Abstract
本发明公开了土鳖虫多糖的用途,将其用于制备钙片,具体过程为:取4‑8份当归、2‑6份枸杞以及10‑12份肉桂混合,水提醇沉法提取,向所得浸膏中加入2‑4份土鳖虫多糖,添加碳酸钙制成钙片。有益效果为:本发明所公布的钙片制备工艺方法简单,制备时间短,所制备的钙片崩解时间短,口感好,生物利用率高。

技术领域
本发明涉及保健品技术领域,尤其是涉及土鳖虫多糖的用途。
背景技术
近几年来,随着分子生物学的发展,人们逐渐认识到多糖、蛋白质与核算一样是涉及生命活动本质的三类生物大分子之一。多糖是一类具有广泛生活活性的生物大分子,又称多聚糖。它不仅是人体生命必需的成分,而且也存在于一切细胞膜结构中,幷参与多种生命功能活动。
发明内容
本发明的目的是提供土鳖虫多糖在制备钙片中的用途,制备方法简单,时间短,所制备的钙片口感好,生物利用率高。
为了达到上述目的本发明提供的技术方案为:土鳖虫多糖的用途,具体过程为:取4-8份当归、2-6份枸杞以及10-12份肉桂混合,水提醇沉法提取,向所得浸膏中加入2-4份土鳖虫多糖,添加碳酸钙制成钙片。方法简单,制备时间短,所制备的钙片崩解时间短,口感好,生物利用率高。
作为优选,土鳖虫多糖的制备包括以下步骤:
脱脂:对土鳖虫进行粉碎,加入石油醚,浸泡,除去石油醚,将所得土鳖虫置于40-45℃干燥,通过脱脂处理,避免了脂质对后续提取和纯化过程的影响,提高了所制备的土鳖虫多糖的纯度;
提取:取干燥的土鳖虫,加入蒸馏水,调pH为4-5,加入木瓜蛋白酶提取,然后升温至100℃灭酶活,抽滤,将所得滤液浓缩得土鳖虫多糖粗体物,通过酶解法提取土鳖虫多糖,能够有效破坏蛋白质与多糖之间的结合键,使土鳖虫多糖能够被充分提取,提高土鳖虫的资源利用率;
纯化:将所得土鳖虫多糖粗提物用Sevage法进行除蛋白,接着加入95%的乙醇醇沉24h,得土鳖虫多糖,该条件下,多糖的损失率低,对多糖活性的破坏小。
作为优选,土鳖虫与石油醚的料液比为1g:3.5-4mL。该料液比提供了恰当的浓度梯度,增加了土鳖虫所含的脂质向石油醚中扩散的动力,从而提高了脂质的去除率,且该条件下石油醚消耗较少,节约了资源,具有良好的环境保护效益。
作为优选,浸泡时间为9-10h。该条件下,土鳖虫中所含的脂质可以被石油醚充分溶出,且所用的时间较短,缩短了制备工艺的周期,提高了工艺的经济价值。
作为优选,木瓜蛋白酶的加入量占土鳖虫质量的0.01%-0.02%,提取温度为45-55℃,提取时间为3-4h。该条件下,蛋白酶的活性高,酶解反应速度快,且木瓜蛋白酶与底物结合的机率高,使蛋白酶能够有效破坏土鳖虫多糖与蛋白质之间的氢键,使土鳖虫多糖能够被充分提取。
与现有技术相比,本发明的有益效果为:本发明所公布的制备工艺方法简单,制备时间短,所制备的钙片崩解时间短,口感好,生物利用率高;通过脱脂处理,避免了脂质对后续提取和纯化过程的影响,提高了所制备的土鳖虫多糖的纯度;通过酶解法提取土鳖虫多糖,能够有效破坏蛋白质与多糖之间的结合键,使土鳖虫多糖能够被充分提取,提高土鳖虫的资源利用率。
具体实施方式
以下通过实施例进一步详细描述:
实施例1:土鳖虫多糖的用途,包括以下步骤:
1)脱脂:对土鳖虫进行粉碎,按料液比1g:3.5mL加入石油醚,浸泡9h,除去石油醚,将所得土鳖虫置于40℃干燥,通过脱脂处理,避免了脂质对后续提取和纯化过程的影响,提高了所制备的土鳖虫多糖的纯度,且该料液比提供了恰当的浓度梯度,增加了土鳖虫所含的脂质向石油醚中扩散的动力,从而提高了脂质的去除率,且该条件下石油醚消耗较少,节约了资源,具有良好的环境保护效益;

