Abstract
本发明公开了一株植物乳杆菌新菌株及其应用。所述的植物乳杆菌新菌株,命名为581,保藏于中国微生物菌种保藏管理委员会普通微生物保藏中心,其微生物保藏号是CGMCC No.13121,保藏日期为2016年10月18日。该菌株可适用于多种果蔬汁的发酵,提高果蔬汁中有机酸、游离氨基酸、维生素等有益物质含量,对部分果蔬汁的不愉悦风味也具有改善作用。同时该菌株具有良好的耐酸、耐胆盐特性,对小肠上皮细胞具有良好的黏附特性,可以顺利到达肠道、与肠道上皮细胞发生黏附。体内小鼠实验结果表明,该菌株具有肠道改善功能,对便秘模型小鼠具有良好的润肠通便功效。

背景技术
果蔬汁是时下年经人以及关注健康的中老年人热衷选择的一种饮品,因其具有丰富的营养、新鲜健康而备受青睐。采用益生菌生物发酵技术,对果蔬汁进行深加工,除了可以增加果蔬汁的风味或改善一些蔬菜汁的不良风味外,更可以提升果蔬汁的保健功能,提高果蔬制品的附加值。益生菌通过代谢果蔬汁中的碳水化合物、蛋白质等物质,产生有机酸、肽类、酶类等有益物质,同时,果蔬汁中本身含有的维生素、膳食纤维等营养物质仍然存在,结合益生菌株本身的功能特性,将为消费者提供更多的有益因素。
随着现代人饮食结构的变化、心理因素和社会因素的影响,便秘的发病率有逐渐增高的趋势。严重便秘可导致电解质和酸碱平衡紊乱,引起痔疮、下肢静脉曲张等疾病,影响人们生活。此外,由于肠道排便不畅导致的毒素或有害代谢物的积累与多种慢性疾病的发生发展息息相关。早在1908年,诺奖获得者梅切尼科夫就提出了慢性疾病肠源性假说,近年来的研究更证实了这一观点。益生菌在发挥其润肠通便等肠道调节功能时,具有安全、无副作用的明显优势。植物乳杆菌在肠道中通过代谢产生的乳酸等有机酸,能够维持肠道的生态平衡、抑制有害细菌的生长,并能促进肠道蠕动,增加粪便含水量,作用温和,对维持肠道正常的生理功能具有重要作用。
适用于果蔬汁发酵的益生菌株除需具备益生功能外,还需具备一定的发酵特性。果蔬汁相比常作为益生菌发酵基料的乳类制品,营养组成相对贫瘠或存在缺失。此外,一些果蔬汁或浓缩果蔬汁还存在pH环境偏低、渗透压环境偏高等特性,果蔬汁发酵专用菌株还需具备一定的耐酸、耐高糖或耐盐特性。
发明内容
本发明的目的在于提供一种可用于果蔬发酵的植物乳杆菌新菌株,且所述菌株具有改善肠道功能的益生作用,可以顺利到达肠道、与肠道上皮细胞发生黏附,并发挥润肠通便功能。
本发明的另一个目的在于提供上述植物乳杆新菌株在发酵果蔬汁,以及其他食品、饮料、保健食品、药品、饲料中的应用。
为了达到上述目的,本发明采用了以下技术手段:
本发明菌株分离自四川泡菜样品,经MRS接种培养、分离纯化后分离获得的一株乳酸菌株,经鉴定其属于植物乳杆菌(Lactobacillus plantarum),命名为581,分类命名为植物乳杆菌(Lactobacillus plantarum),保藏在中国微生物菌种保藏管理委员会普通微生物中心,其微生物保藏号是CGMCC No.13121,保藏日期为2016年10月18日。
该菌株的生物学性质如下:
(1)形态学特征:在MRS培养基上形成明显的圆形、乳白色菌落,边缘整齐,隆起,不透明,表面光滑湿润,直径在0.2-2mm之间。革兰氏染色典型阳性,显微镜观察下细胞形状为杆状,无鞭毛,不产芽孢,不运动。
(2)培养特征:最适生长温度为30-37℃,需氧或兼性厌氧,生长于MRS培养基中。培养基成分:蛋白胨10g,牛肉膏10g,酵母膏5g,葡萄糖20g,乙酸钠5g,K2HPO4 2g,MgSO4·7H2O 0.2g,MnSO4·4H2O 0.2g,
本发明提供的植物乳杆菌(Lactobacillus plantarum)581可以适应果蔬汁中相对贫瘠的生长环境,在多种PH较低的果蔬汁环境下也能够良好生长,适用于多种果蔬汁的发酵,并对部分果蔬汁的不愉悦风味具有改善作用。所述果蔬汁的原料包括但不限于,香蕉、芒果、桑葚、木瓜、梨、蓝莓、葡萄、橙、西柚、哈密瓜、奇异果、胡萝卜、紫薯、甜菜、甘蓝、南瓜、西红柿、沙棘、大蒜、姜、芦荟、枸杞、山药等。
本发明所提供的植物乳杆菌(Lactobacillus plantarum)581除作为发酵剂发酵果蔬汁外,本身也具有益生功能。植物乳杆菌(Lactobacillus plantarum)581菌株具备良好的耐酸耐胆盐特性,在pH=2.0酸环境作用下4h后,活菌数仅降低0.243±0.035个数量级,在0.6%胆盐作用8h后,活菌数仅降低1.378±0.205个数量级;具有良好的黏附特性,在HT-29细胞模型试验中表现出良好的黏附能力,单细胞黏附菌数达12.41±0.97。表明植物乳杆菌581菌株能够顺利到达肠道,与肠道上皮细胞发生黏附。小鼠体内试验表明,服用植物乳杆菌581菌株可以缩短便秘小鼠的首次排便时间,增加小鼠的排便粒数、排便质量,并提高小鼠的小肠墨汁推进速率,表现出一定的润肠通便效果。

