Background
Grape white rot, grape anthracnose, apple ring rot and apple anthracnose are one of the important diseases in the main production area of fruit trees. At present, chemical bactericides are still the main means for preventing and treating diseases of fruit trees, but the problems of increased prevention and treatment cost, overproof fruit residues, environmental pollution, development of drug resistance and the like are caused by unreasonable large amount of chemical pesticides for a long time. The close attention of the public on food safety and the increasing call for global environmental protection require researchers to find effective and safe control methods, so that development and utilization of pesticide resources which are compatible with the environment and harmless to humans are increasingly favored by various countries.
The biogenic pesticide is also called biological pesticide, and refers to pesticide developed by utilizing biological resources, and the active ingredients of the biogenic pesticide are naturally existing, and have the characteristics of low toxicity, no pollution, strong specificity, easy degradation, small possibility of accumulating toxicity in ecological environment, environmental compatibility and the like. The pesticide has the advantages of ensuring the organic food and the green food with continuously increasing demands, avoiding or lightening the negative effect caused by applying chemical pesticides, ensuring that pathogenic bacteria are difficult to generate drug resistance and the like, and being an ideal pesticide type.
The zhongshengmycin is an antibiotic produced by Hainan variety of Streptomyces lavendulae, and belongs to N-glycoside alkaline water-soluble substances. The action mechanism is mainly that the bacteria are inhibited from synthesizing mycoprotein, so that the thallus is dead; for fungi, the strain deforms filamentous hyphae, inhibits spore germination and can directly kill spores. The zhongshengmycin has contact killing and penetrating functions, high activity on bacterial diseases and partial fungal diseases of crops and certain yield increasing effect. The development of resistance to crop pathogens has not been retrieved.
The botanical pesticide eugenol exists in clove oil, cinnamon leaf oil, cinnamon bark oil, camphor oil, nutmeg oil and the like, and can be separated from the clove oil. The eugenol is absorbed by leaves, stems and roots of plants, has an upward conduction function, is safe, environment-friendly, free of residues, rapid in drug effect and long in lasting period. After the crops with diseases are sprayed, the bacterial spores are immediately deformed and dissolved and disappear. In recent years, eugenol is concerned by clinicians, scientific research departments and patients, and is also used as a pesticide for popularization and application by scientific research institutions and manufacturers. The compound of eugenol and zhongshengmycin for preventing and treating apple ring rot, apple anthracnose, grape white rot and grape anthracnose is not reported yet.
Disclosure of Invention
The invention aims to provide a Zhongshengmycin and eugenol single agent and a compound composition thereof for preventing and treating important fruit tree diseases such as apple ring spot, grape white rot and the like. The invention discusses the application of the compound composition of zhongshengmycin and eugenol in the agricultural field, and is particularly completed by research on prevention and treatment of important diseases such as apple ring rot, grape white rot and the like.
The invention is realized by the following technical scheme:
the compound composition of the zhongshengmycin and the eugenol is characterized in that the weight ratio of the zhongshengmycin to the eugenol as effective components is 70: 1-1: 70;
the further improvement of the compound composition of the zhongshengmycin and the eugenol is that the synergistic weight ratio of the zhongshengmycin to the eugenol as effective components is 40: 1-1: 10;
the further improvement of the compound composition of the zhongshengmycin and the eugenol is that the optimal proportion of the zhongshengmycin and the eugenol in the effective components is 10:1, and the synergistic coefficient is 2.18;
the total weight of the zhongshengmycin and the eugenol in the effective components of the compound composition accounts for 2.2 percent of the compound composition, and the balance is conventional auxiliary material additives.
The compound composition of the zhongshengmycin and the eugenol is applied to preventing and treating apple ring spot, apple anthracnose, grape white rot and grape anthracnose.
The invention also provides a use method of the compound composition of the zhongshengmycin and the eugenol, which is characterized in that the compound composition of the zhongshengmycin and the eugenol is diluted by 600-fold and 800-fold and has better control effect on apple ring spot, apple anthracnose, grape white rot and grape anthracnose.
The composition of the invention has the following advantages:
1. has synergistic effect. Indoor toxicity test results show that the zhongshengmycin and the eugenol have synergistic effect on the botrytis cinerea within the range of 40: 1-1: 10, wherein the 10:1 synergistic effect is most obvious, and the synergistic coefficient reaches 2.18.
2. The optimal proportion of the pesticide has prominent field tank-mixing control effect and reduces the dosage of the preparation. The 2.2% zhongshengmycin and eugenol compound composition prepared by the institute has the advantages that the 600-fold and 800-fold effect is remarkable in the pesticide effect test for preventing and treating apple ring spot, apple anthracnose, grape white rot and grape anthracnose in the field, and the prevention and treatment effect is superior to or equivalent to that of a single agent and a control agent. Meanwhile, the dosage of the preparation is greatly reduced, and compared with the direct barrel mixing according to the label concentration, the dosage of the compound composition is reduced by 33 percent and 50 percent respectively by 600 times and 800 times.
3. High safety. The eugenol and the zhongshengmycin have low toxicity to human and livestock, and the content of the prepared compound composition in our hospital is lower than that of the effective component of a single agent in the market, so that the compound composition is safer to crops, environment and fresh fruit eaters.
4. The risk of drug resistance of pathogenic bacteria is reduced. As shown in the advantage 2, the dosage of the biotins and the eugenol in the compound composition is obviously lower than that of a single-agent product sold in the market, and the dosage of the preparation is obviously reduced when the compound composition is used in the field, so that the selection pressure of pathogenic bacteria such as white rot of grape and the like on the two bactericides can be greatly reduced. In addition, the zhongshengmycin and the eugenol have different action mechanisms, and the pathogenic bacteria are not easy to generate drug resistance when the pesticide is used in a tank.

