本发明属于保健品领域,本发明提供了一种粉剂由菊粉、龙眼肉、红枣、枸杞子、熟地黄、核桃仁、管花肉苁蓉、牛磺酸、五味子、和肉桂制备而成,原料全部来源于天然中草药,无辅料添加,成分天然,标签清洁,符合人们追求食品天然健康的理念趋势,且服用剂量小,服食方便,可直接口服或溶于水中服用,冷水可溶,入口即化,口感好,吸收快。本发明所述粉剂制备方法简单,适合大规模生产携带方便,制得的粉剂稳定性好,存放时间长。实验表明本发明所述粉剂可以促进实验动物的生长,增加动物精子活力,增进性反应时间与次数,提高性激素水平及性器官脏器系数比,具有壮阳功能,效果显著,可用于制备具有壮阳功能的保健食品。
Description
技术领域
本发明属于保健品领域,具体涉及一种粉剂及其制备方法与应用,尤其 是以天然中草药为主要原料制备的具有壮阳功能的保健粉剂。
背景技术
阳虚体质就是指人体的阳气不足,气是构成人体和维持人体生命活动的 最基本物质。阳气不足就会产生一系列症状,表现为疲倦怕冷,四肢冰冷、 唇色苍白,少气懒言,嗜睡乏力,男性遗精,女性白带清稀,易腹泻,排尿 次数频繁,性欲衰退等。阳虚体质的人平素畏冷,手足不温,易出汗;喜热 饮食,精神不振,睡眠偏多。
近年来人民生活水平明显提高人们的消费观念、健康观念发生了较大变 化。为规避不健康带来的各种不利影响,使人们越来越重视营养保健品的使 用。目前壮阳类保健食品主要以补充维生素、矿物质及补益类药材为主,大 多以口服液的形态存在。口服液剂携带不便,稳定性差,存放时间短。片剂 在压片时大都需要加入淀粉、羟甲基纤维素等大剂量辅料,崩解时间长。同 时对吞咽有困难的人群服用不方便。且补益类药材煎煮、携带、服用不便。
发明内容
有鉴于此,本发明的目的在于针对现有技术存在的缺陷提供一种粉剂及 其制备方法与应用。本发明所述粉剂以天然中草药为主要原料制备,具有壮 阳功能,同时不含辅料,成分天然,服用方便。
为实现本发明的目的,本发明采用如下技术方案:
一种粉剂,由菊粉、龙眼肉、红枣、枸杞子、熟地黄、核桃仁、管花肉 苁蓉、牛磺酸、五味子和肉桂制备而成。
其中,优选的,所述菊粉、龙眼肉、红枣、枸杞子、熟地黄、核桃仁、 管花肉苁蓉、牛磺酸、五味子和肉桂的质量比为(10~100):(1~10):(1~10):(1~ 10):(1~10):(1~10):(0.1~5):(0.1~10):(0.1~5):(0.1~5)。
在一些实施方案中,所述菊粉、龙眼肉、红枣、枸杞子、熟地黄、核桃 仁、管花肉苁蓉、牛磺酸、五味子和肉桂的质量比为10:3:3:2:2:2:1:0.3:0.5:0.5。
在一些实施方案中,所述菊粉、龙眼肉、红枣、枸杞子、熟地黄、核桃 仁、管花肉苁蓉、牛磺酸、五味子和肉桂的质量比为50:2.5:5:2.5:6.25:3.75:0.75: 0.25:2.5:2.5。
在另一些实施方案中,所述菊粉、龙眼肉、红枣、枸杞子、熟地黄、核 桃仁、管花肉苁蓉、牛磺酸、五味子和肉桂的质量比为20:2.5:1:4:1:4:1.5: 5:1:0.1。
本发明还提供了所述粉剂的制备方法,将所述龙眼肉、红枣、核桃仁、 枸杞子、熟地黄、五味子、肉桂和肉苁蓉加水提取,提取液浓缩,加入菊粉、 牛磺酸后干燥、粉碎。
其中,所述的加水提取优选为加水煎煮提取。
进一步的,所述的加水煎煮提取优选为提取1-2次,每次加3~20倍的水 提取0.5~3小时。
在一些实施方案中,所述加水煎煮提取为加20倍水煎煮提取3小时。
在一些实施方案中,所述加水煎煮提取为第一次加5倍~20倍的水提取 0.5~3小时,第二次加3倍到15倍的水提取0.5~2小时。在一些实施例中,第 一次加入15倍水提取2小时,第二次加入10倍水,提取1小时。在另一些 实施例中,第一次加5倍水,提取1小时,第二次加5倍的水提取0.5小时。
本发明所述制备方法在加水提取后收集所有提取液,进行浓缩。所述浓 缩优选为高温罐浓缩或反渗透浓缩。
进一步的,本发明所述制备方法在浓缩后进行干燥。所述干燥优选为喷 雾干燥、冷冻干燥、带式干燥、微波干燥或真空干燥。
优选的,本发明所述制备方法干燥后还包括粉碎的步骤。所述粉碎粒度 优选控制在40目到80目。
制粒,包装。
在某一具体实施方案中,本发明通过灌胃给予本发明所述粉剂和对照样 品,观察本发明所述粉剂对雄性大鼠的影响,结果显示,本发明所述粉剂可 以促进实验动物的生长,增加动物精子活力,增进性反应时间与次数,提高 性激素水平及性器官脏器系数比。
因此,本发明提供了所述粉剂在制备具有壮阳功能的保健食品中的应用。

