Description
技术领域
本发明涉及益生菌双歧杆菌菌株的基因组、由该基因组编码的蛋白质以及由该蛋白质制备的胞外多糖。双歧杆菌是存在于人类结肠微生物区系中的若干种优势可培养细菌中的一种。
背景技术
认为双歧杆菌是益生菌,因为它们是当足量摄入时,除基本的营养成分外还提供健康有益效果的活体生物体。为了提供益生菌效应,高水平摄入的双歧杆菌必须到达它们的作用位点。建议使用的最低含量为大约106-107个活体双歧杆菌/g肠内容物(Bouhnik,Y.,Lait 1993)。有文献报道在成人和婴儿中完成的体内研究指示一些双歧杆菌菌株能够在通过胃肠道后依然存活。已经观察到了在不同双歧杆菌菌株的耐酸和胆盐的能力之间的显著差异,这指示存活能力是选择潜在的益生菌菌株的重要判据。
摄取双歧杆菌能够改善肠胃传输并可预防或有助于治疗疾病,所述疾病可由缺乏微生物区系或有害的微生物区系引起,诸如胃肠道(GIT)感染、便秘、肠易激综合征(IBS)、炎性肠病(IBD)–克隆氏病和溃疡性结肠炎、食物过敏、抗生素诱发的腹泻、心脏血管疾病和某些癌症(如结肠直肠癌)。
由于它们感觉到的促健康活性,近年来双歧杆菌已经受到了更多的科学研究,包括对多个菌株的全基因组测序(参见Liu等人,2005)。这些基因组序列将提供研究这些微生物与其人类宿主相互作用并发挥其益生菌功能的分子机制的基因学平台。
发明内容
根据本发明,提供分离和纯化的长双歧杆菌(Bifidobacterium longum)菌株,其中所述菌株:
a)表达含有6-脱氧塔罗糖(6-deoxy tallose)的胞外多糖;并且
b)包含核酸序列SEQ ID NO:86或与其具有至少95%的序列同源性的核酸序列,其编码蛋白质-酪氨酸-磷酸酶。
排除以下菌株:
35624(NCIMB 41003)、
BL1207(PTA-9608)、
AH121A(NCIMB 41675)、
AH1714(NCIMB 41676)、和
AH1206(NCIMB 41382)。

c)在至少两种来源于选自包括以下项的组的核酸序列的引物存在下扩增所提取的核酸:SEQ ID No:62至SEQ ID No:86或与其具有至少95%的序列同源性的核酸序列;
d)鉴定表达胞外多糖的细菌菌株。
在一种情况下,所述引物包含来自选自包括SEQ ID NO:62至SEQ ID NO:86的组的核酸序列的至少10个连续碱基。
所述引物可以包含选自包括以下项的组的核酸序列:SEQ ID NO:12至SEQ ID NO:61或与其具有至少95%的序列同源性的核酸序列。

根据本发明的实施方案的长双歧杆菌菌株可表达或生产EPS,其产量为约1mg/L至约1000mg/L的细菌培养物,取决于生长条件而定。

本发明基于长双歧杆菌NCIMB 41003(35624TM)的全基因组序列。基因组序列列于所附序列表的SEQ ID NO:1中,并包含2,264056个碱基对。基因组序列分析鉴定了具有如下表1所示的开放阅读框的1,734个基因。

实施例
下列实施例进一步描述和证明了本发明范围内的实施方案。这些实施例仅为了例证目的而给出并且不可被理解为是对本发明的限制,因为在不脱离本发明的实质和范围的情况下可以有许多变型。
实施例1:长双歧杆菌35624的分离
