Abstract
本发明公开了一种具有辅助降血糖作用的复方茶及其制备方法,属于保健品技术领域。所述复方茶的原料按重量份数计包括:武夷岩茶黄片提取物20~60份,黄精水提物7~17份,超微铁皮石斛粉末3~9份,枸杞水提物0.5~2份,炙甘草水提物0.5~1份,茶多糖1~4份。该复方茶由武夷岩茶黄片与名贵中药复合而成,能有效作用于人体多个器官而发挥协同作用,具有明显降血糖作用,在制备治疗糖尿病的保健品中具有潜在应用。

技术领域
本发明涉及一种具有辅助降血糖作用的复方茶及其制备方法,属于保健食品行业。
背景技术
糖尿病是一类由于胰岛素绝对或相对缺乏或胰岛素生物活性降低,或者以上二者共同引起的体内代谢失调及高血糖状态。临床上主要分为2种类型,即1型糖尿病或2型糖尿病。胰岛素绝对不足多见于1型糖尿病,相对不足多见于2型糖尿病,其中2型糖尿病患者占糖尿病总人数的90%左右。2型糖尿病的病理机制比较复杂,包括胰岛β细胞数量减少,胰岛分泌功能丧失、胰岛素受体敏感度降低、自由基增加及代谢信号传导受阻等,最终导致糖脂代谢紊乱。从胰岛素的起始合成到最终与靶细胞内特异受体结合而引发细胞内代谢的整个过程中,任一环节发生问题都会诱发糖尿病。糖尿病是一种常见而又难以根治的慢性病,其患病率有逐年上升的趋势,目前尚无较为理想的治疗方法。在我国,对糖尿病的防治刻不容缓!
在武夷岩茶初制加工过程中,由于鲜叶走水不足而产生“黄片”。“黄片”,条索欠紧结,色泽乌黑,质量轻飘,但其富含茶多糖、茶多酚、氨基酸等物质,具有极佳的降血糖效果,直接废弃既污染环境亦导致资源浪费。因此开发武夷岩茶黄片保健食品不仅解决了茶叶资源的浪费问题,同时还给糖尿病患者带来了福音。
发明内容
本发明的目的在于针对现有技术不足,提供一种具有辅助降血糖作用的复方茶及其制备方法。
为实现上述目的,本发明采用如下技术方案:
一种具有辅助降血糖作用的复方茶,按重量份数计,所述复方茶的原料包括:武夷岩茶黄片水提物20~60份,黄精水提物7~17份,超微铁皮石斛粉末3~9份,枸杞水提物0.5~2份,炙甘草水提物0.5~1份,茶多糖1~4份。
所述的武夷岩茶黄片水提物采用以下方法制备:将干燥的武夷岩茶黄片粉碎过20目筛,加入质量倍数为30-50倍量的水,在70-90℃的水浴锅中提取2次,每次提取20-40min,真空抽滤后合并两次滤液,再减压浓缩干燥得到武夷岩茶黄片水提物。
所述的黄精水提物采用以下方法制备:将干燥的黄精粉碎过10目筛,加入质量倍数为10-30倍量的水,在70-90℃的水浴锅中提取2次,每次提取20-40min,真空抽滤后合并两次滤液,减压浓缩干燥得到黄精水提物。
所述的炙甘草水提物采用以下方法制备:将干燥的炙甘草粉碎过10目筛,加入质量倍数为20-40倍量的水,在70-90℃的水浴锅中提取2次,每次提取20-40min,真空抽滤后合并两次滤液,减压浓缩干燥得到炙甘草水提物。
所述的枸杞水提物采用以下方法制备:将干燥的枸杞粉碎过10目筛,加入质量倍数为20-50倍量的水,在70-90℃的水浴锅中提取2次,每次提取20-40min,真空抽滤后合并两次滤液,减压浓缩干燥得到枸杞水提物。
所述的超微铁皮石斛粉末采用以下方法制备:将铁皮石斛进行超微粉碎,得到不低于200目的超微铁皮石斛粉末。
所述的茶多糖:由市场购得。