制备方法:将龙眼肉、红枣、枸杞子、熟地黄、核桃仁、管花肉苁蓉、 五味子和肉桂经水煎煮提取2次,第一次加5倍水,提取1小时,第二次加5 倍的水提取0.5小时,合并2次提取液,反渗透浓缩,浓缩温度低于40℃, 加入菊粉、牛磺酸进行冷冻干燥,预冻温度-30℃,维持3-4小时,干燥,迅 速升温至60℃,保持1小时,随后再缓慢升温至80℃,保持10-12小时,得 冻干粉,粉碎即得所述保健粉剂。
试验例1、壮阳功能检验
1、材料与仪器
供试品:实施例2所述保健粉剂
试验动物:SPF级雄性大鼠,适应期结束,体重200±20g。首选SD大鼠。 每组8-10只。
将按体重随机分为正常对照组,阳性对照组,供试品低、中、高剂量组。 适应性喂养一周后,每天一次,连续灌胃给予四周,正常对照组给予等体积 生理盐水。
供试品低、中、高剂量组的配制方法:
供试品高剂量组(0.3g/ml的供试品溶液):精密称取13.5g供试品,加 45ml超纯水,配置成浓度为0.3g/ml的溶液,即为供试品高剂量组(1.5g/kg, 0.5ml/100g),相当于人体推荐量的30倍;
供试品中剂量组(0.1g/ml的供试品溶液):从高剂量组取15ml加30ml 超纯水,即为供试品中剂量组(0.5g/kg,0.5ml/100g),相当于人体推荐量 的10倍;
供试品低剂量组(0.05g/ml的供试品溶液):从中剂量组取15ml加15ml 超纯水,即为供试品低剂量组(0.25g/kg,0.5ml/100g),相当于人体推荐量 的5倍。
阳性对照品的配制方法:
精密称取东革阿里药材粉末100g至500ml烧瓶中,加去离子水提取3 次,每次400ml提取2小时。提取液合并浓缩,定容相当于0.4g/ml(药材/ 水)。精密量取6.625ml浓度为0.4g/ml的东革阿里原液,加超纯水至50毫 升,配置成浓度为0.053g/ml的溶液,即为东革阿里阳性对照组(31.8/3mg/kg, 0.5ml/100g),相当于人体推荐量的5倍。
2、试验方法
2.1大鼠体重称量
给药前,将大鼠称重后随机分组。给药后每周固定时间称重一次,实验 结束绘制体重变化曲线。
2.2各组大鼠阴茎勃起指数(PEI值)测定
适应性喂养5天后,每天一次,连续灌胃给药四周,对照组给予等体积 生理盐水。每组动物每周进行两次性行为学观察。给药完毕,每组动物分为3 批,3-4只老鼠/笼,以雌鼠在笼外做诱导,人工观察大鼠阴茎勃起时间(阴茎 勃起潜伏期:从观察时间开始,到阴茎首次勃起需要的时间,共测九次。第 一次是给药前测的,以后每周两次,共四周给药),并用摄像头连续记录90min 内大鼠阴部理毛、爬背次数和参与动物数,并计算PEI值。
PEI=每组大鼠相关性行为反应总次数×每组中有相关性行为反应的大 鼠数/每组大鼠数量
2.3血清睾酮水平检测
连续给药四周后,眼角静脉丛采血1.5ml,静置1h,3000转离心30min,取 血清,ELISA试剂盒检测血清睾酮水平。
2.4精子活力评定及脏器系数比
用1%戊巴比妥钠(0.2ml/100g)腹腔麻醉,下腹正中切口,提出睾丸, 找出睾丸和附睾,分离出睾丸和附睾,称重,然后用眼科剪剪下附睾,将一 侧附睾从中间剪断,取附睾尾端,置于2ml PBS中(37℃,预温),用眼科剪剪 成3段,温育10min,待附睾精子充分游离后显微镜下计数,计算精子活力:
精子活力=(计数室总精子数-不能前进运动的总精子数)÷计数室总精 子数。
最后将动物安乐死,再取精囊腺和提肛肌,将所有脏器称重,计算脏器 系数比。
2.4结果
2.4.1体重增长曲线
给药3周以后,供试品低剂量组体重增长略低于正常。中剂量组大鼠体 重增加明显,第四周后体重增长到339.4±16.9g,体重增长率达到119.8%,显 著高于正常组(体重329.8±21.7g,体重增长率93.8%)和东革阿里组。其他 组体重增长与正常组相比变化不大。

