背景技术
糖尿病是是由于胰岛素分泌缺陷引起的以高血糖为特征的代谢紊乱性疾病,是一种慢性终身的代谢疾病。糖尿病伴随的慢性高血糖会对人体的组织器官,尤其是眼睛、肾脏、神经、心脏和心血管等组织,造成长期的损害,引起功能障碍。目前,用于治疗糖尿病的药物,多有不良反应,且对糖尿病并发症预防和治疗效果也不理想。
随着海洋药物的兴起,海藻活性物质的研究已成为国内外研究的热点,特别是海藻多糖其含量丰富,具有多种生理活性,作为天然的高分子化合物在控制血糖和慢性并发症发展方面,具有显著效果和优势。海藻中浒苔、马尾藻、墨角藻、裙带菜、麒麟菜等均具有降血糖活性,能有效调节胰岛素含量及其敏感性。但海藻类食物,通常腥味浓郁,很多人接受不了。

一种含浒苔改善糖尿病的保健食品,包括如下重量份数的组分:
浒苔 15-20份, 鹿角菜 10—15份, 裙带菜 10—15份, 马尾藻 5—10份,
墨角藻 5—10份, 麒麟菜 20-40份, 苦瓜 15-20份, 芡实 10—15份,
冬麦 10—15份, 杞子 10—20份, 核桃 2—5份, 薏仁 2—5份, 生地 5—10份。
上述含浒苔改善糖尿病的保健食品,制备方法包括如下步骤:
步骤1,取浒苔、鹿角菜、裙带菜、马尾藻、墨角藻、麒麟菜,用清水洗涤3次,以除去其中的盐分等杂物, 60℃烘干备用;称取15-20份浒苔、10-15份鹿角菜、10-15份裙带菜、5-10份马尾藻、5-10份墨角藻、15-20份苦瓜、10-15份芡实、10-15份冬麦、10-20份杞子、2-5份核桃、2-5份薏仁、5-10份生地,分别超微粉碎,过80目筛;
步骤2,取步骤1制备的浒苔、鹿角菜、裙带菜、马尾藻、墨角藻超微粉末混合,上述混合粉末与蒸馏水按重量体积比g/mL为1:9混合,浸泡2h后于100℃煎煮30min,过滤取上清滤液,滤渣依照上述步骤操作2次,合并滤液,抽滤浓缩成海藻水提物浸膏,冷冻干燥后粉碎过80目筛,制备得到海藻复合物;
步骤3,取步骤1制备的苦瓜、芡实、冬麦、杞子、核桃、薏仁、生地超微粉末混合,上述混合粉末与蒸馏水按重量体积比g/mL为1:9混合,于100℃条件下煎煮30min,过滤取上清滤液,滤渣依照上述步骤操作2次,合并滤液,抽滤浓缩成其水提物浸膏,冷冻干燥后粉碎过80目筛,制备得到陆源的复合物;
步骤4,将步骤2制备得到的海藻复合物和步骤3制备得到的陆源的复合物按照重量比1:1均匀混合后,按混合物与蒸馏水重量体积比g/mL为1:4混合均匀,制成复合物浸膏;
步骤5,取步骤1中处理好的麒麟菜20-40份,切割成碎块,投入质量浓度为10%的NaOH溶液中,麒麟菜和碱液的重量比为1: 9,浸泡24小时,并每隔8小时搅拌一次;
步骤6,将经步骤5处理得到的麒麟菜捞起,挤干碱液,用清水充分浸洗直到洗出水接近中性,除去杂质,麒麟菜沥干水份,按麒麟菜与蒸馏水重量体积比g/mL为1:20混合,于100℃煎煮60min,趁热用80目分样筛过滤除去残渣,冷却至室温;
步骤7,将步骤6中产物加入4倍体积的95%乙醇进行沉淀,胶液内容物交联,过滤得到麒麟菜凝胶;
步骤8,将步骤7制备的凝胶中加入步骤4制备的混合的复合物浸膏,等体积混匀,制成软材,后制成胶囊剂、颗粒剂、片剂、丸剂或溶液剂。
本发明的优点在于:
(1)本发明提供的功能食品是海源与陆源药食两用材料的有效提取物复配制备而成,利用其中各成分的协同作用,制成胶囊剂、颗粒剂、片剂、丸剂或溶液剂等剂型。经药效学验证,此功能食品具有改善胰岛素抵抗的功效,能够降低糖尿病空腹血糖值,预防和辅助治疗糖尿病。
(2)此发明也为海藻产品的多元化开发提供了新的思路。所用原料均为天然物质及其生物活性成分,制备条件温和,最大限度地保留了原料的有效成分及营养物质,无副作用,健康安全,极具开发空间。
本发明以浒苔、鹿角菜、裙带菜、马尾藻、墨角藻、麒麟菜、苦瓜、冬麦、杞子、核桃、芡实、薏仁、生地的提取物为主的保健食品,经药效学验证,可显著降糖尿病空腹血糖值,预防和辅助治疗糖尿病。海源和陆源食材中功效成分的复配可产生协同及增效作用,营养互补,降血糖效力得以协同增效,风味得以相互掩蔽、优化和加强,进一步改善食品的品质。
具体实施方式
实施例1
(1)、取浒苔、鹿角菜、裙带菜、马尾藻、墨角藻、麒麟菜,用清水洗涤3次,以除去其中的盐分等杂物, 60℃烘干备用;称取15份浒苔、10份鹿角菜、10份裙带菜、5份马尾藻、5份墨角藻、15份苦瓜、10份芡实、10份冬麦、10份杞子、2份核桃、2份薏仁、5份生地分别超微粉碎,过80目筛;
(2)、取步骤(1)制备的浒苔、鹿角菜、裙带菜、马尾藻、墨角藻超微粉末混合,上述混合粉末与蒸馏水按重量体积比g/mL为1:9混合,浸泡2h后于100℃煎煮30min,过滤取上清滤液,滤渣依照上述步骤操作2次,合并滤液,抽滤浓缩成海藻水提物浸膏,冷冻干燥后粉碎过80目筛,制备得到海源的海藻复合物;
(3)、取步骤(1)制备的苦瓜、芡实、冬麦、杞子、核桃、薏仁、生地超微粉末混合,上述混合粉末与蒸馏水按重量体积比g/mL为1:9混合,于100℃条件下煎煮30min,过滤取上清滤液,滤渣依照上述步骤操作2次,合并滤液,抽滤浓缩成其水提物浸膏,冷冻干燥后粉碎过80目筛,制备得到陆源的复合物;
(4)、将步骤(2)制备得到的海源的海藻复合物和步骤(3)制备得到的陆源的复合物按照重量比1:1均匀混合后,按混合物与蒸馏水重量体积比g/mL为1:4混合均匀,制成其复合物浸膏;
(5)、取步骤(1)中处理好的麒麟菜取20份,切割成碎块,投入质量浓度为10%的NaOH溶液中,麒麟菜和碱液的重量比为1: 9,浸泡24小时,并每隔8小时搅拌一次;
(6)、将经步骤5处理得到的麒麟菜捞起,挤干碱液,用清水充分浸洗直到洗出水接近中性,除去杂质,麒麟菜沥干水份,按麒麟菜与蒸馏水重量体积比g/mL为1:20混合,于100℃煎煮60min,趁热用80目分样筛过滤除去残渣,冷却至室温;
(7)、将步骤(6)中产物加入4倍体积的95%乙醇进行沉淀,胶液内容物交联,过滤得到凝胶;
(8)、将步骤(7)制备的凝胶中加入步骤(4)制备的混合的复合物浸膏,等体积混匀,制成软材,后制成胶囊剂、颗粒剂、片剂、丸剂或溶液剂。
实施例2
(1)取浒苔、鹿角菜、裙带菜、马尾藻、墨角藻、麒麟菜,用清水洗涤3次,以除去其中的盐分等杂物, 60℃烘干备用;称取20份浒苔、15份鹿角菜、15份裙带菜、6份马尾藻、6份墨角藻、20份苦瓜、12份芡实、12份冬麦、10份杞子、3份核桃、3份薏仁、6份生地分别超微粉碎,过80目筛;
(2)取步骤(1)制备的浒苔、鹿角菜、裙带菜、马尾藻、墨角藻超微粉末混合,按粉末与蒸馏水重量体积比g/mL为1:9,浸泡2h后于100℃煎煮30min,过滤取上清液,滤渣依照上述步骤操作2次,合并煎液,抽滤浓缩成海藻水提物浸膏,冷冻干燥后粉碎过80目筛,制备得到海源的海藻复合物;