如上所述的一种具有辅助降血糖作用的复方茶的制备方法:取武夷岩茶黄片水提物20~60份,黄精水提物7~17份,超微铁皮石斛粉末3~9份,枸杞水提物0.5~2份,炙甘草水提物0.5~1份,茶多糖1~4份,混合均匀后,装袋,每袋2-6克包装即为复方袋装茶。
如上所述的一种具有辅助降血糖作用的复方茶的另一种制备方法:取明胶置于烧杯中,加入50-80℃的水搅拌使其完全溶解,随后依次加入武夷岩茶黄片水提物、黄精水提物、超微铁皮石斛粉末、枸杞水提物、炙甘草水提物、茶多糖和甘露醇,搅拌均匀制成软材,将所得软材在制粒机中制粒、过筛,装袋,每袋8-12克包装即为复方颗粒剂。
本发明中各原料药理、功效如下:
武夷岩茶:提神益思,消除疲劳、生津利尿、解热防暑、杀菌消炎、祛寒解酒、解毒防病、消食去腻、减肥健美,突出表现在防癌症、降血糖血脂、抗衰老等功效。
黄精:养阴润肺;补脾益气,滋肾填精。主阴虚劳嗽;肺燥咳嗽;脾虚乏力;食少口干;消渴;肾亏腰膝酸软;阳痿遗精;耳鸣目暗;须发早白;体虚赢瘦;风癞癣疾。用于脾胃虚弱,体倦乏力,口干食少,肺虚燥咳,精血不足,内热消渴。
茶多糖:降血糖、降血脂、增强免疫力、降血压、减慢心率、增加冠脉流量、抗凝血、抗血栓和耐缺氧等,近些年来发现茶多糖还具有治疗糖尿病的功效。
铁皮石斛:《本草纲目》记载“石斛除痹下气,补五脏虚劳赢瘦,强阴益精,久服,厚肠胃,补内绝不足,平胃气,长肌肉,逐皮肤邪热痱气,脚膝疼冷痹弱,定志除惊,轻身延年,益气除热,治男子腰膝软弱,健阳,逐皮肤风痹,骨中久冷,补肾益力,壮筋骨,暖水休,益智清气,治发热自汗,痈疽排脓内塞。”
枸杞:抗氧化、抗衰老、抗肿瘤、抗脂肪肝、降血脂、增强肌体免疫作用等与其他中药协调入药服用,作为降血糖亦与其他具降糖作用的中药合用。
炙甘草:炙甘草含有甘草甜素、甘草次酸、甘草多糖等多种化甘草学成分,具有抗炎及抗变态反应作用,可调节机体免疫功能,抗肿瘤和降血糖作用。
本发明的有益效果在于:
(1)本发明以武夷岩茶黄片为主要原材料,将黄精、铁皮石斛、茶多糖做为辅料,制备方法简单易行、对设备要求不高,并且健康、绿色、环保;
(2)茶水提物与中药水提物具有协同作用,动物实验表明:根据本发明的制备方法制得的复方茶颗粒剂降血糖功效明显优于单独茶叶水提物,这为茶与中药复合降血糖提供了实验依据。
具体实施方式
以下结合具体实施例对本发明做进一步说明,但本发明不仅仅限于这些实施例。
实施例1
(1)取干燥的武夷岩茶黄片,并粉碎过20目筛,加入30倍量的水,在70℃的水浴锅中提取2次,每次提取30min,真空抽滤后合并两次滤液,再减压浓缩干燥得到武夷岩茶黄片水提物;
(2)取干燥的黄精,并粉碎过10目筛,加入10倍量的水,在70℃的水浴锅中提取2次,每次提取30min,真空抽滤后合并两次滤液,减压浓缩干燥得到黄精水提物;
(3)取干燥的炙甘草,并粉碎过10目筛,加入20倍量的水,在70℃的水浴锅中提取2次,每次提取30min,真空抽滤后合并两次滤液,减压浓缩干燥得到炙甘草水提物;
(4)取干燥的枸杞,并粉碎过10目筛,加入20倍量的水,在70℃的水浴锅中提取2次,每次提取30in,真空抽滤后合并两次滤液,减压浓缩干燥得到枸杞水提物;
(5)将铁皮石斛用中药超微粉碎机进行超微粉碎,得到不低于200目的超微铁皮石斛粉末;