2.4.2脏器系数比
给药完毕,摘取各组大鼠的睾丸、附睾、提肛肌及精囊腺称重,比较各 组间脏器系数比。结果如表3所示。

结果显示,各组间除睾丸脏器系数比无明显差别外,阳性对照组及供试 品各组均出现了脏器系数比值的增加,其中精囊腺的增加最为明显。供试品 中高剂量组作用与阳性对照组相当。
2.4.2大鼠PEI值
给药后,东革阿里药材及供试品组大鼠爬背、阴部理毛及阴茎勃起等性 反应时间与强度均增强。如表4所示,东革阿里药材平均PEI值显著高于空白 组,供试品低、中、高剂量组平均PEI值均高于正常,与正常组比较,统计学 处理有显著性差异。与东革阿里组比较无统计学差异。

2.4.3阴茎勃起潜伏期
与给药前相比,给药2周后东革阿里药材及供试品组大鼠的阴茎勃起潜伏 期均缩短(表5),部分组别供试品组效果强于阳性对照组。中剂量组表现尤 其突出。

2.4.4血清睾酮结果
血清睾酮检测结果(表6)显示,低、中、高剂量的供试品组均提高血清 睾酮水平,其中低、中剂量组作用优于东革阿里药材。

2.4.5精子活力结果
精子活力检测结果如表7,供试品低、中、高剂量组均能增加雄性大鼠精 子活力,具有剂量依赖性关系,其中高剂量组作用与阳性对照相当。

按照上述方法对实施例1和实施例3所述粉剂进行检验,结果与实施例2结 果相当。
综上所述,本发明所述粉剂可以促进实验动物的生长,增加动物精子活 力,增进性反应时间与次数,提高性激素水平及性器官脏器系数比,作用与 阳性对照药相当,因而具有较好的壮阳功效。
Claims (10)
1.一种粉剂,其特征在于,由菊粉、龙眼肉、红枣、枸杞子、熟地黄、核桃仁、管花肉苁蓉、牛磺酸、五味子、和肉桂制备而成。
2.根据权利要求1所述的粉剂,其特征在于,所述菊粉、龙眼肉、红枣、枸杞子、熟地黄、核桃仁、管花肉苁蓉、牛磺酸、五味子和肉桂的质量比为(10~100):(1~10):(1~10):(1~10):(1~10):(1~10):(0.1~5):(0.1~10):(0.1~5):(0.1~5)。
3.根据权利要求1所述的粉剂,其特征在于,所述菊粉、龙眼肉、红枣、枸杞子、熟地黄、核桃仁、管花肉苁蓉、牛磺酸、五味子和肉桂的质量比为10:3:3:2:2:2:1:0.3:0.5:0.5。
4.根据权利要求1所述的粉剂,其特征在于,所述菊粉、龙眼肉、红枣、枸杞子、熟地黄、核桃仁、管花肉苁蓉、牛磺酸、五味子和肉桂的质量比为50:2.5:5:2.5:6.25:3.75:0.75:0.25:2.5:2.5。
5.根据权利要求1所述的粉剂,其特征在于,所述菊粉、龙眼肉、红枣、枸杞子、熟地黄、核桃仁、管花肉苁蓉、牛磺酸、五味子和肉桂的质量比为20:2.5:1:4:1:4:1.5:5:1:0.1。
6.权利要求1-5任意一项所述的粉剂的制备方法,其特征在于,将所述龙眼肉、红枣、枸杞子、熟地黄、核桃仁、管花肉苁蓉、五味子和肉桂加水提取,提取液浓缩,加入菊粉、牛磺酸后干燥、粉碎。
7.根据权利要求6所述的制备方法,其特征在于,所述的加水提取为加水煎煮提取。
8.根据权利要求7所述的制备方法,其特征在于,所述的加水煎煮提取为提取1-2次,每次加3~20倍的水提取0.5~3小时。
9.根据权利要求6所述的制备方法,其特征在于,所述浓缩为高温罐浓缩或反渗透浓缩;所述干燥为喷雾干燥、冷冻干燥、带式干燥、微波干燥、真空干燥。
10.权利要求1-5任意一项所述粉剂在制备具有壮阳功能的保健食品中的应用。
