Abstract
本发明涉及归脾制剂在提高乙醇脱氢酶和乙醛脱氢酶活性中的应用,属于中药技术领域。该归脾制剂能有效通过提高乙醇脱氢酶和乙醛脱氢酶活性,从而加快乙醇的代谢,达到解酒护肝的目的,减轻乙醇对人体的毒害作用,该制剂药食两用,可以长期服用,对身体无毒副作用。

技术领域
本发明涉及中药技术领域,尤其涉及一种归脾制剂在提高乙醇脱氢酶和乙醛脱氢酶活性中的应用。
技术背景
随着社会经济的发展和人们生活水平的提高,以及酒文化在我国的影响,饮酒在我国已经非常普遍,但大量的饮酒可引起恶心、呕吐、记忆力减退,注意力不集中,严重时甚至可因呼吸肌麻痹导致死亡。长期饮酒可引起精神障碍,胃溃疡以及酒精肝、脂肪肝、肝硬化等疾病,大量饮酒已经成为当今社会影响人类健康的重要因素之一。解酒可有多个方式,一是用胃肠吸收抑制剂抑制乙醇的胃肠吸收,加快其在胃肠道的流过速度,从而降低血液中的乙醇浓度;另一方面用代谢增强剂使药物直接作用于肝代谢酶系,提高肝内乙醇脱氢酶(ADH)、乙醛脱氢酶(ALDH)的活力,加速乙醇及其代谢产物的消除速率,减轻其对组织和细胞的损害。
目前市场上的解酒药的成分多数只含有兴奋剂、维生素与氨基酸等成分,提供的只是安慰、清醒和缓解头痛的一些作用,而无实际解酒效用或效果不明显,另外市场上还有一些解酒产品主要是由葛花、葛根、灵芝、板蓝根等中药材料制成,主要的作用机理是通过抗氧化清除自由基的保肝作用,在体内吸收后有可能加快乙醇在体内的代谢,因此作用比较缓慢。市场上的解酒药,大多是在醉酒后服用,其实是一种下策,因为此时过量的酒精已经进入肝脏,令肝脏降低肝醣的合成,促进脂肪积聚,就会形成脂肪肝,对人体造成了伤害。
乙醇脱氢酶(ADH)和乙醛脱氢酶(ALDH)是代谢乙醇过程中的主要解酒酶,人饮酒后,大部分乙醇在乙醇脱氢酶的作用下氧化分解为乙醛,再经过乙醛脱氢酶转化为乙酸并与辅酶A反应生成乙酰辅酶A,最后经三羧酸循环被氧化成二氧化碳和水。解酒酶活性的变化可以影响到血中乙醇的浓度。若该酶活性下降或受到抑制,可使机体对乙醇的生物利用度提高,进而加重乙醇对脑、肝脏等器官的危害作用,提高解酒酶的活性可缓解乙醇对机体的毒害作用。
发明内容
本发明的目的涉及一种归脾制剂在提高乙醇脱氢酶和乙醛脱氢酶活性中的应用,能有效通过提高乙醇脱氢酶和乙醛脱氢酶活性,从而加快乙醇的代谢,达到解酒护肝的目的,减轻乙醇对人体的毒害作用。该制剂药食两用,可以长期服用,对身体无毒副作用。
该制剂为归脾制剂,本发明为提供该归脾制剂的一种新用途,即归脾制剂在提高乙醇脱氢酶和乙醛脱氢酶活性中的应用。
本发明实施例提供如下技术方案:
归脾制剂在提高乙醇脱氢酶和乙醛脱氢酶活性中的应用,其特征在于所述归脾制剂的药效成分由下列重量比的组份制得,党参70-280g,炒制白术140-560g,蜜炙黄芪70-280g,蜜炙甘草35-140g,茯苓140-560g,制远志140-560g,炒制酸枣仁70-280g,龙眼肉140-560g,当归140-560g,木香35-140g,去核大枣35-140g。