(3)取步骤(1)制备的苦瓜、芡实、冬麦、杞子、核桃、薏仁、生地超微粉末混合,按粉末与蒸馏水重量体积比g/mL为1:9,于100℃条件下煎煮30min,过滤取上清液,滤渣依照上述步骤操作2次,合并煎液,抽滤浓缩成其水提物浸膏,冷冻干燥后粉碎过80目筛,制备得到陆源的复合物;
(4)将步骤(2)制备得到的海源的海藻复合物和步骤(3)制备得到的陆源的复合物按照重量比1:1均匀混合后,按混合物与蒸馏水重量体积比g/mL为1:4混合均匀,制成其复合物浸膏;
(5)取步骤(1)中处理好的30份麒麟菜,切割成碎块,投入质量浓度为10%的NaOH溶液中,麒麟菜和碱液的重量比为1: 9,浸泡24小时,并每隔8小时搅拌一次;
(6)将步骤(5)处理得到的麒麟菜,按麒麟菜藻体与蒸馏水重量体积比g/mL为1:20,100℃煎煮60min,趁热用80目分样筛过滤除去残渣,冷却至室温;
(7)将步骤(6)中产物加入4倍体积的95%乙醇进行沉淀,胶液内容物交联,过滤得到凝胶;
(8)将步骤(7)制备的凝胶中加入步骤(4)混合的复合物浸膏,等体积混匀,制成软材,后制成胶囊剂、颗粒剂、片剂、丸剂或溶液剂。
实施例3
(1)取浒苔、鹿角菜、裙带菜、马尾藻、墨角藻、麒麟菜,用清水洗涤3次,以除去其中的盐分等杂物, 60℃烘干备用;称取18份浒苔、12份鹿角菜、12份裙带菜、5份马尾藻、5份墨角藻、20份苦瓜、10份芡实、10份冬麦、10份杞子、5份核桃、5份薏仁、10份生地分别超微粉碎,过80目筛;
(2)取步骤(1)制备的浒苔、鹿角菜、裙带菜、马尾藻、墨角藻超微粉末混合,按粉末与蒸馏水重量体积比g/mL为1:9混合,浸泡2h后于100℃煎煮30min,过滤取上清滤液,滤渣依照上述步骤操作2次,合并滤液,抽滤浓缩成海藻水提物浸膏,冷冻干燥后粉碎过80目筛,制备得到海源的海藻复合物;
(3)取步骤(1)制备的苦瓜、芡实、冬麦、杞子、核桃、薏仁、生地超微粉末混合,按粉末与蒸馏水重量体积比g/mL为1:9混合,于100℃条件下煎煮30min,过滤取上清滤液,滤渣依照上述步骤操作2次,合并滤液,抽滤浓缩成其水提物浸膏,冷冻干燥后粉碎过80目筛,制备得到陆源的复合物;
(4)将步骤(2)制备得到的海源的海藻复合物和步骤(3)制备得到的陆源的复合物按照重量比1:1均匀混合后,按混合物与蒸馏水重量体积比g/mL为1:4混合均匀,制成其复合物浸膏;
(5)取步骤(1)中处理好的麒麟菜40份,切割成碎块,投入质量浓度为10%的NaOH溶液中,麒麟菜和碱液的重量比为1: 9,浸泡24小时,并每隔8小时搅拌一次;
(6)将步骤(5)处理得到的麒麟菜,按麒麟菜藻体与蒸馏水重量体积比g/mL为1:20混合,100℃煎煮60min,趁热用80目分样筛过滤除去残渣,冷却至室温;
(7)将步骤(6)中产物加入4倍体积的95%乙醇进行沉淀,胶液内容物交联,过滤得到凝胶;
(8)将步骤(7)制备的凝胶中加入步骤(4)混合的复合物浸膏,等体积混匀,制成软材,后制成胶囊剂、颗粒剂、片剂、丸剂或溶液剂。
实验:四氧嘧啶高血糖模型小鼠降血糖试验
