Abstract
本发明公开了一种基于荧光和比色双重模式检测碱性磷酸酶活性的方法、制备的传感器及应用,将碱性磷酸酶溶液投入到过量的L‑抗坏血酸‑2‑磷酸溶液中,然后将混合物置于碱性缓冲溶液中共同孵育,生成L‑抗坏血酸;加入双蒸水、石墨烯量子点和硝酸银溶液进一步混合孵育;银纳米粒子逐步沉积在石墨烯量子点表面,石墨烯量子点表面的荧光强度逐步降低,石墨烯量子点表面沉积的银纳米粒子的吸光度逐渐增加;根据荧光强度或吸光度的变化,进行碱性磷酸酶活性的定量检测。本发明可以通过肉眼对目标物碱性磷酸酶进行可视化鉴别,也可用比色法和荧光法对碱性磷酸酶进行定量检测。双重模式的检测通过两种方法输出测定结果,减小了环境波动引起的误差,保证了测定结果的可靠性。

背景技术

(2)将碱性磷酸酶溶液投入到过量的L-抗坏血酸-2-磷酸溶液中,然后将混合物置于碱性缓冲溶液中共同孵育,生成L-抗坏血酸;
(3)往步骤(2)的混合物中加入双蒸水、石墨烯量子点和硝酸银溶液进一步混合孵育;
(4)银纳米粒子逐步沉积在石墨烯量子点表面,石墨烯量子点表面的荧光强度逐步降低,石墨烯量子点表面沉积的银纳米粒子的吸光度逐渐增加;
(5)根据荧光强度或吸光度的变化,进行碱性磷酸酶活性的定量检测。
所述步骤(1)中,石墨烯量子点的直径在10nm以下,平均直径为1~3nm,其荧光光谱的最大激发波长Ex和最大发射波长Em分别为362nm和461nm。
石墨烯量子点可通过柠檬酸加热得到,反应温度为220℃,反应时间为20min,然后逐滴加入到0.25M的氢氧化钠溶液中,充分搅拌10min,再用0.22μm的微孔滤膜过滤,3.5kDa的透析袋透析24h,真空冷冻干燥制得粉末。
所述步骤(2)中,共同孵育的条件是温度为25~40℃,时间为10~20min。
所述步骤(2)中,碱性缓冲溶液中含有无水硫酸镁,pH为9.8,所述碱性缓冲溶液选自三羟甲基氨基甲烷-硝酸缓冲溶液、二乙醇胺-硝酸缓冲溶液、甘氨酸-氢氧化钠缓冲溶液、碳酸钠-碳酸氢钠缓冲溶液中的至少一种,所述硫酸镁的浓度为0.2~2mM。
所述L-抗坏血酸-2-磷酸的溶液,用量为50μL,浓度为10~40mM;目标物碱性磷酸酶溶液用量为20μL,缓冲液用量为200μL。
加入多余的双蒸水的目的是将反应体系的最终体积定在2mL,用作溶剂和定容。
所述步骤(3)中,混合孵育的条件为:温度为20~40℃,时间为20~60min,pH大于6。
双蒸水体积为1580μL,量子点溶液的浓度为6~10mg/mL,用量为50μL,硝酸银溶液的浓度为10~40mM,用量为100μL。

