退热,生津,透疹,升阳止泻的功效。主治:外感发热头痛、高血压颈项强痛、口渴、消渴、麻疹不透、热痢、泄泻。
上述提取物功效的评价方法如下:
(1)实验动物饲养及模型建立:高血压模型大鼠是通过腹腔注射一氧化氮合成酶抑制剂L-NNA(剂量为15mg/(kg*d),连续3周)诱导的。对照组给予1mL生理盐水腹腔注射,每日1次,连续3周。
(2)高血压大鼠模型建立成功后,将大鼠随机分为3组:模型组、提取物干预组、对照组,每组15只。期间各组大鼠给药方法如下所示:
对照组和模型组给予2ml生理盐水灌胃,每日1次,连续4周;
提取物干预组按100mg/(kg*d)分别灌胃实施例1中最佳提取条件制备的提取物,连续4周。
(3)药物干预4周后,用BP-6无创血压仪测量大鼠的血压,保证动物基本在同一个时间测定,避免日间自身血压波动的影响,记录实验结果,每组取平均值,比较实验结果。
具有如下有益效果:
本发明提供的是一种调节血压的新型压片糖果及其制备方法,
(1)通过设计单因素实验和五因素四水平的正交实验,以总黄酮含量以及提取物功效两项作为指标,对原料青钱柳、金花茶、辣木叶、蒲公英、葛根的配比进行了筛选,得到原料的最佳配比。单因素实验结果,得到了每个原料不同配比对总黄酮含量以及提取物功效的影响,为正交实验四个水平的参数设计提供了参考。五因素四水平的正交实验,高效率、快速地对原料配比进行了筛选。总黄酮是提取物的主要活性物质,对调节血压发挥着关键作用。功效评价是对提取物治疗血压效果好坏以及有无的准确评价。
(2)通过设计五因素四水平的正交实验,以吸湿率作为指标考察稳定性,对辅料青葡萄糖粉、麦芽糊精、柠檬酸、硬脂酸镁、薄荷醇的配比进行了筛选,得到辅料的最佳配比。吸湿率反映的是辅料在75%湿度环境下的稳定性。吸湿率过高,会导致片剂容易变质、保质期缩短、不方便保存等问题的出现。所以,吸湿率是考察辅料种类及配比的一项重要指标。
(3)通过设计三因素两水平的正交实验,以总黄酮含量作为指标,对提取原料的溶剂体积、提取时间、提取次数进行了筛选,得到最佳提取条件,优化了提取工艺,提高了提取物中总黄酮的含量。
综上,本发明提供的一种调节血压的新型压片糖果及其制备方法,制备工艺设计合理,制得的成品活性物质含量高、治疗效果显著、稳定性好,不易吸潮,方便储存,保质期长,从整体上提高了调节血压的新型压片糖果的质量。本发明提供的一种调节血压的新型压片糖果具有降血压、增强免疫力的多重功效,效果显著,简单方便,无毒副作用。
具体实施方式
为了使本发明的目的、技术方案及有益效果更加清楚明白,以下结合具体实施例,对本发明进行进一步详细说明。应当理解,此处所描述的具体实施例仅用以解释本发明,而不构成对本发明的限制。
实施例1
制备调节血压的新型压片糖果
1原料配比的筛选
1.1提取:将青钱柳、金花茶、辣木叶、蒲公英、葛根分别清洗干净,80℃烘干6h,加入1L水,混合均匀,提取4次,每次提取2h,浓缩滤液,测量相应指标。以总黄酮含量和提取物功效作为指标,通过单因素实验方法确定影响总黄酮含量和提取物功效的原料种类及对应的重量份。提取物功效采用大鼠的收缩压和舒张压表示。
1.2正交实验因素表,见表1。