用健康成年雄性ICR小鼠建立高血糖模型。造型前随机取15只小鼠禁食3h测血糖,作为基础血糖。小鼠禁食24h(自由饮水)后,尾静脉注射四氧嘧啶75mg/kg,5天后禁食3h筛选血糖值大于10mmol/L的小鼠45只,按血糖水平随机分为1个阳性药物对照组、1个高血糖模型对照组和1个本发明功能食品组,每组15只,功能食品组给予150mg/kg的受试物,阳性药物对照组给予二甲双胍,高血糖模型对照组给予蒸馏水,连续灌胃28天,测空腹血糖值(禁食同试验前),比较各组动物血糖值,观察复合物对四氧嘧啶高血糖模型小鼠空腹血糖的影响。

本发明的功能食品对四氧嘧啶高血糖模型小鼠空腹血糖的影响:造模前随机取15只动物测空腹血糖为5.6±1.3mmol/L,由表1可见,造模后各组动物空腹血糖明显升高,与造模前比较差异有显著性(P<0.01),说明模型成立。给予150mg/kg本发明功能食品28天,功能食品组小鼠空腹血糖显著低于模型组(P<0.01),说明本发明功能食品对四氧嘧啶高血糖模型小鼠有降低空腹血糖作用。
上述实施例和实验仅作为对本发明的说明,凡依本发明申请专利范围所做的均等变化与修饰,皆应属本发明的涵盖范围。
Claims (2)
1.一种含浒苔改善糖尿病的保健食品,其特征是:包括如下重量份数的组分:
浒苔 15-20份, 鹿角菜 10—15份, 裙带菜 10—15份, 马尾藻 5—10份,
墨角藻 5—10份, 麒麟菜 20-40份, 苦瓜 15-20份, 芡实 10—15份,
冬麦 10—15份, 杞子 10—20份, 核桃 2—5份, 薏仁 2—5份, 生地 5—10份。
2.根据权利要求1所述一种含浒苔改善糖尿病的保健食品,其制备方法包括如下步骤:
步骤1,取浒苔、鹿角菜、裙带菜、马尾藻、墨角藻、麒麟菜,用清水洗涤3次,以除去其中的盐分等杂物, 60℃烘干备用;称取15-20份浒苔、10-15份鹿角菜、10-15份裙带菜、5-10份马尾藻、5-10份墨角藻、15-20份苦瓜、10-15份芡实、10-15份冬麦、10-20份杞子、2-5份核桃、2-5份薏仁、5-10份生地,分别超微粉碎,过80目筛;
步骤2,取步骤1制备的浒苔、鹿角菜、裙带菜、马尾藻、墨角藻超微粉末混合,上述混合粉末与蒸馏水按重量体积比g/mL为1:9混合,浸泡2h后于100℃煎煮30min,过滤取上清滤液,滤渣依照上述步骤操作2次,合并滤液,抽滤浓缩成海藻水提物浸膏,冷冻干燥后粉碎过80目筛,制备得到海藻复合物;
步骤3,取步骤1制备的苦瓜、芡实、冬麦、杞子、核桃、薏仁、生地超微粉末混合,上述混合粉末与蒸馏水按重量体积比g/mL为1:9混合,于100℃条件下煎煮30min,过滤取上清滤液,滤渣依照上述步骤操作2次,合并滤液,抽滤浓缩成其水提物浸膏,冷冻干燥后粉碎过80目筛,制备得到陆源的复合物;
步骤4,将步骤2制备得到的海藻复合物和步骤3制备得到的陆源的复合物按照重量比1:1均匀混合后,按混合物与蒸馏水重量体积比g/mL为1:4混合均匀,制成复合物浸膏;
步骤5,取步骤1中处理好的麒麟菜20-40份,切割成碎块,投入质量浓度为10%的NaOH溶液中,麒麟菜和碱液的重量比为1: 9,浸泡24小时,并每隔8小时搅拌一次;
步骤6,将经步骤5处理得到的麒麟菜捞起,挤干碱液,用清水充分浸洗直到洗出水接近中性,除去杂质,麒麟菜沥干水份,按麒麟菜与蒸馏水重量体积比g/mL为1:20混合,于100℃煎煮60min,趁热用80目分样筛过滤除去残渣,冷却至室温;