筛选在重建手术期间获取的人G.I.T.的大肠和小肠的部分和阑尾以得到益生菌细菌菌株。将所有样品在手术后立即储存在-80℃的无菌容器中。将冷冻组织解冻、称重并放置在含半胱氨酸(0.05%)的四分之一强度林格液(Ringers’solution)中。轻柔振荡每个样品以移除松散附着的微生物。在转移到第二体积的林格液中后,将样品涡旋振荡7分钟以移除牢固附着的细菌。为了分离组织内部的细菌,也将样品在Braun共混机中进行均质化。将溶液连续稀释并铺开涂布(100μl)到以下琼脂培养基上:RCM(增强的梭菌培养基)和RCM,使用乙酸将其调整至pH 5.5;TPY(胰酶解酪蛋白、蛋白胨和酵母提取物),Chevalier,P.等人(1990)。MRS(deMann,Rogosa和Sharpe);ROG(Rogosa乙酸盐培养基(SL));LLA(Lapiere的肝-乳糖琼脂);BHI(脑心浸液琼脂);LBS(乳杆菌选择性琼脂)和TSAYE(补充有0.6%酵母提取物的胰蛋白胨大豆琼脂)。所有琼脂培养基(除TPY琼脂外)由Oxoid Chemicals供应。在37℃下,将板在厌氧广口瓶(BBL,Oxoid)中使用CO2发生试剂盒(Anaerocult A,Merck)温育2-5天。
将革兰氏阳性、过氧化氢酶阴性的杆形或分叉/多形的细菌分离物在复合非选择培养基(TPY)上划线培养以纯化。除非另外说明,否则通常将分离物在37℃、厌氧条件下在TPY培养基中培养。将推定的双歧杆菌菌种储存在40%的甘油中,并且储存在-20℃和-80℃。
选择来自不同样品的大约一千五百种过氧化氢酶阴性的细菌分离物,并且按照它们的革兰氏反应、细胞大小和形态、在15℃和45℃的生长以及葡萄糖的发酵终产物进行表征。大于百分之六十的分离物经测试为革兰氏阳性的,同型发酵的球菌,它们四个一组、链状或串状排列。百分之十八的分离物是革兰氏阴性的杆菌和异型发酵的球杆菌。
剩余的分离物(百分之二十二)主要是同型发酵的球杆菌。对三十八个菌株进行较详细的表征。所有三十八个分离物经测试为硝酸盐还原阴性和从色氨酸中生产吲哚阴性。
选择这组菌株中的长双歧杆菌35624进行全基因组测序,这是由于其在结肠炎小鼠模型中已被证明的抗炎活性(McCarthy等人,2004)以及它在被肠易激综合征(IBS)患者口服后的免疫调节效应(O’Mahony等人,2005)。
实施例2-长双歧杆菌35624的基因组测序
如前所述分离来自双歧杆菌的染色体DNA(O'Riordan和Fitzgerald,1998)。使用商业测序服务提供商Agencourt Bioscience(Beverly,MA)和Eurofins MWG Operon(德国)使用Roche 454 FLX Titanium仪器对35624的基因组序列进行测序,然后组装,然后使用PCR产物的Sanger测序闭合剩余的间隙。序列读取最初使用以下进行组装:Phred(Ewing和Green,1998;Ewing等人,1998)、Phrap(P.Green,华盛顿大学;http://www.phrap.org/)、RepeatMasker(AFA.Smit、R.Hubley和P.Green;www.repeatmasker.org/)和Staden软件包(Staden等人,2000)。使用Newbler v 2.3(http://454.com/products/analysis-software/index.asp)验证35624的最终组装。35624基因组序列的登录号是CP013673。
实施例3-分析长双歧杆菌35624的基因组
