Abstract
本發明屬於保健品領域,本發明提供了一種粉劑由菊粉、龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、牛磺酸、五味子、和肉桂製備而成,原料全部來源於天然中草藥,無輔料添加,成分天然,標籤清潔,符合人們追求食品天然健康的理念趨勢,且服用劑量小,服食方便,可直接口服或溶於水中服用,冷水可溶,入口即化,口感好,吸收快。本發明所述粉劑製備方法簡單,適合大規模生產攜帶方便,製得的粉劑穩定性好,存放時間長。實驗表明本發明所述粉劑可以促進實驗動物的生長,增加動物精子活力,增進性反應時間與次數,提高性激素水準及性器官臟器係數比,具有壯陽功能,效果顯著,可用於製備具有壯陽功能的保健食品。
Description
本發明屬於保健品領域,具體涉及一種粉劑及其製備方法與應用,尤其是以天然中草藥為主要原料製備的具有壯陽功能的保健粉劑。
陽虛體質就是指人體的陽氣不足,氣是構成人體和維持人體生命活動的最基本物質。陽氣不足就會產生一系列症狀,表現為疲倦怕冷,四肢冰冷、唇色蒼白,少氣懶言,嗜睡乏力,男性遺精,女性白帶清稀,易腹瀉,排尿次數頻繁,性欲衰退等。陽虛體質的人平素畏冷,手足不溫,易出汗;喜熱飲食,精神不振,睡眠偏多。
近年來人民生活水準明顯提高人們的消費觀念、健康觀念發生了較大變化。為規避不健康帶來的各種不利影響,使人們越來越重視營養保健品的使用。目前壯陽類保健食品主要以補充維生素、礦物質及補益類藥材為主,大多以口服液的形態存在。口服液劑攜帶不便,穩定性差,存放時間短。片劑在壓片時大都需要加入澱粉、羥甲基纖維素等大劑量輔料,崩解時間長。同時對吞咽有困難的人群服用不方便。且補益類藥材煎煮、攜帶、服用不便。
有鑑於此,本發明的目的在於針對現有技術存在的缺陷提供一種粉劑及其製備方法與應用。本發明所述粉劑以天然中草藥為主要原料製備,具有壯陽功能,同時不含輔料,成分天然,服用方便。
為實現本發明的目的,本發明採用如下技術方案:一種粉劑,由菊粉、龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、牛磺酸、五味子和肉桂製備而成。
其中,優選地,所述菊粉、龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、
管花肉蓯蓉、牛磺酸、五味子和肉桂的質量比為(10~100):(1~10):(1~10):(1~10):(1~10):(1~10):(0.1~5):(0.1~10):(0.1~5):(0.1~5)。
在一些實施方案中,所述菊粉、龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、牛磺酸、五味子和肉桂的質量比為10:3:3:2:2:2:1:0.3:0.5:0.5。
在一些實施方案中,所述菊粉、龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、牛磺酸、五味子和肉桂的質量比為50:2.5:5:2.5:6.25:3.75:0.75:0.25:2.5:2.5。
在另一些實施方案中,所述菊粉、龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、牛磺酸、五味子和肉桂的質量比為20:2.5:1:4:1:4:1.5:5:1:0.1。
本發明還提供了所述粉劑的製備方法,將所述龍眼肉、紅棗、核桃仁、枸杞子、熟地黃、五味子、肉桂和肉蓯蓉加水提取,提取液濃縮,加入菊粉、牛磺酸後乾燥、粉碎。
其中,所述的加水提取優選為加水煎煮提取。
進一步地,所述的加水煎煮提取優選為提取1-2次,每次加3~20倍的水提取0.5~3小時。
在一些實施方案中,所述加水煎煮提取為加20倍水煎煮提取3小時。
在一些實施方案中,所述加水煎煮提取為第一次加5倍~20倍的水提取0.5~3小時,第二次加3倍到15倍的水提取0.5~2小時。在一些實施例中,第一次加入15倍水提取2小時,第二次加入10倍水,提取1小時。在另一些實施例中,第一次加5倍水,提取1小時,第二次加5倍的水提取0.5小時。
本發明所述製備方法在加水提取後收集所有提取液,進行濃縮。所述濃縮優選為高溫罐濃縮或反滲透濃縮。
進一步地,本發明所述製備方法在濃縮後進行乾燥。所述乾燥優選為噴霧乾燥、冷凍乾燥、帶式乾燥、微波乾燥或真空乾燥。
優選地,本發明所述製備方法乾燥後還包括粉碎的步驟。所述粉碎細微性優選控制在40目到80目。
製粒,包裝。
在某一具體實施方案中,本發明藉由灌胃給予本發明所述粉劑和對照樣品,觀察本發明所述粉劑對雄性大鼠的影響,結果顯示,本發明所述粉劑可以促進實驗動物的生長,增加動物精子活力,增進性反應時間與次數,提高性激素水準及性器官臟器係數比。
因此,本發明提供了所述粉劑在製備具有壯陽功能的保健食品中的應用。

