Technical Field
The invention relates to a compound composition for preventing and treating diseases of fruit trees such as apples, grapes and the like, belonging to the technical field of bactericides.
Background
Apple ring rot, apple anthracnose, grape white rot and grape anthracnose are one of the important diseases in the main production area of fruit trees. In the foreseeable future, the chemical bactericide is still a main means for preventing and treating the diseases of fruit trees, but the problems of increased prevention and treatment cost, overproof fruit residues, environmental pollution, development of drug resistance and the like caused by unreasonable large quantities of chemical pesticides for a long time are solved, and the system drug resistance measurement result in our hospital shows that part of ring rot germs have low-level drug resistance to common bactericides such as tebuconazole, carbendazim, difenoconazole and the like. The reasonable field barrel-mixed chemical pesticide is an important way for solving the problem that the current pesticide is not scientifically and reasonably used.
The zhongshengmycin serving as a protective bactericide is an antibiotic produced by Hainan variety of Streptomyces lavendulae, and belongs to N-glycoside alkaline water-soluble substances. The action mechanism is mainly that the bacteria are inhibited from synthesizing mycoprotein, so that the thallus is dead; for fungi, the strain deforms filamentous hyphae, inhibits spore germination and can directly kill spores. The zhongshengmycin has contact killing and penetrating functions, high activity on bacterial diseases and partial fungal diseases of crops and certain yield increasing effect. The development of resistance to crop pathogens has not been retrieved. Imazalil is a systemic broad-spectrum bactericide, has a control effect on a plurality of fungal diseases of fruits, vegetables and ornamental plants, particularly has an excellent control effect on common penicilliosis and green mildew of fruits and vegetables, and is recommended by the Ministry of agriculture to be one of the bactericides preferentially used in green agriculture. The action mechanism is that the permeability, physiological function and lipid anabolism of the cell membrane are influenced, so that the cell membrane of the pathogenic bacteria is damaged, and the formation of pathogenic bacteria spores is inhibited. Imazalil is a novel bactericide which is newly popularized, and zhongshengmycin with different action sites is selected to be compounded with the imazalil, so that the drug resistance of pathogenic bacteria to the imazalil can be prevented, and the using amount of field preparations can be reduced.
Disclosure of Invention
The invention aims to provide the zhongshengmycin and imazalil single agent for preventing and controlling important fruit tree diseases such as apple ring spot, grape white rot and the like and the compound composition thereof, the compound composition can effectively prevent and control the important fruit tree diseases such as apple and grape diseases, the field dosage of a chemical bactericide is reduced, the optimal proportion of the compound bactericide is screened out through indoor and outdoor tests, the synergy is obvious, the environmental pollution is reduced, and the generation and development of the pathogenic bacterium resistance are well controlled.
The invention is realized by the following technical scheme:
the compound composition for preventing and treating the fruit tree diseases is characterized in that the weight ratio of the zhongshengmycin to the imazalil in the effective components is 60: 1-1: 60;
the compound composition for preventing and treating the fruit tree diseases is further improved in that the weight ratio of the zhongshengmycin to the imazalil in the effective components is 4: 1-1: 20;
the compound composition for preventing and treating the fruit tree diseases is further improved in that the optimal weight of the zhongshengmycin and the imazalil in the effective components is 1:4, and the synergistic coefficient is 2.05;
the compound composition for preventing and treating the diseases of the fruit trees comprises the active ingredients of the zhongshengmycin and the imazalil, wherein the total weight of the zhongshengmycin and the imazalil in the whole compound composition accounts for 10 percent of the compound composition, and the balance is a conventional auxiliary material additive.
The compound composition for preventing and treating the fruit tree diseases is applied to preventing and treating apple ring rot, apple anthracnose, grape white rot and grape anthracnose.
The compound composition for preventing and treating the fruit tree diseases has better prevention effect when being applied to preventing and treating apple ring spot, apple anthracnose, grape white rot and grape anthracnose under the dilution of 800-1000 times.
The compound composition for preventing and treating the fruit tree diseases has the following advantages:
1. the bactericidal composition has a synergistic effect, and indoor toxicity measurement results show that the zhongshengmycin and the imazalil have a synergistic effect on the botrytis cinerea in a range of 4:1 to 1:20, wherein the synergistic effect of 1:4 is most obvious, and the synergistic coefficient reaches 2.05;
2. the optimal proportion of the field barrel-mixing has outstanding control effect, the dosage of the preparation is reduced, the prepared 10 percent Zhongshengmycin-imazalil compound composition 800 times has outstanding performance in the drug effect test for controlling apple ring rot, apple anthracnose, grape white rot and grape anthracnose in the field, the performance is superior to or equivalent to the control effect of a single agent and a contrast agent, simultaneously, the dosage of the preparation is greatly reduced, and compared with the barrel-mixing directly according to the label concentration, the dosage of the compound composition is respectively reduced by 50 percent and 60 percent by 800 times and 1000 times;
3. the safety is high, the toxicity of the zhongshengmycin and the imazalil to human and livestock is low, the content of the prepared compound composition is lower than that of the effective component of a single agent in the market, and the compound composition is safer for crops, environment and fresh fruit eaters;
4. the risk of drug resistance of pathogenic bacteria is reduced, as described in the item 2, the dosage of the zhongshengmycin and the imazalil in the compound composition is obviously lower than that of a single agent product sold in the market, and the dosage of the preparation is obviously reduced when the compound composition is used in the field, so that the selection pressure of pathogenic bacteria such as ring rot of apple and the like on the two bactericides can be greatly reduced, in addition, the zhongshengmycin and the imazalil have different action mechanisms, and the pathogenic bacteria are not easy to generate drug resistance when the compound composition is.