所述步骤(5)中,碱性酶活性的定量检测过程具体如下:配置一系列不同浓度的碱性磷酸酶标准品溶液加入到反应体系中,监测反应体系吸光度或相对荧光强度的变化,根据吸光度的变化ΔA415nm或相对荧光强度(F0-F)/F0随碱性磷酸酶标准品溶液活性的变化绘制标准曲线,得到碱性磷酸酶活性的标准曲线方程,将待测样品加入到反应体系中,监测反应体系吸光度的变化ΔA415nm或相对荧光强度的变化(F0-F)/F0,根据碱性磷酸酶活性的标准曲线方程,推算出碱性磷酸酶的用量,从而实现待测样品中碱性磷酸酶活性的定量检测。
碱性磷酸酶用于催化L-抗坏血酸-2-磷酸(AA-P)反应得到L-抗坏血酸(AA),L-抗坏血酸(AA)与硝酸银(AgNO3)反应生成纳米银粒子,纳米银粒子沉积在量子点表面得到纳米复合结构。L-抗坏血酸-2-磷酸溶液是过量的,石墨烯量子点和硝酸银的量是已知的。
所述步骤(5)中,利用紫外可见光谱的比色法检测,碱性磷酸酶活性在0.3~10U/L范围内具有线性关系,检测限为0.1U/L;利用荧光光谱的荧光法检测,碱性磷酸酶活性在0.05~2.5U/L范围内具有线性关系,检测限为0.02U/L。
一种使用所述的检测碱性磷酸酶活性的方法制备的传感器。
如所述的一种传感器在碱性磷酸酶抑制剂中的应用。
本发明的测定原理是:利用酶促反应和GQDs@Ag纳米复合结构的光学特性。碱性磷酸酶催化L-抗坏血酸-2-磷酸(AA-P)去磷酸化,生成具有还原性的L-抗坏血酸(AA),可与硝酸银(AgNO3)发生氧化还原反应,生成银纳米粒子沉积于石墨烯量子点表面,随着碱性磷酸酶浓度的增加,银纳米壳的生成增加,检测液的颜色在可见光下从无色变为黄色,在365nm紫外灯下,量子点的荧光从明亮的蓝色荧光到荧光逐渐淬灭。紫外可见分光光度计下,随着纳米银粒子的生成增加,溶液在415nm处的吸光度逐渐增加。在荧光分光光度计下,随着纳米银粒子在量子点表面的沉积增加,量子点的荧光淬灭程度增加,量子点在Ex=362nm,Em=461nm处的荧光强度减弱。因此,利用比色法和荧光法可以达到定量检测的目的。基于紫外吸收增强和荧光淬灭的程度作为输出信号,碱性磷酸酶活性可被定量检测,同时能够制成生物传感器并将其应用在碱性磷酸酶抑制剂的研究及其筛选中。
本发明相比现有技术具有以下优点:
(1)本发明基于碱性磷酸酶的酶促反应、石墨烯量子点的荧光“开-关”效应以及纳米银的紫外吸收特性建立了高灵敏度的双重模式的碱性磷酸酶的检测方法。不仅可以通过肉眼对目标物碱性磷酸酶进行可视化鉴别,也可用比色法和荧光法对碱性磷酸酶进行定量检测。双重模式的检测通过两种方法输出测定结果,减小了环境波动引起的误差,保证了测定结果的可靠性,更适合实际应用;
(2)相比之前报道的测定碱性磷酸酶的双重传感器存在的检测限不够低或者反应时间过长的问题,该方法得到的碱性磷酸酶双重传感器可以在较短的时间内完成高灵敏度的检测;
(3)采用石墨烯量子点(GQDs)作为探针,具有合成简单、造价低廉、绿色无毒、理化性质稳定和生物相容性良好等优势;GQDs@Ag纳米复合结构在反应过程中生成,不需要预先合成,因此简化了操作过程。首次利用纳米银粒子沉积于石墨烯量子点表面引起体系紫外吸收和荧光强度同时发生改变,来构建碱性磷酸酶的双重传感器;
(4)同时能将该生物传感器成功应用于碱性磷酸酶抑制剂的研究和筛选。

