Abstract
本发明公开了一种采用静电纺丝法制备纳米纤维敷料的方法,以六氟异丙醇为溶剂配制0.1g/mL的I型胶原蛋白溶液,加入利多卡因,得到外层纺丝液;以二氯甲烷为溶剂配制0.2 g/mL的聚己内酯溶液,并加入莫匹罗星,得到内层纺丝液;将外层纺丝液和内层纺丝液分别加入2个注射器中,由同轴针头连接形成同轴结构进行高压静电纺丝,将得到的敷料真空干燥后浸泡在京尼平的乙醇溶液中,用乙醇洗涤后干燥,得到纳米纤维敷料。制备的纳米纤维敷料具有更高溶胀率和力学强度,更适合作为湿性敷料使用并在吸收伤口渗出液,在耐长期使用性和抗菌性等方面具有优势。
Description
技术领域
本发明属于医用敷料的制备领域,具体涉及一种采用静电纺丝法制备纳米纤维敷料的方法。
背景技术
医用敷料是重要的辅助医疗用品,主要用于外科创伤、烧伤和各种慢性病引起的皮肤溃烂的治疗。早期的敷料仅具有简单保护和防止感染的单一功能。随着医疗技术的不断发展,具有减轻患者痛苦、可吸收渗出液和抗菌以防止感染、减少疤痕与收缩、促使愈合等作用的功能性敷料应运而生,并在临床上承担起越来越重要的作用。
电纺丝技术在敷料制备中具有出色的可调控性,通过电纺丝技术制备的敷料也具有透气及比表面积大等优点,已逐渐成为医用敷料制备的主流技术之一。目前,现有的医用敷料在作为湿性敷料时,强度较弱,抗菌效果不佳。
发明内容
本发明提供一种采用静电纺丝法制备纳米纤维敷料的方法,制备的敷料具有核壳结构,作为湿性敷料时仍然具有较好的效果。
一种采用静电纺丝法制备纳米纤维敷料的方法,包括以下步骤:
(1)以六氟异丙醇为溶剂配制0.1g/mL的I型胶原蛋白溶液,加入利多卡因,得到外层纺丝液;
(2)以二氯甲烷为溶剂配制0.2 g/mL的聚己内酯溶液,并加入莫匹罗星,得到内层纺丝液;
(3)将步骤(1)和步骤(2)得到的外层纺丝液和内层纺丝液分别加入2个注射器中,由同轴针头连接形成同轴结构进行高压静电纺丝,纺丝参数:正电压17kV,负电压-3 kV,内层PCL流速为3mL/h,外层胶原为1 mL/h,接收距离为15cm,将得到的敷料于室温下真空干燥24h;
(4)将步骤(3)得到的敷料浸泡在0.1g/mL的京尼平的乙醇溶液中,反应3 h,用乙醇洗涤敷料3次,于室温下真空干燥24 h,得到纳米纤维敷料。
步骤(1)中利多卡因的纯度为98%,加入量为0.015g/mL。
步骤(2)中莫匹罗星的加入量为0.08 g/mL。
本发明提供的方法制备的纳米纤维敷料具有核壳纤维结构,在敷料纤维的外壳和内核之中分别负载了利多卡因和莫匹罗星,利多卡因是临床常用的局部麻药,吸收后可以有效抑制人体的疼痛感,莫匹罗星则可以用于预防和治疗各种细菌性皮肤感染,对伤口的感染起到良好的预防作用,内核的聚己内酯可以提供机械强度和维持敷料使用时的形状完整性,制备的纳米纤维敷料具有更高溶胀率和力学强度,更适合作为湿性敷料使用并在吸收伤口渗出液,在耐长期使用性和抗菌性等方面具有优势。
具体实施例方式
实施例1。
一种采用静电纺丝法制备纳米纤维敷料的方法,包括以下步骤:
(1)以六氟异丙醇为溶剂配制0.1g/mL的I型胶原蛋白溶液,加入利多卡因,得到外层纺丝液;
(2)以二氯甲烷为溶剂配制0.2 g/mL的聚己内酯溶液,并加入莫匹罗星,得到内层纺丝液;
(3)将步骤(1)和步骤(2)得到的外层纺丝液和内层纺丝液分别加入2个注射器中,由同轴针头连接形成同轴结构进行高压静电纺丝,纺丝参数:正电压17kV,负电压-3 kV,内层PCL流速为3mL/h,外层胶原为1 mL/h,接收距离为15cm,将得到的敷料于室温下真空干燥24h;
(4)将步骤(3)得到的敷料浸泡在0.1g/mL的京尼平的乙醇溶液中,反应3 h,用乙醇洗涤敷料3次,于室温下真空干燥24 h,得到纳米纤维敷料。

Claims (3)
1.一种采用静电纺丝法制备纳米纤维敷料的方法,其特征在于:包括以下步骤:
(1)以六氟异丙醇为溶剂配制0.1g/mL的I型胶原蛋白溶液,加入利多卡因,得到外层纺丝液;
(2)以二氯甲烷为溶剂配制0.2 g/mL的聚己内酯溶液,并加入莫匹罗星,得到内层纺丝液;
(3)将步骤(1)和步骤(2)得到的外层纺丝液和内层纺丝液分别加入2个注射器中,由同轴针头连接形成同轴结构进行高压静电纺丝,纺丝参数:正电压17kV,负电压-3 kV,内层PCL流速为3mL/h,外层胶原为1 mL/h,接收距离为15cm,将得到的敷料于室温下真空干燥24h;
(4)将步骤(3)得到的敷料浸泡在0.1g/mL的京尼平的乙醇溶液中,反应3 h,用乙醇洗涤敷料3次,于室温下真空干燥24 h,得到纳米纤维敷料。
2.根据权利要求1所述的一种采用静电纺丝法制备纳米纤维敷料的方法,其特征在于:步骤(1)中利多卡因的纯度为98%,加入量为0.015g/mL。
3. 根据权利要求1所述的一种采用静电纺丝法制备纳米纤维敷料的方法,其特征在于:步骤(2)中莫匹罗星的加入量为0.08 g/mL。
