本发明涉及妇科技术领域,具体涉及一种抗菌肽妇科凝胶的制备方法。
背景技术
健康女性阴道在内分泌激素的作用下,上皮细胞增生,表层细胞含有丰富的糖原,有利于乳酸杆菌的生长,这种细菌占阴道的90%以上。这种乳酸杆菌大量存在,就抑制了其他致病菌的生长,在阴道形成了一个正常的生态平衡。
当外界病原体大量侵入,破坏阴道的防御功能,正常寄生在阴道内的菌群微生态平衡失调,阴道内乳酸杆菌等有益菌减少而其他细菌大量繁殖,厌氧菌繁殖同时产生胺类物质,碱化阴道,使阴道分泌物增多并有臭味,最终导致细菌性阴道炎。
目前妇科凝胶采用纳米银离子,具有消炎、杀菌、止痒作用,对金黄色葡萄球菌、大肠杆菌、淋球菌、白色念珠球菌、真菌、酵母菌等病原体微生物的生长有明显抑制作用主要采用抗生素治疗,但也杀灭了有益菌群(乳酸杆菌,阴道双歧杆菌),破坏了阴道菌落的生态平衡。并且制造成本高,接触反应强烈,不适合预防使用。
CN101099815A公开了妇科抗菌凝胶及其制备方法,具体公开了以芦茶、磺胺嘧啶银干粉、冰片等为特征组分的应用于妇科疾患抑菌清洗的药用组合物;制备方法为将羧甲基纤维素溶于水,加入芦荟叶肉胶、磺胺嘧啶银干粉、冰片混合形成凝胶,添加的磺胺嘧啶银干粉和冰片杀菌效果很强,也快速地杀灭了有益菌群,适用于短暂紧迫的疾患治疗,但不适合吸收平和、持效长久的预防。所以,目前缺乏成本低、吸收平和、疗效持久、调节阴道生态平衡的妇科凝胶。
发明内容
本发明所要解决的技术问题是,克服以上现有技术的缺陷,提供一种吸收平和、疗效持久、调节阴道生态平衡的抗菌肽妇科凝胶的制备方法。
本发明的技术解决方案如下:一种抗菌肽妇科凝胶的制备方法,包括以下步骤:
1)取蜜蜂,50~70℃加热1~2h,烘干全虫,粉碎,得到蜜蜂粉末;
2)将蜜蜂粉末放入含精氨酸的甘油-醋酸溶液中,浸泡12~20h,超声振动10~20min,得到混合液;
3)将混合液过滤除去滤渣,滤液80~100℃加热40~60min,浓缩,得到蜜蜂提取液;
4)向蜜蜂提取液内加入丝瓜汁、芦荟粉,30~50℃搅拌20~40min,再加入0.6~1.2mmol/L的MgCl2溶液,搅拌后冻干,在相对湿度为60~80%的空气下恢复常温,即得。
优选地,所述蜜蜂为意大利蜜蜂、蜂王浆高产蜜蜂、安纳托利亚蜜蜂、高加索蜜蜂、卡尼鄂拉蜜蜂、喀尔巴阡蜜蜂中的一种。
优选地,所述蜜蜂粉末的粒径为1~5mm。
优选地,所述步骤2)甘油-醋酸溶液中甘油的体积分数为15~30%。保持弱酸性环境,发挥阴道的自洁功能。

蜜蜂接触外界多病毒多细菌的环境,能分泌多种抗菌肽,且免疫能力优于人体娇弱的阴道,故而分泌的抗菌肽天然、抗菌效果高效。经过步骤1)、2)、3),粉碎、浸泡提取、过滤,得到的蜜蜂提取物包含大量抗菌效果优异的抗菌肽,这些抗菌肽成分能够杀灭致病菌群(如加特纳菌、类杆菌),为有益菌群乳酸杆菌创造健康平和的生长繁殖条件,调节阴道炎环境下失调的阴道菌群平衡。产品抗菌肽成分有利于阴道平和地吸收,无刺激性,坚持适当使用3~6个疗程(连续使用1周为一疗程)明显缓解阴道炎病症,分泌物减少,外阴没有瘙痒和灼热感的症状,白带异味消失。
本发明的有益效果是:
1)烘干,水分蒸发,加快粉碎速度,去除蜜蜂翅、足、胸等硬质壳体,提取粉末,方便加工;
2)将蜜蜂粉末浸泡于含精氨酸的甘油-醋酸缓冲液中,最大限度的提取出蜜蜂粉末内的抗菌肽,使得抗菌肽成分快速渗透到缓冲液中,精氨酸有助于增加抗菌肽成分在缓冲液中的溶解度和吸收率,保持抗菌肽的效用;
3)蜜蜂提取液经步骤1)~3)快速高效的提取,简单方便,无需发酵、分子筛分离等复杂工艺;
4)过滤除去固体杂质,再经加热浓缩,增加蜜蜂提取液的浓度,保持蜜蜂体内抗菌肽的抗菌品质;
