Description

Compound mango leaf extract skin cream and preparation method thereof
Technical Field
The invention belongs to the field of medical care and daily chemical products, and particularly relates to a compound mango leaf extract skin cream taking a mango leaf extract as a main active component and a preparation method thereof.
Background
Mango (Mangifera indica L.) is a common tropical fruit in southern regions of China. The mango planting area in the Panxi region in Sichuan is more than 30 ten thousand mu, the mango planting area has the characteristics of high yield and good quality, the mango planting area occupies the domestic late-maturing mango market, and the mango industry becomes the leading industry of local agriculture. Long-term test and analysis of the inventor show that the content of mangiferin (mangiferin) in mango leaves in the Panxi region is as high as 2-8%, which is much higher than that of other plants such as rhizoma anemarrhenae and the like, and is also much higher than that of mangiferin in mango leaves in other production regions in China.
Mangiferin, also known as guan Zhi Ning, pale gray yellow needle crystal (50% ethanol), has good curative effect in treating chronic bronchitis, is the main effective component of Tibetan capillary artemisia for treating hepatitis, and is the anti-virus active component in rhizome of rhizoma anemarrhenae. Modern medical research has proved that mangiferin also has the pharmacological actions of antioxidation, anti-inflammation, bacteriostasis, radiation resistance, preventing and treating skin aging, reducing blood sugar, reducing uric acid, anti-tumor, immunoregulation, cholagogue, inhibiting central nervous system, anti-inflammation, anti-herpes simplex virus HSV-2, relieving sacral radiculopathy caused by HSV-2 virus, and the like. In pharmacopoeia of China and quality standards of various traditional Chinese medicinal materials and Chinese patent medicines, the content of mangiferin is often used as a quality control index. The raw materials of the novel health care products or the medicines which are developed by taking mangiferin as a main component at present are mostly pure mangiferin products and are generally oral preparations. Because mangiferin has extremely low bioavailability to human bodies, the clinical trial effect of the oral preparation products is not ideal. References are as follows: [1] li Y J, Sui Y J, Dai YH, et a1.LC determination and pharmacologics determination of mangiferin in plantations and tissues [ J ]. Chromatographasia, 2008, 67(11-l 2): 957-; [2] research on in vitro metabolic transformation of mangiferin by human intestinal flora [ J ] research on mangiferin metabolism by chinese traditional medicine, journal of pith of Tantan treasure, Deng Jia, etc. [ 2011, 36 (4): 443-445.
the inventor of the invention develops a new method, and prepares a compound external preparation by taking a mango leaf extract (with the mangiferin content of 60-70%) as a main active ingredient, so that the defect of low bioavailability of the mangiferin for oral administration can be overcome, the drug effect can be fully exerted, mango leaf resources in mango planting fields can be fully utilized, the production cost of products is reduced, and the economic benefit of the mango industry is improved.
Disclosure of Invention
The invention aims to provide a compound mango leaf extract skin cream taking a mango leaf extract as a main raw material and a preparation method thereof. The skin cream has good prevention and treatment effects on skin itching and red swelling caused by skin furuncle and mosquito bite.
The main active ingredients of the compound mango leaf extract skin cream are mango leaf extract, arbutin, salicylic acid and titanium dioxide; the weight ratio of the mango leaf extract to the white pigment is 1-2 parts of mango leaf extract, 0.8-1.2 parts of arbutin, 0.8-1.2 parts of salicylic acid and 0.8-1.2 parts of titanium dioxide; the mangiferin content in the mango leaf extract is 60-70%.
In the compound mango leaf extract skin cream, the active component mangiferin has strong anti-inflammatory and antioxidant effects and is a core functional component; arbutin can cooperate with mangiferin to play the roles of resisting inflammation and oxidation, and also has the effects of whitening and removing freckles; salicylic acid is a traditional external anti-inflammatory drug and can enhance the anti-inflammatory effect of mangiferin; the three components jointly become the active components of the product; the titanium dioxide can cover the yellow color of mangiferin, and can generate a certain sunscreen effect on skin due to low ultraviolet transmittance.
Furthermore, on the basis of the main active ingredients of the skin cream, conventional auxiliary materials are added to prepare the external cream. Particularly, because the mango leaf extract has poor water solubility and lipid solubility and certain color, and the physical and appearance properties of the skin-care cream prepared by the conventional cream preparation method hardly meet the use requirements of products, the inventor especially develops the specific auxiliary material combination to play the effect of the active ingredient of the invention and meets the appearance property requirements of the products when in use.
In order to improve the water solubility and the fat solubility of mangiferin, the invention adopts a cream formulation, and the weight ratio is as follows:
1) the main active ingredients are as follows: 1-2 parts of mango leaf extract, 0.8-1.2 parts of arbutin, 0.8-1.2 parts of salicylic acid and 0.8-1.2 parts of titanium dioxide; the mangiferin content in the mango leaf extract is 60-70%.
2) Auxiliary materials: 4-6 parts of stearic acid, 0.8-1.2 parts of glycerin monostearate, 3-5 parts of butyl stearate, 1-3 parts of cetyl alcohol, 6-10 parts of glycerol, 0.8-1.2 parts of liquid paraffin, 0.2-0.3 part of potassium hydroxide, 0.4 part of preservative and 40-50 parts of distilled water or deionized water.
Furthermore, essence can be added into the auxiliary materials, and the adding amount of the essence is 0.1-0.3 part by weight for increasing aroma and seasoning.
Preferably, the essence can adopt common flavoring essences such as jasmine essence, rose essence, osmanthus essence and the like.
