本发明公开基于金枪鱼活性提取物的复配物,成分包含金枪鱼活性提取物和免疫调节肽,免疫调节肽添加量为金枪鱼活性提取物重量的0.5~1wt%,金枪鱼活性提取物经脱腥、脱脂、中性蛋白酶水解制得。有益效果为:本发明基于金枪鱼活性提取物的复配物抗氧化活性强,能够清除人体的自由基、抑制油脂氧化,保护细胞遭到自由基的破坏,保护人体的机能;原料来源于金枪鱼下脚料,避免资源的浪费,提高低值鱼产品的附加值,属于纯天然的提取物,对人体健康不会造成损害;能调节免疫细胞的信号转导系统,调节人体免疫力,易于吸收消化,口感好、无腥味,可设计成全自动化操作,生产效率高。

本发明涉及鱼类产品深加工技术领域,尤其是涉及基于金枪鱼活性提取物的复配物及其制备方法。
背景技术
金枪鱼类属鲈形目鲭科又叫鲔鱼,华人世界又称为“吞拿(鱼)”。是大洋暖水性洄游鱼类,主要分布于低中纬度海区,在太平洋、大西洋、印度洋都有广泛的分布。我国东海、南海也有分布。同金枪鱼最相似的是鲣属鱼类,最简单的区分方法是鲣属腹部有4-6条黑色纵带,其它相近鱼种如舵鲣、狐鲣等有暗色纵带等。而金枪鱼类,鱼体无任何黑斑,或深色纵纹,金枪鱼腹鳍明显比舵鲣、狐鲣的长。蛋白质含量高达20%,但脂肪含量很低,俗称海底鸡,营养价值高。鱼肉中脂肪酸大多为不饱和脂肪酸,所含氨基酸齐全,人体所需8种氨基酸均有,还含有维生素、丰富的铁、钾、钙、镁、碘等多种矿物质和微量元素。目前,金枪鱼渔获物主要用于加工制作生鱼片,而加工过程产生的下脚料(如鱼鳞、鱼皮、鱼内脏、鱼头、暗色肉等)约占鱼总质量50-70%,这些下脚料中的胶原蛋白质含量高,却未被充分利用甚至被废弃。
鱼类活性提取物是从鱼中提取的一类纯天然营养物质,相对分子质量较小,容易被吸收,且吸收效率非常高,一方面提供人体生长发育所需的营养物质,另一方面对人体生命活动发挥着极其重要的作用,作为高档营养食品、特殊医学用途配方食品和保健食品基料,市场前景非常广阔。目前,中国金枪鱼加工水平较低,多以初加工形式输出给日本,下脚料作为加工的副产物大多被废弃,若能从将金枪鱼下脚料提取出活性物质并且分离纯化,不仅能开发新型的具有生物活性的产品,还能提高金枪鱼皮的利用效率。
现有技术如授权公告号为CN 103073621 B的中国发明专利,公开了一种金枪鱼碎肉蛋白抗氧化肽及其制备方法和用途,该抗氧化肽的氨基酸序列为Tyr-Glu-Asn-Gly-Gly,制备时以脱脂金枪鱼碎肉作为原料,加入磷酸盐缓冲液,调节pH到5.0~7.0,得混合液;将混合液搅拌预热,加入木瓜蛋白酶,得酶解产物;将酶解产物先灭酶再依次经脱盐、超滤和层析,得到抗氧化肽。该制备工艺科学合理,酶解过程易监控,制得的抗氧化肽具有安全无毒副作用、抗氧化活性强和易于消化吸收等优点,但该蛋白抗氧化钛口感较差。
发明内容
本发明的目的在于提供一种抗氧化活性强、提高人体免疫力,易于吸收消化,安全无毒副作用,口感好的基于金枪鱼活性提取物的复配物及其制备方法。
本发明针对上述技术中提到的问题,采取的技术方案为:基于金枪鱼活性提取物的复配物,成分包含金枪鱼活性提取物和鱼蛋白粉,金枪鱼活性提取物的添加量为鱼蛋白粉重量的5~10wt%。该复配物可用作保健食品的添加物质,不仅能够丰富保健品的营养物质,为机体提供其必需的蛋白质,而且易于吸收消化,安全无毒副作用,口感好、无腥味,能够提高机体免疫力,增强机体抵抗病毒害的损伤,原料来源于金枪鱼下脚料,避免资源的浪费,提高低值鱼产品的附加值。
基于金枪鱼活性提取物的复配物的制备方法,具体操作步骤为:
脱腥处理:将金枪鱼下脚料在0.5~1MPa下蒸煮5~10min,然后在微波功率为460~480W的条件下处理3~5min,然后用绞肉机搅碎得金枪鱼糜,高压蒸煮脱腥与微波脱腥相结合,可达到良好的效果好,可使下脚料色泽达到最佳,对营养成分无影响,且脱腥过程属于物理脱腥,操作简单方便,未用到化学试剂,不能产生对环境有影响的物质;
脱脂:将金枪鱼糜与乙醚以料液比为1:2~4(W/V)浸泡脱脂65~75h,然后用浓度为0.2~0.3mol/L的氢氧化钠溶液在2~5℃下脱杂蛋白2~4h,该步骤除去鱼糜的脂肪和杂蛋白,提高蛋白质的含量,降低杂质对活性提取物浓度的影响,提高提取物的纯度和得率,且该处理方法对提取物的成分及功效无任何影响;
金枪鱼活性提取物的制备:调pH至6~7,在温度为45~55℃下,添加900~1100U/g中性蛋白酶酶解3~5h,用截留分子量为5~10KDa的超滤膜在0.5~1MPa超滤分离,冷冻干燥,即得金枪鱼活性提取物,该提取物含有一种免疫调节肽,其氨基酸序列为HSHACASYYSVCGYYVSHRGRCYCRCLRCRVLHPGKLCVCVNCSR,免疫调节肽在机体内能增强免疫细胞的吞噬功能,具有体外刺激吞噬细胞吞噬绵羊红细胞的功能,达到提高机体免疫力的功效,且在机体消化道内不易被降解,可作为一个免疫因子调节免疫细胞的信号转导系统,调节人体免疫力;
复配物配制:按配方称取金枪鱼活性提取物和鱼蛋白粉,混合均匀即可得基于金枪鱼提取物的复配物,该复配物可用作保健食品的添加物质,不仅能够丰富保健品的营养物质,为机体提供其必需的蛋白质,而且易于吸收消化,安全无毒副作用,口感好、无腥味,能够提高机体免疫力,增强机体抵抗病毒害的损伤。
与现有技术相比,本发明的优点在于:1)本发明基于金枪鱼活性提取物的复配物能增强免疫细胞的吞噬功能,具有体外刺激吞噬细胞吞噬绵羊红细胞的功能,达到提高机体免疫力的功效;2)在机体消化道内不易被降解,可作为一个免疫因子调节免疫细胞的信号转导系统,调节人体免疫力;3)原料来源于金枪鱼下脚料,避免资源的浪费,提高低值鱼产品的附加值,属于纯天然的提取物,对人体健康不会造成损害,可作为食品中添加的抗氧化剂;4)易于吸收消化,安全无毒副作用,口感好、无腥味,可设计成全自动化操作,生产效率高。
具体实施方式
下面通过实施例对本发明方案作进一步说明:
实施例1:
基于金枪鱼活性提取物的复配物,成分包含金枪鱼活性提取物和鱼蛋白粉,金枪鱼活性提取物的添加量为鱼蛋白粉重量的8wt%。
基于金枪鱼活性提取物的复配物的制备方法,具体操作步骤为:
1)脱腥处理:将金枪鱼下脚料在0.6MPa下蒸煮8min,然后在微波功率为460W的条件下处理4min,然后用绞肉机搅碎得金枪鱼糜,高压蒸煮脱腥与微波脱腥相结合,可达到良好的效果好,可使下脚料色泽达到最佳,对营养成分无影响,且脱腥过程属于物理脱腥,操作简单方便,未用到化学试剂,不能产生对环境有影响的物质;
2)脱脂:将金枪鱼糜与乙醚以料液比为1:3(W/V)浸泡脱脂70h,然后用浓度为0.25mol/L的氢氧化钠溶液在3℃下脱杂蛋白3.5h,该步骤除去鱼糜的脂肪和杂蛋白,提高蛋白质的含量,降低杂质对活性提取物浓度的影响,提高提取物的纯度和得率,且该处理方法对提取物的成分及功效无任何影响;
3)金枪鱼活性提取物的制备:调pH至7,在温度为50℃下,添加1100U/g中性蛋白酶酶解3h,用截留分子量为6KDa的超滤膜在0.8MPa超滤分离,冷冻干燥,即得金枪鱼活性提取物,该提取物含有一种免疫调节肽,其氨基酸序列为HSHACASYYSVCGYYVSHRGRCYCRCLRCRVLHPGKLCVCVNCSR,免疫调节肽在机体内能增强免疫细胞的吞噬功能,具有体外刺激吞噬细胞吞噬绵羊红细胞的功能,达到提高机体免疫力的功效,且在机体消化道内不易被降解,可作为一个免疫因子调节免疫细胞的信号转导系统,调节人体免疫力;