使用PRODIGAL预测软件(http://prodigal.ornl.gov/)进行推定的开放阅读框(ORF)的预测,并由BLASTX[58]比对支持。将Prodigal/BLASTX的结果手动组合,并且基于BLASTP分析对由National Center for Biotechnology(http://www.ncbi.nlm.nih.gov/)提供的非冗余蛋白质数据库进行ORF的初步鉴定)。使用ORF发现输出和相关的BLASTP结果,Artemis(Rutherford等人,2000)被用于可视化和手动编辑,以便验证并在必要时重新定义每个预测的编码区域的开始,或去除或增加编码区域。手动进行将蛋白质功能分配到预测的编码区域。另外,修改后的基因/蛋白质数据集的个别成员是针对蛋白质家族(Pfam)(Punta等人,2012)和COG(Tatusov等人,2000)COG数据库进行搜索。分别使用RNAMMER(http://www.cbs.dtu.dk/services/RNAmmer/)和tRNA-scanSE(http://lowelab.ucsc.edu/tRNAscan-SE/)检测核糖体RNA(rRNA)和转运RNA(tRNA)基因。
实施例4-鉴定长双歧杆菌35624的基因组中的独特基因
我们鉴定了来自SEQ ID NO:1的碱基编号448412至474640(包括端值)的区域,我们将该区域命名为胞外多糖(EPS)区域1(SEQ ID NO:2)。EPS区域1编码基因簇B624_0342至B624_0366(表2)。
可逆的蛋白质磷酸化是调节所有活生物体中基本信号传导事件的主要机制。酪氨酸磷酸化是当今公认的细菌生理学的关键调控装置,与胞外多糖产生、毒力、应激反应和DNA代谢有关(Grangeasse等人,2012)。酪氨酸磷酸酶与酪氨酸激酶一起负责控制多糖的生物合成和输出,但其潜在机制尚不清楚(Grangeasse等人,2012)。

实施例5–从粪便样品中分离并筛选产生EPS的双歧杆菌菌株
粪便样品制备
受试者使用Kendall精密马桶样本收集系统收集粪便样品。在处理样品前将收集的样品冷藏在冰袋中。仅仅使用制备时间不超过二十四小时的样品进行评估。
将10.0g混合粪便样品置于包含90ml生理盐水的塑料胃袋中。将悬浮液均质化2分钟。使悬浮液滤过包含在一次性漏斗中的网垫。在过滤后,将45ml过滤后的粪便匀浆转移到50ml的一次性离心管中。该粪便悬浮液进一步用于DNA提取或细菌分离。
使用三次TaqMan实时PCR测定筛选粪便样品。
粪便样品DNA制备。用微型离心机将粪便匀浆的2.0ml等分试样离心沉淀。将沉淀物重悬在20mg/ml的溶菌酶中,在37℃放置2小时。用QIAamp DNA Stool Mini Kit(Qiagen)提取DNA。通过Pico Green测定法(Molecular Probes)测量DNA浓度。一旦测量出DNA浓度,将DNA储存在4℃。
TaqMan实时PCR反应。将试验样品稀释到DNA浓度为2ng/ul使得5μl样品包含总计10ng DNA。通过总计三个单独的测定来测定样品。
制备以下反应混合物:
制备一定量的待测定样品的批量混合物。将45μl等分试样分配到96孔微滴定板的每个孔中,然后每个孔加入5μl DNA。短时间地旋转所述微滴定板,将其置于热循环仪(ABI7900 HT)中。使用标准TaqMan热循环规程。
TaqMan RT-PCR程序。标准TaqMan定量PCR热循环规程如下所示:
步骤1:95℃持续10分钟(以活化AmpliTaq Gold聚合酶)
步骤2:95℃持续15秒(变性步骤)
步骤3:60℃持续60秒(引发/聚合步骤)
重复步骤2和步骤340次。在步骤3收集荧光数据。