如無特殊說明,本發明實施例中所涉及的試劑均為市售產品,均可以藉由商業管道購買獲得。
配方:
製備方法:將龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、五味子、和肉桂加20倍水煎煮提取3小時,提取液濃縮,濃縮溫度控制在95℃-100℃,加入菊粉、牛磺酸進行噴霧乾燥,得噴乾粉,粉碎即得所述保健粉劑。
配方:
製備方法:將龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、五味子和 肉桂經水煎煮提取2次,第一次加入15倍水提取2小時,第二次加入10倍水,提取1小時,合併2次提取液,濃縮,濃縮溫度控制在95℃-100℃,加入菊粉、牛磺酸進行帶式乾燥,得帶乾粉,粉碎即得所述保健粉劑。
配方:
製備方法:將龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、五味子和肉桂經水煎煮提取2次,第一次加5倍水,提取1小時,第二次加5倍的水提取0.5小時,合併2次提取液,反滲透濃縮,濃縮溫度低於40℃,加入菊粉、牛磺酸進行冷凍乾燥,預凍溫度-30℃,維持3-4小時,乾燥,迅速升溫至60℃,保持1小時,隨後再緩慢升溫至80℃,保持10-12小時,得凍乾粉,粉碎即得所述保健粉劑。
1、材料與儀器
供試品:實施例2所述保健粉劑
試驗動物:SPF級雄性大鼠,適應期結束,體重200±20g。首選SD大鼠。每組8-10只。
將按體重隨機分為正常對照組,陽性對照組,供試品低、中、高劑量 組。適應性餵養一周後,每天一次,連續灌胃給予四周,正常對照組給予等體積生理鹽水。
供試品低、中、高劑量組的配製方法:供試品高劑量組(0.3g/ml的供試品溶液):精密稱取13.5g供試品,加45ml超純水,配置成濃度為0.3g/ml的溶液,即為供試品高劑量組(1.5g/kg,0.5ml/100g),相當於人體推薦量的30倍;供試品中劑量組(0.1g/ml的供試品溶液):從高劑量組取15ml加30ml超純水,即為供試品中劑量組(0.5g/kg,0.5ml/100g),相當於人體推薦量的10倍;供試品低劑量組(0.05g/ml的供試品溶液):從中劑量組取15ml加15ml超純水,即為供試品低劑量組(0.25g/kg,0.5ml/100g),相當於人體推薦量的5倍。
陽性對照品的配製方法:精密稱取東革阿裡藥材粉末100g至500ml燒瓶中,加去離子水提取3次,每次400ml提取2小時。提取液合併濃縮,定容相當於0.4g/ml(藥材/水)。精密量取6.625ml濃度為0.4g/ml的東革阿裡原液,加超純水至50毫升,配置成濃度為0.053g/ml的溶液,即為東革阿裡陽性對照組(31.8/3mg/kg,0.5ml/100g),相當於人體推薦量的5倍。
2、試驗方法
2.1 大鼠體重稱量
給藥前,將大鼠稱重後隨機分組。給藥後每週固定時間稱重一次,實驗結束繪製體重變化曲線。
2.2 各組大鼠陰莖勃起指數(PEI值)測定
適應性餵養5天後,每天一次,連續灌胃給藥四周,對照組給予等體積生理鹽水。每組動物每週進行兩次性行為學觀察。給藥完畢,每組動物分為3批,3-4只老鼠/籠,以雌鼠在籠外做誘導,人工觀察大鼠陰莖勃起時間(陰莖勃起潛伏期:從觀察時間開始,到陰莖首次勃起需要的時間,共測九次。第一次是給藥前測的,以後每週兩次,共四周給藥),並用攝像頭連續記錄90min內大鼠陰部理毛、爬背次數和參與動物數,並計算PEI值。