1.3以总黄酮含量和提取物功效作为指标,五因素四水平的正交实验及极差分析,见表2。

从表2可知,实验4的总黄酮含量最高,为72%,收缩压16KPa、舒张压12KPa,血压正常,即各原料重量为青钱柳15g、金花茶55g、辣木叶55g、蒲公英25g、葛根115g,也就是说原料最佳配比为青钱柳:金花茶:苦瓜:山楂:栀子=15:55:55:25:115。另外,从表2的极差分析结果看出,葛根配比的变化对提取物中总黄酮含量的影响最大,其次是辣木叶、蒲公英,青钱柳、金花茶的影响较小。葛根配比的变化对提取物的功效影响最大,其次是辣木叶,蒲公英、青钱柳、金花茶的影响较小。
1.4验证:将青钱柳15g、金花茶55g、辣木叶55g、蒲公英25g、葛根115g,分别清洗干净,80℃烘干6h,加入1L水,混合均匀,提取4次,每次提取2h。总黄酮含量为72%,大鼠收缩压为16KPa,舒张压为11.2KPa。确定青钱柳:金花茶:辣木叶:蒲公英:葛根=15:55:55:25:115为最佳重量份配比。
2辅料配比的筛选
2.1按照上述确定的最佳重量份配比的原料与辅料葡萄糖粉、麦芽糊精、柠檬酸、硬脂酸镁、薄荷醇按照表3,混合均匀,放在75%的恒湿密闭容器中2h,测量吸湿前后的质量,并记录,计算吸湿率。
2.2正交实验因素表,见表3。

2.3以吸湿率为指标,进行五因素四水平的正交实验及极差分析,见表3。

从表4可知,实验1的吸湿率最低,为1.7%,即各辅料重量为葡萄糖粉10g、麦芽糊精10g、柠檬酸2g、硬脂酸镁10g、薄荷醇2g,也就是说辅料最佳配比为葡萄糖粉:麦芽糊精:柠檬酸:硬脂酸镁:薄荷醇=10:10:2:10:2。另外,从表4的极差分析结果看出,麦芽糊精配比的变化对吸湿率的影响最大,其次是葡萄糖粉,柠檬酸、硬脂酸镁、薄荷醇的影响较小。
3提取条件的筛选
3.1称取青钱柳15g、金花茶55g、辣木叶55g、蒲公英25g、葛根115g,分别清洗干净,80℃烘干6h,水提取,浓缩滤液。提取条件的正交实验因素表见表5。

3.2以总黄酮含量为指标,进行三因素两水平的正交实验及极差分析,见表6。

从表6可知,实验3的总黄酮含量最高,为74%,即溶剂体积为1.5L、每次提取1h、提取4次为最佳提取条件。另外,从表6的极差分析结果看出,提取时间的变化对吸湿率的影响最大,其次是溶剂体积,提取次数的影响较小。
4干燥工艺的筛选:将上述最佳原料配比得到的提取物合并滤液,浓缩,设计两组平行实验,分别进行热风干燥、冷冻干燥处理,制成干浸膏。考察两种干燥工艺对干浸膏总黄酮含量的影响。
结果见表7。

从表3可知,二者比较,冷冻干燥工艺制得的干浸膏总黄酮含量高,为74%,所以冷冻干燥工艺为最佳干燥工艺。
5压片
将上述制得的干浸膏粉碎后,加入10g葡萄糖粉、10g麦芽糊精、2g柠檬酸混合,加入20g水,放入制粒机,筛大小为20目。温度设定为55℃,干燥至糖粉水分在3%~6%之间。将干燥好的糖粉颗粒过20目筛,加入2g薄荷醇和10g硬脂酸镁,在V型混合机中混合5min后,装袋备用。然后,将糖粉放入进料斗,压片机压力设为20N,压片,制得压片糖果成品。测量压片糖果的硬度为100N,溶出度为95%。
实施例2
(1)实验动物饲养及模型建立:高血压模型大鼠是通过腹腔注射一氧化氮合成酶抑制剂L-NNA(剂量为15mg/(kg*d),连续3周)诱导的。对照组给予1mL生理盐水腹腔注射,每日1次,连续3周。
(2)高血压大鼠模型建立成功后,将大鼠随机分为四组:模型组、提取物干预组、对照组、阳性药物干预组,每组15只。期间各组大鼠给药方法如下所示:
对照组和模型组给予2mL生理盐水灌胃,每日1次,连续4周;
阳性药物干预组以15mg/(kg*d)的剂量连续灌胃卡托普利,每日1次,连续4周。
提取物干预组按100mg/(kg*d)分别灌胃实施例1中最佳提取条件制备的提取物,连续4周。
(3)药物干预4周后,用BP-6无创血压仪测量大鼠的血压,保证动物基本在同一个时间测定,避免日间自身血压波动的影响,记录实验结果,每组取平均值,比较实验结果。结果见表8。