(6)称37.5g取明胶置于烧杯中,加入50℃的水溶解15min,然后在50℃的水浴锅中加热搅拌使之完全溶解,并依次加入3.6g茶多糖、53g武夷岩茶黄片水提物、13g黄精水提物、6.5g超微铁皮石斛粉末、1.7g枸杞水提物、0.7g炙甘草水提物和37.5g甘露醇,用玻璃棒搅拌均匀,制成软材,过3号筛(50目筛)制粒,在50℃条件下的电热恒温鼓风干燥箱中烘干后整粒,过3号筛但不能通过5号筛的颗粒即成品。
实施例2
(1)取干燥的武夷岩茶黄片,并粉碎过20目筛,加入50倍量的水,在90℃的水浴锅中提取2次,每次提取35min,真空抽滤后合并两次滤液,再减压浓缩干燥得到武夷岩茶黄片水提物;
(2)取干燥的黄精,并粉碎过10目筛,加入30倍量的水,在90℃的水浴锅中提取2次,每次提取35min,真空抽滤后合并两次滤液,减压浓缩干燥得到黄精水提物;
(3)取干燥的炙甘草,并粉碎过10目筛,加入40倍量的水,在90℃的水浴锅中提取2次,每次提取35min,真空抽滤后合并两次滤液,减压浓缩干燥得到炙甘草水提物;
(4)取干燥的枸杞,并粉碎过10目筛,加入40倍量的水,在90℃的水浴锅中提取2次,每次提取35min,真空抽滤后合并两次滤液,减压浓缩干燥得到枸杞水提物;
(5)将铁皮石斛用中药超微粉碎机进行超微粉碎,得到不低于200目的超微铁皮石斛粉末;
(6)称10.4g取明胶置于烧杯中,加入80℃的水溶解5min,然后在50℃的水浴锅中加热搅拌使之完全溶解,并依次加入1g茶多糖、20g武夷岩茶黄片水提物、7g黄精水提物、3g超微铁皮石斛粉末、0.5g枸杞水提物、0.5g炙甘草水提物和10.4g甘露醇,用玻璃棒搅拌均匀,制成软材,过3号筛(50目筛)制粒,在80℃条件下的电热恒温鼓风干燥箱中烘干后整粒,过3号筛但不能通过5号筛的颗粒即成品。
实施例3
(1)取干燥的武夷岩茶黄片,并粉碎过20目筛,加入40倍量的水,在80℃的水浴锅中提取2次,每次提取20min,真空抽滤后合并两次滤液,再减压浓缩干燥得到武夷岩茶黄片水提物;
(2)取干燥的黄精,并粉碎过10目筛,加入20倍量的水,在80℃的水浴锅中提取2次,每次提取20min,真空抽滤后合并两次滤液,减压浓缩干燥得到黄精水提物;
(3)取干燥的炙甘草,并粉碎过10目筛,加入30倍量的水,在80℃的水浴锅中提取2次,每次提取20min,真空抽滤后合并两次滤液,减压浓缩干燥得到炙甘草水提物;
(4)取干燥的枸杞,并粉碎过10目筛,加入35倍量的水,在80℃的水浴锅中提取2次,每次提取20min,真空抽滤后合并两次滤液,减压浓缩干燥得到枸杞水提物;
(5)将铁皮石斛用中药超微粉碎机进行超微粉碎,得到不低于200目的超微铁皮石斛粉末;
(6)称41.7g取明胶置于烧杯中,加入65℃的水溶解10min,然后在65℃的水浴锅中加热搅拌使之完全溶解,并依次加入4g茶多糖、60g武夷岩茶黄片水提物、17g黄精水提物、9g超微铁皮石斛粉末、2g枸杞水提物、1g炙甘草水提物和41.7g甘露醇,用玻璃棒搅拌均匀,制成软材,过3号筛(50目筛)制粒,在65℃条件下的电热恒温鼓风干燥箱中烘干后整粒,过3号筛但不能通过5号筛的颗粒即成品。
实施例4
(1)取干燥的武夷岩茶黄片,并粉碎过20目筛,加入45倍量的水,在85℃的水浴锅中提取2次,每次提取40min,真空抽滤后合并两次滤液,再减压浓缩干燥得到武夷岩茶黄片水提物;
(2)取干燥的黄精,并粉碎过10目筛,加入25倍量的水,在85℃的水浴锅中提取2次,每次提取40min,真空抽滤后合并两次滤液,减压浓缩干燥得到黄精水提物;