Preferably, sorbic acid, potassium sorbate and calcium propionate are used as the preservative, and potassium sorbate is preferably used.
The skin cream of the invention adopts stearic acid and potassium hydroxide as auxiliary materials, so that potassium stearate is produced through reaction, and the potassium stearate and the components such as glycerin monostearate, butyl stearate, cetyl alcohol and the like play a good emulsifying role together, so that the product has uniform and smooth texture. The glycerol is used as a water retention agent and a lubricant; the liquid paraffin plays a role in solidifying in the product, so that the product is more stable in properties, and has a certain barrier effect and a certain lubricating effect on skin; the potassium sorbate is a common preservative for cosmetics, and the safety and the using dosage are verified for a long time. Repeated tests prove that the skin cream with ideal properties can be obtained by adopting the components and the corresponding proportion because the mangiferin has poor water solubility and lipid solubility.
The preparation method of the skin cream provided by the invention comprises the following steps of weighing the main active ingredients and the auxiliary materials according to the weight ratio:
A. preparation of a two-phase solution:
1) mixing stearic acid, glycerol, glyceryl monostearate, butyl stearate, cetyl alcohol and liquid paraffin, stirring and heating to 90-100 ℃, and maintaining for 20-40 min to obtain an oil phase solution;
2) mixing and dissolving water and potassium hydroxide, heating to 90-100 ℃ while stirring, and maintaining for 10-15 min to obtain an aqueous phase solution;
B. stirring and emulsifying: and C, controlling the temperature of the oil phase solution obtained in the step A to be 85-95 ℃, then adding the water phase solution obtained in the step A, stirring for 10-20min, then sequentially adding the mango leaf extract, arbutin, salicylic acid and titanium dioxide, stirring uniformly, and gradually reducing the temperature to 55-60 ℃ to obtain a uniform paste.
C. Standing, cooling, and packaging.
The inventor tests prove that the adding sequence of the materials cannot be reversed, otherwise, the finally obtained product cannot easily meet the requirement of satisfactory appearance properties. The product disclosed by the invention is prepared into cream by matching mangiferin, arbutin, salicylic acid and titanium dioxide with corresponding auxiliary materials, the cream can be quickly melted and disappeared after being smeared on skin, the skin has lubrication, moistening and luster feeling, and the cream has obvious effects of relieving itching, diminishing inflammation and promoting skin recovery aiming at skin pruritus caused by skin furuncle and mosquito bite.
Detailed Description
The advantageous effects of the present invention are illustrated by the following preparation examples of the present invention.
The mango leaf extract with the mangiferin content of 60-70% is prepared by a method for extracting and separating the mangiferin from the mango leaves with Chinese patent application number 201720057903.7, and specifically the mango leaf extract with the mangiferin content of 60-70% is obtained by extraction and separation from the mango leaves, and comprises the following steps:
A. preparing raw materials: drying mango leaves serving as a raw material, crushing, and sieving with a 20-40-mesh sieve to obtain mango leaf powder;
B. ester extraction and impurity removal: weighing mango leaf powder, adding 2-4L ethyl acetate into each kilogram of mango leaf powder, sealing and soaking for more than or equal to 4h, shaking at room temperature for more than or equal to 10min, and filtering; repeatedly extracting for 3-5 times until the green color of the extractive solution is almost disappeared to obtain mango leaf residue;
C. methanol extraction: taking the mango leaf residue extracted in the step B, adding 2-4L of methanol into each kilogram of mango leaf residue, sealing and soaking for more than or equal to 4 hours at the temperature of 30-40 ℃, stirring and extracting for 0.5-1 hour at the temperature of 50-60 ℃, filtering while hot, repeatedly extracting for 4-6 times, combining extracting solutions, precipitating at room temperature for more than or equal to 8 hours, and filtering to obtain a filtrate;
D. concentration and washing: concentrating the filtrate obtained in the step C into a thick extract, adding 2-4L of methanol into each kilogram of extract, stirring and washing at 50-60 ℃, cooling, filtering, and repeatedly washing for 3-5 times until filter residues are yellow-white particles; drying the filter residue to obtain a mangiferin crude product with the purity of 60-70%.
Wherein, fresh mango leaves are adopted as the mango leaves in the step A; in particular, fresh mango tender leaves are adopted as the mango leaves in the step A. However, in view of the collection of the raw materials, young leaves of mango, old leaves at the base of the shoot, and a mixture thereof are generally used. This is because the inventors found: the mango leaf content in mango leaves of different varieties and old tender degrees is greatly different, for example, the content of Kate, Haiton, Jilu and other varieties with large planting areas is generally 2-4%, the content of traditional old varieties such as Miscanthus sinensis and the like can be as high as 6-8%, and the content of tip tender leaves of the same variety is far higher than that of old leaves at the base of branches. Therefore, the raw materials used in the extraction method mainly comprise tender leaves of various mangos, old leaves at the base of branches and mixtures thereof, and fresh mango leaves can be collected as mixed raw materials during flower thinning and fruit thinning or during pruning after mango harvesting.
Preferably, the drying in the step A is natural drying or drying at 45-65 ℃ until the moisture content is less than 8%.
Preferably, the room temperature precipitation in the step C is filtered for more than or equal to 8 hours, and a filter screen with more than or equal to 200 meshes is adopted for fine filtration.
Preferably, the filtrate obtained in the step D is concentrated by vacuum rotary evaporation at the temperature of 5-65 ℃.
Preferably, the step D is carried out for stirring and washing for 10-20min at 50-60 ℃.
Preferably, the filtering in the step D adopts a filter screen with the mesh size of more than or equal to 200.
Preferably, the drying in the step D is drying at 50-60 ℃.