具体实施方式
下面对本发明的实施例作详细说明,本实施例在以本发明技术方案为前提下进行实施,给出了详细的实施方式和具体的操作过程,但本发明的保护范围不限于下述的实施例。
实施例1
制备石墨量子点
称取2g柠檬酸白色固体加热至220℃,柠檬酸从白色固体逐渐转变为无色黏稠液体、淡黄色液体,最后成橘黄色液体;随后将橘黄色液体逐滴添加到50ml的0.25M氢氧化钠溶液并充分搅拌10min;再用0.22μm的微孔滤膜过滤,3.5kDa的透析袋透析24h,再用真空冷冻干燥的方法制得粉末。如图1A所示,合成的量子点在Ex=362nm,Em=461nm处对应的荧光强度最大,如图1B所示,Ex=362nm对应的发射光谱和Em=461nm对应的激发光谱呈镜像对称的关系。
实施例2
将实施例1制备得到的石墨量子点进行稳定性考察。
如图2A所示,本实施例中将量子点放在20~100℃的温度下孵育1h后测定荧光强度,发现量子点在温度低于50℃时稳定,高于50℃荧光强度显著下降,因此,纳米银在石墨烯量子点表面沉积的反应温度不得超过50℃。
如图2B所示,将量子点放在3~13的pH条件下孵育1h后测定荧光强度,发现量子点在pH高于6时稳定,低于6时荧光强度显著下降,因此,纳米银在石墨烯量子点表面沉积的pH不得低于6。
如图2C所示,对于两个不同批次量子点间荧光强度的比较,发现不同批次合成的量子点荧光光谱大小形状、最大发生波长以及最大波长处的荧光强度很接近。
如图2D所示,两个不同批次量子点应用于荧光法测定碱性磷酸酶活性的线性的也很接近,说明该量子点合成方法重现性好。

实施例3
本实施例将纳米银沉积在石墨烯量子点表面。如图3所示,基于纳米银在石墨烯量子点表面沉积的比色法和荧光法双模式的碱性磷酸酶活性分析方法的原理中显示,在碱性磷酸酶的存在下,L-抗坏血酸-2-磷酸的磷酸基团被水解,生成的抗坏血酸还原银离子生成纳米银沉积在量子点表面,导致反应溶液颜色逐渐加深,紫外吸收增强,石墨烯量子点的荧光减弱。基于这个原理,对碱性磷酸酶活性可以进行比色法和荧光法双模式的定量分析。

实施例4
如图4A所示,为了进一步研究碱性磷酸酶传感器的机制,本实施例用紫外可见分光光度计和荧光分光光度计对不同体系的光学性质进行验证。试剂添加分为如下三组:实验组AA-P+ALP+GQDs+AgNO3(a),阳性对照组AA+GQDs+AgNO3(b),阴性对照组包括AA-P+ALP+AgNO3(c),AA-P+GQDs+AgNO3(d),ALP+GQDs+AgNO3(e)和AA-P+ALP+GQDs(f)。与阳性对照组(曲线b)相比,阴性对照组(曲线b c d e)并没有导致紫外和荧光强度的显著改变,实验组(曲线a)能够导致显著的紫外吸光度增强和荧光淬灭,其与阳性对照组结果类似。一系列结果表明磷酸化抗坏血酸经过酶促反应产生抗坏血酸,随后与硝酸银反应生成银纳米粒子,沉积于石墨烯量子点表面,从而导致体系紫外和荧光发生光学改变。另外,如图4B和图4C所示,照片显示的颜色变化与紫外和荧光的光学变化基本保持一致,进一步表明银纳米粒子沉积于石墨烯量子点表面而形成GQD@Ag纳米复合结构。
通过透射电镜观察反应前后石墨烯量子点的形态变化,如图5A所示,反应前石墨烯量子点的粒径在2nm左右,如图5B所示,反应后粒径增大到13nm左右。通过原子力显微镜观察,如图5C所示,可以看到当仅含有量子点时,量子点的高度在0.4nm左右,如图5D所示,当纳米银沉积在量子点表面时,高度增加至13nm左右,因此推测生成的银纳米粒子沉积于石墨烯量子点表面致使粒径变大。通过能量色散x射线元素图观察反应后量子点表面的形貌和元素,如图6A、6B和6C所示,可以看到反应后碳元素和银离子同时存在,纳米银沉积在量子点表面。通过能量色散谱图观察反应前后量子点表面的元素变化,如图6D所示,当仅含有量子点时,可以看到碳元素的峰存在而观察不到银元素的峰,如图6E所示,反应后可以看到量子点表面碳元素和银元素的峰同时存在,因此,通过能量色散x射线元素图和能量色散谱图可以进一步证明银纳米粒子在石墨烯量子点表面的沉积。