三次TaqMan RT-PCR测定的引物和探针。使用EPS基因特异性测定法和两个长双歧杆菌35624基因特异性测定法(UNK1和UNK2)筛选粪便样品DNA。使用的特异性基因和它们的TaqMan引物组如下表3所示。

通过长双歧杆菌35624EPS基因特异性测定法显示高DNA浓度,但是使用长双歧杆菌35624基因特异性测定法为阴性反应的粪便样品进一步用于分离潜在的产生EPS的细菌。
实施例6-来自粪便样品的菌株长双歧杆菌AH0097和菌株长双歧杆菌AH1172的分 离和表征。
将1毫升细菌悬浮液(参见上文实施例5)转移到9.0ml的无菌磷酸盐缓冲盐水中,这形成10-1稀释液。将1毫升该10-1稀释液转移到9.0ml的无菌磷酸盐缓冲盐水中,这是10-2稀释液。持续该过程直至制备出10-10稀释液。然后将0.1ml的每个稀释液置于强化梭状芽胞杆菌琼脂(Reinforced Clostridial Agar,RCA)平板(BD或等同物)和乳杆菌Man-RogosaSharpe琼脂(MRSA)平板(BD或等同物)的表面上。将平板在厌氧条件下(COY厌氧室),在33℃±2℃下温育48-72小时。
在温度之后,从RCA和MRSA平板上采集单个菌落(总计大约100个菌落)并进一步划线接种到新平板上用于分离物纯化。将带有划线接种菌落的平板在厌氧条件下(COY厌氧室),在33℃±2℃温育48至72小时。在温育之后,观察平板上的纯菌落,然后将其用于DNA提取。
使用三次TaqMan实时PCR测定筛选粪便分离物。

使用ABI Full Gene PCR试剂盒(Applied Biosystems,Foster City,CA)扩增并测序16S rRNA基因片段。

PCR扩增在GeneAmp PCR System 9700热循环仪上进行,使用以下程序:
初始保持:95℃持续10分钟
30个循环:95℃保持30秒(变性)
60℃保持30秒(退火)
72℃保持45秒(延伸)
最终延伸:72℃保持10分钟
(2).16S rRNA基因测序
进一步在热循环仪上进行测序,使用以下程序:
25个循环:96℃持续10秒(变性)
50℃持续5秒(退火)
60℃持续4分钟(延伸)
最终步骤 保持在4℃
使用DyeEXTM2.0 spin试剂盒进一步纯化测序的PCR产物,并且使用3130 xl遗传分析仪(Applied Biosystems,Foster City,CA)测序。
(3)序列数据分析:
通过使用Basic Local Alignment Search Tool(BLAST)比较共有序列与GenBank序列。GenBank搜索表明,长双歧杆菌35624 EPS基因特异性阳性,但长双歧杆菌35624基因特异性阴性的分离物均为长双歧杆菌。这些菌株是由Alimentary Health命名,AH0097和AH1172。
实施例7-分离并筛选表达EPS的长双歧杆菌菌株
分离细菌菌株
使用如上文实施例1所述的方法从肠组织和/或粪便样品中分离细菌。具体而言,从健康成年人受试者的粪便样品中分离长双歧杆菌AH0097和长双歧杆菌菌株AH1172。
EPS基因簇筛选
使用下表4中列出的引物,筛选细菌菌株是否存在的EPS簇1(上表2)中的基因。简而言之,使用以下方法筛选PCR EPS基因簇:
用细菌菌株的新生菌落无菌接种10ml改良Rogosa肉汤培养基(+0.05%半胱氨酸),并且在37℃厌氧培养至浑浊(约16小时至约24小时)。将肉汤培养物离心并使用SigmaTM提取过程从所得沉淀中分离DNA。使用nanodrop确定样品中的DNA浓度,并且使用DEPC水将样品稀释到最终浓度为每个样品50ng/μl DNA。在单个PCR反应中使用模板DNA样品,在以下条件下使用下表4中列出的引物组:

引物经过特别设计,以扩增EPS簇1的25个基因的大约500个碱基对的PCR产物。在用适当的DNA梯子(DNA ladder)参照大小进行琼脂糖凝胶电泳后,可视化PCR产物。下表5指示存在(是)或不存在(否)500bp的PCR产物。

预测的eps624基因对eps624结构的作用。
eps624簇通过所谓的Wzx/Wzy依赖型路径(其通常使用引发性糖基转移酶(pGT)、一种或多种糖基转移酶(GH)、翻转酶和聚合酶以产生细胞外杂多糖)编码许多预测为EPS产生所需的关键酶(Schmid等人,2015)。预测eps624基因簇的第一个基因(相应于基因座标签BL_0342,在此称为pgt624)编码pGT,其将第一个单糖添加到作为低聚糖亚基生物合成的一部分的细胞质膜结合载体分子十一异戊烯(Salazar等人,2009)。eps624簇编码5个另外的GT(对应于基因座标签BL_0345、BL_0346、BL_0349和BL_0352;),预测它们每个向载体分子添加一个单糖以便完成低聚糖亚基,然后通过翻转酶(预测由对应于基因座标签BL_0355的基因编码)向膜的外侧输出,并且随后由聚合酶(推定由基因座标签BL_0353指定)使用来产生EPS聚合物。有趣的是,eps624簇的对应于B624_0347和118B624_0348的两个相邻基因预测为编码UDP-葡萄糖醛酸5'-差向异构酶和UDP-葡萄糖6-脱氢酶,这表明EPS引入的一个单糖是葡萄糖醛酸的差向异构体,例如,半乳糖醛酸或甘露糖醛酸。位于eps624簇内的三个基因(对应于基因座标签B624_0360至B624_0362)编码已知参与dTDP-L-鼠李糖生物合成的酶(Marumo等人,1992),而B624_0350和B624_0357的推导的蛋白质产物预测为编码NAD依赖型还原酶/差向异构酶。已经示出这些酶参与了向dTDP-D-岩藻糖或dTDP-6-脱氧-L-塔罗糖产生的dTDP-L-鼠李糖生物合成路径的重选路由(Gaugler和Gabriel,1973;Nakano等人,2000;Yoshida等人1999)。eps624簇也编码两种预测的乙酰转移酶,类似于先前已经显示在多糖中对特定糖组分(例如6-脱氧-L-塔罗糖)进行O-乙酰化反应的酶(Knirel等人,2002)。此外,具有基因座标签B624_0344和B624_0366的基因分别代表与控制EPS输出和聚合有关的推定的酪氨酸激酶和磷酸酪氨酸蛋白磷酸酶活性(Grangeasse等人,2012)。因此,根据eps624簇的基因内容物,我们预测35624产生的EPS由6个单糖组成的重复亚基组成,其中一个是葡萄糖醛酸差向异构体,另外的一个或两个是D-岩藻糖或6-脱氧-塔罗糖,且其中一些可以是经O-乙酰化的。
位于eps624簇内的基因(对应于基因座标签B624_0342至B624_0366)是产生长双歧杆菌35624亚种longum特异性EPS所需的。由细菌产生EPS最低限度需要引发性糖基转移酶(pGT)、一种或多种糖基转移酶(GH)、翻转酶和聚合酶以产生细胞外杂多糖。糖基转移酶的数目可以变化,从而影响低聚糖亚基的长度。此外,乙酰转移酶的存在可能导致一些单糖的O-乙酰化。
蛋白质-酪氨酸-磷酸酶