5)MgCl2促进蜜蜂提取物内抗菌肽的表达,加快杀死致病菌群,芦荟粉进一步消除炎症,溶于丝瓜汁阴道对抗菌肽的充分吸收和保持,增加湿润度,为乳酸杆菌提供优良的繁殖条件;
6)蜜蜂提取液与丝瓜汁、芦荟粉、MgCl2溶液混合好的溶液,经过冻干处理,杀菌消毒,液态变为易凝胶化的固态,在湿润环境中膨胀为柔软适中、伸展压缩自如、胶黏稳定的凝胶胶体,方便使用;
7)各组分通过协同作用,经过多步骤加工制作,成本低,取自有益昆虫蜜蜂,得到的抗菌肽妇科凝胶能够吸收平和、疗效持久、调节阴道生态平衡,有效治疗阴道炎。

以下所提供的实施例并非用以限制本发明所涵盖的范围,所描述的步骤也不是用以限制其执行顺序。本领域技术人员结合现有公知常识对本发明做显而易见的改进,亦落入本发明要求的保护范围之内。
实施例一
一种抗菌肽妇科凝胶的制备方法,包括以下步骤:
1)取蜜蜂,50℃加热2h,烘干全虫,粉碎,得到蜜蜂粉末;所述蜜蜂为意大利蜜蜂;所述蜜蜂粉末的粒径为1mm;
2)将蜜蜂粉末放入含精氨酸的甘油-醋酸溶液中,浸泡12h,超声振动10min,得到混合液;所述甘油-醋酸溶液中甘油的体积分数为15%;所述精氨酸在甘油-醋酸溶液的浓度为0.5mol/L;
3)将混合液过滤除去滤渣,滤液80℃加热60min,浓缩至密度为1.4g/ml,得到蜜蜂提取液;
4)按如下质量份数比例配制:蜜蜂提取液4份、丝瓜汁10份、粒径为0.12mm的芦荟粉8份、MgCl2溶液1份;向蜜蜂提取液内加入质量浓度为10%的丝瓜汁、芦荟粉,30℃搅拌40min,再加入0.6mmol/L的MgCl2溶液,搅拌后冻干,在相对湿度为80%的空气下恢复常温冷却,即得。
实施例二
一种抗菌肽妇科凝胶的制备方法,包括以下步骤:
1)取蜜蜂,70℃加热1h,烘干全虫,粉碎,得到蜜蜂粉末;所述蜜蜂为蜂王浆高产蜜蜂;所述蜜蜂粉末的粒径为5mm;
2)将蜜蜂粉末放入含精氨酸的甘油-醋酸溶液中,浸泡20h,超声振动20min,得到混合液;所述甘油-醋酸溶液中甘油的体积分数为30%;所述精氨酸在甘油-醋酸溶液的浓度为1mol/L;
3)将混合液过滤除去滤渣,滤液100℃加热40min,浓缩至密度为1.6g/ml,得到蜜蜂提取液;
4)按如下质量份数比例配制:蜜蜂提取液8份、丝瓜汁15份、粒径为0.18mm的芦荟粉10份、MgCl2溶液2份;向蜜蜂提取液内加入质量浓度为30%的丝瓜汁、芦荟粉,50℃搅拌20min,再加入1.2mmol/L的MgCl2溶液,搅拌后冻干,在相对湿度为60%的空气下恢复常温冷却,即得。
实施例三
一种抗菌肽妇科凝胶的制备方法,包括以下步骤:
1)取蜜蜂,60℃加热1.5h,烘干全虫,粉碎,得到蜜蜂粉末;所述蜜蜂为安纳托利亚蜜蜂;所述蜜蜂粉末的粒径为3mm;
2)将蜜蜂粉末放入含精氨酸的甘油-醋酸溶液中,浸泡15h,超声振动15min,得到混合液;所述甘油-醋酸溶液中甘油的体积分数为22%;所述精氨酸在甘油-醋酸溶液的浓度为0.8mol/L;
3)将混合液过滤除去滤渣,滤液90℃加热50min,浓缩至密度为1.5g/ml,得到蜜蜂提取液;
4)按如下质量份数比例配制:蜜蜂提取液6份、丝瓜汁12份、粒径为0.16mm的芦荟粉9份、MgCl2溶液1.5份;向蜜蜂提取液内加入质量浓度为20%的丝瓜汁、芦荟粉,40℃搅拌30min,再加入0.8mmol/L的MgCl2溶液,搅拌后冻干,在相对湿度为70%的空气下恢复常温冷却,即得。
产品测试
一、抑菌效果测试
a)培养试菌:将致病菌群(加特纳菌、类杆菌)和有益菌群(乳酸杆菌,阴道双歧杆菌)活化,挑取单个菌落至5ml相应液体培养基中,置于37℃培养12h;取100μL,稀释10倍后取100μL菌悬液至固体培养基平板上,并涂匀。