从表6可知,提取物干预组能够降低高血压大鼠的收缩压和舒张压,使之恢复正常水平,且降压的效果最好,优于卡托普利。
当然,上述说明并非是对本发明的限制,本发明也并不限于上述举例,本技术领域的普通技术人员,在本发明的实质范围内作出的任何变化、添加或替换,也应属于本发明的保护范围。
Claims (10)
1.一种调节血压的新型压片糖果,其特征在于,包括青钱柳15~25重量份、金花茶35~55重量份、辣木叶35~55重量份、蒲公英15~25重量份、葛根85~115重量份,葡萄糖粉10~30重量份、麦芽糊精10~30重量份、柠檬酸2~6重量份、硬脂酸镁10~15重量份、薄荷醇2~6重量份。
2.根据权利要求1所述调节血压的新型压片糖果,其特征在于,包括青钱柳15~20重量份、金花茶50~55重量份、辣木叶50~55重量份、蒲公英20~25重量份、葛根100~115重量份,葡萄糖粉10~15重量份、麦芽糊精10~15重量份、柠檬酸2~4重量份、硬脂酸镁10~12重量份、薄荷醇2~4重量份。
3.根据权利要求2所述调节血压的新型压片糖果,其特征在于,包括青钱柳15重量份、金花茶55重量份、辣木叶55重量份、蒲公英25重量份、葛根115重量份,葡萄糖粉10重量份、麦芽糊精10重量份、柠檬酸2重量份、硬脂酸镁10重量份、薄荷醇2重量份。
4.根据权利要求1所述调节血压的新型压片糖果的制备方法,其特征在于,包括以下步骤:
(1)干浸膏的制备
a提取:称取权利要求1所述重量份的青钱柳、金花茶、辣木叶、蒲公英、葛根,加1~1.5L水,每次提取时间为1~2h,提取次数为2~4次;
b浓缩:合并滤液,将滤液进行蒸馏,收集馏分;
c干燥:将收集的馏分分别进行热风干燥、冷冻干燥处理,制成干浸膏;
(2)压片的制备
将干浸膏粉碎后与权利要求1所述重量份的葡萄糖粉、麦芽糊精、柠檬酸混合,加入水,放入制粒机;干燥,将干燥好的糖粉颗粒过20~40目筛,加入权利要求1所述重量份的薄荷醇和硬脂酸镁,在V型混合机中混合5~10min后,放入进料斗,压片机压力设为5N~20N,制得压片糖果成品。
5.根据权利要求4所述调节血压的新型压片糖果的制备方法,其特征在于,所述提取的水体积为1.5L,所述每次提取时间为1h,所述提取次数为4次。
6.根据权利要求4所述调节血压的新型压片糖果的制备方法,其特征在于,所述浓缩可以采用常压蒸馏或减压蒸馏的方法。
7.根据权利要求4所述调节血压的新型压片糖果的制备方法,其特征在于,所述冷冻干燥工艺为干浸膏的最佳干燥工艺。
8.根据权利要求4所述调节血压的新型压片糖果的制备方法,其特征在于,所述V型混合机中混合时间为5min。
9.根据权利要求4~8所述调节血压的新型压片糖果的制备方法,其特征在于,所述压片糖果硬度为70N~120N。
10.根据权利要求9所述调节血压的新型压片糖果的制备方法,其特征在于,所述压片糖果硬度为100N。