Example 1
Indoor toxicity test and compounding ratio screening
1.1 test agent
12% of a zhongshengmycin mother drug, a product of Fujian Kaili biological products Co.Ltd;
98% imazalil technical, product of Israeli Mark chemical company.
1.2 test strains
Grape white rot (Coniothyrium diplodiella) is obtained by separating field diseased fruits, and the first generation strain obtained by single spore culture and purification is stored in a refrigerator at 4 ℃ for later use.
1.3 instrumentation
The instrument mainly comprises an electronic balance, an ultra-clean workbench, a biological incubator, a culture dish, a pipettor and the like. 1.4 Experimental methods
The FAO-recommended cell growth rate method (Mycelium growth rate test) was used.
1.4.1 compounding of medicaments

1.4.2 preparation of drug-containing plates
Adding the medicine into PDA culture medium dissolved quantitatively according to the medicine concentration gradient set at 1.4.1 to obtain a medicine-containing plate, and repeating for 3 times each concentration with the plate without medicine as blank control.
1.4.3 inoculum culture
And (3) punching a bacterium disc by using a puncher with the diameter of 0.6 cm along the edge of the cultured bacterial colony, reversely transplanting the bacterium disc onto a flat plate by using a disinfection inoculation needle, and placing the flat plate at the constant temperature of 25 ℃ for culturing for 6 days.
1.4.4 investigation of results
And measuring the expansion diameter of the bacterial colony by a ruler cross measurement method, and calculating the inhibition rate of the medicament on the growth of hyphae compared with the expansion diameter of a control bacterial disc without applying the medicament.
1.5 statistical analysis
Regression analysis is carried out according to the logarithm value of the concentration of each medicament and the corresponding hyphal growth inhibition rate value, and a toxicity regression equation, a correlation coefficient (r) and EC of each medicament are calculated50。

The synergistic effect of the compounded medicament is evaluated according to the synergistic coefficient (SR), namely, SR <0.5 is antagonistic action, SR <0.5 and less than or equal to 1.5 are additive action, and SR >1.5 is synergistic action. 1.6 analysis of results
Indoor toxicity experiments show that the weight ratio of the zhongshengmycin to the imazalil is 30: 1-10: 1, the weight ratio of the zhongshengmycin to the imazalil is 30: 1-40: 1, the weight ratio of the zhongshengmycin to the imazalil is additive, the synergistic effect is in the range of 4: 1-1: 20, 1:4 is the optimal compounding ratio, and the synergistic coefficient is 2.05.

Example 2
Plot experiment in the field
According to an indoor compounding ratio screening experiment, the Zhongshengmycin and imazalil are screened out to be in the optimal ratio of 1:4, and a 10% Zhongshengmycin and imazalil compound composition is prepared. According to the pesticide test rule (II) of pesticide field efficacy test of Ministry of agriculture, part 13, 121 and 181, 10% Zhongshengmycin-imazalil compound composition is designed for field efficacy tests on apple ring rot, apple anthracnose, grape white rot and grape anthracnose. 2.1 test Agents
1000 times of 20% imazalil aqueous emulsion, a product of one-sail biotechnology group limited company;
600 times of 3% zhongshengmycin wettable powder, a product of Shandong Mgan-Fengchen Biotech limited company;
10% Zhongshengmycin-imazalil is 800 times, 1000 times and 1200 times, and is prepared by a tobacco platform farm institute.
2.2 control Agents
430g/L tebuconazole 4000 times of suspending agent, Bayer crop science (China) products.
2.3 cell design
The test is provided with 8 treatments, each treatment is repeated for 4 times, 32 cells are provided, and 6 adult trees are provided in each cell. The cells are randomly arranged. The pesticide application instrument adopts a Gongnong 26 type high-pressure motor sprayer, and all the branch tip blades in the tree crown are uniformly applied with pesticide until water drops.
2.4 investigation time and method
3-4 plants are selected in the center of each cell in the harvesting period, all fruits are investigated, the total number of fruits and the number of diseased fruits are recorded, and the control effect is calculated according to the disease fruit rate.

