Abstract
本发明提供了一种具有缓解疲劳功能的中药组合物,包括红景天、枸杞、西洋参、黄芪、当归、肉苁蓉、黄精、五味子、大枣等多种中药材的提取物,其中对红景天提取物通过发酵后,再提取,提高了有效成分的含量,药物利用力更高。本发明从整体上调节人体内环境,扶正固本,协调脏腑经络机能,进而纠正阴阳偏盛偏衰,使机体恢复到阴平阳秘的正常状态,在此基础制备的保健品口感好,抗疲劳活性高,市场前景广阔。

本发明涉及中药保健品领域,具体涉及一种具有缓解疲劳功能的中药组合物及其应用。
背景技术
随着社会进步和经济发展,快节奏的生活和工作压力,人们经常发现自己精力不足,记忆力下降,思维迟钝,继而感到全身疲劳,就是通常所说的亚健康状态。如果处于亚健康状态的人群不及早采取措施消除疲劳,它将损害机体健康,导致疾病,使机体早衰,甚至危及生命。
现代医学关于疲劳发生的机制可大致归纳为以下几个说法:能源物质耗竭学说、代谢物质堆积学说、内环境稳定性失调学说、保护性抑制学说、内分泌失调学说、自由基影响理论学说以及“突变”学说等。任何一个学说都不能独立解释所有的疲劳现象,人体疲劳不是固定由某一种或几种原因引起的,它是多种致疲劳因素总和的结果。
传统中医对疲劳的认识是在“整体观念”和“阴阳、藏象、经络、营卫气血”等基础理论指导下,较深刻揭示疲劳的本质,并对不同疲劳症候进行辩证分型论治。《素问上古天真论》曰:“形劳而不倦,气从以顺”,即疲劳过度则能耗气而虚。《素问宣明五气篇》曰:“五劳所伤,久视伤血,久卧伤气,久坐伤肉,久立伤骨,久行伤筋,是谓五劳之伤”,阐明了疲劳与脏腑机能的密切关系。《素问调经篇》说:“阴虚生内热,奈何”这些论述,阐明了疲劳或疲倦等症耗损精气和肺脾肾等脏腑功能的密切关系,充分体现了中医的整体观念。《理虚元鉴》中指出“七情不损,五劳不成”,指疲劳的病机主要是精血不足,水火不济所致。因此中医上将疲劳大致分为三种类型和九种常见疲劳症,三个类型:“形体疲劳”、“神志疲劳”、“脏腑疲劳”,九种常见症状:筋肉疲劳酸痛症、关节骨疲劳证、脾胃功能失调症、肾气不足症、月经失常症、阴虚(液、津亏)证、阴阳两虚证、气血两虚证和失眠证。
目前市面所售的抗疲劳保健品多为营养物质补充类,这些物质主要包括糖、脂肪、蛋白质、氨基酸(支链氨基酸、芳香族氨基酸、谷氨酰胺、牛磺酸、肌酸)、B族维生素、Vc、VE、左旋肉碱、微量元素(钙、磷、钾、钠、铁、硒、锌等)、水和电解质、丙酮酸等,由于这些营养素以补充机体的损失为主要目的,所以添加这类营养素的抗疲劳运动食品又称为营养强化食品,并不能称为真正意义上的保健食品。
中医药在消除疲劳和增强运动能力方面有其独特的优势。现已证实,中医中药和食疗药物不仅可以增加人体内的能量物质,而且有调节神经内分泌系统、加强代谢等作用,并可刺激体内激素的分泌与释放,增强心血管系统、消化系统以及造血系统、骨骼系统等功能,延缓过度疲劳的产生。
红景天系蔷薇目景天科红景天属多年生草本或亚灌性木野生植物,因花、根、茎的浸液均呈红色而得名。遗传上红景天对恶劣环境具有很强的适应,可在高寒、干燥、缺氧、强紫外线照射、昼夜温差大等极其恶劣而多变的自然环境中生长,具备其它植物所没有的特殊适应性,造就了其独特的抗辐射、耐缺氧、抗疲劳、延缓人体衰老等功效,被誉为“高原人参”。
世界共有红景天属植物90多种,主要分布于北半球高寒地带,多分布于喜马拉雅地区、亚洲西部至北部,经中国、朝鲜、日本至北美洲。中国是红景天属植物的分布中心,储量居世界之首,约有73种,2亚种、7变种,分布较为广泛。从东北到西南呈人字型分布,主要分布在东北、新疆、四川、西藏等省。
我国红景天植物资源丰富,且人工种植红景天技术已取得了显著进展。人们通过现代研究发现红景天具有与人参、刺五加类似的“适应原样”作用,而且不会出现人参的过度兴奋作用和刺五加容易导致便秘的作用,因而越来越受到人们的广泛重视。