实施例5
为了得到最佳性能,优化了体系的相关参数包括磷酸化抗坏血酸浓度、硝酸银浓度、缓冲溶液的浓度、体系的孵育时间以及酶促反应孵育时间。如图7A和7B所示,随着磷酸化抗坏血酸和硝酸银浓度增加,相对荧光强度逐渐增加,在磷酸化抗坏血酸浓度达到0.6mM和硝酸银浓度达到1.2mM,反应体系达到动态平衡,相对荧光强度不再增加,因此分别选择0.6mM磷酸化抗坏血酸和1.2mM硝酸银作为反应体系的最佳条件。如图7C所示,Tris浓度在0~7mM范围内被研究,发现Tris浓度在1mM时石墨烯量子点的相对荧光强度最大,因此选择1mM的Tris作为缓冲溶液的最优浓度。另外,反应体系的孵育时间、酶促反应孵育时间也被优化。如图7D所示,体系孵育时间在0~45min范围内,随着时间增加其相对荧光强度也增加,随后达到动态平衡。因此,选择45min作为体系的最佳孵育时间。如图8所示,酶促反应孵育时间在1~30min范围内,随着时间增加其相对荧光也增加,15min后达到动态平衡,因此选择15min作为酶促反应的最佳孵育时间。
基于GQDs@Ag纳米复合物的紫外吸光和相对荧光强度的改变作为输出信号,在最优的反应条件下,采用不同浓度碱性磷酸酶标准溶液利用比色法和荧光法来检测碱性磷酸酶传感器的线性范围和检测限。如图9A所示,GQDs@Ag纳米复合物的紫外吸收峰随着碱性磷酸酶活性增加而不断增大,图中由下至上,浓度依次由0增加到50U/L,如图9B所示,ΔA 415nm与碱性磷酸酶活性的响应曲线,如图9B插图,碱性磷酸酶活性在0.3~10U/L范围呈现良好的线性关系(Y=0.0862X-0.0068,R2=0.9961),检测限为0.1U/L。另外,如图9C所示,石墨烯量子点荧光强度随着碱性磷酸酶活性的增加而逐渐下降,图中由上至下,浓度依次由0增加到5.0U/L。如图9D所示,相对荧光强度(F0-F)/F0与碱性磷酸酶活性的响应曲线,如图9D插图所示,碱性磷酸酶活性在0.05~2.5U/L范围内呈良好的线性关系(Y=0.1526X+0.0547,R2=0.9899),检测限为0.02U/L。

实施例6
选择性是评价本生物传感器发明性能的一个重要的参数。因此,选择了其他一些可能有干扰的生物酶和蛋白质来评价该碱性磷酸酶生物传感器的选择性。用八组示例分别用荧光和吸光强度做对比实验,如图10中1-8所示,第一组为空白组无任何酶,第二组加入谷丙转氨酶(ALT,100U/L),第三组加入谷草转氨酶(AST,100U/L),第四组加入胰蛋白酶(Try,100U/L),第五组加入葡萄糖氧化酶(GOx,100U/L),第六组加入人血清白蛋白(HSA,1.0mg/mL),第七组加入胎牛血清白蛋白(BSA,1.0mg/mL),第八组为碱性磷酸酶(ALP,10U/L)。将第二组至第七组分别加入到含有碱性磷酸酶(ALP,10U/L)的反应体系中,测定反应体系吸光度和相对荧光强度的变化。如图10A所示,与不含有其他生物酶或蛋白质的碱性磷酸酶反应体系相比,包含了其他生物酶和蛋白的碱性磷酸酶反应体系可以引起相似的吸光度和相对荧光强度的变化。它们之间的差异可以被忽略,因此可以说明该发明对碱性磷酸酶的选择性良好。另外,如图10B所示,加入了其他生物酶和蛋白的碱性磷酸酶反应体系和单独的碱性磷酸酶反应体系一样,在可见光下观察发现溶液由无色变为黄色,在365nm紫外光下观察发现溶液荧光明显减弱,进一步证明了本发明对碱性磷酸酶的高选择性检测。