党参140g,炒制白术280g,蜜炙黄芪140g,蜜炙甘草70g,茯苓280g,制远志280g,炒制酸枣仁140g,龙眼肉280g,当归280g,木香70g,去核大枣70g。
归脾制剂为固体制剂或液体制剂。
所述固体制剂包括胶囊剂、颗粒剂、丸剂、片剂或软胶囊剂,所述液体制剂为口服液。
所述归脾制剂的药效成分为其组分提取物。
所述归脾制剂的制备方法为:党参、炒制白术、蜜炙黄芪、蜜炙甘草、茯苓、制远志、炒制酸枣仁、龙眼肉、当归、木香、去核大枣粉碎或切碎成颗粒,用水微沸动态回流提取两次,合并药液,滤过,滤液浓缩至50℃时相对密度1.05—1.15的浸膏,滤过,滤液加入辅料,制成制剂。
所述制备方法中所述辅料选自:水,糊精,蜂蜜,蔗糖,乳糖,甘草酸,甜菊苷,山梨酸,苯甲酸,苯甲酸钠,尼泊金甲酯,尼泊金乙酯,尼泊金丙酯中的一种或几种。
本发明的归脾制剂由党参、白术、黄芪(蜜炙)、甘草(蜜炙)、茯苓、远志、酸枣仁、龙眼肉、当归、木香、大枣组成;党参补中益气、健脾益肺;白术、茯苓均可健脾除湿,常配伍使用;黄芪补气固表、利尿托毒、排脓;当归补血活血;木香行气止痛;远志、酸枣仁、龙眼肉共奏补益心脾、养血安神、养肝益肝的疗效;大枣补中益气、养血安神。实验表明,该制剂可以提高乙醇脱氢酶和乙醛脱氢酶的活性,从而加快乙醇的代谢,降低乙醇对人体的毒害作用。
该制剂的使用方法为,于饮酒前30分钟喝下,连续服用一周效果更佳。
具体实施方式
下面结合实施例对本发明做进一步的详细说明。
1、制备归脾制剂
实施例1:
以上十一味,粉碎或切碎成适宜颗粒,用水微沸动态回流提取二次,第一次加10倍量水提取1小时,第二次加5倍量水提取0.5小时,合并药液,滤过。滤液减压浓缩至相对密度1.05-1.15(50℃)的浸膏,滤过,滤液加入糊精适量,混匀,制成颗粒1000g,即得。
实施例2:
以上十一味,粉碎或切碎成适宜颗粒,用水微沸动态回流提取二次,第一次加10倍量水提取1小时,第二次加5倍量水提取0.5小时,合并药液,滤过。滤液减压浓缩至相对密度1.05-1.15(50℃)的浸膏,滤过,滤液加入糊精适量,混匀,制成颗粒1000g,即得。
实施例3:
以上十一味,粉碎或切碎成适宜颗粒,用水微沸动态回流提取二次,第一次加10倍量水提取1小时,第二次加5倍量水提取0.5小时,合并药液,滤过。滤液减压浓缩至相对密度1.05-1.15(50℃)的浸膏,滤过,滤液加入适量的水、蜂蜜、糊精、混匀,泛制成1000丸,干燥即得。
2、药效试验
归脾制剂对乙醇脱氢酶和乙醛脱氢酶活性的影响
一.实验动物
ICR小鼠,体重20~22g
二、实验方法
ICR小鼠60只,雌雄各半,随机分为4组,即模型对照组、实施例1高、低剂量组、阳性对照组。实施例1高、低剂量组将实施例1所得解酒制剂按照给药剂量进行灌胃(给药剂量:实施例1低剂量组1.5g/kg;实施例1高剂量组3g/kg);模型对照组灌胃等体积的生理盐水,连续给药一周;阳性对照组以海王金樽给药,给药剂量为0.7g/kg,仅在解剖当天进行给药。各实验组最后一天给药30min后,均按0.25ml/20g灌胃56%二锅头酒,观察小鼠的活动情况(以小鼠翻正反射完全消失作为醉酒指标,计算各组小鼠的平均睡眠潜伏期),灌酒50min后,解剖各组小鼠取肝脏,匀浆,按试剂盒说明书进行乙醇脱氢酶(ADH)和乙醛脱氢酶(ALDH)测定。