(3)取干燥的炙甘草,并粉碎过10目筛,加入35倍量的水,在85℃的水浴锅中提取2次,每次提取40min,真空抽滤后合并两次滤液,减压浓缩干燥得到炙甘草水提物;
(4)取干燥的枸杞,并粉碎过10目筛,加入45倍量的水,在85℃的水浴锅中提取2次,每次提取40min,真空抽滤后合并两次滤液,减压浓缩干燥得到枸杞水提物;
(5)将铁皮石斛用中药超微粉碎机进行超微粉碎,得到不低于200目的超微铁皮石斛粉末;
(6)称54g取明胶置于烧杯中,加入70℃的水溶解5-15min,然后在50-80℃的水浴锅中加热搅拌使之完全溶解,并依次加入2.5茶多糖、40g武夷岩茶黄片水提物、12g黄精水提物、6g超微铁皮石斛粉末、1.25g枸杞水提物、0.75g炙甘草水提物和54g甘露醇,用玻璃棒搅拌均匀,制成软材,过3号筛(50目筛)制粒,在70℃条件下的电热恒温鼓风干燥箱中烘干后整粒,过3号筛但不能通过5号筛的颗粒即成品。
实施例5
(1)取干燥的武夷岩茶黄片,并粉碎过20目筛,加入45倍量的水,在85℃的水浴锅中提取2次,每次提取40min,真空抽滤后合并两次滤液,再减压浓缩干燥得到武夷岩茶黄片水提物;
(2)取干燥的黄精,并粉碎过10目筛,加入25倍量的水,在85℃的水浴锅中提取2次,每次提取40min,真空抽滤后合并两次滤液,减压浓缩干燥得到黄精水提物;
(3)取干燥的炙甘草,并粉碎过10目筛,加入35倍量的水,在85℃的水浴锅中提取2次,每次提取40min,真空抽滤后合并两次滤液,减压浓缩干燥得到炙甘草水提物;
(4)取干燥的枸杞,并粉碎过10目筛,加入45倍量的水,在85℃的水浴锅中提取2次,每次提取40min,真空抽滤后合并两次滤液,减压浓缩干燥得到枸杞水提物;
(5)将铁皮石斛用中药超微粉碎机进行超微粉碎,得到不低于200目的超微铁皮石斛粉末;
(6)取武夷岩茶黄片水提物25g份,黄精水提物9g,超微铁皮石斛粉末4.5g,枸杞水提物1g,炙甘草水提物0.6g,茶多糖1.5g,混合均匀后,装袋,每袋2-6克包装即为复方袋装茶。
本发明的辅助降低血糖的颗粒剂进行的有关动物实验:
实验材料:本发明的辅助降低血糖复方茶颗粒剂。
1.试验方法:
采用“保健食品功能学评价程序和检验方法”中“辅助降血糖功能评价程序及检验方法”。
2.实验动物:体重为200±20g的Wistar大鼠,雄性。
3.实验分组及剂量选择
将动物适应性喂养1周后将实验鼠分为6组,分别是:正常对照组、模型对照组、高中低复方茶剂量组、武夷岩茶组。将本发明的降血糖复方茶颗粒剂按照成人每日每公斤体重推荐摄入量的10倍、20倍、30倍剂量配制成溶液按老鼠体重灌胃,其中正常组对照组和模型对照组给予灌胃同体积的0.9%生理盐水,茶叶剂量组给予灌胃相同体积的茶叶水提物。
4.实验检测项目及方法
高血糖大鼠造模:取正常大鼠50只,随机分为5组,每组10只,将大鼠禁食3-5个小时,用血糖仪检测大鼠尾静脉血糖,作为该批次动物基础血糖值。随后动物禁食24大时(自由饮水),按剂量30mg/kg腹腔注射STZ溶液(用前新鲜配制,临用前溶解在pH=4.5的0.1mmol/L柠檬酸缓冲液内,1次/d,连续2d),正常对照组注射等量柠檬酸缓冲液,1次/d,连续2d。3天后动物禁食3-5小时,测尾静脉血糖,血糖值≧16.7mmol/L并且有多饮多食多尿等症状即认定为高血糖模型成功动物。