实施例7
利用上述碱性磷酸酶传感器,可筛选出潜在的碱性磷酸酶抑制剂。磷酸二氢钾,一个典型的抑制剂,被用于测试该传感器筛选碱性磷酸酶抑制剂的能力。将磷酸化抗坏血酸、碱性磷酸酶和不同浓度的磷酸二氢钾溶液在三羟甲基氨基甲烷溶液中37℃孵育15min后,再加入1580μL的水、50μL量子点和100μL硝酸银室温继续孵育45min。将磷酸化抗坏血酸、碱性磷酸酶和硝酸银的终浓度分别为0.6mM、50U/L、1.2mM。如图11所示,磷酸二氢钾的终浓度范围为0~1000μM,随着磷酸二氢钾浓度的增加碱性磷酸酶被抑制的程度呈现增强趋势,达到600μM后逐渐趋于平衡,磷酸二氢钾的半数抑制效应IC50=194.51μM。实验结果表明:该传感器可应用于碱性磷酸酶抑制剂的研究和筛选。

实施例8
本实施例用于测定未知浓度碱性磷酸酶标准品的浓度:配置不同浓度的碱性磷酸酶标准品溶液,将50μL浓度为24mM磷酸化抗坏血酸溶液和20μL不同浓度的碱性磷酸酶标准品溶液或未知浓度碱性磷酸酶标准品溶液置于200μL含0.5mM无水硫酸镁的pH9.8的三羟甲基氨基甲烷-硝酸缓冲溶液或二乙醇胺-硝酸缓冲溶液或甘氨酸-氢氧化钠缓冲溶液或碳酸钠-碳酸氢钠缓冲溶液体系中37℃孵育15min,再加入1580μL的双蒸水、50μL的量子点和的100μL浓度为24mM硝酸银室温继续孵育45min。用紫外可见分光光度计测定溶液在415nm处吸光度,利用吸光度随碱性磷酸酶标准溶液浓度的变化绘制标准曲线,从而测定出细胞裂解液中碱性磷酸酶的浓度。或者用荧光分光光度计测定溶液在Ex=362nm,Em=461nm处对应的荧光强度,利用相对荧光强度随碱性磷酸酶标准溶液的浓度的变化绘制标准曲线,从而测定出未知浓度碱性磷酸酶标准品的浓度。

实施例9
本实施例用于测定细胞里碱性磷酸酶的浓度:配置不同浓度的碱性磷酸酶标准品溶液,将50μL浓度为24mM磷酸化抗坏血酸溶液和20μL不同浓度的碱性磷酸酶标准品溶液或细胞裂解液置于200μL含0.5mM无水硫酸镁的pH9.8的三羟甲基氨基甲烷-硝酸缓冲溶液或二乙醇胺-硝酸缓冲溶液或甘氨酸-氢氧化钠缓冲溶液或碳酸钠-碳酸氢钠缓冲溶液体系中37℃孵育15min,再加入1580μL的水、50μL的量子点和的100μL浓度为24mM硝酸银室温继续孵育45min。用紫外可见分光光度计测定溶液在415nm处吸光度,利用吸光度随碱性磷酸酶标准溶液的浓度的变化绘制标准曲线,从而测定出细胞裂解液中碱性磷酸酶的浓度。或者用荧光分光光度计测定溶液在Ex=362nm,Em=461nm处对应的荧光强度,利用荧光强度随碱性磷酸酶标准溶液的浓度的变化绘制标准曲线,从而测定出细胞裂解液中碱性磷酸酶的浓度。再利用BCA试剂盒测定细胞裂解液中BSA蛋白的浓度,碱性磷酸酶的浓度和BSA蛋白的浓度的比值即为单位细胞内碱性磷酸酶的浓度。

