背景技术
世界卫生组织将缺铁性贫血定为4大营养缺乏症之一。世界卫生组织2008年资料显示,全球缺铁性贫血发病率为37%,人口数达到20亿3千万。我国2002全国第四次营养与健康调查数据显示我国贫血发病率为20.1%,贫血人口数超过2.6亿。中、晚期妊娠的孕妇铁缺乏的患病率在50%左右,2岁以内婴幼儿和60岁以上老年人贫血患病率也较高,分别为31.1%、29.1%,而男性为15.8%,女性为23.3%。可知,缺铁性贫血的人群数量庞大。
中医认为贫血属于“血虚”症,因“气为血帅,血为气母”,血虚均伴不同程度的气虚症状。西医认为造血不良或红细胞破坏过多是贫血的核心因素,而造血原料是决定造血不良的主要因素之一。造血原料包括铁、叶酸和维生素B12等营养素。贫血时,血红蛋白减少、红细胞携带氧的能力降低,全身组织和器官发生缺氧变化,故其所产生的危害涉及到全身各个系统。轻度贫血一般会导致疲倦、乏力、头晕、耳鸣、记忆力减退、注意力不集中等症状。贫血可引起机体免疫功能和抗感染力下降等问题。

一种可补血和增强免疫力的含铁营养组合物,所述组合物中包含焦磷酸和和红枣提取物,其中,焦磷酸铁与红枣提取物的质量比为1:4~18。
本发明中,通过选择将焦磷酸铁与红枣提取物进行复配,并限定两者的质量比,意外的发现两者具有协同的促进人体的补血以及增强人体免疫力的功效。但两者的添加量需严格控制,红枣提取物添加量过少时,则起不到协同补血的作用,添加量过多,则组合物起不到协同改善免疫力的作用。根据本发明的优选实施方式,所述焦磷酸铁与红枣提取物的质量比为1:5~15。此时,得到的营养组合物的补血以及增强人体免疫力的功效更佳。根据本发明的更优选实施方式,所述焦磷酸铁与红枣提取物的质量比为1:5~10,此时前述效果最佳。
根据本发明的优选实施方式,所述组合物中还包含桂圆提取物,其中,焦磷酸铁与桂圆提取物的质量比为1:0.5~10。添加一定量的桂圆提取物不仅能起到促进补血的作用,对得到的营养组合物的改善人体免疫力效果也有进一步的促进作用。例如,示例的焦磷酸铁与桂圆提取物的质量比包括但不限于为1:1~5、1:1~3、1:2等。
根据本发明的优选实施方式,所述红枣提取物采用水提法制得,其中,每20份红枣提取得到1份的红枣提取物。红枣具有健脾养血的功效。
根据本发明的优选实施方式,所述桂圆提取物采用水提法制得,其中,每20份桂圆提取得到1份的桂圆提取物。
为达到上述第二个目的,本发明提供一种具有增强免疫力、补血的食品,该食品包含上述含铁营养组合物。
根据本发明的优选实施方式,所述食品为口服制剂,剂型为粉剂、颗粒剂、口服液、饮料、软糖、胶囊或片剂。具体的,还可根据实际要得到的口服制剂的形态,加入本领域常规的食品可接受的载体、稀释剂、矫味剂等。
根据本发明的优选实施方式,所述口服制剂可根据其具体的不同形态,按照本领域的常规手段制备即可获得。根据本发明的优选实施方式,当所述口服制剂为口服液时,口服制剂的制备包括配料、将各物料混合均匀、灭菌、灌装、巴氏灭菌即可得到产品。
根据本发明的优选实施方式,所述口服制剂为片剂时,该片剂的制备包括配料、混合、过筛,压片。
根据本发明的优选实施方式,所述口服制剂为胶囊时,该胶囊的制备包括配料、混合、装胶囊。
根据本发明的优选实施方式,所述口服制剂为粉剂或颗粒剂时,制备方法包括配料、混合、过筛即可。
根据本发明的优选实施方式,所述口服制剂为软糖时,该软糖的制备包括前处理、熬糖、混料调配、成型。
根据本发明的优选实施方式,所述口服制剂为饮料时,该饮料的制备包括配料、调配、均质、灭菌、灌装、巴氏灭菌即可得到产品。
本发明的有益效果如下:
1.本发明中将中西医理论结合,创造性的将红枣提取物与焦磷酸铁按特定的配比结合,在改善贫血以及增强免疫力方面具有协同的明显的促进作用。更进一步的,当组合物中含有一定配比的桂圆提取物时,得到的组合物的补血及增强免疫力效果更佳。
2.本发明中的含铁营养组合物可根据实际的产品形态需要而应用在食品中,适合包括儿童、女性和孕妇等各类人群的日常保健食用。
具体实施方式
