Abstract
本发明提供了一种香菇菌棒的转色菌皮提取物的制备方法,将香菇菌棒转色菌皮从香菇菌棒取下,然后进行干燥,干燥后粉碎过40‑200目筛;称取转色菌皮粉,按质量比为l:20~50的比例加入蒸馏水,80~100℃的水浴条件下提取1~4小时,重复提取2~5次,合并滤液,然后进行抽滤获得提取液;将提取液置于‑90~‑50℃的条件下冷冻12‑24小时,然后用真空冷冻干燥机干燥,浓缩,得到得香菇菌棒的转色菌皮提取物。还提供了上述的香菇菌棒的转色菌皮提取物在制备治疗抗菌药物或者保健制品中的应用。以革兰氏阳性菌金黄色葡萄球菌为受试菌,采用体外抑菌圈法对其抗菌效果进行了检测,具有显著抑制效果。
Description
技术领域
本发明属于生物医药领域,涉及一种香菇,具体来说是一种香菇菌棒的转色菌皮提取物的制备方法及其应用。
背景技术
抗菌药物是临床应用最广的一类药物,但是目前这类药物的应用存在着许多问题,如耐药性、毒副作用等。尤其是细菌的耐药性发展比较快,使得对这类菌所致的严重感染没有有效的药物可以使用,对人类健康造成较大威胁。因此,目前人们从天然资源中发展新的抗菌药物作为抗菌药物研究的一个重点。
香菇[Lentinula edodes(Berk.)Pegler.],俗称冬菇,是世界第二大食用菌,其栽培始源于中国,至今已有800年以上的历史。在1989年,中国香菇总产量首次超过日本,一跃成为世界香菇生产第一大国。香菇防治癌症的范围广泛,已用于临床治疗。香菇还含有多种维生素、矿物质,对促进人体新陈代谢,提高机体适应力有很大作用。实际生产过程中,香菇栽培周期至少可以分为四个重要阶段,分别为营养菌丝生长阶段、菌丝后熟与转色形成阶段、原基形成阶段、子实体生长发育阶段。迄今,其中香菇菌丝后熟与转色形成阶段的菌棒表层的转色菌皮具体功效包括体外抑菌活性方面未见报道。
发明内容
本发明的目的在于提供一种香菇菌棒的转色菌皮提取物的制备方法及其应用,所述的这种香菇菌棒的转色菌皮提取物的制备方法及其应用要解决现有技术中的抗生素在治疗过程中容易产生耐药性的技术问题,同时要解决香菇菌棒的转色菌皮没有得到有效利用的技术问题。
本发明提供了一种香菇菌棒的转色菌皮提取物的制备方法,包括如下步骤:
1)将香菇菌棒转色菌皮从香菇菌棒取下,然后进行干燥,干燥后粉碎过40-200目筛;
2)称取转色菌皮粉,按质量比为l:20~50的比例加入蒸馏水,80~100℃的水浴条件下提取1~4小时,重复提取2~5次,合并滤液,然后进行抽滤获得提取液;
3)将提取液置于-90~-50℃的条件下冷冻12-24小时,然后用真空冷冻干燥机干燥,浓缩,得到得香菇菌棒的转色菌皮提取物。
本发明还提供了上述制备方法获得的香菇菌棒的转色菌皮提取物。
本发明还提供了上述香菇菌棒的转色菌皮提取物在制备抗菌药物、抗菌试剂、防腐剂、食品或者保健制品中的应用。
进一步的,所述的细菌为金黄色葡萄球菌。
本发明还提供了一种药物组合物,含有上述的香菇菌棒的转色菌皮提取物。
本发明以废弃的香菇栽培菌棒的菌皮为原料,经过刮取粉碎、热水提取、减压浓缩(抽滤)、干燥等步骤,得到一种具有高抗菌活性作用的香菇菌棒菌皮提取物。以革兰氏阳性菌金黄色葡萄球菌为受试菌,采用体外抑菌圈法对其抗菌效果进行了检测,结果显示,香菇菌棒转色菌皮的水提取物具有较强的抗黄色金葡萄球菌的作用。其抗菌效果与香菇菌棒的非菌皮提取物进行比较,具有显著抑制效果。本发明的香菇菌棒的转色菌皮提取物可作为抗菌类药物制剂的原料,制成片剂、胶囊、复方制剂等,应用于食品、药品等行业。
本发明和已有技术相比,其技术效果是积极和明显的。本发明以香菇菌棒表层的转色菌皮为原料,进行抗菌提取物的研制,可以作为天然的食品和药品抗菌防腐剂,实现香菇菌棒的综合利用。
具体实施方式
以下通过抑菌活性实验来进一步证明本发明的有益效果:
实施例1
将干燥的香菇菌棒转色菌皮粉碎过的40目筛,称取20g,加入1000ml的蒸馏水,用100℃水进行浸提2小时,重复3次,抽滤得到提取液,-80℃冷冻12小时后用真空冷冻干燥机干燥24小时,得到干燥品。
采用微量稀释法测定香菇菌棒的转色菌皮提取物的最小抑菌浓度(MICs)。其中菌皮提取物设定的梯度,即2048,1024,512,256,128,64,32,16,8,4,2,1,0.5和0.25mg.L-1。
步骤如下:
1)将PA接种于LB液体培养基中,在37℃,220r/min条件下,培养24h。
2)2500r/min离心5min,倾掉上清液,用PBS稀释菌液至0.5麦氏比色管。
3)将0.5麦氏比色的菌液用LB液体培养基再稀释200倍备用。
4)在96孔中每个梯度设为8个复孔,每孔分别加入100μL的含香菇菌棒转色菌皮提取物的培养液和100μL的3)步的菌液,混合后37℃恒温过夜培养24h来测定最小抑菌浓度。实验重复三次。以香菇菌棒转色菌皮提取物的抑制待测菌生长的最低浓度为最小抑菌浓度(MIC)。实验结果如下。
抑菌作用结果如表1所示。

由表1可知,香菇菌棒转色菌皮对金黄色葡萄球菌有较好的抑菌效果,其对对金黄色葡萄球菌的MIC为256mg/L。
Claims (1)
1.一种香菇菌棒的转色菌皮提取物在制备抗金黄色葡萄球菌的药物中的用途,所述的香菇菌棒的转色菌皮提取物的制备步骤为:
1)将香菇菌棒转色菌皮从香菇菌棒取下,然后进行干燥,干燥后粉碎过40-200目筛;
2)称取转色菌皮粉,按质量比为l:20~50的比例加入蒸馏水,80~100℃的水浴条件下提取1~4小时,重复提取2~5次,合并滤液,然后进行抽滤,获得提取液;
3)将提取液置于-90~-50℃的条件下冷冻12-24小时,然后用真空冷冻干燥机干燥,浓缩,得到得香菇菌棒的转色菌皮提取物。