高血糖Wistar大鼠血糖实验:将高血糖模型大鼠随机分为模型对照组、武夷岩茶组和3个复方茶剂量组。分组完成后,每天上午8点给药,高、中、低三剂量组分别给予灌胃剂量为3.3g/kg、6.7g/kg、10g/kg的辅助降糖复方茶颗粒剂,正常组对照组和模型对照组给予灌胃同体积的0.9%生理盐水,连续30天,测定空腹血糖(禁食同上)。
高血糖Wistar大鼠糖耐量实验:6组大鼠连续给药7天后,第8天禁食5h,自由饮水,测定血糖值,灌胃给予25%葡萄糖溶液(2.0g/kg),分别于给予葡萄糖后0、30、60、120min再次取血测定血糖值并计算血糖曲线下面积。血糖曲线下面积S(mmol/L)=(a+2b+3c+2d)/4 ,其中a、b、c、d分别为灌胃葡萄糖溶液0、30、60、120min后的血糖值。
5.实验结果
(1)复方茶颗粒剂降低高血糖大鼠空腹血糖实验

武夷岩茶复方颗粒剂高、中、低剂量组与武夷岩茶组大鼠在连续给药30d后,血糖值基本没多大变化,与对照组相比显著性不差异。表明长期灌胃武夷岩茶复方颗粒剂与武夷岩茶的对正常血糖值没多大影响。见表1。
(2)复方茶颗粒剂对高血糖大鼠糖耐量实验

表2表明高血糖大鼠在给予灌胃25%葡萄糖溶液后,血糖值在0-30min时间段内迅速升高并达到顶峰值,之后血糖逐渐降低,模型对照组大鼠血糖于糖负荷后逐渐升高,与糖符合前血糖浓度相差不大。复方茶颗粒剂高、中、低剂量组与武夷岩茶组在糖负荷后0-30min时间段内迅速升高并达到顶峰值,120min后血糖值在糖负荷前血糖值附近,并且与模型组血糖比较具有显著性差异。各给药组的曲线下面积与模型组相比都具有显著性差异,表明复方茶颗粒剂与武夷岩茶能改善糖尿病大鼠糖耐量的作用。
以上实验表明,本复方茶颗粒剂对STZ糖尿病大鼠有明显降血糖作用,与模型对照组相比具有显著性差异。

1.一种具有辅助降血糖作用的复方茶,其特征在于:按重量份数计,所述复方茶的原料包括:武夷岩茶黄片水提物20~60份,黄精水提物7~17份,超微铁皮石斛粉末3~9份,枸杞水提物0.5~2份,炙甘草水提物0.5~1份,茶多糖1~4份。
2.根据权利要求1所述的具有辅助降血糖作用的复方茶,其特征在于:所述的武夷岩茶黄片水提物采用以下方法制备:将干燥的武夷岩茶黄片粉碎过20目筛,加入质量倍数为30-50倍量的水,在70-90℃的水浴锅中提取2次,每次提取20-40min,真空抽滤后合并两次滤液,再减压浓缩干燥得到武夷岩茶黄片水提物。

5.根据权利要求1所述的具有辅助降血糖作用的复方茶,其特征在于:所述的枸杞水提物采用以下方法制备:将干燥的枸杞粉碎过10目筛,加入质量倍数为20-50倍量的水,在70-90℃的水浴锅中提取2次,每次提取20-40min,真空抽滤后合并两次滤液,减压浓缩干燥得到枸杞水提物。
6.根据权利要求1所述的具有辅助降血糖作用的复方茶,其特征在于:所述的超微铁皮石斛粉末采用以下方法制备:将铁皮石斛进行超微粉碎,得到不低于200目的超微铁皮石斛粉末。
7.一种制备如权利要求1所述的具有辅助降血糖作用的复方茶的方法,其特征在于:取武夷岩茶黄片水提物20~60份,黄精水提物7~17份,超微铁皮石斛粉末3~9份,枸杞水提物0.5~2份,炙甘草水提物0.5~1份,茶多糖1~4份,混合均匀后,装袋,每袋2-6克包装即为复方袋装茶。