具体实施方式
实施例1
本发明菌株的分离与鉴定
本发明菌株从四川泡菜样品中分离获得,采用MRS固体平板经倾注平板法培养样品稀释液后,挑取单克隆的典型乳酸菌菌落进行反复划线纯化培养,获得该菌株的纯培养物。菌株的菌落特征及光学显微镜下的形态学特征见图1。
1)生理生化特征试验结果
该菌株氧化酶、接触酶反应均为阴性。碳水化合物产酸实验(API50CH)结果如下表所示,结合《伯杰氏细菌鉴定手册》,初步推断为植物乳杆菌。

PCR反应体系如表2所示。PCR扩增条件:94℃预变性5min;94℃变性30s,55℃退火30s,72℃延伸1min,30个循环;最后在72℃下延伸10min,终止反应。经测序后所得有效序列于NCBI上进行比对后,结果显示与植物乳杆菌同源性最高。

实施例2
植物乳杆菌新菌株在酸环境中的生长特性
本发明的植物乳杆菌581保存于甘油管中,经活化后,按1%接种量接种于不同pH梯度的液体MRS培养基中,测定其生长特性。采用紫外-分光光度计分别于接种后8h、16h、24h测定菌液浊度(OD600),结果如下表所示。植物乳杆菌581在pH=3.0的环境下即有轻微生长,培养24h后OD600达0.4770±0.0481,菌数为1.5×108cfu/mL;在pH=3.5的环境下能良好生长,培养24h后OD600达1.7020±0.0495,菌数为8.0×108cfu/mL;在pH≧4.0的环境下,培养24h后OD600≧2.20,菌数≧1.5×109cfu/mL。具有非常强的在酸环境中的生长能力。
实施例3
植物乳杆菌新菌株的产酸特性
本发明的植物乳杆菌581保存于甘油管中,经活化后,按1%接种量接种于液体MRS培养基中,测定其生长特性与产酸特性。581菌株于接种后16h达到生长稳定期,于接种后的7-8h产酸速率达到高峰,于接种后的16h产酸趋于稳定。
采用高效液相色谱法测定植物乳杆菌581的产酸量与产酸种类。使用安捷伦20RBAX SB-C18色谱株(250mm×4.6mm,5μm)作为固定相;流动相为0.01M NH4H2PO4,10min;流速1.0ml/min,进样体积20μL;DAD检测器210nm进行检测。以酒石酸、苹果酸、乳酸、乙酸、柠檬酸、富马酸为混标,外标法计算产酸量。结果表明,581菌株属于同型发酵,产酸种类为乳酸,于接种后16h产酸量达15.4055±0.1110g/L。
实施例4
植物乳杆菌发酵果蔬汁的制备
本发明菌株可用于多种果蔬汁原料的发酵,包括香蕉、芒果、桑葚、木瓜、梨、蓝莓、葡萄、橙、西柚、哈密瓜、奇异果、胡萝卜、紫薯、甜菜、甘蓝、南瓜、西红柿、沙棘、大蒜、姜、芦荟、枸杞、山药等。以下提供几个具体的实施例。
分别取胡萝卜、芒果、香蕉、紫薯、大蒜浓缩汁,稀释至合适浓度后,于95℃下灭菌5min。待料液冷却至30℃后,将本发明菌株(植物乳杆菌581)的冷冻干燥菌粉以0.002%(107cfu/mL)的添加量接种到发酵基料中,37℃下进行发酵。在接种后20h内测定基料的pH变化曲线,在发酵后16h测定菌数,结果如图2与下表所示。对发酵后果蔬汁进行感官品尝,经植物乳杆菌581发酵后,对果蔬汁本身的体系与色泽无显著影响,未观察到任何不良风味,其中对于胡萝卜汁和大蒜汁,其发酵料液原本带有的生冲味道明显减少,口感更加柔和绵厚,醇香可口,同时发酵产生的乳酸以及其他风味物质也为多款果蔬汁增加了清新、酸爽的口感。
实施例5
植物乳杆菌新菌株的耐酸耐胆盐特性
将本发明菌株与市售菌株均培养至对数生长末期后,获取菌悬液,将菌悬液分成2组,分别进行如下操作:①用盐酸分别调整至pH=2.0、pH=2.5的MRS溶液,37℃静置培养0、1、2、4h;②加入胆盐浓度分别为0.3、0.6%的MRS溶液,37℃静置培养0、4、8h;完成上述操作后,采用MRS固体培养基经稀释涂布法进行计数,结果如下表所示。L.plantarum 581耐酸特性明显优于市售菌株,在pH=2.0酸环境作用下,4h后活菌数仅降低0.243±0.035个数量级,市售菌株4h后活菌数降低3.554±0.778个对数值。L.plantarum 581耐胆盐特性在高浓度胆盐作用下更为明显,市售菌株在0.6%胆盐作用4、8h后,已无检出,本发明菌株在0.6%胆盐作用8h后,菌数仅降低1.378±0.205个数量级。
实施例6
植物乳杆菌新菌株的黏附特性
建立HT-29细胞培养体系,细胞生长于含10%胎牛血清的DMEM培养基中(含青霉素100U/ml,链霉素100mg/ml)。待细胞传至第三代后,采用0.25%的胰酶(含EDTA)消化后进行重悬,以1×106细胞/孔的密度接种于已放置细胞爬片的6孔细胞培养板中,于37℃,含5%CO2浓度的培养箱中进行培养,构建出模拟体内乳杆菌黏附肠上皮细胞的黏附模型。将本发明菌株与市售同类型菌株(对照菌1、对照菌2)均培养至对数生长末期后,进行黏附实验,结果如下表所示。L.plantarum 581表现出高黏附能力,优于市售的同类型菌株。