b)参照说明书制备本发明的抗菌肽妇科凝胶样品A。
c)将滤纸用打孔机打成半径为3mm的圆片,120℃高压蒸气灭菌30min,烘干,得到无菌滤纸片。
d)在无菌滤纸片表面涂抹凝胶样品A,涂抹厚度均为10μm,涂抹面贴在含菌平板上,置于37℃培养12h,测定滤纸片抑菌圈的半径,结果见表1。

抑菌圈即表示抗菌效果,即抑菌圈半径越大,抗菌效果越好。由表1可以看出,本发明的抗菌肽妇科凝胶样品A对致病菌群(加特纳菌、类杆菌)由优良的抗菌效果,对有益菌群(乳酸杆菌,阴道双歧杆菌)不会造成强烈的破坏,本发明的抗菌肽妇科凝胶能够高效的杀灭致病菌群(加特纳菌、厌氧菌),并为有益菌群乳酸杆菌创造健康平和的生长繁殖条件,从而调节阴道炎环境下失调的阴道菌群平衡。
二、各组分协同作用测试
1、培养试菌:将致病菌群(加特纳菌、类杆菌)活化,挑取单个菌落至5ml相应液体培养基中,置于37℃培养12h;取100μL,稀释10倍后取100μL菌悬液至固体培养基平板上,并涂匀。
2、制备不同的凝胶样品A、A1、A2、A3,其中A为本发明的抗菌肽妇科凝胶,参照说明书制备;A1为本发明步骤4)中取消丝瓜汁制成的凝胶;A2为本发明步骤4)中取消芦荟粉制成的凝胶;A3为本发明步骤4)中取消MgCl2溶液制成的凝胶。
3、将滤纸用打孔机打成半径为3mm的圆片,120℃高压蒸气灭菌30min,烘干,得到无菌滤纸片。
4、在无菌滤纸片表面涂抹不同的凝胶样品A、A1、A2、A3,涂抹厚度均为10μm,涂抹面贴在含菌平板上,置于37℃培养12h,测定滤纸片抑菌圈的半径,具体结果见表2。

由表2可以看出,A1、A2、A3是取消部分组分后的测试结果,加特纳菌、类杆菌的抑菌圈半径都较A的小,说明取消丝瓜汁、芦荟粉或MgCl2溶液对致病菌群的抗菌效果都有所降低。采用本发明A将蜜蜂提取液与丝瓜汁、芦荟粉、MgCl2溶液混合,制成的凝胶抗菌效果最好。
Claims (8)
1.一种抗菌肽妇科凝胶的制备方法,其特征在于,包括以下步骤:
1)取蜜蜂,50~70℃加热1~2h,烘干全虫,粉碎,得到蜜蜂粉末;所述蜜蜂粉末的粒径为1~5mm;
2)将蜜蜂粉末放入含精氨酸的甘油-醋酸溶液中,浸泡12~20h,超声振动10~20min,得到混合液;
3)将混合液过滤除去滤渣,滤液80~100℃加热40~60min,浓缩,得到蜜蜂提取液;
4)向蜜蜂提取液内加入丝瓜汁、芦荟粉,30~50℃搅拌20~40min,再加入0.6~1.2mmol/L的MgCl2溶液,搅拌后冻干,在相对湿度为60~80%的空气下恢复常温,即得;所述步骤4)中各组分的质量份数按如下比例配制:蜜蜂提取液4~8份、丝瓜汁10~15份、芦荟粉8~10份、MgCl2溶液1~2份。
2.根据权利要求1所述的抗菌肽妇科凝胶的制备方法,其特征在于,所述蜜蜂为意大利蜜蜂、蜂王浆高产蜜蜂、安纳托利亚蜜蜂、高加索蜜蜂、卡尼鄂拉蜜蜂、喀尔巴阡蜜蜂中的一种。
3.根据权利要求1所述的抗菌肽妇科凝胶的制备方法,其特征在于,所述甘油-醋酸溶液中甘油的体积分数为15~30%。
4.根据权利要求1所述的抗菌肽妇科凝胶的制备方法,其特征在于,所述精氨酸在甘油-醋酸溶液的浓度为0.5~1mol/L。
5.根据权利要求1所述的抗菌肽妇科凝胶的制备方法,其特征在于,所述步骤3)浓缩至密度为1.4~1.6g/ml。
6.根据权利要求1所述的抗菌肽妇科凝胶的制备方法,其特征在于,所述丝瓜汁的质量浓度为10~30%。
7.根据权利要求1所述的抗菌肽妇科凝胶的制备方法,其特征在于,所述芦荟粉的粒径为0.12~0.18mm。
8.根据权利要求1所述的抗菌肽妇科凝胶的制备方法,其特征在于,所述步骤4)冻干指先在-10~-30℃冷冻,然后在2~10Pa真空条件下脱水干燥。