The field efficacy test result shows that the 10% zhongshengmycin and imazalil have good control effect on apple ring spot. The 800-time and 1000-time control effects are respectively 89.51 percent and 88.47 percent, are obviously higher than the control effects of two single agents of 20 percent imazalil aqueous emulsion 1000 times, 3 percent zhongshengmycin wettable powder 600 times and a contrast agent of 430g/L tebuconazole suspending agent 4000 times, and have no obvious difference with the 20 percent imazalil aqueous emulsion 1000 times and 3 percent zhongshengmycin wettable powder 600 times in tank mixing (Table 4).

The 10% zhongshengmycin and imazalil compound composition has 800-1200 times of prevention and control effect on apple anthracnose, the 800-1000 times of prevention effect is respectively 87.02% and 86.29%, the prevention effect is not significantly different from that of a single 20% imazalil aqueous emulsion by 1000 times, but is significantly higher than that of a single 3% zhongshengmycin wettable powder by 600 times and that of a contrast medicament 430g/L tebuconazole suspending agent by 4000 times, and the prevention effect is not significantly different from that of a 20% imazalil aqueous emulsion by 1000 times and that of a 20% zhongshengmycin wettable powder by 600 times in tank mixing (Table 5). The 1200-time control effect of the 10% Zhongshengmycin and imazalil compound composition is not obviously different from the 4000-time control effect of the 430g/L tebuconazole suspending agent serving as a contrast medicament, but is obviously higher than the 600-time control effect of a single-dose 3% Zhongshengmycin wettable powder.

The field test result of 800-1200 times prevention and treatment of grape white rot of 10% zhongshengmycin and imazalil compound composition shows that the 800-time and 1000-time prevention effect has no significant difference with 100 times of a single 20% imazalil aqueous emulsion, 600 times of tank-mixed with 1000 times of 20% imazalil aqueous emulsion and 4000 times of 600 times of a 20% zhongshengmycin wettable powder and 4000 times of a contrast agent 430g/L tebuconazole suspending agent, but is significantly higher than the prevention effect of 600 times of the single 3% zhongshengmycin.

800 times and 1000 times of control effects of the 10% zhongshengmycin and imazalil compound composition for preventing and treating grape anthracnose are 90.77% and 89.44%, respectively, the control effects are not obviously different from the control effects of 1000 times of a single-dose 20% imazalil aqueous emulsion, but are obviously higher than 4000 times of the control effects of a contrast medicament 430g/L tebuconazole suspending agent. The 1200-time control effect of the compound composition has no significant difference with the 4000-time control effect of the 430g/L tebuconazole suspending agent as a contrast medicament.

Example 3
Safety test
The content of the zhongshengmycin and the imazalil in the compound composition is less than that of a commercial product, so that the toxicity to human and livestock and the safety of crops are in accordance with requirements. The toxicity test result of the compound bactericide by a affirming organization shows that the compound bactericide belongs to a slightly toxic pesticide:
rat acute oral toxicity LD50 of more than 5000mg/kg and male parent of more than 5000 mg/kg;
rat acute percutaneous toxicity LD50 of greater than 5000mg/kg and male parent of greater than 5000 mg/kg;
the integral of the rabbit eye irritation is 0(1h), and the rabbit eye irritation is diluted by 0(1:100) after 48h, so that the rabbit eye irritation is not irritant to eyes;
the integral of the rabbit eye irritation is 0(4h), and the rabbit eye irritation is not irritant to skin.
According to the invention, the effects of the antibiotic zhongshengmycin and imazalil on apple ring spot, apple anthracnose, grape white rot and grape anthracnose after being used independently and compounded through indoor toxicity measurement and field tests are used, the optimal ratio of the two pesticides is determined for production and use, and the purposes of reducing the environment pollution of the pesticides and controlling the generation and development of drug resistance of germs are achieved.