三、实验结果

睡眠潜伏时间:实施例1高、低剂量组睡眠潜伏时间均明显高于模型对照组,说明本发明归脾制剂可通过加快乙醇在体内的代谢,缓解醉酒症状,具有一定的解酒功效。
乙醇脱氢酶(ADH)、乙醛脱氢酶(ALDH):实施例1高、低剂量组的乙醇脱氢酶、乙醛脱氢酶活性均显著高于模型对照组,说明归脾制剂在提高乙醇脱氢酶、乙醛脱氢酶活性的应用方面具有积极的效果。
ADH和ALDH是机体乙醇代谢关键酶,乙醇经ADH催化氧化成乙醛,乙醛再经过ALDH催化氧化成乙酸,后者以酰CoA的形式进入三羧酸循环,最后氧化成CO2和H2O排出体外。由上表可知,实施例1高、低剂量组、阳性对照组的乙醇脱氢酶和乙醛脱氢酶活性显著高于模型组。从上述数据分析,本发明制剂具有提高解酒酶活性的作用,可通过提高解酒酶活性从而加快乙醇代谢,从而达到解酒护肝的作用。
3、临床资料
以下通过临床试验例进一步阐述本发明制剂的有益效果。
①实验人群:选取经常饮酒的男性60名,年龄30-60岁
②服用方法:口服实施例1所述方法制备的归脾制剂9g,于饮酒前30min服用,连续服用一周。
③疗效评定:
无效:服用归脾制剂后饮酒,醉酒症状无缓解或效果不明显,饮酒后恶心、呕吐、头晕、睡眠不好等症状未得到改善。
有效:服用归脾制剂后饮酒,醉酒症状得到缓解,饮酒后恶心、呕吐、头晕、睡眠不好等症状得到改善。
④治疗结果见下表:

1.归脾制剂在提高乙醇脱氢酶和乙醛脱氢酶活性中的应用,其特征在于所述归脾制剂的药效成分由下列重量比的组份制得,党参70-280g,炒制白术140-560g,蜜炙黄芪70-280g,蜜炙甘草35-140g,茯苓140-560g,制远志140-560g,炒制酸枣仁70-280g,龙眼肉140-560g,当归140-560g,木香35-140g,去核大枣35-140g。
2.根据权利要求1所述的应用,其中党参140g,炒制白术280g,蜜炙黄芪140g,蜜炙甘草70g,茯苓280g,制远志280g,炒制酸枣仁140g,龙眼肉280g,当归280g,木香70g,去核大枣70g。
3.根据权利要求1所述的应用,所述归脾制剂为固体制剂或液体制剂。
4.根据权利要求3所述的应用,所述固体制剂包括胶囊剂、颗粒剂、丸剂、片剂或软胶囊剂,所述液体制剂为口服液。
5.根据权利要求1所述的应用,所述归脾制剂的药效成分为其组分提取物。
6.根据权利要求5所述的应用,所述归脾制剂的制备方法为:党参、炒制白术、蜜炙黄芪、蜜炙甘草、茯苓、制远志、炒制酸枣仁、龙眼肉、当归、木香、去核大枣粉碎或切碎成颗粒,用水微沸动态回流提取两次,合并药液,滤过,滤液浓缩至50℃时相对密度1.05—1.15的浸膏,滤过,滤液加入辅料,制成制剂。
7.根据权利要求6所述的应用,所述制备方法中所述辅料选自:水,糊精,蜂蜜,蔗糖,乳糖,甘草酸,甜菊苷,山梨酸,苯甲酸,苯甲酸钠,尼泊金甲酯,尼泊金乙酯,尼泊金丙酯中的一种或几种。