实施例7
菌株在实验动物体内的肠道改善功能试验
7.1小鼠排便实验
1)实验动物与分组
实验动物采用远交系ICR雄性小鼠150只,18-22g,由浙江中医药大学动物实验中心提供。
动物饲养保持环境温度为21±2℃,湿度为30-70%,12h光照交替,自由饮水,自由摄入小鼠维持饲料。实验小鼠自购入起适应1周后,按体重进行随机分组,分为5组。
对照组1:正常对照组,灌胃等体积生理盐水
对照组2:模型对照组,灌胃等体积生理盐水
对照组3:灌胃市售通便药物,灌胃剂量参照人体日服用量,为2g/kg小鼠体重;
对照组4:灌胃市售同类型菌株悬液,灌胃剂量为3×109cfu/kg小鼠体重;
实验组:灌胃本发明菌株悬液,灌胃剂量为3×109cfu/kg小鼠体重。
2)指标测定
受试样品给喂7d后,各组小鼠禁食不禁水16h。对照组1给喂蒸馏水,对照组2-4及实验组给予复方地芬诺酯悬液(10mg/kg BW),给药0.5h后,对照组1-2小鼠灌胃墨汁0.2ml,对照组3-4和实验组小鼠给喂含受试样品的墨汁0.2ml。灌胃墨汁后,每只小鼠单笼饲养,观察并记录每只小鼠首粒排黑便时间,以及6h内排黑便的粒数及重量。
3)实验结果
经植物乳杆菌(Lactobacillus plantarum)581灌胃7天后的小鼠,其首次排黑便时间(201.5±33.8vs.353.8±59.3,p<0.01)、6h排黑便粒数(6.9±3.7vs.2.40±1.35,p<0.01)、6h排黑便质量(0.2149±0.2043vs.0.0717±0.0559,p<0.01)三项指标均显著优于对照组2(模型对照组)及对照组1(未造模的正常对照组)小鼠,且优于市售通便药物及市售同类型菌株。

7.2小鼠小肠运动性实验
1)实验动物与分组

3)实验结果
与模型组(对照组2)小鼠相比,灌胃植物乳杆菌(Lactobacillus plantarum)581可显著增加受试小鼠的小肠墨汁推进率(39.88±9.10vs.29.27±5.11,p<0.01),作用效果与市售通便药物及市售同类型菌株相似。
