实施例3葡萄糖氧化酶耐热两点组合突变体的获得
采用实施例1中所述筛选并获得突变体的方法,对葡萄糖氧化酶GOD的第64位和第415位耐热位点进行突变组合筛选。进一步,对获得的突变体分别进行热稳定性实验。
结果显示:耐热性比上述单点突变体更好的两点突变组合包括:A64R+A415K,或A64R+A415R,或A64R+A415N,或A64L+A415K,或A64L+A415R,或A64L+A415N,或A64K+A415K,或A64K+A415R,或A64K+A415N,或A64M+A415K,或A64M+A415R,或A64M+A415N,或A64F+A415K,或A64F+A415R,或A64F+A415N,或A64N+A415K,或A64N+A415R,或A64N+A415N,或A64Q+A415K,或A64Q+A415R,或A64Q+A415N。
所述两点突变体的葡萄糖氧化酶酶活残留率比上述单点突变体普遍提高了30%~235%。而其中耐热性最强的两点突变组合包括:A64R+A415K,或A64R+A415R,或A64L+A415K,或A64L+A415R。将所述四个两点突变体分别命名为GOD11,GOD12,GOD13,GOD14,其编码核苷酸序列分别为SEQ ID NO:13,SEQ ID NO:14,SEQ ID NO:15,SEQ ID NO:16。
所述葡萄糖氧化酶突变体具体的耐热性能见表2。

综上所述,与野生型相比,本发明筛选获得的葡萄糖氧化酶突变体具有更强的耐热性,更适合用作饲料添加剂,有利于葡萄糖氧化酶在饲料中的广泛应用。