Example 1
Indoor toxicity test and compounding ratio screening
1.1 test agent
A12% zhongshengmycin wettable powder, a product of Fujian Kaili biological products Co., Ltd, is dissolved in water to prepare a mother solution of 1000mg/L for later use.
0.3 percent of eugenol soluble solution, a product of Yijia agricultural chemical Limited company in Shandong province, is dissolved in water to prepare 1000mg/L mother solution for standby.
1.2 test strains
Grape white rot (Coniothyrium diplodiella) is obtained by separating field diseased fruits, and the first generation strain obtained by single spore culture and purification is stored in a refrigerator at 4 ℃ for later use.
1.3 instrumentation
The instrument mainly comprises an electronic balance, an ultra-clean workbench, a biological incubator, a culture dish, a pipettor and the like. 1.4 Experimental methods
The FAO-recommended cell growth rate method (Mycelium growth rate test) was used.

And (3) punching a bacterium disc by using a puncher with the diameter of 0.6 cm along the edge of the cultured bacterial colony, reversely transplanting the bacterium disc onto a flat plate by using a disinfection inoculation needle, and placing the flat plate at the constant temperature of 25 ℃ for culturing for 6 days.
1.4.4 investigation of results
And measuring the expansion diameter of the bacterial colony by a ruler cross measurement method, and calculating the inhibition rate of the medicament on the growth of hyphae compared with the expansion diameter of a control bacterial disc without applying the medicament.
1.5 statistical analysis
Regression analysis is carried out according to the logarithm value of the concentration of each medicament and the corresponding hyphal growth inhibition rate value, and a toxicity regression equation, a correlation coefficient (r) and EC of each medicament are calculated50。

The synergistic effect of the compounded medicament is evaluated according to the synergistic coefficient (SR), namely, SR <0.5 is antagonistic action, SR <0.5 and less than or equal to 1.5 are additive action, and SR >1.5 is synergistic action. 1.6 analysis of results
The results of indoor toxicity experiments show that the two single-dose indoor inhibition effects on grape white rot germs are good, and EC is503.1942mg/L and 0.7089mg/L respectively. The effective component proportion of both zhongshengmycin and eugenol is 40: 1-1: 10, the synergy is most remarkable particularly when the proportion of the effective component proportion of the zhongshengmycin to the eugenol is 10:1, and the EC of the eugenol is50The concentration is 1.1098mg/L, and the synergistic coefficient is 2.18. The Zhongshengmycin-eugenol 10:1 compound bactericide has high bactericidal activity on grape white rot.