为了更清楚地说明本发明,下面结合优选实施例对本发明做进一步的说明。本领域技术人员应当理解,下面所具体描述的内容是说明性的而非限制性的,不应以此限制本发明的保护范围。
实施例1
1.备料
一种可补血和增强免疫力的含铁营养软糖,原料(以原料总量为1000g计)为:
其中,上述红枣提取物是通过水提法得到,每20份红枣原料提取得到1份的提取物。
2.上述含铁营养软糖的制备方法,包括如下步骤:
1)前处理:将明胶与适量的水,置于溶胶罐中混合,在75℃温度下加热,使明胶充分溶解,将得到的胶液保温备用;将葡萄糖浆、白砂糖、山梨糖醇液适量的水置于溶糖罐中,在120℃温度下加热,使其充分溶解,得糖浆溶液;在调配罐中将充分溶解后得到的胶液和糖浆溶液混合均匀,得混合液;
2)混合、调配:将焦磷酸铁溶于剩余的水中,得焦磷酸铁的水溶液,再在混合液中加入焦磷酸铁的水溶液、红枣提取物、红糖、柠檬酸、苹果酸、食品用香精,搅拌均匀得到料液;
3)成型:将淀粉磨盘干燥至水分含量为6%,将料液输送至浇注机,浇注于淀粉磨盘中成型,将浇注完成后的磨盘送至干燥房进行干燥,干燥温度为25℃,干燥时间为72小时,将干燥后的糖进行脱模、上砂糖粉,包装即得成品。
实施例2
重复实施例1,区别在于,焦磷酸铁6g,红枣提取物84g,桂圆提取物6g。
实施例3
取焦磷酸铁8g,红枣提取物80g,加入口服液常用辅料,按常规的制备口服液的方法将物料混匀、灭菌、灌装、巴氏灭菌制成口服液。
实施例4
取焦磷酸铁10g,红枣提取物60g,加入常用辅料,过筛,制成粉剂。
实施例5
取焦磷酸铁12g,红枣提取物96g,加入常用辅料,混合,制成胶囊。
实施例6
取焦磷酸铁5g,红枣提取物60g,加入常用辅料,混合,制成软胶囊。
实施例7
取焦磷酸铁12g,红枣提取物108g,加入必要辅料,过筛,制成颗粒剂。
上述各实施例中,桂圆提取物是通过水提法得到,每20份红枣原料提取得到1份的提取物;焦磷酸铁的含铁量为24%。
对比例1
重复实施例1,区别在于,将焦磷酸铁的添加量改为84g,不添加红枣提取物,其余条件不变,制备得到软胶囊。
对比例2
重复实施例1,区别在于,将红枣提取物的添加量改为84g,不添加焦磷酸铁,其余条件不变,制备得到粉剂。
对比例3
重复实施例1,区别在于,将焦磷酸铁换成同等添加量的硫酸亚铁(含铁量为16.5%),其余条件不变,制备得到营养软糖。
对比例4
重复实施例3,区别在于,将红枣提取物的添加量改为8g,焦磷酸铁的添加量保持为8g,其余条件不变,制备得到营养软糖。
对比例5
重复实施例3,区别在于,将红枣提取物的添加量改为160g,焦磷酸铁的添加量保持为8g,其余条件不变,制备得到营养软糖。
测试各实施例及对比例得到的组合物对改善贫血效果的功效
1.实验材料
1.1试验样品
将焦磷酸铁、红枣提取物分别按照实施例1~7以及对比例1~5中的配比进行混合,依次编号为样品1~7以及对比样品1~5,空白对照组样品:生理盐水(N.S)。
1.2实验动物
SPF级小鼠,体重18~22g,购于广东省医学实验动物中心。
1.3实验设备
EXFA6030型全自动动物血液细胞分析仪(南京普朗医用设备有限公司);
TG16-W型微量高速离心机(湖南湘仪实验室仪器开发有限公司);
DJ-5000C型电子天平(中外合资亚太电子天平厂)。
表1:给药剂量设计
注:以上小鼠剂量按照60kg成人日服用量的10倍折算。焦磷酸铁原料含铁量24%,硫酸亚铁原料中铁含量为16.5%。
2.试验方法
全部小鼠自造模之日起隔日由眼底后静脉丛放血0.3mL,同时改变其正常饮食量,饲料控制在每日70g/kg体重,自由饮水,连续7d。
将小鼠随机分为13组,每组11只。各给药组小鼠于造模前7d给相应药物,每日1次,连续灌胃至造模第24d(即给药第31d)。空白对照组给予等体积生理盐水。
在造模第0、24d每只小鼠尾静脉取血20μL,检测外周血血红细胞计数(RBC)和血红蛋白(Hb)浓度。
3.试验结果
3.1在给药31d(造模第24d)时外周血血红细胞计数(RBC)见表2。
注:与同期空白对照组比较*P<0.05、**P<0.01。
3.2在给药31d(造模第24d)时外周血血红蛋白(Hb)含量见表3。