红景天作为传统的藏药,药用历史久远,藏医《四部医典》记载红景天“性凉,清热,滋补元气”;《神农本草经》记载“长生不老草,主养命以应天,无毒,多服久服不伤身,轻身益气”;《本草纲目-草部》称红景天为“草本上品”,具有“扶正固本,补气养血”等功效。卫生部1991年已批准红景天为新食品资源。
药理学研究发现,红景天具有抗衰老、抗缺氧、抗辐射等不良刺激、对机体双向调节、抗病毒及抗肿瘤、抗疲劳及改善学习记忆等许多新的功能。也用于治疗脑血管疾病,老年性心肌能衰竭,糖尿病等症。特殊环境中的作业人员服用红景天制剂后,机体抵抗力和适应性均明显增强。目前红景天在医疗、食品、保健品和化妆品诸多方面得到了广泛的应用,特别是在老年医学、军事医学、航天医学及运动医学上有着更为重要的药用价值。
西洋参:性凉,味甘、微苦。归心、肺、肾经,具有补气养阴,清热生津的功效。用于气虚阴亏,内热,咳喘痰血,虚热烦倦,口燥喉干。古语云:“西洋参性凉而补,凡欲用人参而不受人参之温者皆可用之”。故补而不燥是西洋参的特别之处。临床上应用包括:强化心肌及增强心脏之活力;强壮中枢神经,安定身心并恢复疲劳,有镇静及解酒作用,增强记忆能力;对血压有调整作用;能调节胰岛之分泌,因此对糖尿病有功效;对肝脏有调节毒素之分泌,促进新陈代谢的作用;能增强体力并对运动员具极大助益;抑制癌细胞生长,增加免疫功能;助消化,对慢性胃病和肠胃衰弱有助效。
枸杞子:味甘、性平,入肝、肾经,有补益肝肾、养血补血、益精明目、润肺止咳的作用,主要用于肝肾两虚、精血不足之腰膝酸软、头晕耳鸣、遗精耳鸣、自汗乏力;肾阳不足之阳痿、不育;阴精亏损、肝血不足之眼目昏花以及阴虚劳嗽等证,枸杞子能够促进肝细胞再生,增强非特异性免疫功能的作用。
当归:味甘、辛,性温,入肝、心、脾经,具有补血活血,调经止痛,润肠通便的功效。主治心血不足之头晕目眩,倦怠乏力,心悸气短,血海空虚,冲任虚寒或淤血阻滞之月经不调,及其他血瘀作痛及阴虚血少肠燥便秘等证,能改善子宫的血液循环、减轻盆腔充血,缓解痛经。
肉苁蓉:味甘、咸,性温,入肾、大肠经,有补肾阳、益精血,润肠通便的作用,用于肾阳虚衰,精血不足之阳痿早泄,遗精;白浊;尿频余沥;腰痛脚弱;耳鸣目花;月经衍期;宫寒不孕;肠燥便秘及产后血虚、病后津液不足等证。
黄精:味甘、性平,入脾、肺、肾经,具有补气养阴、健脾、润肺、益肾的功效,可安神、活气血、降气宽肠、清热肿风痛、祛积化滞、清肠润肠、健脾润肺、止咳平喘、抗炎、抗过敏、强心等功效。黄精性味甘甜,食用爽口。其肉质根状茎肥厚,含有大量淀粉、糖分、脂肪、蛋白质、胡萝卜素、维生素和多种其他营养成分,生食、炖服既能充饥,又有健身之用,可令人气力倍增、肌肉充盈、骨髓坚强,对身体十分有益。黄精根状茎形状有如山芋,山区老百姓常把它当作蔬菜食用。黄精集七大营养素于一身,是能当饭吃的食品,有着卓越的营养保健价值和非凡的食疗功效。
大枣:味甘,性温,入脾、胃经,具有补气健脾、养血安神、调和营卫、缓和药性的功效,用于脾胃虚弱、体倦乏力、食少便溏、气血不足、头痛发热、汗出热风等证。大枣含有三萜类化合物及环磷酸腺苷,有较强的抑癌、抗过敏作用;枣中含有抗疲劳作用的物质,能增强人的耐力;枣还具有减轻毒性物质对肝脏损害的功效;枣中的黄酮类化合物,有镇静降血压的作用。
五味子:味酸、甘,性温,归肺、心、肾经。具有收敛固涩,益气生津,补肾宁心的功效,用于肺虚喘咳,梦遗滑精,遗尿尿频,久泻不止,自汗盗汗,津伤口渴,内热消渴,心悸失眠等证。
