Abstract

本发明涉及到一种载有家蚕抗菌肽的可促进创伤愈合的复合纤维膜。该复合纤维膜包括由聚合物制成的支架和具有抗菌的家蚕抗菌肽BmceropinB1。该复合纤维膜通过释放支架上载有的家蚕抗菌肽BmceropinB1,与细菌的细胞膜结合,改变细胞膜表面电势,破坏细菌细胞膜,影响细菌正常的生物学功能,抑制并杀死细菌,而对于正常细胞不产生伤害作用。该复合纤维膜可应用于医药和临床医学,在防止伤口愈合时的感染及促进伤口愈合的过程中发挥重要作用。

技术领域
本发明涉及一种可促进创伤愈合的纤维膜的制备方法,特别涉及用家蚕抗菌肽制备可促进创伤愈合的纤维膜的方法。
背景技术
由细菌引起的普通伤口感染和手术后伤口感染是医学上常见的问题。频繁的使用抗生素来解决伤口感染的问题已经造成细菌耐药性的不断增强。
为了研究出具有促进创伤愈合的材料,一些具有抗菌作用的纳米颗粒,例如银(Ag)、氧化锌(ZnO),通过各种方法载入到天然或合成聚合物材料中,结果表明,合成的材料具有明显的抗菌作用,但是,这些材料也不可避免地会对正常细胞产生一定的毒性。
抗菌肽是自然界中存在的一类小分子多肽。抗菌肽普遍带有正电荷,具有广谱抗菌性,热稳定性好,对正常细胞无毒,不诱导细菌产生耐药性等特点。现今为止,抗菌肽被认为是抗生素的最有可能代替者。
将家蚕抗菌肽BmcecropinB1与PLLA在溶液中混合,再通过静电纺丝制备成纤维膜,应用于医药和临床医学,在预防细菌引起的伤口感染,促进伤口愈合过程中发挥有效的作用。
发明内容
本发明所要解决的技术问题是,针对以上现有抗菌剂存在的缺点,提出用家蚕抗菌肽BmcecropinB1制备可促进创伤愈合的纤维膜的方法。本发明步骤简单,操作方便,无毒无害,抗菌效果好,能防止伤口感染,有效促进伤口愈合,具有很大的发展潜力。
为了实现上述目的,本发明采取的解决方案为:一种用家蚕抗菌肽BmceropinB1制备可促进创伤愈合的纤维膜的方法,其特征在于,包括如下步骤:
(1)家蚕抗菌肽BmceropinB1溶液的制备:将家蚕抗菌肽BmceropinB1加入缓冲溶液中,制备家蚕抗菌肽BmceropinB1溶液,然后在其中加入聚左旋聚乳酸(PLLA),搅拌均匀后得到含有PLLA和家蚕抗菌肽BmceropinB1的分散液;
(2)将步骤(1)所得到的分散液在高压静电纺丝仪器上进行纺丝;
(3)将静电纺丝得到的纳米纤维膜烘干。
本发明的有益效果在于,(1)静电纺丝制备的纤维膜中家蚕抗菌肽BmceropinB1与PLLA混合均匀,BmceropinB1可从纤维膜中缓释,达到持续抗菌效果;(2)静电纺丝制备的纤维膜比表面积高,孔隙率高,在作用于伤口过程中不影响伤口的透气;(3)家蚕抗菌肽BmceropinB1不会诱导细胞产生耐药性,对正常细胞没有毒害作用;(4)本发明操作简单易行、周期短、高效、操作设备成本低。
具体实施方式
以下通过特定的具体实例说明本发明的实施方式,本领域技术人员可由本说明书所揭露的内容轻易地了解本发明的其他优点与功效。本发明还可以通过另外不同的具体实施方式加以实施或应用,本说明书中的各项细节也可以基于不同观点与应用,在没有背离本发明的精神下进行各种修饰或改变。
实施例1
(1)家蚕抗菌肽BmceropinB1溶液的制备:将家蚕抗菌肽BmceropinB1加入pH6.5的缓冲溶液中,制备成0.01%(W/W)的家蚕抗菌肽BmceropinB1溶液,然后在其中加入1%(W/W)聚左旋聚乳酸(PLLA),搅拌均匀后得到含有PLLA和家蚕抗菌肽BmceropinB1的分散液;
(2)将步骤(1)所得到的分散液在高压静电纺丝仪器上进行纺丝,针头到转轴的距离为10厘米,电压15KV,溶液流速0.33mL/min;
(3)将静电纺丝得到的纳米纤维膜烘干。
实施例2
(1)家蚕抗菌肽BmceropinB1溶液的制备:将家蚕抗菌肽BmceropinB1加入pH7.0的缓冲溶液中,制备成0.1%(W/W)的家蚕抗菌肽BmceropinB1溶液,然后在其中加入1%(W/W)聚左旋聚乳酸(PLLA),搅拌均匀后得到含有PLLA和家蚕抗菌肽BmceropinB1的分散液;
(2)将步骤(1)所得到的分散液在高压静电纺丝仪器上进行纺丝,针头到转轴的距离为12厘米,电压18KV,溶液流速0.43mL/min;
(3)将静电纺丝得到的纳米纤维膜烘干。
实施例3
(1)家蚕抗菌肽BmceropinB1溶液的制备:将家蚕抗菌肽BmceropinB1加入pH7.0的缓冲溶液中,制备成0.2%(W/W)的家蚕抗菌肽BmceropinB1溶液,然后在其中加入1%(W/W)聚左旋聚乳酸(PLLA),搅拌均匀后得到含有PLLA和家蚕抗菌肽BmceropinB1的分散液;
(2)将步骤(1)所得到的分散液在高压静电纺丝仪器上进行纺丝,针头到转轴的距离为15厘米,电压21KV,溶液流速0.50mL/min;
(3)将静电纺丝得到的纳米纤维膜烘干。

1.一种用家蚕抗菌肽BmceropinB1制备可促进创伤愈合的纤维膜的方法,其特征在于,包括如下步骤:
(1)家蚕抗菌肽BmceropinB1溶液的制备:将家蚕抗菌肽BmceropinB1加入缓冲溶液中,制备家蚕抗菌肽BmceropinB1溶液,然后在其中加入聚左旋聚乳酸(PLLA),搅拌均匀后得到含有PLLA和家蚕抗菌肽BmceropinB1的分散液;
(2)将步骤(1)所得到的分散液在高压静电纺丝仪器上进行纺丝;
(3)将静电纺丝得到的纳米纤维膜烘干。
2.根据权利要求1所述的一种家蚕抗菌肽BmceropinB1,其特征在于所述家蚕抗菌肽BmceropinB1氨基酸序列为:Ala-Pro-Glu-Pro-Arg-Trp-Lys-Ile-Phe-Lys-Lys-Ile-Glu-Lys-Met-Gly-Arg-Asn-Ile-Arg-Asp-Gly-Ile-Val-Lys-Ala-Gly-Pro-Ala-Ile-Glu-Val-Leu-Gly-Ser-Ala-Lys-Ala-Ile- Gly-Lys。
