Abstract

本发明公开了一种医用复合敷料及其制备方法。该医用复合敷料是将无纺布或棉布等织物作为载体材料,浸渍于由羧甲基壳聚糖溶液、麻油、蜂蜡组成的混合液,取出,封装于内包装袋,最后辐照灭菌而得。医用复合敷料具有以下优点:(1)含有羧甲基壳聚糖、麻油和蜂蜡等促愈合成分,促创面愈合功效显著;(2)载体材料赋予膜敷料一定的柔韧度和强度,换药时,易揭易贴,操作方便,不会和创面发生粘连,不产生二次损伤;(3)具有保持湿润、抗菌、减少疤痕形成等功效。该医用复合敷料为一种能明显促进创面愈合的湿性复合敷料,适用于手术创口、擦伤、烧伤、供皮区、褥疮、糖尿病溃疡以及其他难愈合创面的治疗。

一种医用复合敷料及其制备方法
技术领域
本发明涉及一种医用复合敷料及其制备方法,属于生物医用材料领域。
背景技术
创伤是正常皮肤(组织)在外界致伤因子作用下所导致的损害。常见的创伤有手术切开、擦伤、烧伤、供皮区、褥疮、下肢血管性溃疡、糖尿病溃疡以及其他难愈合性创伤等。创面的形成和存在,对机体的局部乃至全身的功能将产生重大的影响。为了加速创伤的愈合,减少创面的感染,一般采用敷料覆盖和保护的方法进行治疗。
理想的医用敷料应具备以下性能:(1)保持创面湿润环境,利于伤口愈合;(2)能够有效吸收创面渗出物,减少感染;(3)不但具有良好的通透性,而且能够有效阻隔细菌和有害粒子的入侵;(4)去除敷料时不会与伤口发生粘连,以免造成二次损伤;(5)具有抗菌、消炎作用,储存方便,使用方便;(6)最好有止血、止痛等作用,可促进肉芽生长和皮肤再生,加速愈合,减少疤痕。
目前国内外市场上涌现出各种各样的医用敷料产品,按照材料分类,大致可分为:天然材料类、合成高分子材料类、无机材料类和复合材料类。天然材料又包括棉纤维、甲壳素/壳聚糖类、海藻酸盐、明胶、胶原、动物皮、同种自体/异体皮等。合成分子材料类,包括聚氨酯、聚乙烯醇、聚丙烯酰胺、聚乙烯、聚丙烯醇、聚己内酯、聚乳酸、聚四氟乙烯、硅橡胶、多氨基甲酸乙酯和硅氧烷弹性体等。无机材料类,包括石墨、无机活性玻璃等。由于创面愈合过程是一个复杂的过程,不同的创面和同一个创面不同的阶段对敷料有不同的要求,所以目前任何一种单一材料都不能满足创面愈合过程的复杂需要。例如,天然材料制成的敷料大部分吸收性好,具有良好的生物相容性及生物学活性,但是机械性能差;合成高分子材料隔绝性能好,机械强度好,但是吸收性能相对天然材料差些。所以,通过对材料的复合,兼具多种材料的优势,实现创面愈合的要求,是目前开发新型敷料的趋势。
目前,“湿性愈合”理论已是一种公认的治疗创面的观点,市场上出现了许多湿性敷料,多以藻酸盐、羧甲基纤维素钠、生物多糖衍生物等作为主要成分。在众多湿性敷料中,含有壳聚糖或其衍生物成分(尤其是羧甲基壳聚糖)因具有止痛、止血、促进伤口愈合、减小疤痕、抑菌、良好的生理相容性和生物可降解性等优异的性能,成为了目前国内外开发治疗创面产品的热点。临床应用证实,羧甲基壳聚糖本身或与其他材料复合后,制成粉剂、膜、无纺布、胶带、绷带、溶液、水凝胶、干凝胶、棉纸、洗液、喷剂等多种形式的伤口敷料,对烧伤,烫伤、切割伤、皮肤溃疡等具有良好的治疗效果。
现代药理研究证实麻油性凉甘,具有滋润、去腐生肌的功效。麻油可为创面提供组织生长所需要的营养成分,如不饱和脂肪酸、芝麻素和维生素E等成分,能改善慢性创面负氮平衡,促进创面愈合。另外,麻油具有强大的保湿功能,使创面处于近似于生理环境条件,通过无损伤的液化排除坏死组织,有利于创面上皮细胞的生长,减少瘢痕形成。
在中医药学上,蜂蜡自古就有解毒、生肌、定痛的功效。经过近代西方医学的研究发现,蜂蜡具有消炎抑菌、局部麻醉、降血脂和预防动脉粥样硬化等作用。蜂蜡熔点为62-67℃,可增加药膏的稠度,且有较弱的吸水性,吸水后形成油包水型(W/O)乳剂。在应用于创面治疗时,蜂蜡起到了框架的作用,将存在于蜂蜡框架内部的羧甲基壳聚糖和麻油渗透入创面深部,达到促伤口愈合的作用。而蜂蜡则留在了创面的外边,形成一层膜,对创面起到保护和湿润的作用,减少了细菌感染的机会。
发明内容