4)钙片的制备:取8份当归、6份枸杞以及12份肉桂混合,水提醇沉法提取,向所得浸膏中加入4份土鳖虫多糖,添加碳酸钙制成钙片。
实施例3:土鳖虫多糖的用途,包括以下步骤:
1)脱脂:对土鳖虫进行粉碎,按料液比1g:3.6mL加入石油醚,浸泡9h,除去石油醚,将所得土鳖虫置于42℃干燥,通过脱脂处理,避免了脂质对后续提取和纯化过程的影响,提高了所制备的土鳖虫多糖的纯度,且该料液比提供了恰当的浓度梯度,增加了土鳖虫所含的脂质向石油醚中扩散的动力,从而提高了脂质的去除率,且该条件下石油醚消耗较少,节约了资源,具有良好的环境保护效益;
2)提取:取干燥的土鳖虫,加入蒸馏水,调pH为4,加入占土鳖虫质量的0.015%木瓜蛋白酶,再加入占木瓜蛋白酶质量0.02%的3,5-二氯苯甲酸和占木瓜蛋白酶质量0.023%的3,5-二羟基苯乙酮,在温度为47℃提取4h,然后升温至100℃灭酶活,抽滤,将所得滤液浓缩得土鳖虫多糖粗体物,通过酶解法提取土鳖虫多糖,能够有效破坏蛋白质与多糖之间的结合键,使土鳖虫多糖能够被充分提取,提高土鳖虫的资源利用率,且该条件下蛋白酶的活性高,酶解反应速度快,且木瓜蛋白酶与底物结合的机率高,使蛋白酶能够有效破坏土鳖虫多糖与蛋白质之间的氢键,使土鳖虫多糖能够被充分提取,所加入的3,5-二羟基苯乙酮、3,5-二氯苯甲酸与木瓜蛋白酶肽链中的Asn-158具有协同作用,能够促进His-159的咪唑环的摆放,使得Cys-25的去质子化过程更加顺畅,提高了Cys-25与羰基碳之间的亲核反应速率,从而提高了蛋白酶的酶解速率,缩短了土鳖虫多糖的提取时间;
3)纯化:将所得土鳖虫多糖粗提物用Sevage法进行除蛋白,接着加入95%的乙醇醇沉24h,得土鳖虫多糖;
4)钙片的制备:取5份当归、4份枸杞以及11份肉桂混合,水提醇沉法提取,向所得浸膏中加入3份土鳖虫多糖,添加碳酸钙制成钙片。

1.土鳖虫多糖的用途,其特征在于将土鳖虫多糖用于制备钙片中的用途,所述制备的具体过程为:取4-8份当归、2-6份枸杞以及10-12份肉桂混合,水提醇沉法提取,向所得浸膏中加入2-4份土鳖虫多糖,添加碳酸钙制成钙片。
2.根据权利要求1所述的土鳖虫多糖的用途,其特征在于:所述土鳖虫多糖的制备方法包括以下步骤:
1)脱脂:对土鳖虫进行粉碎,加入石油醚,浸泡,除去石油醚,将所得土鳖虫置于40-45℃干燥;
2)提取:取干燥的土鳖虫,加入蒸馏水,调pH为4-5,加入木瓜蛋白酶提取,然后升温至100℃灭酶活,抽滤,将所得滤液浓缩得土鳖虫多糖粗体物;
3)纯化:将所得土鳖虫多糖粗提物用Sevage试剂进行除蛋白,接着加入95%的乙醇醇沉24h,得土鳖虫多糖。
3.根据权利要求2所述的土鳖虫多糖的用途,其特征在于:所述步骤1)中土鳖虫与石油醚的料液比为1g:3.5-4mL。
4.根据权利要求2所述的土鳖虫多糖的用途,其特征在于:所述步骤1)中浸泡时间为9-10h。
5.根据权利要求2所述的土鳖虫多糖的用途,其特征在于:所述步骤2)中提取温度为45-55℃,提取时间为3-4h。
6.根据权利要求2所述的土鳖虫多糖的用途,其特征在于:所述步骤2)中木瓜蛋白酶的加入量占土鳖虫质量的0.01%-0.02%。