黄芪:性温,味甘,归肺、脾经。具有益气固表、敛汗固脱、托疮生肌、利水消肿之功效。用于治疗气虚乏力,中气下陷,久泻脱肛,便血崩漏,表虚自汗,痈疽难溃,久溃不敛,血虚萎黄,内热消渴,慢性肾炎,蛋白尿,糖尿病等。黄芪和人参均属补气良药,人参偏重于大补元气,回阳救逆,常用于虚脱、休克等急症,效果较好。而黄芪则以补虚为主,常用于体衰日久、言语低弱、脉细无力者。现代医学研究表明,黄芪有增强机体免疫功能、保肝、利尿、抗衰老、抗应激、降压和较广泛的抗菌作用。能消除实验性肾炎蛋白尿,增强心肌收缩力,调节血糖含量。黄芪不仅能扩张冠状动脉,改善心肌供血,提高免疫功能,而且能够延缓细胞衰老的进程。
发明内容
本发明以发酵红景天提取物为主料,再辅以西洋参、枸杞、黄芪、当归等8种中药的提取物为辅料,进行科学配置,各组分发挥协同作用,得到一种具有明显抗疲劳效果的中药组合物,也可应用到保健食品中。
本发明采用如下技术方案:
一种具有缓解疲劳功能的中药组合物,其特征在于,由下列重量份的组分原料组成:红景天提取物50-85份,枸杞子提取物10-20份,西洋参提取物4-6份,黄芪提取物5-10份,当归提取物5-10份,五味子提取物5-20份,肉苁蓉提取物5-10份、黄精提取物10-20份,大枣提取物8-12份,所述的红景天提取物为发酵红景天提取物。
所述的具有缓解疲劳功能的中药组合物,其特征在于,由下列重量份的组分原料组成:红景天提取物80份,西洋参提取物5份,枸杞子提取物15份,黄芪提取物8份,当归提取物8份,五味子提取物15份,肉苁蓉提取物8份,黄精提取物15份,大枣提取物10份,所述的红景天提取物为发酵红景天提取物。
所述的具有缓解疲劳功能的中药组合物,其特征在于,由下列重量份的组分原料组成:红景天提取物70份,西洋参提取物5份,枸杞子提取物20份,黄芪提取物5份,当归提取物10份,五味子提取物20份,肉苁蓉提取物5份,黄精提取物20份,大枣提取物10份,所述的红景天提取物为发酵红景天提取物。
所述的具有缓解疲劳功能的中药组合物在保健饮料中的应用。
所述的具有缓解疲劳功能的中药组合物在保健口服液中的应用。
所述的发酵红景天提取物的制备方法,包括以下步骤:
a、将红景天药材粉碎成细粉,过60目筛,向粗粉中加入粗粉质量4-6%的蜂蜜、1%的大麦芽粉、1%的桑葚粉、120%的纯净水,配制培养基,并于121℃灭菌20min,冷却后,在按接种量4%接种酵母,在温度为28-32℃的条件下,发酵20h,发酵结束后,在45-50℃条件下干燥,得到发酵红景天粉;
b、向发酵红景天粉加入15-25倍量的50-90%乙醇溶液进行提取,温度60-85℃,提取时间1-3h,抽滤,收集滤液和滤渣;将滤渣以同样方法再次提取,过滤,合并滤液,向滤液中加入1-3%的明胶和壳聚糖作为絮凝剂对提取液进行精制,离心,取上清液干燥,得到红景提取物。
本发明的有益效果:
本发明以发酵红景天提取物为主料,再辅以西洋参、枸杞、黄芪、当归等8种中药的提取物为辅料,进行科学配置,各组分发挥协同作用,得到一种具有明显抗疲劳效果的中药组合物。本发明将红景天发酵后再提取有效成分,提高了红景天的有效成分含量,同时有效成分更易被吸收,在此基础上,再复配西洋参、黄芪、当归、黄精、大枣、五味子、肉苁蓉等药材的提取物组成一个有机复方,从补气活血,健脾胃,补肾虚多方面来调节人体的内环境,达到改善血液循环,增加细胞携氧量,有效的提高抗疲劳的效果。本组方从整体上调节人体内环境,扶正祛邪,扶正固本,协调脏腑经络机能,进而纠正阴阳偏盛偏衰,使机体恢复到阴平阳秘的正常状态,在此基础制备的保健品口感好,抗疲劳活性高,市场前景广阔。

下面通过具体的实施例对本发明做进一步的详细描述,不是对本发明的限制。
实施例1
一种具有缓解疲劳功能的中药组合物,由下列质量份的组分混合制成:红景天提取物80份,西洋参提取物5份,枸杞子提取物15份,黄芪提取物8份,当归提取物8份,五味子提取物15份,肉苁蓉提取物8份,黄精提取物15份,大枣提取物10份。