可逆的蛋白质磷酸化是调节所有活生物体中基本信号传导事件的主要机制。酪氨酸磷酸化是当今公认的细菌生理学的关键调控装置,与胞外多糖产生、毒力、应激反应和DNA代谢有关(Grangeasse等人,2012)。酪氨酸磷酸酶与酪氨酸激酶一起负责控制多糖的生物合成和输出,但其潜在机制尚不清楚(Grangeasse等人,2012)。在长双歧杆菌中,亚种longum35624基因组B624_0344编码酪氨酸激酶,而B624_0366编码酪氨酸磷酸酶。B624_0366先前未被识别为作为WO2010/055499A公布的专利申请PCT/IE2009/000079中的eps624簇的一部分。
刚果红琼脂筛选
使用刚果红琼脂筛选用于EPS表达细菌菌株的表型筛选。简而言之,用细菌菌株的新生菌落无菌接种到10ml改良Rogosa肉汤培养基(+0.05%半胱氨酸),并且在37℃厌氧培养直至浑浊为止(约16小时至约24小时)。将肉汤培养物无菌划线接种到刚果红琼脂平板上,并且在37℃厌氧温育48小时。据信EPS作为某些菌株生长和/或代谢的副产物而产生,EPS阻止刚果红染料的摄取,导致奶油状/白色的菌落形态。产生较少EPS的菌株容易吸收刚果红染料,产生粉红色/红色的菌落形态。不生产EPS的菌株染上红色,在红色琼脂背景中看上去几乎是透明的。

在多个水平发挥炎性疾病的控制。控制因子包括激素、前列腺素、反应氧和氮中间体、白三烯和细胞因子。细胞因子是低分子量生物活性蛋白,它们涉及免疫和炎性响应的产生和控制。许多类型的细胞产生这些细胞因子,中性白细胞、单核细胞和淋巴细胞是炎性反应中的主要来源,这是因为它们在受伤位置大量存在。
在炎性位置生成的细胞因子存在多个机制影响炎症应答。趋化性刺激炎性细胞聚集到受伤位置,而某些细胞因子促进细胞渗透进入组织。在受伤组织中释放的细胞因子导致炎性渗透物的活化。大多数细胞因子是多效性的,并且表达多个生物重叠活性。因为不受控制的炎症应答能够导致疾病诸如IBD,有合理理由认为在个体中错误的细胞因子生产影响这些疾病。
长双歧杆菌35624、长双歧杆菌AH0097和长双歧杆菌AH1172诱导PBMC中几乎相同 的细胞因子谱。
使用BD Vacutainer CPT管(BD目录号362761),按照制造商的说明书,从新鲜人外周血中分离外周血单个核细胞(PBMC)。洗涤PBMC并将其重悬在Dulbecco's MEM(Gibco目录号10569-010)加25mM HEPES、10%胎牛血清(Sigma目录号F4135)、和1%青霉素/链霉素(Sigma目录号P0781)。将2×105个PBMC(在200μl DMEM中)铺板于96孔培养板的每个孔中。
细菌在Difco MRS培养基中生长,并且在进入稳定期后收获。所有细胞在37℃厌氧条件下生长。构造每个生长条件下的生长曲线(OD对#活细胞),并且通过细胞数目在添加到PBMC前将洗涤细胞归一化。
将细菌(20μl,在磷酸盐缓冲盐水(PBS)中)添加到PBMC的每个孔中以提供每个实验指示的细菌总数。测试五个不同量的细菌:2E07、1E07、5E6、2E6和1E6,将它们添加到PBMC的单独的孔中。也进行无细菌的对照实验。所有测定重复三次。在37℃温育2天后,将板以300xg旋转,并且去除上清液并冷藏在-80℃直至分析。
使用来自Meso Scale Discovery(Gaithersburg,MD;目录号K15008B-1)的96孔检测试剂盒测定培养上清液中的细胞因子。测定人白介素10(Il-10)和肿瘤坏死因子α(TNFa)的量并报告皮克每毫升。每个样品重复测定两次。
图3至图6示出代表性实验的结果。对于所示的每种细胞因子,长双歧杆菌35624、长双歧杆菌AH0097和长双歧杆菌AH1172诱导几乎相同水平的细胞因子。这些水平与由与之进行比较的其它双歧杆菌菌株诱导的水平差异很大。