实施例10
本实施例用于测定血浆里碱性磷酸酶的浓度:配置不同浓度的碱性磷酸酶标准品溶液,将50μL浓度为24mM磷酸化抗坏血酸溶液和20μL不同浓度的碱性磷酸酶标准品溶液或血浆置于200μL含0.5mM无水硫酸镁的pH9.8的三羟甲基氨基甲烷-硝酸缓冲溶液或二乙醇胺-硝酸缓冲溶液或甘氨酸-氢氧化钠缓冲溶液或碳酸钠-碳酸氢钠缓冲溶液体系中37℃孵育15min,再加入1580μL的水、50μL的量子点和的100μL浓度为24mM硝酸银室温继续孵育45min。用紫外可见分光光度计测定溶液在415nm处吸光度,利用吸光度随碱性磷酸酶标准溶液的浓度的变化绘制标准曲线,从而测定出血浆中碱性磷酸酶的浓度。或者用荧光分光光度计测定溶液在Ex=362nm,Em=461nm处对应的荧光强度,利用荧光强度随碱性磷酸酶标准溶液的浓度的变化绘制标准曲线,从而测定出血浆中碱性磷酸酶的浓度。

Claims (5)
1.一种基于荧光和比色双重模式检测碱性磷酸酶活性的方法,其特征在于,包括以下步骤:
(1)制备石墨烯量子点;
(2)将碱性磷酸酶溶液投入到过量的L-抗坏血酸-2-磷酸溶液中,然后将混合物置于碱性缓冲溶液中共同孵育,生成L-抗坏血酸;
(3)往步骤(2)的混合物中加入双蒸水、石墨烯量子点和硝酸银溶液进一步混合孵育;
(4)银纳米粒子逐步沉积在石墨烯量子点表面,石墨烯量子点表面的荧光强度逐步降低,石墨烯量子点表面沉积的银纳米粒子的吸光度逐渐增加;
(5)根据荧光强度或吸光度的变化,进行碱性磷酸酶活性的定量检测;
所述步骤(1)中,石墨烯量子点的直径在10nm以下,平均直径为1~3nm,其荧光光谱的最大激发波长Ex和最大发射波长Em分别为362nm和461nm;
所述步骤(2)中,共同孵育的条件是温度为25~40℃,时间为10~20min;
所述步骤(3)中,混合孵育的条件为:温度为20~40℃,时间为20~60min,pH大于6;
所述L-抗坏血酸-2-磷酸、石墨烯量子点和硝酸银溶液的体积比为1~3:1~3:1~6;
所述步骤(3)中,纳米银粒子沉积在量子点表面得到纳米复合结构,所述纳米复合结构的直径在100nm以下,平均直径在10~20nm,在415nm波长处有最大紫外吸收。
2.根据权利要求1所述的一种基于荧光和比色双重模式检测碱性磷酸酶活性的方法,其特征在于,所述步骤(5)中,碱性酶活性的定量检测过程具体如下:配置一系列不同浓度的碱性磷酸酶标准品溶液加入到反应体系中,监测反应体系吸光度或相对荧光强度的变化,根据吸光度的变化ΔA415nm 或相对荧光强度(F0-F)/F0随碱性磷酸酶标准品溶液活性的变化绘制标准曲线,得到碱性磷酸酶活性的标准曲线方程,将待测样品加入到反应体系中,监测反应体系吸光度的变化ΔA415nm或相对荧光强度的变化(F0-F)/F0,根据碱性磷酸酶活性的标准曲线方程,推算出碱性磷酸酶的用量,从而实现待测样品中碱性磷酸酶活性的定量检测。
3.根据权利要求1所述的一种基于荧光和比色双重模式检测碱性磷酸酶活性的方法,其特征在于,所述步骤(5)中,利用紫外可见光谱的比色法检测,碱性磷酸酶活性在0.3~10U/L范围内具有线性关系,检测限为0.1U/L;利用荧光光谱的荧光法检测,碱性磷酸酶活性在0.05~2.5U/L范围内具有线性关系,检测限为0.02U/L。
4.一种使用如权利要求1所述的检测碱性磷酸酶活性的方法制备的传感器。
5.如权利要求4所述的一种传感器在碱性磷酸酶抑制剂中的应用。