实施例2
一种具有缓解疲劳功能的中药组合物,由下列质量份的组分混合制成:
红景天提取物70份,西洋参提取物5份,枸杞子提取物20份,黄芪提取物5份,当归提取物10份,五味子提取物20份,肉苁蓉提取物5份,黄精提取物20份,大枣提取物10份。
实施例3
一种具有缓解疲劳功能的中药组合物,由下列质量份的组分混合制成:红景天提取物50份、枸杞子提取物10份、西洋参提取物5份、黄芪提取物10份、当归提取物5份、五味子提取物5份、肉苁蓉提取物5份、黄精提取物10份,大枣提取物10份。
实施例4
一种具有具有缓解疲劳功能的保健饮料,
配方:
红景天提取物60份、枸杞子提取物15份、西洋参提取物5份、黄芪提取物5份、当归提取物10份、五味子提取物15份、肉苁蓉提取物8份、黄精提取物15份,大枣提取物10份,
制备方法:
将配方量的原料混合后加入10%甜味剂和0.2%柠檬酸,混匀,加入固体物料总质量10-20倍纯水进行均质处理后过滤,滤液灭菌后在无菌条件下灌装。
上述发酵红景天提取物的制备方法,包括以下步骤:
a、将红景天药材粉碎成细粉,过60目筛,向粗粉中加入粗粉质量4-6%的蜂蜜、1%的大麦芽粉、1%的桑葚粉、120%的纯净水,配制培养基,并于121℃灭菌20min,冷却后,在按接种量4%接种酵母,在温度为28-32℃的条件下,发酵20h,发酵结束后,在45-50℃条件下干燥,得到发酵红景天粉;
b、向发酵红景天粉加入20倍量的70%乙醇溶液进行提取,温度75℃,提取时间2h,抽滤,收集滤液和滤渣;将滤渣以同样方法再次提取,过滤,合并滤液,向滤液中加入2%的明胶和壳聚糖作为絮凝剂对提取液进行精制,离心,取上清液干燥,得到发酵红景天提取物。
为了表明本发明的中药组合物药物疗效,进行如下药效学实验:
抗疲劳试验方法示例
首先将实施例1、2、3的组合物加入15倍纯净水充分溶解,加入10%甜味剂和0.2%柠檬酸,混匀。
参照中华人民共和国卫生部颁发的《保健食品检验与评价技术规范》(卫法监发[2003]42号,2003-02-14)的方法,进行抗疲劳活性实验研究。
选用BALB/c小鼠,20±2g,饲养一周,环境温度控制在(25±2)℃,光照12/24h。然后分为对照组和低、中、高剂量组,分别灌胃一个月。高、中、低三个剂量组给予160、80、40mL/kg·d的复合红景天口服液,对照组为生理盐水。每日一次,连续灌胃30天。测定各组小鼠力竭性游泳时间、血清尿素、血乳酸含量及肝糖原水平。
力竭性游泳时间:末次给予受试物30min后,置小鼠在游泳箱(大小约50cm×50cm×40cm)中游泳,水深不少于30cm,水温25℃±0.5℃,鼠尾根部负荷5%体重的铅皮。记录小鼠自游泳开始至死亡的时间,作为小鼠游泳时间。
血清尿素氮含量测定:末次给予受试物30min后,无负重在温度为30℃的水中游泳90min,采血测定。在测定管加入0.05mL血清,在标准管加入0.05mL 10mmol/L尿素标准液,在空白管中加入0.05mL蒸馏水。再各加入2.5mL 1g/L二乙酰—肟溶液、2.5mL酸溶液,充分混匀,置沸水浴精确15min,立即用自来水冷却。以空白管调零,读取各管在波长520nm下的吸光度(A值)。
尿素含量(mmol/L)=Au×10/As
式中:Au—测定管吸光度,As—标准管吸光度。
肝糖原含量测定:
样本制备:
末次给样后30min,无负重在温度为30℃的水箱中游泳90min,立即处死,称取肝脏200mg,加入4mLTCA溶液(5%三氯醋酸,用蒸馏水配),匀浆1min,将匀浆液倒入离心管,以3000r/min离心15min,将上清液转移至另一试管内。在沉淀中再加入4mLTCA溶液,匀浆1min,再次离心15min,取上清液,并与第一次离心的上清液合并,充分混匀。