Example 2
Plot experiment in the field
According to an indoor compounding ratio screening experiment, the Zhongshengmycin and eugenol which are 10:1 are screened out to be the optimal proportion, and 2.2 percent of Zhongshengmycin and eugenol compound composition is prepared. According to the pesticide test rule (II) of pesticide field efficacy test of Ministry of agriculture, part 13, 121 and 181, 2.2% Zhongshengmycin and eugenol compound composition is designed for field efficacy tests of apple ring spot, apple anthracnose, grape white rot and grape anthracnose.
2.1 test Agents
0.3 percent of eugenol is 600 times of soluble solution, and is a product of Yijia agricultural chemical Limited company in Shandong province;
600 times of 3% zhongshengmycin wettable powder, a product of Shandong Mgan-Fengchen Biotech limited company;
2.2 percent of zhongshengmycin and eugenol 600 times, 800 times and 1000 times, and prepared by a tobacco platform farm academy.
2.2 cell design
The test has 7 treatments, each treatment has 4 repetitions, 28 cells are arranged, and 6 adult trees are arranged in each cell. The cells are randomly arranged. The pesticide application instrument adopts a Gongnong 26 type high-pressure motor sprayer, and all the branch tip blades in the tree crown are uniformly applied with pesticide until water drops.
2.3 investigation time and method
3-4 plants are selected in the center of each cell in the harvesting period, all fruits are investigated, the total number of fruits and the number of diseased fruits are recorded, and the control effect is calculated according to the disease fruit rate.
2.4 method of calculating drug efficacy
2.5 test results
600 and 800 times of 2.2 percent of zhongshengmycin and eugenol compound bactericide has good control effect on grape white rot, and the control effect is 88.57 percent and 85.30 percent respectively. The compound bactericide 600-fold liquid is obviously superior to the control effect of two single agents of 3% Zhongshengmycin 600-fold and 0.3% eugenol 600-fold on the level of 5%, and has no obvious difference with 600-fold barrelling mixing of 3% Zhongshengmycin wettable powder and 0.3% eugenol soluble solution 600-fold. The 800-time liquid control effect of the compound bactericide is obviously lower than that of the tank mix on the level of 5%, but the control effect is also more than 85%, and the compound bactericide has no obvious difference with 600 times of 3% zhongshengmycin wettable powder, and is obviously better than 600 times of 0.3% eugenol soluble solution on the level of 5%. Fully indicates that the 4.2 percent zhongshengmycin and eugenol compound bactericide has stronger bactericidal activity on the white rot of grapes. (Table 4).

The control effect of the 2.2% zhongshengmycin and eugenol compound bactericide of the compound composition on grape anthracnose is respectively 87.62% and 85.30% in 600 times and 800 times. The 600-time control effect of the compound bactericide is obviously superior to that of a 3% Zhongshengmycin wettable powder by 600 times and that of a 0.3% eugenol soluble solution by 600 times on the level of 5%, and the compound bactericide has no obvious difference from that of the 3% Zhongshengmycin wettable powder by 600 times and that of the 0.3% eugenol soluble solution by 600 times in tank mixing. The 800-time liquid control effect of the compound bactericide is not obviously different from 600-time control effect of 3% zhongshengmycin wettable powder, and is obviously superior to 600-time control effect of 0.3% eugenol soluble solution on the 5% level (Table 5).

The field control result of the 2.2% zhongshengmycin and eugenol compound bactericide on the apple ring spot shows that: the compound bactericide has a control effect of 600 and 800 times, is remarkably superior to that of a single preparation of 600 times of 3% Zhongshengmycin wettable powder and 600 times of 0.3% eugenol soluble solution on the level of 5%, and has no remarkable difference from the barrelling of 600 times of 3% Zhongshengmycin wettable powder and 600 times of 0.3% eugenol soluble solution (Table 6).

The 2.2% zhongshengmycin and eugenol compound bactericide has 600 times and 800 times of control effects on apple anthracnose of 88.61% and 85.66% respectively, is remarkably superior to the control effects of 600 times of 3% zhongshengmycin wettable powder and 600 times of single dose of 0.3% eugenol soluble solution on the 5% level, and has no remarkable difference with 600 times of 3% zhongshengmycin wettable powder and 600 times of 0.3% eugenol soluble solution in tank mixing.

Example 3
Safety test
The content of the zhongshengmycin and the eugenol in the compound composition is less than that of a product sold on the market, so that the toxicity to human and livestock and the safety of crops meet the requirements. The toxicity test result of the compound bactericide by a affirming organization shows that the compound bactericide belongs to a slightly toxic pesticide:
rat acute oral toxicity LD50 of more than 5000mg/kg and male parent of more than 5000 mg/kg;
rat acute percutaneous toxicity LD50 of greater than 5000mg/kg and male parent of greater than 5000 mg/kg;
the integral of the rabbit eye irritation is 0(1h), and the rabbit eye irritation is diluted by 0(1:100) after 48h, so that the rabbit eye irritation is not irritant to eyes;
the integral of the rabbit eye irritation is 0(4h), and the rabbit eye irritation is not irritant to skin.
According to the invention, the optimal proportion of the two pesticides is determined for production and use through the effect of the single use and the compound of the zhongshengmycin and the eugenol in the biological source pesticide on the main diseases of grapes and apples by indoor toxicity measurement and field tests, so that the purposes of reducing the environmental pollution of the pesticides and controlling the generation and development of the drug resistance of germs are achieved.