本发明的目的在于提供一种医用复合敷料,具体地说是,提供一种含有羧甲基壳聚糖、麻油和蜂蜡等促愈合成分,能明显促进创面愈合的湿性复合敷料,适用于手术创口、擦伤、烧伤、供皮区、褥疮、糖尿病溃疡以及其他难愈合创面的治疗。
本发明的目的通过以下技术方案来实现:
一种医用复合敷料及其制备方法,是将无纺布或棉布等织物作为载体材料,浸渍于由羧甲基壳聚糖溶液、麻油、蜂蜡组成的混合液,取出,封装于内包装袋,最后辐照灭菌而得。
所述的医用复合敷料的载体材料,可以是水刺无纺布、针刺无纺布、棉布和纱布等,优选水刺无纺布,更进一步地,优选卫生用水刺无纺布。
所述的医用复合敷料的载体材料,其厚度在0.2-4mm之间,其长宽可按照临床需求进行裁剪。
所述的载体材料浸渍于由羧甲基壳聚糖溶液、麻油、蜂蜡组成的混合液的时间为2-10min。
所述的羧甲基壳聚糖溶液、麻油、蜂蜡组成的混合液的配制过程,是先分别配制羧甲基壳聚糖溶液和麻油/蜂蜡混合液,之后再将羧甲基壳聚糖溶液和麻油/蜂蜡混合液以一定的比例混合,搅拌均匀。
所述的羧甲基壳聚糖溶液中羧甲基壳聚糖的浓度为5-20%。
所述的麻油/蜂蜡混合液中,蜂蜡与麻油的比例为5-20∶100。
所述的羧甲基壳聚糖溶液与麻油/蜂蜡混合液的比例为5-50∶100。
所述的医用复合敷料,适用于各种创面的治疗,包括手术创口、擦伤、烧伤、供皮区、褥疮、糖尿病溃疡以及其他难愈合性创面。
本发明获得的医用复合敷料具有以下优点:(1)含有羧甲基壳聚糖、麻油和蜂蜡等促愈合成分,促创面愈合功效显著;(2)载体材料赋予膜敷料一定的柔韧度和强度,换药时,易揭易贴,操作方便,不会和创面发生粘连,不产生二次损伤;(3)具有保持湿润、抗菌、减少疤痕形成等功效。
具体实施方式
现结合下列实施例来进一步描述本发明,下列实施例旨在进一步举例说明本发明的制备方法,这些实施例不以任何方式限制本发明待批权利要求的范围。
实施例1
(1)羧甲基壳聚糖溶液的配制:称取75g羧甲基壳聚糖,加入425g纯化水,搅拌至溶解,配制成15%羧甲基壳聚糖溶液。
(2)麻油/蜂蜡混合液的配制:称取100g蜂蜡和1000g麻油,两者混合,加热、搅拌至蜂蜡溶解。
(3)羧甲基壳聚糖溶液、麻油、蜂蜡混合液的配制:将步骤(1)配制的15%羧甲基壳聚糖溶液和步骤(2)配制的麻油/蜂蜡混合液,两者混合,加热,搅拌均匀。
(4)将厚度为0.5mm的水刺无纺布浸渍到羧甲基壳聚糖溶液、麻油、蜂蜡的混合液中,5min后取出,封装于内包装袋,最后进行辐照灭菌。
实施例2
(1)羧甲基壳聚糖溶液的配制:称取25g羧甲基壳聚糖,加入475g纯化水,搅拌至溶解,配制成5%羧甲基壳聚糖溶液。
(2)麻油/蜂蜡混合液的配制:称取200g蜂蜡和1000g麻油,两者混合,加热、搅拌至蜂蜡溶解。
(3)羧甲基壳聚糖溶液、麻油、蜂蜡混合液的配制:将步骤(1)配制的5%羧甲基壳聚糖溶液和步骤(2)配制的麻油/蜂蜡混合液,两者混合,加热,搅拌均匀。
(4)将厚度为2mm的水刺无纺布浸渍到羧甲基壳聚糖溶液、麻油、蜂蜡的混合液中,10min后取出,封装于内包装袋,最后进行辐照灭菌。

1.一种医用复合敷料及其制备方法,其特征在于,是将无纺布或棉布等织物作为载体材料,浸渍于由羧甲基壳聚糖溶液、麻油、蜂蜡组成的混合液,取出,封装于内包装袋,最后辐照灭菌而得。
2.根据权利要求1所述的医用复合敷料,其特征在于,所述的载体材料,可以是水刺无纺布、针刺无纺布、棉布和纱布等,优选水刺无纺布,更进一步地,优选卫生用水刺无纺布。
3.根据权利要求1所述的医用复合敷料,其特征在于,所述的载体材料,其厚度在0.2-4mm之间,其长宽可按照临床需求进行裁剪。
4.根据权利要求1所述的医用复合敷料,其特征在于,载体材料浸渍于由羧甲基壳聚糖溶液、麻油、蜂蜡组成的混合液的时间为2-10min。
5.根据权利要求1所述的医用复合敷料,其特征在于,羧甲基壳聚糖溶液、麻油、蜂蜡组成的混合液的配制过程,是先分别配制羧甲基壳聚糖溶液和麻油/蜂蜡混合液,之后再将羧甲基壳聚糖溶液和麻油/蜂蜡混合液以一定的比例混合,搅拌均匀。