取1mL上清液放入10mL离心管中,加入4mL 95%乙醇,充分混匀不留界面。用干净塞子塞上,37℃水浴3h。沉淀完全后,将试管于3000r/min离心15min。小心倒掉上清液并使试管倒立放置10min。
用2mL蒸馏水溶解糖原,加水时将管壁的糖原洗下。如管底的糖原不立即溶解,振荡管子直到完全溶解。
制作试剂空白和标准管:空白管:吸2mL蒸馏水到干净离心管;标准管:吸0.5mL葡萄糖标准液(含100mg/dL葡萄糖)和1.5mL蒸馏水放入同样的管子。
测定样品糖原含量:将10mL蒽酮试剂(溶液中含0.05%的蒽酮,1%的硫脲,用72%H2SO4配制)加入各管,充分混合。从管子中注入蒽酮试剂时起,将管子放在冷水龙头下冲凉。在所有管子都达到凉水温度后,将其浸于沸水浴15min,然后移到冷水浴,冷却到室温。将管内液体移入比色管,在波长620nm下,用空白管调零后测定吸光度。并根据标准曲线计算出糖原含量,根据所称取的肝脏重量换算成肝糖原含量(以mg/g肝表示)。
每克肝组织中糖原的毫克数=DU/DS×0.5×提取液体积/肝组织克数×0.9
DU:样品管吸光度;DS:标准管吸光度;提取液体积:8mL;肝组织克数:0.2g。
血乳酸含量测定:末次给样30min后,负重2%体重在温度30℃的水中游泳60min后停止,安静15min后采血备用。在5mL试管中加入0.48mL 1%NaF溶液,准确吸取全血20μL加入试管底部。用试管上部清液清洗微量吸管数次,再加入1.5mL蛋白沉淀剂,振荡混匀,于3000r/min离心10min,取上清液。在空白管加入0.5mL沉淀剂-NaF混合液、标准管加入0.5mL乳酸标准液、测定管加入0.5mL上清液,再各加入0.1mL 4%CuSO4溶液、3mL浓硫酸,充分混匀,置沸水浴加热5min,取出后放入冰水浴冷却10min。各加入0.1mL 1.5%对羟基联苯,摇匀,置30℃水浴30min(每隔10min振摇一次)。取出后放入沸水浴中加热90s,取出冷却至室温,在波长560nm处用5mm光径比色皿比色,空白管调零。计算血乳酸含量。

实验结果:

通过对小鼠负重游泳时间、血清尿素氮、血乳酸和肝糖原含量的测定,表明提取物组合物能够显著延长小鼠游泳时间,降低血清尿素氮和血乳酸的含量,增加肝糖原的含量,具有很好的抗疲劳活性。
同时,我们也对单独的红景天提取物进行了抗疲劳活性实验研究,结果表明发酵后的红景天提取物和未发酵的红景天提取物都具有一定的抗疲劳效果,且发酵后的提取物效果明显优于未发酵的提取物,但是,二者的抗疲劳效果都明显低于本发明的复方实验结果。

1.一种具有缓解疲劳功能的中药组合物,其特征在于,由下列重量份的组分原料组成:红景天提取物50-85份,枸杞子提取物10-20 份,西洋参提取物4-6 份,黄芪提取物5-10份,当归提取物5-10份,五味子提取物5-20份,肉苁蓉提取物5-10份、黄精提取物10-20份,大枣提取物8-12份,所述的红景天提取物为发酵红景天提取物。
2.如权利要求1所述的具有缓解疲劳功能的中药组合物,其特征在于,由下列重量份的组分原料组成:红景天提取物80份,西洋参提取物5份,枸杞子提取物15 份,黄芪提取物8份,当归提取物8份,五味子提取物15份,肉苁蓉提取物8份,黄精提取物15份,大枣提取物10份,所述的红景天提取物为发酵红景天提取物。
3.如权利要求1所述的具有缓解疲劳功能的中药组合物,其特征在于,由下列重量份的组分原料组成:红景天提取物70份,西洋参提取物5 份,枸杞子提取物20 份,黄芪提取物5份,当归提取物10份,五味子提取物20份,肉苁蓉提取物5份,黄精提取物20份,大枣提取物10份,所述的红景天提取物为发酵红景天提取物。