从上表2和表3中可知,本发明实施例中焦磷酸铁复合红枣提取物具有显著改善贫血的作用。从实施例1以及对比例1和2的对比可知,本发明软糖中的焦磷酸铁与红枣提取物间具有协同的促进改善机体贫血的作用。同时,进一步结合对比例4和对比例5可知,仅在特定的比例范围内,本发明的组合物才具有显著协同改善贫血的作用。结合对比例3可知,本发明中的焦磷酸铁的作用是不可用其它的铁剂所替代的。
测试各实施例及对比例得到的组合物对增强免疫力效果的功效
1.实验材料
1.1试验样品
将焦磷酸铁、红枣提取物分别按照实施例1~7以及对比例1~3中的配比进行混合,依次编号为样品1~7以及对比样品1~5,空白对照组样品:生理盐水(N.S)。
1.2实验动物

分析天平(AB265-S瑞士梅特勒一托利多仪器责任有限公司);

722分光光度计(SP-722,上海光谱仪器有限公司);
恒温水浴箱(TZL-5007,苏州珀西瓦尔实验设备有限公司);
酶标仪(MK3,百思科有限公司);
显微镜(eclipse 80i,日本尼康)等。

2.试验方法
实验动物随机分为3大组,每组130只,分别进行。分别进行迟发型变态反应实验(I大组);ConA诱导的小鼠淋巴细胞转化实验(II大组);抗体生成细胞数的测定(III大组)。3大组再随机分为实施例组、对比例组和空白对照组,每组10只。各实施例组和对比例组小鼠给相应样品,对照组给予等体积蒸馏水,每天一次,连续灌胃30天后测定免疫指标。
3.试验结果
表5结果表明,焦磷酸铁复合红枣提取物能显著增强小鼠的细胞免疫功能。

从上表5和表6中可知,本发明实施例中焦磷酸铁复合红枣提取物具有显著增强免疫力的作用。从实施例1以及对比例1和2的对比可知,本发明软糖中的焦磷酸铁与红枣提取物间具有协同的增强免疫力的作用。同时,进一步结合对比例4和对比例5可知,仅在特定的比例范围内,本发明的组合物才具有显著协同增强免疫力的作用。结合对比例3可知,本发明中的焦磷酸铁的作用是不可用其它的铁剂所替代的。