白介素-10(IL-10)是一种抗炎细胞因子,它由多种类型的细胞生产,包括单核细胞、巨噬细胞、树突状细胞、巨细胞和淋巴细胞(尤其是T调节细胞)。IL-10下调抗原递呈细胞上的促炎Th1细胞因子、MHC II类抗原、和共刺激分子的表达。它也促进B细胞的存活、增殖、和抗体生产。该细胞因子能够阻碍NF-κB活性,并且涉及JAK-STAT信号传导途径的调控。小鼠基因敲除研究已经证明IL-10在免疫调节中的主要作用是作为IL-10KO小鼠发育严重结肠炎。此外,细菌是IL-10的强诱导剂,这已经证明促进T调节细胞的体内分化,因此有助于免疫自稳(O’Mahony等人,AJP 2006;O’Mahony等人,PLoS Pathogens 2008)。
肿瘤坏死因子α(TNFα)是一个关键的促炎症细胞因子,原因在于它激活细胞因子的级联反应以及导致炎症状态的生物学效应。因此,抑制TNFα的药剂例如英夫利昔(infliximab)现在通常被用来治疗炎性疾病。
用于配制产生EPS菌株的组合物和方法
组合物
下表显示了引入表达EPS的冷冻干燥细菌的组合物。
为确保组合物具有足够的储存寿命,可以使用以下干燥处理过程。混合和填充操作可以在RH可以保持在50%以下,或者40%以下的湿度受控的环境中进行。可以将冷冻干燥的细菌预浓缩至所需的CFU/g,以基于待投配的混合粉末的总量和冷冻干燥的细菌级分在每个剂量单位中达到所需的总量。冷冻干燥的细菌可以与适量的稳定赋形剂诸如微晶纤维素、马铃薯淀粉等一起加入到例如Pharmatech混合器的混合腔中。稳定赋形剂可以被预干燥以具有低水含量,例如小于10%,或者小于6%的水含量。混合腔内的顶部空间可以用诸如氮气的干燥气体冲洗,使得相对湿度保持在低水平。然后可以以60rpm的混合速度将粉末混合20分钟以确保良好的混合。一旦混合完成,粉末可以在低Rh环境中处理和储存,例如低于50%RH或低于40%RH。
然后可以在干燥环境下,将为达到每个剂量单位所需CFU的适当量的所得干共混粉末填充到适当大小的胶囊中,例如明胶或羟丙基甲基纤维素(HPMC)胶囊。另选地,可在干燥环境下,将适当量的所得粉末填充到小袋中并密封。所得胶囊或小袋可以在室温(20-30℃)下储存,或者在降低的温度(4℃)下储存以延长储存寿命。
另选地,可以使用标准技术将冷冻干燥的细菌掺入乳制品诸如酸奶中或掺入咀嚼剂型中。
使用方法
本文描述的组合物旨在用作预防性、治疗性或非治疗性治疗以减轻影响动物,优选哺乳动物和人类的疾病和病症。所述组合物可作为药物、OTC或补充剂,例如DSHEA产品施用给受试者。预防性、治疗性或非治疗性治疗的非限制性示例可以用于炎性疾病或病症或调节免疫系统。疾病和病症可以包括炎性肠病、肠易激综合征、由于一系列原因诸如旅行或抗生素治疗、变态反应、哮喘、发烧控制、营养失调、维持或改善胃肠道或皮肤状况引起的腹泻、和呼吸障碍或感染。特别地,这些组合物可以用于治疗或预防胃肠道的不利状况。
典型地,组合物作为剂量方案的一部分提供给受试者。剂量方案可以根据正在治疗或预防的特定状态和单位剂量形式而变化。例如,每剂量施用的细菌总量可以在1×106至1×1012CFU/剂量,优选1×108至1×1010CFU/剂量的范围内。当作为胶囊提供时,单位剂量可以直接用流体或不用流体吞服。包装在小袋中的粉末可以直接摄取,或与液体如奶或果汁混合或与食物诸如酸奶混合。如果已经掺入食物补充剂诸如酸奶中,或者已经掺入咀嚼型食物中,则可以通过食用来服用该剂量。剂量方案可以规定每月至少一次、每周至少一次、每天至少一次或每天一次以上施用。