1)脱腥处理:将金枪鱼下脚料在1MPa下蒸煮5min,然后在微波功率为470W的条件下处理3min,然后用绞肉机搅碎得金枪鱼糜,高压蒸煮脱腥与微波脱腥相结合,可达到良好的效果好,可使下脚料色泽达到最佳,对营养成分无影响,且脱腥过程属于物理脱腥,操作简单方便,未用到化学试剂,不能产生对环境有影响的物质;
2)脱脂:将金枪鱼糜与乙醚以料液比为1:2(W/V)浸泡脱脂65h,然后用浓度为0.2mol/L的氢氧化钠溶液在3℃下脱杂蛋白4h,该步骤除去鱼糜的脂肪和杂蛋白,提高蛋白质的含量,降低杂质对活性提取物浓度的影响,提高提取物的纯度和得率,且该处理方法对提取物的成分及功效无任何影响;
3)金枪鱼活性提取物的制备:调pH至7,在温度为55℃下,添加1000U/g中性蛋白酶酶解3h,用截留分子量为7KDa的超滤膜在1MPa超滤分离,冷冻干燥,即得金枪鱼活性提取物,该提取物含有一种免疫调节肽,其氨基酸序列为HSHACASYYSVCGYYVSHRGRCYCRCLRCRVLHPGKLCVCVNCSR,免疫调节肽在机体内能增强免疫细胞的吞噬功能,具有体外刺激吞噬细胞吞噬绵羊红细胞的功能,达到提高机体免疫力的功效,且在机体消化道内不易被降解,可作为一个免疫因子调节免疫细胞的信号转导系统,调节人体免疫力;

1.基于金枪鱼活性提取物的复配物,其特征在于:所述的复配物包含金枪鱼活性提取物和鱼蛋白粉。
2.根据权利要求1所述的基于金枪鱼活性提取物的复配物,其特征在于:所述的复配物中金枪鱼活性提取物的添加量为鱼蛋白粉重量的5~10wt%。
3.根据权利要求1所述的一种低值鱼蛋白钙肽的加工方法,其特征在于:所述的金枪鱼活性提取物含有一种免疫调节肽,其氨基酸序列为HSHACASYYSVCGYYVSHRGRCYCRCLRCRVLHPGKLCVCVNCSR。
4.基于金枪鱼活性提取物的复配物的制备方法,其特征在于:所述的制备方法的具体操作步骤为:
1)脱腥处理:将金枪鱼下脚料在蒸煮,然后微波处理;
2)脱脂:将金枪鱼糜与乙醚浸泡脱脂,然后用氢氧化钠溶液脱杂蛋白;
3)金枪鱼活性提取物的制备:添加中性蛋白酶酶解,超滤分离,冷冻干燥;
4)复配物配制:按配方称取金枪鱼活性提取物和免疫调节肽,混合均匀。
5.根据权利要求4所述的基于金枪鱼活性提取物的复配物的制备方法,其特征在于:所述的脱腥处理步骤中蒸煮压力为0.5~1MPa,时间为5~10min;微波功率为460~480W,时间为3~5min。
6.根据权利要求4所述的基于金枪鱼活性提取物的复配物的制备方法,其特征在于:所述的脱脂步骤中金枪鱼糜与乙醚的重量体积比为1: 2~4,脱脂65~75h。
7.根据权利要求4所述的基于金枪鱼活性提取物的复配物的制备方法,其特征在于:所述的脱脂步骤中氢氧化钠溶液的浓度为0.2~0.3mol/L,脱杂蛋白温度为2~5℃,时间为2~4h。
8.根据权利要求4所述的基于金枪鱼活性提取物的复配物的制备方法,其特征在于:所述的金枪鱼活性提取物的制备步骤中中性蛋白酶的添加量为900~1100U/g。
9.根据权利要求4所述的基于金枪鱼活性提取物的复配物的制备方法,其特征在于:所述的金枪鱼活性提取物的制备步骤中酶解pH为6~7,温度为45~55℃,时间为3~5h。
10.根据权利要求4所述的基于金枪鱼活性提取物的复配物的制备方法,其特征在于:所述的金枪鱼活性提取物的制备步骤中超滤膜的截留分子量为5~10KDa,超滤压力为0.5~1MPa。