2.3 血清睪酮水準檢測
連續給藥四周後,眼角靜脈叢採血1.5ml,靜置1h,3000轉離心30min,取血清,ELISA試劑盒檢測血清睪酮水準。
2.4 精子活力評定及臟器係數比
用1%戊巴比妥鈉(0.2ml/100g)腹腔麻醉,下腹正中切口,提出睪丸,找出睪丸和附睪,分離出睪丸和附睪,稱重,然後用眼科剪剪下附睪,將一側附睪從中間剪斷,取附睪尾端,置於2ml PBS中(37℃,預溫),用眼科剪剪成3段,溫育10min,待附睪精子充分游離後顯微鏡下計數,計算精子活力:精子活力=(計數室總精子數-不能前進運動的總精子數)÷計數室總精子數。
最後將動物安樂死,再取精囊腺和提肛肌,將所有臟器稱重,計算臟器係數比。
2.4 結果
2.4.1體重增長曲線
給藥3周以後,供試品低劑量組體重增長略低於正常。中劑量組大鼠體重增加明顯,第四周後體重增長到339.4±16.9g,體重增長率達到119.8%,顯著高於正常組(體重329.8±21.7g,體重增長率93.8%)和東革阿裡組。其他組體重增長與正常組相比變化不大。

2.4.2 臟器係數比
給藥完畢,摘取各組大鼠的睪丸、附睪、提肛肌及精囊腺稱重,比較各組間臟器係數比。結果如表3所示。
結果顯示,各組間除睪丸臟器係數比無明顯差別外,陽性對照組及供試品各組均出現了臟器係數比值的增加,其中精囊腺的增加最為明顯。供試品中高劑量組作用與陽性對照組相當。
2.4.2 大鼠PEI值
給藥後,東革阿裡藥材及供試品組大鼠爬背、陰部理毛及陰莖勃起等性反應時間與強度均增強。如表4所示,東革阿裡藥材平均PEI值顯著高於空白組,供試品低、中、高劑量組平均PEI值均高於正常,與正常組比較,統計學處理有顯著性差異。與東革阿裡組比較無統計學差異。

2.4.3 陰莖勃起潛伏期
與給藥前相比,給藥2周後東革阿裡藥材及供試品組大鼠的陰莖勃起潛伏期均縮短(表5),部分組別供試品組效果強於陽性對照組。中劑量組表現尤其突出。
2.4.4 血清睪酮結果如圖4
血清睪酮檢測結果(表6)顯示,低、中、高劑量的供試品組均提高血清睪酮水準,其中低、中劑量組作用優於東革阿裡藥材。

2.4.5 精子活力結果
精子活力檢測結果如表7,供試品低、中、高劑量組均能增加雄性大鼠精子活力,具有劑量依賴性關係,其中高劑量組作用與陽性對照相當。
按照上述方法對實施例1和實施例3所述粉劑進行檢驗,結果與實施例2結果相當。
綜上所述,本發明所述粉劑可以促進實驗動物的生長,增加動物精子活力,增進性反應時間與次數,提高性激素水準及性器官臟器係數比,作用與陽性對照藥相當,因而具有較好的壯陽功效。

- 一種粉劑,其係由菊粉、龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、牛磺酸、五味子、和肉桂製備而成,其中所述菊粉、龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、牛磺酸、五味子和肉桂的質量比為(10~100):(1~10):(1~10):(1~10):(1~10):(1~10):(0.1~5):(0.1~10):(0.1~5):(0.1~5);所述粉劑由如下方法製備:將所述龍眼肉、紅棗、枸杞子、熟地黃、核桃仁、管花肉蓯蓉、五味子和肉桂加水提取,提取液濃縮,加入菊粉、牛磺酸後乾燥、粉碎;其中所述的加水提取為加水煎煮提取;所述粉劑具有壯陽功能。
